Abstract
本发明公开了一种反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,涉及胃漂浮制剂技术领域,包括如下步骤:(1)复合载体的制备,(2)复合载体的改性,(3)胃漂浮片的制备。本发明所制缓释型胃漂浮片适用于反刍动物体内寄生虫的防治,无毒无害,不存在使用安全隐患;进入反刍动物胃液后呈漂浮状态,所含体内寄生虫防治剂缓慢释放,有效延长体内寄生虫防治剂在胃中的滞留时间,从而延长药效分子的释放时间,提高药效分子的生物利用度,进而保证对反刍动物体内寄生虫的防治效果,减少用药次数。

技术领域:
本发明涉及胃漂浮制剂技术领域,具体涉及一种反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺。
背景技术:
反刍动物,是偶蹄目中的一个亚目。反刍是指进食经过一段时间以后将在胃中半消化的食物返回嘴里再次咀嚼,反刍动物就是有是有反刍这种消化方式的动物。通常是一些食草动物,因为植物的纤维是比较难消化的。反刍动物采食一般比较匆忙,特别是粗饲料,大部分未经充分咀嚼就吞咽进入瘤胃,经过瘤胃浸泡和软化一段时间后,食物经逆呕重新回到口腔,经过再咀嚼,再次混入唾液并再吞咽进入瘤胃的过程。
反刍动物在生长过程中会遇到很多寄生虫病原,这些病原种类甚多,分布广泛,会导致动物出现消耗性和慢性疾病,严重影响反刍动物的生长与发育,继而给畜牧业带来不可忽视的经济损失。体内寄生虫的存在更会影响反刍动物的摄食量、消化功能,使反刍动物表现出贫血、消瘦、营养不良、发育缓慢、水肿等症状。
目前,反刍动物体内寄生虫的防治主要依赖于药物,而药物主要分为化学药和中药。化学药虽然起效快,但治疗后易复发,且容易产生抗药性,并伴随副反应;而中药虽然使用安全性高,但起效慢,服药时间长,药物利用度低。
发明内容:
本发明所要解决的技术问题在于提供一种工艺重复性强、使用安全性高且防治效果好的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺。
本发明所要解决的技术问题采用以下的技术方案来实现:
反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,包括如下步骤:
(1)复合载体的制备:搅拌下向食品级多聚谷氨酸中滴加40-45℃去离子水直至完全溶解,并滴加5wt%稀硫酸调节溶液pH值于10min内稳定在3-4,再加入泊洛沙姆和聚乙二醇4000,搅拌下补加40-45℃去离子水使泊洛沙姆和聚乙二醇4000完全溶解,并升温至回流状态保温搅拌2-4h,然后以5-10℃/min的冷却速度降温至0-5℃保温搅拌0.5-1h,所得混合物送入冷冻干燥机中,干燥所得固体经超微粉碎机制成微粉,即得复合载体;
(2)复合载体的改性:向所制复合载体中加入食品级聚丙烯酸钠和氢化棕榈油,充分混合后利用微波处理器微波处理5-10min,间隔5-10min后继续微波处理5-10min,间隔5-10min后再次微波处理5-10min,如此反复使微波处理总时间达到15-30min,然后向所得混合物中加入2-3倍重量的去离子水,充分混合后利用微波处理器微波回流处理5-10min,所得混合液送入喷雾干燥机中,干燥所得颗粒经超微粉碎机制成微粉,即得改性复合载体;
(3)胃漂浮片的制备:将体内寄生虫防治剂、改性复合载体、预胶化淀粉、羟丙基纤维素和乳糖按比例充分混合,经湿法制粒、压片后制得胃漂浮片。
