Abstract
本发明提供一种治疗脾虚的药物组合物及其制备方法,药物组合物的重量份组成及制备方法是,将人参3~5份、党参3~5份、黄芪5~10份、当归15~25份混合后加乙醇提取;将白芍15~25份、陈皮8~13份、木香8~13份、山药8~13份、扁豆8~13份、甘草8~13份、川芎15~25份、肉桂5~10份、升麻1~3份、柴胡1~3份、益智仁1~3份、蜀椒1~3份、附子1~3份、干姜1~3份、山茱萸10~15份混合后加纯净水提取;茯苓15~25份、白术15~25份、桂枝5~10份混合后加纤维素酶用水提取,提取液灭活备用,将上述提取液合并,真空冷冻干燥得到药物提取物固体干膏,经超微粉碎,制粒,制成所需要的药物组合物。本发明药物组合物具有药方科学,适合大规模生产、无任何不良反应,药力强、见效快、服用方便的特点。

背景技术
[0002] 脾在五行中属土,在五脏阴阳中属阴中之至阴,脾主运化,统血,升清,输布水谷精 微,为"气血生化之源"。人体出生后,各脏腑组织器官皆依赖脾所化生的水谷精微以濡养, 故称脾为"后天之本"。其与胃、肉、唇、口等构成脾系统。素体脾虚或饮食不节、情志因素、 劳逸失调,药、食损脾或慢性肾病患者湿邪久居,损伤脾气等原因引起脾的功能虚衰、生化 之源不足。脾对食物的消化和吸收起着十分重要的作用,因此几乎所有的胃肠道疾病都可 出现或伴有脾虚。
[0003] 脾虚泛指因脾气虚损引起的一系列脾生理功能失常的病理现象及病证。包括脾气 虚、脾阳虚、中气下陷、脾不统血等证型。其病情虽较繁杂,主要有呕吐、泄泻、水肿、出血、经 闭、带下、四肢逆冷、小儿多涎等。脾虚多因饮食失调、劳逸失度,或久病体虚所引起,脾有运 化食物中的营养物质和输布水液以及统摄血液等作用。脾虚则运化失常,并可出现营养障 碍,水液失于布散而生湿酿痰,或发生失血等症。邹澍《本草经疏》归纳"脾虚十二证,饮食劳 倦,伤脾发热,饮食不消化,属脾气虚;伤食必恶食,停食,为恣饮汤水或冷茶、冷酒所致。水 肿属脾气虚,兼脾阴虚;噎膈属气血两虚,由于血液衰少,而非痰气壅逆所成。脾虚属气虚, 健忘属气血两虚,倦怠、嗜卧属脾气不足。脾虚腹痛,按之则止,属血虚;痞气,属脾气虚及气 郁所致。"
[0004] 从临床现实分析,脾虚中又以脾气虚、脾阳虚更为常见,每有腹胀、痞满、消瘦、肢 乏、泄泻或大便先硬后溏,亦有表现为脾虚便秘者、食减、水肿等症。脾虚分为脾气虚和皮脾 阳虚,脾气虚多因饮食不节,或劳倦过度,或忧思日久,损伤脾土,或抵抗力不足,素体虚弱。 脾阳虚多因脾气虚衰进一步发展而成,也可因饮食失调,过食生冷,或因寒凉药物太过,损 伤脾阳,或肾阳不足,命门火衰,火不生土而致。
[0005] 脾为湿土,喜燥恶湿。湿盛可以导致脾虚,脾虚也可以生湿,往往互为因果。因脾 虚失运,水湿停留,多属本虚标实之证。本虚为主者,治多健脾,佐以化湿;标实为主者,则应 以祛湿为主,兼以运脾。
[0006] 对于脾虚症状,临床上大多采用中医调理脾胃的运化,来达到治疗的目的,但是中 药方剂繁多,各个方剂的针对性不同,而且中药汤剂的煎煮和服用都十分不便,煎煮时间和 服用不当会给治疗带来很大的影响,因此需要一种既能够有效治疗脾虚症状,又能够方便 服用的药物来解决患者问题。

