Abstract
本发明涉及一种降尿酸利尿组合物及其制备方法,该组合物包括以下重量份的原料:芹菜籽40‑60份、绞股蓝10‑20份、菊苣根40‑60份、灵芝27‑32份、蒲公英27‑32份、玉米须27‑32份。制备方法为:取芹菜籽、绞股蓝,加入乙醇,搅拌混合均匀,煮沸提取2‑4次,每次提取时间为1‑3h,过滤,得提取液I;将提取液I使用旋转蒸发仪去除乙醇后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物I;取菊苣根、灵芝、蒲公英、玉米须,加入纯化水,搅拌混合均匀,煮沸提取1‑3次,每次提取时间为1‑3h,过滤,得提取液II;将提取液II煮沸浓缩后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物II;将干燥物I和干燥物II混合均匀制成颗粒即可。本发明的组合物具有显著利尿和降低血尿酸的功效。
Description
技术领域
本发明涉及保健食品领域,具体涉及一种降尿酸利尿组合物及其制备方法。
背景技术
芹菜籽体形呈卵形,非常小,颜色多为棕褐色或深棕色,含桉油醇、磷甲氧基酚、伞花内酯、芹菜素、亚麻酸、挥发性油脂、黄酮、硼,矿物质如:钙、镁、钠、锌。芹菜籽所含降压、降脂成分是芹菜的50倍,短期服用起到降血压,降血脂的作用,长期服用远远优于其他中西药品,无任何毒副作用。芹菜籽的药理作用非常广泛,有较高的使用价值。现代药理研究表明芹菜籽具有降血压、降血脂的作用。由于芹菜籽的钙磷含量较高,所以他有一定镇静和保护血管的作用,又可增强骨骼,预防小儿软骨病。降尿酸利尿组合物中主要是芹菜籽油和黄酮,芹菜籽油中富含不饱和脂肪酸、氨基酸以及钾镁等矿物质。
菊苣根系维吾尔族习用药材。为菊科植物毛菊苣及菊苣干燥的根。菊苣为菊科植物菊苣的地上部分,菊苣的功效是清热解毒、利尿消肿,菊苣的作用是治湿热黄疸、肾炎水肿、胃脘胀痛、食欲不振。根含山莴苣素,野莴苣甙,山莴苣醇,即是蒲公英甾醇,菊苣内酯A,菊苣萜甙B、C,8-脱氧山莴苣素,苦苣菜甙A、C,假还阳参甙B等。菊苣根对于肝脏总脂质、胆固醇、三酰甘油等等有降低的效果。
玉米须为禾本科植物玉蜀黍的花柱和柱头。玉米须常集结成疏松团簇,花柱线状或须状,完整者长至30厘米,直径0.5毫米,淡绿色、黄绿色至棕红色,有光泽,略透明,柱头2裂,叉开,质柔软,气无,味淡。以柔软、有光泽者为佳。夏、秋果实成孰时收集,除去杂质。鲜用或晒干生用。味甘、淡,性平。有利尿消肿;清肝利胆的功效。在中药里,玉米须有广泛的预防保健用途。中医认为能利水消肿,泄热,平肝利胆,还能抗过敏,治疗肾炎水肿、肝炎、高血压、胆囊炎、胆结石、糖尿病、鼻窦炎、乳腺炎等。开水冲泡代茶饮,可以消暑清热。常饮有减肥作用,对防治动脉粥样硬化、高血压病大有裨益。高血脂、高血压、高血糖的病人喝了,可以降血脂、血压、血糖。
蒲公英属多年生草本植物。根圆锥状,表面棕褐色,皱缩,叶边缘有时具波状齿或羽状深裂,基部渐狭成叶柄,叶柄及主脉常带红紫色,花葶上部紫红色,密被蛛丝状白色长柔毛.蒲公英植物体中含有蒲公英醇、蒲公英素、胆碱、有机酸、菊糖等多种健康营养成分。性味甘,微苦,寒。归肝、胃经。有利尿、缓泻、退黄疸、利胆等功效。
发明内容
本发明的目的在于:提供一种降尿酸利尿组合物及其制备方法。
为实现上述目的,本发明提供如下技术方案:
一种降尿酸利尿组合物,包括以下重量份的原料:芹菜籽40-60份、绞股蓝10-20份、菊苣根40-60份、灵芝27-32份、蒲公英27-32份、玉米须27-32份。
优选地,一种降尿酸利尿组合物,包括以下重量份的原料:芹菜籽50份、绞股蓝15份、菊苣根50份、灵芝29.5份、蒲公英29.5份、玉米须29.5份。
一种降尿酸利尿组合物的制备方法,具体步骤如下:
(1)取原料芹菜籽、绞股蓝,加入乙醇,搅拌混合均匀;
(2)将步骤(1)中的原料煮沸提取2-4次,每次提取时间为1-3h,过滤,得提取液I;
(3)将提取液I使用旋转蒸发仪去除乙醇后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物I备用;
(4)取原料菊苣根、灵芝、蒲公英、玉米须,加入纯化水,搅拌混合均匀;
(5)将步骤(4)中的原料煮沸提取1-3次,每次提取时间为1-3h,过滤,得提取液II;
(6)将提取液II煮沸浓缩后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物II备用;
(7)将干燥物I和干燥物II混合均匀制成颗粒即可。
优选地,步骤(1)中乙醇的质量分数为75-85%,乙醇的加入量为芹菜籽和绞股蓝总重量的8-12倍。
优选地,步骤(4)中纯化水的加入量为菊苣根、灵芝、蒲公英、玉米须总重量的8-12倍。
本发明的有益效果在于:
本发明的组合物具有显著利尿和降低血尿酸的功效。本发明采用生物工程技术提取多种原料,具有提取率高,无毒副作用,环保等优点。
具体实施例
实施例1
一种降尿酸利尿组合物的制备方法,具体步骤如下:
(1)称取原料芹菜籽40份、绞股蓝10份,加入80%乙醇,乙醇的加入量为原料总重量的10倍,搅拌混合均匀;
(2)将步骤(1)中的原料煮沸提取3次,每次提取时间为1.5h,过滤,得提取液I;
(3)将提取液I使用旋转蒸发仪去除乙醇后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物I备用;
(4)称取原料菊苣根40份、灵芝27份、蒲公英27份,玉米须27份,加入纯化水,纯化水的加入量为原料总重量的10倍,搅拌混合均匀;
(5)将步骤(4)中的原料煮沸提取2次,每次提取时间为2h,过滤,得提取液II;
(6)将提取液II煮沸浓缩后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物II备用;
(7)将干燥物I和干燥物II混合均匀制成颗粒即可。
实施例2
一种降尿酸利尿组合物的制备方法,具体步骤如下:
(1)称取原料芹菜籽50份、绞股蓝15份,加入80%乙醇,乙醇的加入量为原料总重量的10倍,搅拌混合均匀;
(2)将步骤(1)中的原料煮沸提取3次,每次提取时间为1.5h,过滤,得提取液I;