所述体内寄生虫防治剂由如下重量份数的原料制成:鲜藿香叶15-20份、鲜茼蒿10-15份、干槐花5-10份、胖大海1-5份、鲜紫草1-5份、白茅根1-5份、决明子1-5份、覆盆子1-5份、砂仁1-5份、紫苏籽0.5-2份、黑胡椒0.5-2份,其制备方法为:将胖大海、白茅根、决明子、覆盆子、砂仁经粉碎制成粗粉后加热至70-80℃保温翻炒10-15min,所得混合粉末加入3-5倍重量无水乙醇中,并加热至回流状态保温搅拌1-3h,过滤,所得滤液经减压浓缩后制成固含量35-45%的浸膏;将鲜藿香叶、鲜茼蒿、干槐花、鲜紫草、紫苏籽、黑胡椒混合后加入3-5倍总重的40-45℃水中,浸泡15-30min后转入功率1.5kW磨浆机中,磨浆15-30min,过滤,所得滤液加入浸膏中,充分混合后制得物料I;磨浆所得滤渣送入冷冻干燥机中,充分干燥后经超微粉碎机制成微粉,即得物料II;将物料I和物料II充分混合,并于50-55℃下干燥至恒重,即得体内寄生虫防治剂。
所述食品级多聚谷氨酸、泊洛沙姆和聚乙二醇4000的质量比为10-15:5-10:1-5。
所述复合载体、食品级聚丙烯酸钠和氢化棕榈油的质量比为15-20:1-5:1-5。
所述体内寄生虫防治剂、改性复合载体、预胶化淀粉、羟丙基纤维素和乳糖的质量比为5-10:10-15:10-15:5-10:1-5。
所述泊洛沙姆选自泊洛沙姆188、泊洛沙姆237、泊洛沙姆338、泊洛沙姆407中的一种。
所述食品级聚丙烯酸钠的分子量在800万,pH值9-10。
所述氢化棕榈油经过改性处理,其改性方法为:将氢化棕榈油以3-5℃/min的升温速度加热至熔融状态保温搅拌5min,再加入医药级聚乙烯醇和燕麦β-葡聚糖,充分混合后利用微波处理器微波处理5min,间隔5min后继续微波处理5min,间隔5min后再次微波处理5min,所得混合物转入0-5℃环境中密封静置1h,转出后自然恢复至室温,最后经超微粉碎机制成微粉,即得改性棕榈油。
所述氢化棕榈油、医药级聚乙烯醇和燕麦β-葡聚糖的质量比为5:1:0.5。
所述微波处理器的工作条件为微波频率2450MHz、输出功率700W。
所述聚乙烯醇的醇解度在88±2%,平均聚合度在500-600。
体内寄生虫防治剂各原料的常用功效与配伍关系:
鲜藿香叶:味辛,性温,常用功效是止呕消嗳、止泄、疗头痛头晕、明目、健脾、和胃、发汗解表;君药。
鲜茼蒿:味辛、甘,性平,常用功效是消食开胃、通便利肺、清血养心;臣药。
干槐花:味苦,性微寒,常用功效是凉血止血、清肝泻火;臣药。
胖大海:味甘,性寒,常用功效是清肺化痰、利咽、润肠通便;佐药。
鲜紫草:味苦,性寒,常用功效是凉血活血、清热解毒;佐药。
白茅根:味甘,性寒,常用功效是凉血止血、清热利尿;佐药。
决明子:味甘、苦,性寒,常用功效是清肝明目、润肠通便;使药。
覆盆子:味甘、酸,性温,常用功效是益肾固精缩尿,养肝明;使药。
砂仁:味辛,性温,常用功效是行气调中,和胃,醒脾;使药。
紫苏籽:味辛,性温,常用功效是下气、清痰、润肺、宽肠;使药。
黑胡椒:味辛,性热,常用功效是温中散寒,健胃止痛;使药。
本发明的有益效果是:
(1)本发明以食品级多聚谷氨酸、泊洛沙姆和聚乙二醇4000作为原料,经酯化反应后制得复合载体,复合载体再经改性处理后制得改性复合载体;并与体内寄生虫防治剂、制剂助剂混合后制得缓释型胃漂浮片;
(2)本发明体内寄生虫防治剂由11种中药制成,根据严格的用量组合并遵循君臣佐使的配伍关系,以高效发挥防治反刍动物体内寄生虫的功效,避免使用化学防治剂存在的安全隐患,并实现这一中药配方在防治反刍动物体内寄生虫领域的新应用;
(3)本发明所制缓释型胃漂浮片适用于反刍动物体内寄生虫的防治,无毒无害,不存在使用安全隐患;进入反刍动物胃液后呈漂浮状态,所含体内寄生虫防治剂缓慢释放,有效延长体内寄生虫防治剂在胃中的滞留时间,从而延长药效分子的释放时间,稳定血药浓度,改善药效分子的吸收,提高药效分子的生物利用度,进而保证对反刍动物体内寄生虫的防治效果,减少用药次数。
具体实施方式:
为了使本发明实现的技术手段、创作特征、达成目的与功效易于明白了解,下面结合具体实施例,进一步阐述本发明。
