实施例11:(L)-阿拉伯糖同化的合成代谢途径的体内功能
合成途径基因克隆到操纵子中
在如下构建的IPTG诱导型启动子的控制之下,将编码序列SEQ ID NO:1的酮基己糖激酶(Khk)的C同工型的智人基因khk-C和编码序列SEQ ID NO:9的大肠杆菌果糖-1,6-二磷酸醛缩酶B同工型的fbaB克隆到质粒pEXT20(Dykxhoorn et al.,1996)上的操纵子中。人khk-C基因由Asipu博士(Asipu et al.,2003)提供,并用引物P13和P14(表3)扩增。醛缩酶用引物P28和P29从MG1655株的大肠杆菌基因组DNA进行扩增。
通过加入与相邻片段具有17个核苷酸同源的尾部,设计用于扩增这两个基因的引物而提供能够与Clonetech In-Fusion试剂盒一起使用的PCR片段。使用软件RBS计算器(Salis et al.,2011)选择RBS。因此,fbaB基因处于预测而提供最大表达的序列(TTAGGAGGTATACT)的RBS之后,并且khk-C处于预测而最小化表达的序列(ACAGCTTTTAATTATACACTTTAAGGAGGACAGAC)的RBS之后。通过加入与相邻片段具有15个核苷酸同源性的尾,设计出用新RBS扩增两个基因的引物(P30和P22用于khk-C而P31和P32用于fbaB)而提供能够与融合试剂盒一起使用的PCR片段。质粒pEXT20用限制酶BamHI和SalI消化。使用Clonetech In-Fusion试剂盒通过重组扎结两个PCR片段和线性化的pEXT20而提供质粒pEXT20-khk-C-RBSmax-fbaB。质粒pEXT20-khk-C-RBSmax-fbaB转化于MG1655 ΔaraB中。通过测序检查由此获得的载体pEXT20-khk-C-RBSmax-fbaB。类似地,通过In-Fusion使用引物83/84和33/34(表3)构建质粒pEXT20-khk-C和pEXT20-fbaB。将这些质粒转化到大肠杆菌MG1655 ΔaraB菌株中,其中缺失了编码核酮糖-5激酶的序列SEQ ID88的araB。
通过在(L)-阿拉伯糖上监测的ΔaraB菌株生长测试合成途径
在IPTG存在下,在含有60mM(L)-阿拉伯糖作为唯一碳源的M9液体培养基中测试细菌生长。为了控制缺乏阿拉伯糖同化天然途径的菌株在(L)-阿拉伯糖存在下生长于M9培养基中的能力,测试了以下菌株:MG1655ΔaraB和携带质粒pEXT20-khk-C、pEXT20-fbaB或pEXT20-khk-C-RBSmax-fbaB的MG1655 ΔaraB。在LB+100μM IPTG+2%L-阿拉伯糖中进行预培养过夜,并随后将细胞以OD600nm=0.2和随后以OD600nm=1转移到M9培养基+2%葡萄糖+2%阿拉伯糖+100μM IPTG上,将细胞以OD600nm=0.2转移到M9培养基+2%L-阿拉伯糖+100μMIPTG中。仅分别具有(L)-核酮糖-1激酶和(L)-核酮糖-1P醛缩酶活性的同时表达KhkC和FbaB酶的菌株生长于这些条件中(图13)。这些结果证明了(L)-阿拉伯糖同化合成代谢途径的功能。
