本发明属于药学领域,涉及一种匀浆技术用于中药滴丸剂的制备方法,具体涉及一种匀浆技术运用于六味地黄滴丸的制备方法。本发明的方法能够将六味地黄滴丸熔融物充分研磨均匀,解决丸剂表面粗糙和颜色不匀问题,从而提高六味地黄滴丸的质量。本发明制备工艺方便实用,且易于控制产品质量。

技术领域
本发明属于药学领域,涉及一种匀浆技术用于中药滴丸剂的制备方法,具体涉及一种匀浆技术运用于六味地黄滴丸的制备方法。
背景技术
六味地黄滴丸是由熟地黄、酒萸肉、牡丹皮、山药、茯苓、泽泻提取而成的干膏粉、牡丹皮挥发油加聚乙二醇4000制成,具有滋阴补肾的功效。主要用于头晕耳鸣,腰膝酸软,遗精盗汗。还可用于治疗糖尿病,高血压,肾虚,慢性肾功能衰竭,更年期综合症,肾病综合征等。
现有的六味地黄滴丸制备方法:干膏粉粉碎成细粉,加聚乙二醇4000和牡丹皮挥发油,加热熔化,混匀,滴制,去油,即得。
通过实验反复验证后,我们发现现有的方法主要采用的是加热熔化,混匀的方式进行加工,存在着滴制的丸剂表面粗糙和颜色不匀等一系列问题,严重的影响药品的质量和安全性。
因此,为了能够克服现有技术中存在的问题,我们对现有的制备工艺进行改良,将匀浆技术用于在滴丸剂的制备过程中,很有的解决上述问题。
发明内容
本发明的目的在于提供一种六味地黄滴丸的制备方法。
本发明的方法能够将六味地黄滴丸熔融物充分研磨均匀,解决丸剂表面粗糙和颜色不匀问题,从而提高六味地黄滴丸的质量。本发明制备工艺方便实用,且易于控制产品质量。
具体的,本发明的制备方法,包括以下步骤:
(1)牡丹皮挥发油的制备:牡丹皮加15倍量水蒸馏提取挥发油,蒸馏后的水溶液另器收集;
(2)六味地黄滴丸干膏粉的制备:酒萸肉加8倍量70%乙醇回流提取二次,每次2小时,合并提取液,滤过,滤液备用;熟地黄、山药、泽泻加水煎煮二次,每次加10倍量水,第一次2小时,第二次1小时,合并煎液,滤过,滤液与牡丹皮水溶液合并,减压浓缩至相对密度为1.15~1.20(50℃热测)的清膏,放冷,加乙醇使含醇量达70%,静置48小时,取上清液与上述山茱萸提取液合并,减压回收乙醇至无醇味,备用;茯苓加水煮沸后,于80℃温浸二次,每次1.5小时,合并浸出液,滤过,滤液和上述备用液合并,浓缩至相对密度 1.30-1.40,减压干燥,粉碎,即得;
(3)聚乙二醇4000加热熔融,加入六味地黄滴丸干膏粉、牡丹皮挥发油混匀,用胶体磨研磨20-40分钟,滴制成2000丸(每丸重300mg)或10000丸(每丸重60mg),即得。

本发明的制备方法,相对于现有的六味地黄滴丸的制备方法,采用研体磨匀浆技术将融化后熔融物充分研磨均匀再滴制,这项改进,产生了意想不到的效果:用改进方法制成的六味地黄滴丸,其表面光滑、颜色均一。同时,本发明的方法还明显提高马钱苷和丹皮酚的含量,并能保持其良好的稳定性。
具体实施方式
通过以下具体实施例对本发明进行进一步的说明,但不作为对本发明的限定。
实施例一:
处方:熟地黄480g 酒萸肉240g 牡丹皮180g
山药240g 茯苓180g 泽泻180g
制法:
(1)牡丹皮挥发油的制备:牡丹皮加15倍量水蒸馏提取挥发油,蒸馏后的水溶液另器收集。
(2)六味地黄滴丸干膏粉的制备:酒萸肉加8倍量70%乙醇回流提取二次,每次2小时,合并提取液,滤过,滤液备用。熟地黄、山药、泽泻加水煎煮二次,每次加10倍量水,第一次2小时,第二次1小时,合并煎液,滤过,滤液与牡丹皮水溶液合并,减压浓缩至相对密度为1.15~1.20(50℃热测)的清膏,放冷,加乙醇使含醇量达70%,静置48小时,取上清液与上述山茱萸提取液合并,减压回收乙醇至无醇味,备用;茯苓加水煮沸后,于 80℃温浸二次,每次1.5小时,合并浸出液,滤过,滤液和上述备用液合并,浓缩至相对密度1.30-1.40,减压干燥,粉碎,即得。