实施例1
(1)复合载体的制备:搅拌下向15g食品级多聚谷氨酸中滴加40-45℃去离子水直至完全溶解,并滴加5wt%稀硫酸调节溶液pH值于10min内稳定在3-4,再加入10g泊洛沙姆188和5g聚乙二醇4000,搅拌下补加40-45℃去离子水使泊洛沙姆和聚乙二醇4000完全溶解,并升温至回流状态保温搅拌3h,然后以5℃/min的冷却速度降温至0-5℃保温搅拌30min,所得混合物送入冷冻干燥机中,干燥所得固体经超微粉碎机制成微粉,即得复合载体;
(2)复合载体的改性:向所制15g复合载体中加入2g食品级聚丙烯酸钠和1g氢化棕榈油,充分混合后利用微波处理器微波处理5min,间隔5min后继续微波处理5min,间隔5min后再次微波处理5min,如此反复使微波处理总时间达到30min,然后向所得混合物中加入3倍重量的去离子水,充分混合后利用微波处理器微波回流处理10min,所得混合液送入喷雾干燥机中,干燥所得颗粒经超微粉碎机制成微粉,即得改性复合载体;其中,食品级聚丙烯酸钠的分子量在800万,pH值9-10。
(3)胃漂浮片的制备:将10g体内寄生虫防治剂、15g改性复合载体、15g预胶化淀粉、10g羟丙基纤维素和5g乳糖按比例充分混合,经湿法制粒、压片后制得胃漂浮片。
体内寄生虫防治剂的制备:将5g胖大海、3g白茅根、3g决明子、2g覆盆子、1g砂仁经粉碎制成粗粉后加热至70-80℃保温翻炒15min,所得混合粉末加入5倍重量无水乙醇中,并加热至回流状态保温搅拌2h,过滤,所得滤液经减压浓缩后制成固含量35-45%的浸膏;将15g鲜藿香叶、15g鲜茼蒿、5g干槐花、3g鲜紫草、0.5g紫苏籽、0.5g黑胡椒混合后加入5倍总重的40-45℃水中,浸泡30min后转入功率1.5kW磨浆机中,磨浆30min,过滤,所得滤液加入浸膏中,充分混合后制得物料I;磨浆所得滤渣送入冷冻干燥机中,充分干燥后经超微粉碎机制成微粉,即得物料II;将物料I和物料II充分混合,并于50-55℃下干燥至恒重,即得体内寄生虫防治剂。
实施例2
(1)复合载体的制备:搅拌下向10g食品级多聚谷氨酸中滴加40-45℃去离子水直至完全溶解,并滴加5wt%稀硫酸调节溶液pH值于10min内稳定在3-4,再加入5g泊洛沙姆407和3g聚乙二醇4000,搅拌下补加40-45℃去离子水使泊洛沙姆和聚乙二醇4000完全溶解,并升温至回流状态保温搅拌3h,然后以5℃/min的冷却速度降温至0-5℃保温搅拌30min,所得混合物送入冷冻干燥机中,干燥所得固体经超微粉碎机制成微粉,即得复合载体;
(2)复合载体的改性:向所制20g复合载体中加入3g食品级聚丙烯酸钠和2g氢化棕榈油,充分混合后利用微波处理器微波处理5min,间隔5min后继续微波处理5min,间隔5min后再次微波处理5min,如此反复使微波处理总时间达到30min,然后向所得混合物中加入3倍重量的去离子水,充分混合后利用微波处理器微波回流处理10min,所得混合液送入喷雾干燥机中,干燥所得颗粒经超微粉碎机制成微粉,即得改性复合载体;其中,食品级聚丙烯酸钠的分子量在800万,pH值9-10。
(3)胃漂浮片的制备:将5g体内寄生虫防治剂、10g改性复合载体、10g预胶化淀粉、10g羟丙基纤维素和5g乳糖按比例充分混合,经湿法制粒、压片后制得胃漂浮片。
体内寄生虫防治剂的制备:将5g胖大海、5g白茅根、3g决明子、3g覆盆子、2g砂仁经粉碎制成粗粉后加热至70-80℃保温翻炒15min,所得混合粉末加入5倍重量无水乙醇中,并加热至回流状态保温搅拌2h,过滤,所得滤液经减压浓缩后制成固含量35-45%的浸膏;将20g鲜藿香叶、10g鲜茼蒿、5g干槐花、3g鲜紫草、1g紫苏籽、0.5g黑胡椒混合后加入5倍总重的40-45℃水中,浸泡30min后转入功率1.5kW磨浆机中,磨浆30min,过滤,所得滤液加入浸膏中,充分混合后制得物料I;磨浆所得滤渣送入冷冻干燥机中,充分干燥后经超微粉碎机制成微粉,即得物料II;将物料I和物料II充分混合,并于50-55℃下干燥至恒重,即得体内寄生虫防治剂。
