Abstract
本发明公开了一种含铁皮石斛的中药组合物及其制备方法与应用，其药材原料按重量份计包括：辣根草8‑10份、牛尾菜6‑8份、铁皮石斛3‑5份、水蛭2‑3份。本发明以辣根草活血通络、牛尾菜祛风散湿、铁皮石斛养血滋阴、水蛭化瘀通经为配方原理进行组方，所制得的中药组合物具有良好祛风除湿、温经散寒、活血化瘀、通络止痛等功效，可用于制备关节炎治疗药物。

技术领域
本发明属于医药技术领域，具体涉及一种含铁皮石斛的中药组合物及其制备方法与应用。
背景技术
关节炎是一种常见的慢性疾病，由炎症、感染、创伤或其他因素引起的关节炎性疾病，属于风湿学科疾病，常见的有骨关节炎和类风湿关节炎，其临床表现为关节的红、肿、热、痛、功能障碍及关节畸形，严重者导致关节残疾、影响患者的生活质量。
铁皮石斛为兰科石斛属植物，以新鲜或干燥根茎入药，具有生津养胃、滋阴清热、润肺益肾等多种功效，备受历代医家和医学典籍的推崇，《神农本草经》中更将其列为上品。现代研究证明，铁皮石斛中含有多糖、芪类、酚类、木质素等多种化合物，具有抗肿瘤、抗衰老及提高免疫等作用。
发明内容
本发明的目的在于提供一种含铁皮石斛的中药组合物及其制备方法与应用，其具有良好祛风除湿、温经散寒、活血化瘀、通络止痛等功效，可用于制备关节炎治疗药物。
为实现上述目的，本发明采用如下技术方案：
一种含铁皮石斛的中药组合物，其药材原料按重量份计包括：辣根草8-10份、牛尾菜6-8份、铁皮石斛3-5份、水蛭1-2份。
所述含铁皮石斛的中药组合物的制备方法包括以下步骤：
1）将辣根草、牛尾菜、铁皮石斛干燥，粉碎，过60-80目筛，得药材粉；
2）按重量比1:3-5将水蛭加入100℃的水中，煎煮5-10min后捞出，烘干，研磨成粉，得脱毒水蛭粉；
3）按重量比1:5-8在步骤1）所得药材粉中加入水，80-100℃煎煮0.5-1h后过滤，取滤液A和滤渣备用；
4）将步骤3）所得滤渣与步骤2）所得脱毒水蛭粉混合，按重量比1:3-5加入质量浓度为80%-90%的乙醇溶液，超声提取40-60min后过滤，取滤液B备用；
5）将滤液A、B分别回收溶剂至干后，研磨，过100-200目筛，混匀，即得所述中药组合物。
该中药组合物的药材配方中还可包括血风藤。
本发明的显著优点在于：
（1）本发明以辣根草活血通络、牛尾菜祛风散湿、铁皮石斛养血滋阴、水蛭化瘀通经为组方原则，制备得到一种中药组合物，其具有良好祛风除湿、温经散寒、活血化瘀、通络止痛等功效，对关节炎具有良好的治疗效果。其中，铁皮石斛的使用可起到扶正、调和的作用，可协同提高辣根草、牛尾菜和水蛭对关节炎的治疗功效。
（2）水蛭对关节炎的治疗有较好效果，但其具毒性，故用药安全是限制其应用的一大关键因素。本发明在限定其适宜用量的基础上，通过高温快速灭活及乙醇提取，以达到部分脱毒的效果，提高其用药安全性。
（3）本发明药材组方中还可通过血风藤的加入，进一步促进药效通达经络，达到更好的疗效。
具体实施方式
为了使本发明所述的内容更加便于理解，下面结合具体实施方式对本发明所述的技术方案做进一步的说明，但是本发明不仅限于此。
实施例1
1）将100g辣根草、60g牛尾菜、40g铁皮石斛干燥，粉碎，过60-80目筛，得药材粉；
2）按重量比1:3将20g水蛭加入100℃的水中，煎煮10min后捞出，烘干，研磨成粉，得脱毒水蛭粉；
3）按重量比1:5在步骤1）所得药材粉中加入水，100℃煎煮1h后过滤，取滤液A和滤渣备用；
4）将步骤3）所得滤渣与步骤2）所得脱毒水蛭粉混合，按重量比1:5加入质量浓度为80%的乙醇溶液，超声提取40min后过滤，取滤液B备用；
5）将滤液A、B分别回收溶剂至干后，研磨，过100-200目筛，混匀，即得含铁皮石斛的中药组合物。
实施例2
1）将80g辣根草、80g牛尾菜、50g铁皮石斛干燥，粉碎，过60-80目筛，得药材粉；
2）按重量比1:4将10g水蛭加入100℃的水中，煎煮8min后捞出，烘干，研磨成粉，得脱毒水蛭粉；
3）按重量比1:6在步骤1）所得药材粉中加入水，100℃煎煮1h后过滤，取滤液A和滤渣备用；
4）将步骤3）所得滤渣与步骤2）所得脱毒水蛭粉混合，按重量比1:4加入质量浓度为85%的乙醇溶液，超声提取50min后过滤，取滤液B备用；
5）将滤液A、B分别回收溶剂至干后，研磨，过100-200目筛，混匀，即得含铁皮石斛的中药组合物。
实施例3
1）将90g辣根草、70g牛尾菜、30g铁皮石斛、20g血风藤干燥，粉碎，过60-80目筛，得药材粉；
2）按重量比1:5将10g水蛭加入100℃的水中，煎煮5min后捞出，烘干，研磨成粉，得脱毒水蛭粉；
3）按重量比1:8在步骤1）所得药材粉中加入水，100℃煎煮1h后过滤，取滤液A和滤渣备用；
4）将步骤3）所得滤渣与步骤2）所得脱毒水蛭粉混合，按重量比1:3加入质量浓度为90%的乙醇溶液，超声提取60min后过滤，取滤液B备用；
5）将滤液A、B分别回收溶剂至干后，研磨，过100-200目筛，混匀，即得含铁皮石斛的中药组合物。
功效试验
对照药物：以辣根草110g、牛尾菜90g、水蛭20g为药材配方，按实施例1工艺参数进行提取，制得用作对照的组合物。
1. 对小鼠耳廓微循环的影响
取50只小鼠，适应性喂养一周后随机分为空白组、对照组及试验1-3组，每组10只。各组腹腔注射戊巴比妥钠25mg/kg，麻醉后用眼科剪修剪耳廓毛，将小鼠固定好，平展耳廓并涂少许香柏油于表面，用显微镜观察给药前小鼠耳廓微循环，记录各鼠耳廓细动脉（A）、细静脉（V）的口径及毛细血管开放数量。然后进行灌胃给药，试验1-3组给予2mL实施例1-3所得组合物的生理盐水溶液（0.4mg/mL），对照组给予2mL对照药物的生理盐水溶液（0.4mg/mL），空白组给予等量生理盐水。给药30min后，再次测量各组小鼠耳廓细动脉（A）、耳廓细静脉（V）的口径及毛细血管开放数量，结果见表1。

