Abstract
本发明涉及一种从竹叶中提取竹叶总黄酮的方法。该方法包括以下步骤:首先取竹叶,加水提取,加乙酸乙酯分次萃取,至水液盐酸镁粉测试反应较弱;然后收集乙酸乙酯萃取液,浓缩制干膏,加水溶解,加三氯甲烷,析出的不溶物为目标产品。本发明方法所得竹叶总黄酮产品产率纯度高。总黄酮含量超过50%,产率为0.83%‑1.64%,得到的竹叶总黄酮相对于原料竹叶含有的竹叶总黄酮转移率可达60%以上。且制备工艺简单易行,可实现大规模化工业生产。
Description
技术领域
本发明属于天然产物提取分离技术领域,具体涉及一种从竹叶中制备竹叶总黄酮的方法。
背景技术
竹子是禾本科多年生常绿植物,具有丰富的使用价值。我国的竹林资源约占全国森林总面积的3%,占世界竹林总面积的25%,有“竹子王国”之美称。竹叶在我国具有悠久的药用和食用历史,其医疗保健作用早已为人们所认识。《文经逢原》记载:“竹叶兼行肌表,能疗疮杀虫,竹茹专清胃腑,能止呕除烦”。《中药大辞典》记载:“竹叶清热除烦,生津利尿”。随着现代科学研究的深入,发现竹叶中含有大量对人体有益的活性物质。其中,黄酮是竹叶中一类重要的活性成分,具有抗氧化、清除自由基、抗衰老、抗辐射、抑菌、抗菌防腐、降血脂及保护心脑血管等功效,其显著的功效和广阔的应用前景,引起了研究人员的广泛关注,正成为研究的一个热点。竹叶黄酮具有来源丰富、天然安全等优点,更因其具有优良的保健功能,可用作药品、保健品、香料、化妆品、食品防腐剂、天然抗氧化剂等功能性添加剂。对黄酮类成分进行深层次的开发利用,必将提高竹业生产的经济效益,对于促进竹产业向纵深发展具有重要意义。
现有从竹叶中提取纯化竹叶总黄酮的方法,多为色谱方法,多从纯天然产物化学实验室的角度进行研究,存在方法复杂、不适合工业放大、产率低、效率不高、成本高等问题。因此,有必要寻找一种较为简单、高效的提取分离竹叶总黄酮的方法。
发明内容
为克服以上缺陷,本发明提供一种竹叶总黄酮的提取方法。该方法包括以下步骤:
1、取竹叶,加水提取,加乙酸乙酯分次萃取,至水液盐酸镁粉测试反应较弱;
2、收集乙酸乙酯萃取液,浓缩制干膏,加水溶解,加三氯甲烷,析出的不溶物为目标产品。
作为优选方案:
步骤(1)所述的加水提取的料液比为1:(6-10),次数为2-3次,每次提取时间为30-90min。
步骤(1)所述的加水提取后,水提取液浓缩至相对密度为1.10-1.20,再用乙酸乙酯萃取。
步骤(1)所述的乙酸乙酯用量与竹叶的体积/质量比为1升/5千克,提取次数为2-3次。
步骤(2)所述的加水用量与竹叶的体积/质量比为1升/10千克,所述三氯甲烷用量按体积计与水等同。
步骤(2)中加三氯甲烷后静置12~24h。
作为最优方案包括以下步骤:
1、取竹叶,按料液比1:10加水提取3次,每次30min;水提取液浓缩至相对密度为1.15;再按料液比5kg:1L用乙酸乙酯萃取3次;
2、收集乙酸乙酯萃取液,浓缩制干膏,按料液比10kg:1L加水溶解,再加与水等体积的三氯甲烷,静置24h,析出的不溶物为目标产品。
本发明方法优点在于:工艺简单易行,易实现大规模化工业生产;竹叶总黄酮产品产率高、纯度高。采用《保健食品检验与评价技术规范(2003年版)》所收载总黄酮的测定方法测定,总黄酮含量超过50%,产率在0.83%-1.64%,得到的竹叶总黄酮相对于原料竹叶含有的竹叶总黄酮转移率可达60%以上。
具体实施方式
下面结合具体的实施例对本发明做进一步的详细说明,但本发明并不局限于此。
实施例1竹叶提取工艺优化