实施例3

(3)胃漂浮片的制备:将5g体内寄生虫防治剂、10g改性复合载体、10g预胶化淀粉、10g羟丙基纤维素和5g乳糖按比例充分混合,经湿法制粒、压片后制得胃漂浮片。

(1)复合载体的制备:按照专利CN201510867996.0实施例1所述除阿苯达唑、伊维菌素以外的原料配比,将6g HPMC K100M、2g碳酸氢钠-柠檬酸(质量比1:0.7)、8.4g单硬脂酸甘油酯充分混合,即得复合载体;
(2)胃漂浮片的制备:将5g体内寄生虫防治剂、10g复合载体、10g预胶化淀粉、10g羟丙基纤维素和5g乳糖按比例充分混合,经湿法制粒、压片后制得胃漂浮片。
体内寄生虫防治剂的制备:将5g胖大海、5g白茅根、3g决明子、3g覆盆子、2g砂仁经粉碎制成粗粉后加热至70-80℃保温翻炒15min,所得混合粉末加入5倍重量无水乙醇中,并加热至回流状态保温搅拌2h,过滤,所得滤液经减压浓缩后制成固含量35-45%的浸膏;将20g鲜藿香叶、10g鲜茼蒿、5g干槐花、3g鲜紫草、1g紫苏籽、0.5g黑胡椒混合后加入5倍总重的40-45℃水中,浸泡30min后转入功率1.5kW磨浆机中,磨浆30min,过滤,所得滤液加入浸膏中,充分混合后制得物料I;磨浆所得滤渣送入冷冻干燥机中,充分干燥后经超微粉碎机制成微粉,即得物料II;将物料I和物料II充分混合,并于50-55℃下干燥至恒重,即得体内寄生虫防治剂。
对照例4
(1)复合载体的制备:搅拌下向10g食品级多聚谷氨酸中滴加40-45℃去离子水直至完全溶解,并滴加5wt%稀硫酸调节溶液pH值于10min内稳定在3-4,再加入5g泊洛沙姆407和3g聚乙二醇4000,搅拌下补加40-45℃去离子水使泊洛沙姆和聚乙二醇4000完全溶解,并升温至回流状态保温搅拌3h,然后以5℃/min的冷却速度降温至0-5℃保温搅拌30min,所得混合物送入冷冻干燥机中,干燥所得固体经超微粉碎机制成微粉,即得复合载体;
(2)复合载体的改性:向所制20g复合载体中加入3g食品级聚丙烯酸钠和2g氢化棕榈油,充分混合后利用微波处理器微波处理5min,间隔5min后继续微波处理5min,间隔5min后再次微波处理5min,如此反复使微波处理总时间达到30min,然后向所得混合物中加入3倍重量的去离子水,充分混合后利用微波处理器微波回流处理10min,所得混合液送入喷雾干燥机中,干燥所得颗粒经超微粉碎机制成微粉,即得改性复合载体;其中,食品级聚丙烯酸钠的分子量在800万,pH值9-10。
(3)胃漂浮片的制备:将5g体内寄生虫防治剂、10g改性复合载体、10g预胶化淀粉、10g羟丙基纤维素和5g乳糖按比例充分混合,经湿法制粒、压片后制得胃漂浮片。
体内寄生虫防治剂的制备:将阿苯达唑和伊维菌素以35:1的比例充分混合,即得体内寄生虫防治剂。
对照例5
参照CN201510867996.0实施例1制备胃漂浮片。
实施例4
分别对实施例1-3、对照例1-5所制同规格胃漂浮片进行体外药物释放的测定,测定介质为1000ml肉牛瘤胃液、温度37℃,转速50r/min,测定结果如表1所示。

实施例5
分别对实施例1-3、对照例1-5所制同规格胃漂浮片用于100头肉牛的体内寄生虫防治,每天早晚给药1次,每头每次给药10g,连续给药10天,记录防治效果,结果如表2所示。

Claims (8)
1.反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:包括如下步骤:
(1)复合载体的制备:搅拌下向食品级多聚谷氨酸中滴加40-45℃去离子水直至完全溶解,并滴加5wt%稀硫酸调节溶液pH值于10min内稳定在3-4,再加入泊洛沙姆和聚乙二醇4000,搅拌下补加40-45℃去离子水使泊洛沙姆和聚乙二醇4000完全溶解,并升温至回流状态保温搅拌2-4h,然后以5-10℃/min的冷却速度降温至0-5℃保温搅拌0.5-1h,所得混合物送入冷冻干燥机中,干燥所得固体经超微粉碎机制成微粉,即得复合载体;
(2)复合载体的改性:向所制复合载体中加入食品级聚丙烯酸钠和氢化棕榈油,充分混合后利用微波处理器微波处理5-10min,间隔5-10min后继续微波处理5-10min,间隔5-10min后再次微波处理5-10min,如此反复使微波处理总时间达到15-30min,然后向所得混合物中加入2-3倍重量的去离子水,充分混合后利用微波处理器微波回流处理5-10min,所得混合液送入喷雾干燥机中,干燥所得颗粒经超微粉碎机制成微粉,即得改性复合载体;
(3)胃漂浮片的制备:将体内寄生虫防治剂、改性复合载体、预胶化淀粉、羟丙基纤维素和乳糖按比例充分混合,经湿法制粒、压片后制得胃漂浮片。
2.根据权利要求1所述的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:所述体内寄生虫防治剂由如下重量份数的原料制成:鲜藿香叶15-20份、鲜茼蒿10-15份、干槐花5-10份、胖大海1-5份、鲜紫草1-5份、白茅根1-5份、决明子1-5份、覆盆子1-5份、砂仁1-5份、紫苏籽0.5-2份、黑胡椒0.5-2份,其制备方法为:将胖大海、白茅根、决明子、覆盆子、砂仁经粉碎制成粗粉后加热至70-80℃保温翻炒10-15min,所得混合粉末加入3-5倍重量无水乙醇中,并加热至回流状态保温搅拌1-3h,过滤,所得滤液经减压浓缩后制成固含量35-45%的浸膏;将鲜藿香叶、鲜茼蒿、干槐花、鲜紫草、紫苏籽、黑胡椒混合后加入3-5倍总重的40-45℃水中,浸泡15-30min后转入功率1.5kW磨浆机中,磨浆15-30min,过滤,所得滤液加入浸膏中,充分混合后制得物料I;磨浆所得滤渣送入冷冻干燥机中,充分干燥后经超微粉碎机制成微粉,即得物料II;将物料I和物料II充分混合,并于50-55℃下干燥至恒重,即得体内寄生虫防治剂。
3.根据权利要求1所述的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:所述食品级多聚谷氨酸、泊洛沙姆和聚乙二醇4000的质量比为10-15:5-10:1-5。
4.根据权利要求1所述的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:所述复合载体、食品级聚丙烯酸钠和氢化棕榈油的质量比为15-20:1-5:1-5。
5.根据权利要求1所述的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:所述体内寄生虫防治剂、改性复合载体、预胶化淀粉、羟丙基纤维素和乳糖的质量比为5-10:10-15:10-15:5-10:1-5。
6.根据权利要求1所述的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:所述泊洛沙姆选自泊洛沙姆188、泊洛沙姆237、泊洛沙姆338、泊洛沙姆407中的一种。
7.根据权利要求1所述的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:所述食品级聚丙烯酸钠的分子量在800万,pH值9-10。
8.根据权利要求1所述的反刍动物体内寄生虫防治用缓释型胃漂浮片的制备工艺,其特征在于:所述微波处理器的工作条件为微波频率2450MHz、输出功率700W。
