如本文中所使用,术语“靶向部分”和其相等物是指识别并且结合细胞或组织表面上的互补分子的分子。靶向部分的非限制性实例包括抗体、抗体片段、结合蛋白质和肽、受体和受体配位体。靶向部分的特异性结合伴侣意指所述靶向部分基本上不结合其他结合伴侣。在一些情况下,所述靶向部分/受体复合物或所述靶向部分/特异性结合伴侣复合物可以被细胞内在化。

包含与抑制癌症相关mRNA表达的siRNA和靶向部分两者连接的DNA树枝状聚合物的组合物是本发明的一个方面。一般来说,所述靶向部分将特异性结合癌细胞上的细胞表面受体或细胞表面蛋白,以便将所述siRNA递送至所靶向的癌细胞。任何细胞表面受体或细胞表面蛋白都可以用来结合所述DNA树枝状聚合物组合物的靶向部分。使用结合至在与靶向部分结合之后被内在化的细胞表面受体或细胞表面蛋白的靶向部分有利于将所述siRNA递送至细胞中,它在其中可以更有效地使互补mRNA降解。使用结合至在癌细胞上被过度表达的细胞表面受体和细胞表面蛋白的靶向部分有利于使得siRNA作用对使癌细胞中的互补mRNA降解的特异性相较于也能表达所述细胞表面受体或细胞表面蛋白,但量比肿瘤细胞上低的正常细胞而言有所提高。
所述靶向部分可以通过若干种已知的化学作用中的任一种与所述DNA树枝状聚合物的可用臂连接,这取决于所述靶向部分的化学性质。实例包括形成二硫桥连键、通过N-羟基丁二酰亚胺酯依赖性缩合、通过使用双官能交联反应、通过使所述靶向部分与所述DNA树枝状聚合物的臂直接或间接杂交或者通过使用多聚阳离子化合物经由电荷-电荷相互作用来桥连所述靶向部分与所述DNA树枝状聚合物。关于RNA的这些连接反应描述于WO 2010/017544中,但可以应用于其他适当的靶向部分。还可以如美国专利第8685899号中所描述使用桥连寡核苷酸通过平端连接将核酸靶向部分连接至所述DNA树枝状聚合物。用于使所述靶向部分与所述DNA树枝状聚合物杂交的适当选择包括如WO 02/33125中所描述,对所述靶向部分附加与所述DNA树枝状聚合物的臂的末端序列互补的核酸捕获序列。