实施例四、本发明与现有工艺的比较
原工艺的制备方法:
(1)牡丹皮挥发油的制备:牡丹皮加15倍量水蒸馏提取挥发油,蒸馏后的水溶液另器收集;
(2)六味地黄滴丸干膏粉的制备:酒萸肉加8倍量70%乙醇回流提取二次,每次2小时,合并提取液,滤过,滤液备用;熟地黄、山药、泽泻加水煎煮二次,每次加10倍量水,第一次2小时,第二次1小时,合并煎液,滤过,滤液与牡丹皮水溶液合并,减压浓缩至相对密度为1.15~1.20(50℃热测)的清膏,放冷,加乙醇使含醇量达70%,静置48小时,取上清液与上述山茱萸提取液合并,减压回收乙醇至无醇味,备用;茯苓加水煮沸后,于80℃温浸二次,每次1.5小时,合并浸出液,滤过,滤液和上述备用液合并,浓缩至相对密度 1.30-1.40,减压干燥,粉碎,即得;
(3)聚乙二醇4000加热熔融,加入六味地黄滴丸干膏粉、牡丹皮挥发油混匀,滴制成 2000丸(每丸重300mg)或10000丸(每丸重60mg),即得。
表1:六味地黄滴丸采用匀浆技术制备与原工艺对比试验结果:
从表1可以得出,采用匀浆技术制备的六味地黄滴丸质量明显优于原工艺。
表2:六味地黄滴丸采用匀浆技术研磨时间对比(各三个样检测含量):
从表2可以得出,匀浆技术研磨30分钟含量均一度高于研磨20分钟,研磨40分钟丹皮酚含量会下降,由此选出最优研磨时间30分钟。
表3:六味地黄滴丸采用匀浆技术与原工艺对比加速稳定性试验结果:
从表3加速稳定性试验结果可以得出,采用匀浆技术制备的六味地黄滴丸马钱苷含量和丹皮酚含量均比原工艺更稳定。

Claims (1)

1.一种六味地黄滴丸的制备方法,由以下原料加工制成:
处方:熟地黄480g 酒萸肉240g 牡丹皮180g
山药240g 茯苓180g 泽泻180g
制法:
(1)牡丹皮挥发油的制备:牡丹皮加15倍量水蒸馏提取挥发油,蒸馏后的水溶液另器收集,
(2)六味地黄滴丸干膏粉的制备:酒萸肉加8倍量70%乙醇回流提取二次,每次2小时,合并提取液,滤过,滤液备用,熟地黄、山药、泽泻加水煎煮二次,每次加10倍量水,第一次2小时,第二次1小时,合并煎液,滤过,滤液与牡丹皮水溶液合并,50℃热测,减压浓缩至相对密度为1.15~1.20的清膏,放冷,加乙醇使含醇量达70%,静置48小时,取上清液与上述山茱萸提取液合并,减压回收乙醇至无醇味,备用;茯苓加水煮沸后,于80℃温浸二次,每次1.5小时,合并浸出液,滤过,滤液和上述备用液合并,浓缩至相对密度1.30-1.40,减压干燥,粉碎,即得,
(3)聚乙二醇4000加热熔融,加入六味地黄滴丸干膏粉、牡丹皮挥发油混匀,用胶体磨研磨30分钟,滴制成2000丸,每丸重300mg,或10000丸,每丸重60mg,即得。