2. 对角叉菜胶致小鼠足肿胀的影响
取50只小鼠，适应性喂养一周后将小鼠随机分为空白组、对照组及试验1-3组，每组10只。试验1-3组灌胃给予2mL实施例1-3所得组合物的生理盐水溶液（0.4mg/mL），对照组灌胃给予2mL对照药物的生理盐水溶液（0.4mg/mL），空白组灌胃给予等量生理盐水，每天1次，连续给药3天。末次给药30min后，将每只小鼠右后足趾部皮下注射0.5%角叉菜混悬液40μL进行造模。于造模后3h将小鼠处死，取后双足等同部位精确称重，然后以左右足重量差作为肿胀度，其结果见表2。

由以上实验可以看出，本发明食用菌营养粉能使小鼠耳廓血管口径明显增大，毛细血管开放数量增多，且对炎症状况有明显抑制作用。

1.一种含铁皮石斛的中药组合物，其特征在于：其药材原料按重量份计包括：辣根草8-10份、牛尾菜6-8份、铁皮石斛3-5份、水蛭2-3份。
2.根据权利要求1所述含铁皮石斛的中药组合物，其特征在于：所用药材原料还包括血风藤。
3.一种如权利要求1所述含铁皮石斛的中药组合物的制备方法，其特征在于：包括以下步骤：
1）将辣根草、牛尾菜、铁皮石斛干燥，粉碎，过60-80目筛，得药材粉；
2）按重量比1:3-5将水蛭加入100℃的水中，煎煮5-10min后捞出，烘干，研磨成粉，得脱毒水蛭粉；
3）按重量比1:5-8在步骤1）所得药材粉中加入水，80-100℃煎煮0.5-1h后过滤，取滤液A和滤渣备用；
4）将步骤3）所得滤渣与步骤2）所得脱毒水蛭粉混合，按重量比1:3-5加入乙醇溶液，超声提取40-60min后过滤，取滤液B备用；
5）将滤液A、B分别回收溶剂至干后，研磨，过100-200目筛，混匀，即得所述中药组合物。
4.根据权利要求3所述含铁皮石斛的中药组合物的制备方法，其特征在于：步骤4）中所用乙醇溶液的质量浓度为80%-90%。
5.一种如权利要求1所述中药组合物在制备关节炎治疗药物中的应用。
