Detailed Description
The following detailed description of specific embodiments of the invention is provided, but it should be understood that the scope of the invention is not limited to the specific embodiments.
The information on the purity and supplier of the tiger sex pheromone component and the plant volatile component used in the following examples is shown in table 1.
TABLE 1 Eggera tiger sex pheromone component and synergist components
Example 1Y-type olfactometer determination of a one-component synergist
Preparing a plant-derived volatile single agent: each compound was prepared at 3 concentrations of 0.1, 1, 10. mu.g/. mu.l using n-hexane as a solvent, as a control.
The sensitivity of the tiger is measured by a Y-shaped olfactometer to various reagents. In a room with air cleanness and a temperature of 25 deg.C and a relative humidity of about 70%, a 3 cm-20 cm Y-type olfactometer is selected for biological measurement. Mu.l of the single-component compound of the synergist was dropped onto a filter paper strip, placed in one side arm of a Y-type olfactometer, and the other side arm was placed with an equal amount of n-hexane as a control. After each group is repeated, the positions of the test group and the control group are exchanged, so that the test error is avoided. The filter paper strip is replaced with a new one after each test, each reagent is repeated for 3 times, and a clean Y-shaped tube needs to be replaced when each reagent is replaced. 1 splayed tiger fasted for 24 hours is connected to a position 3cm away from a main arm of the Y-shaped olfactometer, and if the splayed tiger adults stay in any side arm for more than 1min, the selection is considered to be made; if the main arm stays for l0min, no reaction is considered. Repeat 30 tigers in eight characters and 15 male and female heads respectively. And observing the distribution condition of the test insects after 10min, and calculating the reaction rate, the relative reaction rate and the reaction coefficient to perform data analysis.
As can be seen from Table 2, the reaction rates of the selected test insects are all over 80%, and no significant difference exists among treatments, which indicates that the selected test insects are healthy, and the experimental results of the research are reliable. As can be seen from the selective reaction rate, the difference of the reaction values of the black cutworm to different concentrations of the same synergist is not significant. The response values of males and females are not very different for the same synergist. The Sedum octopoda has the highest selective reaction rate (male worm is 0.83 +/-0.04, female worm is 0.84 +/-0.06) to cis-3-hexen-1-ol, which is obviously higher than other synergists with different concentrations. The single-component synergist has attraction activity on the adult athyrium segetaum, wherein the attraction activity of cis-3-hexenyl-1-ol, heptaldehyde and 1-hexanol on the athyrium segetaum is equivalent, and the attraction activity of phenylacetaldehyde and linalool is relatively poor.
TABLE 2 olfactory reaction of the black tiger to a one-component synergist
The data in the table are mean ± sem. Different letters after the same column of data indicate significant differences at P <0.01 levels as tested by Duncan's new repolarization method.
Example 2 synergistic Effect of synergists with different formulations on the sex pheromone of the black tiger
1-hexanol, phenylacetaldehyde, cis-3-hexen-1-ol, heptanal, linalool and the tiger sex pheromone are respectively prepared into n-hexane solution of 500 mu g/mu L. Wherein the star-shaped tiger sex pheromone is obtained by uniformly mixing 9 parts of cis-7-tetradecene acetate, 0.8 part of cis-5-tetradecene acetate and 0.2 part of trans-5-tetradecene acetate.
Sucking a certain amount of each synergist component solution and the splayed cutworm sex pheromone solution by a 200 mu L liquid-transferring gun, and uniformly mixing to obtain a splayed cutworm plant source sex pheromone mixture. The ingredients of the attractant composition are shown in Table 1.
And (3) sucking 200 mu L of the splayed cutworm attractant composition or the splayed cutworm sex pheromone by using a 200 mu L liquid transfer gun, dripping the 200 mu L splayed cutworm attractant composition or the splayed cutworm sex pheromone on a spacer of a rubber plug which is cleaned by n-hexane, dripping 1 ml of dichloromethane for several times after the n-hexane is volatilized, and obtaining the splayed cutworm attractant composition attractant core or the splayed cutworm sex pheromone attractant core after the dichloromethane is volatilized. Each lure contains 100mg of the composition of the eight-character black cutworm attractant or the sex pheromone of the eight-character black cutworm. The prepared lure core is put into an aluminum foil for sealing and is stored at the temperature of minus 20 ℃.
When the trapping test is carried out in the field, the splayed cutworm attractant composition attracting core or the splayed cutworm sex pheromone attracting core is fixed on the general moth trapper by using a thin iron wire. CK1 is blank control containing no core-inducing composition or core-inducing sex pheromone of Gekko swinhonis; CK2 control of tiger sex pheromone lure only in eight characters; groups X1 to X5 are treatments of single component synergist wicks only; group 01 to group 15 are treatments of the lure of the composition containing the one-component black cutworm lure; 16 to 42 groups are the treatment of the core of the splayed cutworm attractant composition containing the two-component synergist; 43 groups to 51 groups are the treatment of the core of the black cutworm attractant composition containing the three-component synergist; the 52 groups to 57 groups are the treatment of the core luring of the splayed cutworm attractant composition containing the four-component synergist; groups 58 to 65 are the treatments of the core luring of the splayed cutworm attractant composition containing the five-component synergist. Each treatment was set up in triplicate, randomly arranged, with two traps adjacent at a spacing of 10 m. And recording the number of the trapped black cutworms in the shape of eight characters in the morning every day from the next day after setting, and removing the black cutworms in time for continuously surveying for 30 days.
The test results are shown in table 3. As can be seen from Table 3, the single-component synergist has an attracting activity to the black cutworm, but the trapping effect is significantly lower than that of the black cutworm sex pheromone itself. However, the trapping effect of the single-component synergist and the tiger sex pheromone after being mixed is obviously higher than that of the single-component synergist. The synergist and the sex pheromone are mixed according to different formula proportions, the trapping effect of the sex pheromone trapping core is higher than that of the sex pheromone, and the using amount of the sex pheromone is greatly reduced.
TABLE 3 field synergism of different synergistic Agents on the eight-character cutworm sex pheromone
The splayed cutworm attractant composition provided by the invention can be accurately applied to the insect condition prediction, mass trapping and mating interference of splayed cutworms, and has important significance for comprehensively treating the splayed cutworms. The splayed cutworm attractant composition is simple to prepare, simple to use, feasible in conditions, strong in trapping capability and good in stability; meanwhile, the composition of the black cutworm attractant has strong specificity, does not generate resistance, has no harm to natural enemies, is nontoxic to people and livestock, and is environment-friendly; the stress resistance is strong, and the wind, rain and sunshine resistance is realized; the duration is long, and the control effect can be maintained for more than 30 days.
The attractant composition is prepared from the star-shaped cutworm sex pheromone and plant volatile substances, and is characterized in that (1) the sex pheromone has the advantages of high efficiency, low toxicity and no pollution; (2) noctuid insects are difficult to generate drug resistance based on noctuid attractants made from host plant volatiles; (3) compared with the traditional pesticide, the plant source noctuid attractant has little dosage and can generate biological activity only with a very small dosage; (4) the insect-attracting spectrum is wide. According to the principle that insects recognize host plant volatile matters and female insects release sex pheromone which has strong attraction capacity to the same kind of males, the formula proportion between the sex pheromone of the female black cutworm and the plant volatile matters can be accurately applied to insect condition observation and report, pest quarantine, mating interference and mass trapping, so that the ideal effect of comprehensively treating the black cutworm is achieved.
Those of ordinary skill in the art will understand that: the invention is not to be considered as limited to the specific embodiments thereof, but is to be understood as being modified in all respects, all changes and equivalents that come within the spirit and scope of the invention.