[0106] 本实施例通过小鼠口服给药药物半数致死量的测定,考察了实施例2制备的中药 组合物对小鼠的急性毒性反应和死亡情况。
[0107] 取小鼠30只,每组10只,分3组,即高、中、低3个剂量组(0. 8、0. 4、0. 2ml/15g) 灌胃给药3次/日,预测药物半数致死量的范围。结果显示,高剂量组测不出半数致死量, 故将药物灌胃给药半数致死量的测定改做最大体积下之最大弄点一最大耐受量的测定。
[0108] 取小鼠30只,空腹12小时后,以预测选定的0. 8ml/15g计量逐只灌胃给药给受试 动物3次/日,连续观察7天。

1. 一种治疗脾虚的药物组合物,其特征在于,包括:人参、党参、黄芪、茯苓、白术、白 芍、陈皮、木香、山药、扁豆、甘草、川芎、桂枝、肉桂、当归、升麻、柴胡、益智仁、蜀椒和附子, 由下述重量份组成:人参3份〜5份、党参3份〜5份、黄苗5份〜10份、获考 :15份〜25 份、白术15份〜25份、白芍15份〜25份、陈皮8份〜13份、木香8份〜13份、山药8份〜 13份、扁見8份〜13份、甘草8份〜13份、川弯15份〜25份、桂枝5份〜10份、肉桂5 份〜10份、当归15份〜25份、升麻1份〜3份、柴胡1份〜3份、益智仁1份〜3份、蜀椒 1份〜3份、附子1份〜3份。
2. 根据权利要求1所述的治疗脾虚的药物组合物,其特征在于,其药物组合物还包括: 干姜1份〜3份、山茱萸10份〜15份。
3. 根据权利要求2所述的治疗脾虚的药物组合物,其特征在于,其药物组合物重量份 组成为:人参3份、党参3份、黄芪5份、茯苓15份、白术15份、白芍15份、陈皮8份、木香 8份、山药8份、扁豆8份、甘草8份、川弯15份、桂枝5份、肉桂5份、当归15份、升麻1份、 柴胡1份、益智仁1份、蜀椒1份、附子1份、干姜1份、山茱萸10份。
4. 一种如权利要求1〜3任一项所述的治疗脾虚的药物组合物,其特征在于,制备方法 如下: 步骤S01,将人参、党参、当归、黄芪混合后加人参、党参、当归、黄芪总重量10倍〜20 倍的70%〜90%的乙醇在70°C〜90°C的条件下提取7小时〜9小时,提取液过滤,滤液备 用; 步骤S02,将白巧、陈皮、木香、山药、扁豆、甘草、川;、肉桂、升麻、柴胡、益智仁、蜀椒、 附子、干姜、山茱萸混合后加混合药材总重量10倍〜20倍的纯净水在70°C〜90°C的条件 下提取7小时〜9小时,提取液过滤,滤液备用; 步骤S03,茯苓、桂枝、白术混合后加纤维素酶用水提取,提取液灭活后过滤,滤液备 用; 步骤S04,将步骤S01、步骤S02和步骤S03得到的提取液合并,真空冷冻干燥得到药物 提取物固体干骨; 步骤S05,将步骤S04制得的药物提取物固体经超微粉碎,添加辅料,制粒,制成所需要 的剂型。
5. 根据权利要求4所述的治疗脾虚的药物组合物的制备方法,其特征在于,在步骤S03 中,所述茯苓、桂枝、白术混合加纤维素酶后的提取方式为:将茯苓、桂枝、白术混合后加茯 苓、桂枝、白术药材混合物重量10倍〜20倍的纯净水在45°C〜50°C的条件下3小时〜5 小时。
6. 根据权利要求4或5所述的治疗脾虚的药物组合物的制备方法,其特征在于,所述纤 维素酶的加入量为茯苓、桂枝、白术药材混合物重量的〇. 08%〜0. 1%。
7. 根据权利要求4所述的治疗脾虚的药物组合物的制备方法,其特征在于,在步骤S03 中,所述灭活的方法为升温到96°C〜100°C加热1小时〜2小时。
8. 根据权利要求4所述的治疗脾虚的药物组合物的制备方法,其特征在于,在步骤 S04中,所述提取液真空冷冻干燥的条件为:预冻温度-50°C〜_40°C,冻结时间6小时〜 8小时;升华干燥方式为直接将搁板温度设定为40°C〜50°C,冷阱中部循环管温度设定 为-501:〜-401:,干燥时间7小时〜9小时 ;包装操作环境湿度10%〜30%,温度151:〜 25。。。