结合至目标细胞类型上的细胞表面受体或细胞表面蛋白的任何靶向部分都可以连接至所述DNA树枝状聚合物。关于所述DNA树枝状聚合物组合物的抗癌治疗用途,一般将需要选择结合至在癌细胞上被特异性表达或在癌细胞上被过度表达的细胞表面受体或细胞表面蛋白的靶向部分。此类靶向部分的实例包括叶酸盐、转铁蛋白和抗间皮素抗体,以及本领域技术人员已知的其他合适的靶向部分。
与目标癌细胞中表达的互补癌症相关mRNA杂交并使其降解的任何siRNA都可以连接至所述DNA树枝状聚合物。此类mRNA的非限制性实例包括HuR,其在卵巢癌细胞中被过度表达。结合至HuR mRNA并且引起其降解的任何siRNA都适用于所述DNA树枝状聚合物组合物中。可以连接至连同有所述靶向部分的所述DNA树枝状聚合物的可用siRNA的具体非限制性实例包括SEQ ID NO:1/SEQ ID NO:3和SEQ ID NO:2/SEQ ID NO:4。在其他非限制性实例中,siRNA SEQ ID NO:1/SEQ ID NO:3或SEQ ID NO:2/SEQ ID NO:4可以连接至具有与所述DNA树枝状聚合物连接的叶酸盐作为所述靶向部分的所述DNA树枝状聚合物。
在上述DNA树枝状聚合物组合物中的任一种中,所述靶向部分和所述siRNA与所述DNA树枝状聚合物的连接可以独立地选自由以下各项组成的组:经由与所述DNA树枝状聚合物的臂连接的捕获寡核苷酸和与所述靶向部分和/或siRNA连接的互补载体寡核苷酸之间的化学缀合和杂交而直接连接至所述DNA树枝状聚合物的臂。
在上述DNA树枝状聚合物组合物中任一种的具体实施方案中,所述靶向部分和/或所述siRNA可以经由与所述DNA树枝状聚合物的臂缔合的捕获寡核苷酸而连接至所述DNA树枝状聚合物。所述捕获寡核苷酸一般与所述DNA树枝状聚合物臂的末端缔合。它通常连接至所述DNA树枝状聚合物的臂的末端,但是它还可以与所述末端杂交并且任选地与其交联或通过使用延伸寡核苷酸而与所述臂缔合,如以下所描述。所述捕获寡核苷酸提供以限定的量存在的特异性限定序列,以便与连接至所述靶向部分和/或所述siRNA的互补载体寡核苷酸杂交。所述捕获寡核苷酸还提供用于控制连接至所述DNA树枝状聚合物的靶向部分和siRNA的数目的手段,因为所述载体寡核苷酸可以在限定的浓度下与所述捕获寡核苷酸杂交,由此使得所期望数目的DNA树枝状聚合物臂被各组分占据。在所述互补载体寡核苷酸杂交后,所述靶向部分和/或siRNA通过沃森-克里克碱基配对连接至所述DNA树枝状聚合物的臂。
上述DNA树枝状聚合物组合物中的任一种都可以与药学上可接受的赋形剂一起配制为药物制剂。此类赋形剂可以包括选自由以下各项组成的组的一种或多种组分:溶剂、分散剂、包覆剂(例如卵磷脂)、表面活性剂(例如羟丙基纤维素)、防腐剂(例如对羟基苯甲酸、苯酚、硫柳汞、山梨酸、氯代丁醇)、乳液、醇(例如,乙醇)、多元醇(例如,甘油、丙二醇)和等张剂(例如,糖、氯化钠)。
包含所述DNA树枝状聚合物组合物的药物制剂还可能适合于本领域中已知的特定施用途径。作为非限制性实例,所述药物制剂可以配制为用于注射或输注的溶液或者为用于口服施用的锭剂。可以例如在水或生理盐水中制备所述DNA树枝状聚合物组合物的溶液或分散液。
包含所述DNA树枝状聚合物组合物的药物制剂可用于治疗患者的癌症、抑制肿瘤细胞增殖和/或抑制肿瘤生长。用于抑制肿瘤细胞增殖和/或抑制肿瘤生长的方法可以在体内应用,用于治疗癌症患者,或在体外应用,诸如用于研究或诊断目的。一般来说,体内施用至癌症患者的药物制剂的量将是有效抑制肿瘤细胞增殖、抑制肿瘤生长和/或治疗患者的癌症的量。有效量将基于施用剂量、施用持续时间、施用方案和施用途径的组合。本领域技术人员将会认识到,可能在医学上有必要或期望基于患者的身体组成或他/她对治疗的反应对剂量进行个性化。
用于治疗患者的癌症的方法可以包括通过肠胃外、腹膜内、静脉内、肿瘤内或口服途径施用至受试者。特别有用的施用途径包括注射、输注或口服施用。为了治疗癌症,典型施用过程可以包括在若干周的过程中连续地或间歇性地进行静脉内施用,随后一段时期通常不进行施用,然后将间歇性施用计划重复至少一次。
实施例
siRNA稳定性、功能性和生物分布的体外测定:
在PBS、无血清培养基和培养基+10%血清中评估siHuR稳定性。siHuR的10%TBUPAGE表明,任何样品中的有义股和反义股都没被降解。
为了评估功能性,制备了与叶酸盐、siHuR序列和荧光标记(Cy3)的不同组合连接的2层DNA树枝状聚合物。通过用以下各物转染细胞来测试siRNA稳定性:siHuR+脂质转染剂、与叶酸盐连接的DNA树枝状聚合物以及与叶酸盐和siHuR两者连接的DNA树枝状聚合物。对照物是未转染的细胞和仅脂质转染剂。
将siHuR(SEQ ID NO:2/SEQ ID NO:4)与脂质转染剂或靶向性DNA树枝状聚合物组合,并且与A2780细胞(人卵巢肿瘤细胞系)一起在含有10%血清的培养基中孵育过夜。将各情况下的HuR mRNA表达与未转染的细胞(100%HuR表达)相比较。结果示于下表中:
相较于仅有siHuR的转染,使用与DNA树枝状聚合物连接的叶酸盐使siHuR靶向卵巢癌细胞显著改善了HuR mRNA敲低。
在携带卵巢肿瘤的小鼠中利用全身施用与CY3和叶酸盐连接的DNA树枝状聚合物进行的生物分布研究表明,卵巢肿瘤中存在非常高的荧光,而正常组织(脑、肝脏、脾脏、肺、心脏和正常卵巢)中存在极少乃至不存在荧光。这些结果表明,对于过度表达叶酸盐受体的癌症来说,可以使用叶酸盐作为连接至所述DNA树枝状聚合物的靶向部分来实现癌症治疗的特异性。
关于对卵巢肿瘤生长的抑制的体内测试:对携带卵巢肿瘤的小鼠每周两次经腹膜内注射2μg siHuR(SEQ ID NO:1/SEQ ID NO:3),所述siHuR呈以下形式:与叶酸盐和siHuR连接的DNA树枝状聚合物、与siHuR连接的DNA树枝状聚合物以及与叶酸盐和阴性对照物siRNA连接的DNA树枝状聚合物。对照物是未衍生化的DNA树枝状聚合物和0.9%生理盐水(未处理)。注射持续四周。
模型系统是经腹膜内注射ID8-Fluc鼠类卵巢癌细胞的C57BL/6雌性小鼠,每个处理组六只小鼠。肿瘤发展遍布整个腹膜腔。注射后大约4周,对携带肿瘤的小鼠进行光学成像以确定基线生物发光,即ID8-luc衍生的肿瘤负载的指标,然后每周两次用四种DNA树枝状聚合物制剂之一或者用0.9%生理盐水处理四周(眼球后注射)(n=5-6只/组)。所述四种DNA树枝状聚合物制剂是:1)与叶酸盐和siHuR连接的DNA树枝状聚合物;2)与siHuR连接的DNA树枝状聚合物;3)与叶酸盐和阴性对照物siRNA连接的DNA树枝状聚合物;和4)未衍生化的DNA树枝状聚合物。每周一次通过光学成像来评估肿瘤对处理的反应。结果示于附图中。使用基线肿瘤负载作为比较指标,在用与叶酸盐和siHuR连接的DNA树枝状聚合物治疗4周的小鼠中,肿瘤生长被抑制约3倍。这是所有处理组中的最高抑制,包括与siHuR连接的DNA树枝状聚合物(p=0.13),并且它实质上比用与叶酸盐和阴性对照物siRNA连接的DNA树枝状聚合物(p=0.22)、未衍生化DNA树枝状聚合物(p=0.10)或0.9%生理盐水(p=0.10)处理的小鼠更佳。这些结果支持使用与叶酸盐和siHuR连接的DNA树枝状聚合物治疗癌症时可以观察到治疗效果有所增强。

Claims (19)