(4)称取原料菊苣根50份、灵芝29.5份、蒲公英29.5份,玉米须29.5份,加入纯化水,纯化水的加入量为原料总重量的10倍,搅拌混合均匀;
(5)将步骤(4)中的原料煮沸提取2次,每次提取时间为2h,过滤,得提取液II;
(6)将提取液II煮沸浓缩后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物II备用;
(7)将干燥物I和干燥物II混合均匀制成颗粒即可。
实施例3
一种降尿酸利尿组合物的制备方法,具体步骤如下:
(1)称取原料芹菜籽60份、绞股蓝20份,加入80%乙醇,乙醇的加入量为原料总重量的10倍,搅拌混合均匀;
(2)将步骤(1)中的原料煮沸提取3次,每次提取时间为1.5h,过滤,得提取液I;
(3)将提取液I使用旋转蒸发仪去除乙醇后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物I备用;
(4)称取原料菊苣根60份、灵芝32份、蒲公英32份,玉米须32份,加入纯化水,纯化水的加入量为原料总重量的10倍,搅拌混合均匀;
(5)将步骤(4)中的原料煮沸提取2次,每次提取时间为2h,过滤,得提取液II;
(6)将提取液II煮沸浓缩后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物II备用;
(7)将干燥物I和干燥物II混合均匀制成颗粒即可。
实施例4
芹菜素检测
配置标准曲线
精密称取干燥至恒重的芹菜素对照品20mg,于50ml容量瓶中定容,作为储备液0.4mg/ml。测量时精密吸取1m至10ml容量瓶中定容(20μg·mL-1)再取5ml稀释至10ml,配置标准曲线。
分组:
a组 实施例1的产物
b组 实施例2的产物
c组 实施例3的产物
将实施例1、2、3的产物分别置于研钵研磨的较分散后备用,精密称取1g待测物,溶于100ml甲醇,搅拌均匀静置1h后取上清稀释150倍后,过针头,在270nm下测定吸光度,然后根据标准曲线得出芹菜素含量如下表1所示:

实施例5
动物学实验
1、实验目的:研究降尿酸利尿组合物对于高尿酸大鼠尿酸的影响
2、实验材料
雄性SD大鼠,体重250g±30g,购于山东济南。盐酸乙胺丁醇片(0.25g*100)上海上药信谊药厂有限公司,别嘌醇片(0.1g*100)上海信谊万象药业股份有限公司,西芹3000由厂家提供,腺苷酸,Solarbio Life Science,降尿酸利尿组合物由厂家提供。T-AOC、MDA、SOD试剂盒购自南京建成生物工程研究所。
3、实验方法
大鼠适应性饲养一周,随机分组,每组15只,分组如下:
A:正常对照组
B:模型对照组
C:西芹3000组
D:别嘌醇组
E:降尿酸利尿组合物实施例1产物组
F:降尿酸利尿组合物实施例2产物组
G:降尿酸利尿组合物实施例3产物组
模型组及各给药组分别给予腺嘌呤100mg/(kg体重*d)+盐酸乙胺丁醇300mg/(kg体重*d)于造成高尿酸动物模型,每天于造模后四小时给予治疗药物。C组给予西芹3000,24mg/100g体重,每100g给予0.5ml体积灌胃;D组给予别嘌醇2.4mg/100g;E、F、G组给予140mg/100g。模型组和对照组分别给予等体积的蒸馏水灌胃。大鼠每周记录体重。
实验的第三周结束后,禁食12h,称重取腹主脉血,离心分离血清,肾功能指标送艾迪康生物检测公司检测,并按照说明书方法测血清SOD、T-AOC活性和MDA水平。
4、实验结果
4.1实验过程中大鼠体重记录:
造模过程中大鼠体重明显降低,药物治疗对大鼠体重无明显改善。

4.2降尿酸利尿组合物对高尿酸血症大鼠肾指数的影响
由表3可知,模型组大鼠肾指数明显增加,药物治疗组队肾脏指数无明显改善,但是实施例2产物组的肾脏指数明显增加。

4.3降尿酸利尿组合物对高尿酸血症大鼠抗氧化作用
由表4可知,模型组大鼠血清超氧化物歧化酶(SOD)活性明显降低(P<0.0001),西芹3000和降尿酸利尿组合物可明显增加SOD活性。模型组大鼠总抗氧化能力明显增加,药物治疗对总抗氧化能力无明显影响。模型组大鼠血清丙二醛(MDA)明显增加(**P<0.01),降尿酸利尿组合物均使MDA明显增加(**P<0.05),而对照药西芹3000和别嘌呤醇对MDA无明显作用。

4.4降尿酸利尿组合物对高尿酸血症大鼠尿酸和肾功能的影响
由表5可知,模型组大鼠尿酸明显增加,肾功能明显降低,表现为SCr和BUN明显增加,别嘌呤醇能明显降低尿酸,降尿酸利尿组合物均能明显降低尿酸水平,作用优于西芹3000。别嘌呤醇和西芹3000对高尿酸导致的大鼠肾功能降低无改善作用,但是降尿酸利尿组合物,尤其是实施例3产物组能明显降低SCr和BUN,提高大鼠肾脏功能。

5、结论
在本实验造模时间内,本发明的降尿酸利尿组合物有明显降低尿酸的作用,以实施例3产物为优,且降尿酸效果和别嘌呤醇相似,优于西芹3000;降尿酸利尿组合物对高尿酸大鼠肾功能有明显保护作用,而别嘌呤醇和西芹3000对肾功能无保护作用;降尿酸利尿组合物有明显抗氧化作用,表现为降低MDA和增加SOD,对总抗氧化作用不明显。所以实施例3是最佳的实施例方案。
Claims (5)
1.一种降尿酸利尿组合物,其特征在于:包括以下重量份的原料:芹菜籽40-60份、绞股蓝10-20份、菊苣根40-60份、灵芝27-32份、蒲公英27-32份、玉米须27-32份。
2.根据权利要求1所述的一种降尿酸利尿组合物,其特征在于:包括以下重量份的原料:芹菜籽50份、绞股蓝15份、菊苣根50份、灵芝29.5份、蒲公英29.5份、玉米须29.5份。
3.一种降尿酸利尿组合物的制备方法,其特征在于:具体步骤如下:
(1)取原料芹菜籽、绞股蓝,加入乙醇,搅拌混合均匀;
(2)将步骤(1)中的原料煮沸提取2-4次,每次提取时间为1-3h,过滤,得提取液I;
(3)将提取液I使用旋转蒸发仪去除乙醇后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物I备用;
(4)取原料菊苣根、灵芝、蒲公英、玉米须,加入纯化水,搅拌混合均匀;
(5)将步骤(4)中的原料煮沸提取1-3次,每次提取时间为1-3h,过滤,得提取液II;
(6)将提取液II煮沸浓缩后置于烘箱内烘至半干,再用真空干燥箱干燥得干燥物II备用;
(7)将干燥物I和干燥物II混合均匀制成颗粒即可。
4.根据权利要求3所述的一种降尿酸利尿组合物的制备方法,其特征在于:步骤(1)中乙醇的质量分数为75-85%,乙醇的加入量为芹菜籽和绞股蓝总重量的8-12倍。
5.根据权利要求3所述的一种降尿酸利尿组合物的制备方法,其特征在于:步骤(4)中纯化水的加入量为菊苣根、灵芝、蒲公英、玉米须总重量的8-12倍。