提取竹叶中所含黄酮类成分,结合其理化性质,选择水煎煮法提取,为选择最佳提取工艺,对影响水煎煮提取效率的三个主要因素加水量(A)、提取次数(B)、提取时间(C),以总黄酮的转移率为考核指标,通过正交试验进行了优化。按三因素三水平,用L9(34)正交表进行正交试验。结果见表1、表2及表3。

在三个因素中,A因素(加水量)、B因素(煎煮次数)对总黄酮的提出有影响,而C因素(加热时间)对其无影响。如想得到最大的总黄酮得量,并从生产周期成本方面考虑,最佳工艺方案是每次提取加水10倍量,每次煎煮0.5小时,共煎煮三次。
实施例2竹叶黄酮的乙酸乙酯萃取分离
取竹叶10千克,煎煮提取3次,每次提取加水10倍量,煎煮0.5小时,滤过,合并滤液,浓缩至稠膏后,平均分成四份,分别加纯净水调匀至相对密度(室温)1.10、1.15、1.20、1.25,用同体积乙酸乙酯萃取。当浓缩太稠时,不利于萃取,太稀,则萃取所用乙酸乙酯量大,耗费溶剂,不经济环保。在1.10、1.15、1.20均萃取得到成功,1.25太稠不易分层。
当相对密度为1.15;乙酸乙酯萃取3次,每次用量0.5升,萃取后的水液用盐酸镁粉反应检测残留黄酮情况,反应已较弱,反映萃取较好。
实施例3竹叶黄酮的氯仿除杂纯化
取竹叶10千克,煎煮提取3次,每次提取加水10倍量,煎煮0.5小时,滤过,合并滤液,浓缩至相对密度为1.15;乙酸乙酯萃取3次,每次用量2升,萃取后的水液用盐酸镁粉反应检测残留黄酮情况,反应已较弱。将乙酸乙酯萃取液合并回收乙酯乙酯,得干膏(乙酸乙酯味较浓),加入1升纯净水,加热搅拌均匀,平均分成两份。放冷,然后分别加入0.5升三氯甲烷,搅拌后,一个放置12小时,一个放置24小时后观察。二者均分为水层和三氯甲烷层,在上层水层和下层三氯甲烷层之间均有棕黄色固体析出,分别滤过,以冷水适量冲洗,干燥,分别得81.2克、85.1克。
采用《保健食品检验与评价技术规范(2003年版)》所收载总黄酮的测定方法测定,总黄酮含量分别为53.21%、54.87%。
实施例4不同产地竹叶总黄酮的制备
对采自广宁县的不同乡镇的竹叶五份,每份10千克,煎煮提取3次,每次提取加水10倍量,煎煮0.5小时,滤过,合并滤液,浓缩至相对密度为1.15;乙酸乙酯萃取3次,每次用量2升,萃取后的水液用盐酸镁粉反应检测残留黄酮情况,反应已较弱。将乙酸乙酯萃取液合并,回收乙酯乙酯,得干膏(乙酸乙酯味较浓),加入1升纯净水,加热搅拌均匀,放冷,分别加入1升三氯甲烷,搅拌后,放置24小时。均在上层水层和下层三氯甲烷层之间有棕黄色固体析出,分别滤过,以冷水适量冲洗,干燥。
采用《保健食品检验与评价技术规范(2003年版)》所收载总黄酮的测定方法,测定原料竹叶与各总黄酮的含量。结果见表4。

Claims (7)
1.一种竹叶总黄酮的提取方法,其特征在于,包括以下步骤:
(1)取竹叶,加水提取,加乙酸乙酯分次萃取,至水液盐酸镁粉测试反应较弱;
(2)收集乙酸乙酯萃取液,浓缩至干膏,加水溶解,加三氯甲烷,析出的不溶物为目标产品。
2.根据权利要求1所述的方法,其特征在于,步骤(1)所述的加水提取的料液比为1:(6-10),次数为2-3次,每次提取时间为30-90min。
3.根据权利要求1所述的方法,其特征在于,步骤(1)所述的加水提取后,水提取液浓缩至相对密度为1.10-1.20,再用乙酸乙酯萃取。
4.根据权利要求1所述的方法,其特征在于,步骤(1)所述的乙酸乙酯用量与竹叶的体积/质量比为1升/5千克,提取次数为2-3次。
5.根据权利要求1所述的方法,其特征在于,步骤(2)所述的加水用量与竹叶的体积/质量比为1升/10千克,所述三氯甲烷用量按体积计与水等同。
6.根据权利要求1所述的方法,其特征在于,步骤(2)中加三氯甲烷后静置12~24h。
7.根据权利要求1所述的方法,其特征在于,包括以下步骤:
