Abstract
本发明公开一种抗肿瘤联合用药物及其在制备抗癌药物中的用途,所述联合用药物包括第一制剂和第二制剂,所述抗肿瘤联合用药物按重量份数计为第一制剂10‑40份,第二制剂0.1‑10份。本发明还公开了绿原酸在制备抗癌药物毒副作用抑制剂中的用途,所述抗癌药物为BCR‑ABL小分子抑制剂。本发明所述技术方案可提高BCR‑ABL小分子抑制剂在慢性粒细胞白血病中的靶向性,降低其毒副作用,并逆转BCR‑ABL小分子抑制剂的耐药性。

技术领域
本发明涉生物医药领域,具体为一种抗肿瘤联合用药物及其在制备抗癌药物中的用途。
背景技术
绿原酸是植物在进行有氧呼吸的过程,经磷酸戊糖途径中间产物合成的一种苯丙素类物质。其提取、合成技术成熟,绿原酸已经被开发应用于食品,保健品,化妆品和药品等多个领域。目前研究结果表明,绿原酸有保护心血管、抗氧化、抗紫外以及抗辐射、抗癌、抗菌、抗病毒以及免疫调节等作用。
白血病是一类常见的严重威胁人类健康的造血系统恶性肿瘤。在我国,白血病每年发病率为30-40万,在成人恶性肿瘤中排第七位,在儿童及青少年恶性肿瘤中排首位。目前,化疗和造血干细胞移植是治疗白血病的主要手段,但是,通过化疗只能取得短暂缓解,而异基因造血干细胞移植治疗白血病5年无病生存率仅为35% -50%。此外,因造血干细胞供体限制,只有部分患者能进行这一治疗,而且还存在移植相关死亡和复发危险性。因此,寻找新的更有效抗白血病药物仍然十分迫切和必要。
目前的研究表明,人体9号染色体上的癌基因c2ABL链接到22号染色体上的断点簇集区(BCR),形成p210BCR2ABL,融合基因和p185BCR2ABL融合基因,形成的断裂点以簇集区(BCR)-艾贝尔逊白血病病毒(ABL)融合基因为主要标志, BCR-ABL融合基因表达BCR-ABL蛋白。这两种融合基因使相应的BCR2ABL酪氨酸激酶持续激活,引起细胞增殖,粘附和生存性质的改变。在过去的20多年里,研究人员发现BCR-ABL激酶在细胞信号转导和转化中起着重要作用,它通过磷酸化和活化一些下游基因,促使白血病细胞无限增殖。BCR-ABL在正常细胞中不表达,所以它是治疗白血病理想的药物靶标。
近年来,BCR-ABL的小分子抑制剂得到了很大的发展,包括BCR-ABL蛋白激酶抑制剂伊马替尼和尼洛替尼;ABL-Sre双重抑制剂达沙替尼和博舒替尼;以及最新开发的帕纳替尼。CML患者在使用BCR-ABL小分子抑制剂时,绝大部分病人 BCR-ABL位点突变产生耐药性。此外BCR-ABL抑制剂还有毒副作用大等问题,使其疗效有限。
发明内容
有鉴于此,本发明提供一种抗肿瘤联合用药物及其在制备抗癌药物中的用途,所述联合用药物包括第一制剂和第二制剂。本发明还公开了绿原酸在制备抗癌药物毒副作用抑制剂中的用途,所述抗癌药物为BCR-ABL小分子抑制剂。本发明所述技术方案可提高BCR-ABL小分子抑制剂在慢性粒细胞白血病中的靶向性,降低其毒副作用,并逆转BCR-ABL小分子抑制剂的耐药性。
为解决以上技术问题,本发明技术方案为采用一种抗肿瘤联合用药物,包括绿原酸和药学上可接收的载体形成的第一制剂,及BCR-ABL小分子抑制剂和药学上可接收的载体形成的第二制剂。
优选的,所述第一制剂10-40份,所述第二制剂0.1-10份。
优选的,所述BCR-ABL小分子抑制剂为BCR-ABL络氨酸激酶抑制剂。
优选的,所述BCR-ABL络氨酸激酶抑制剂包括伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼。
更为优选的,根据上述联合用药物,所述制剂的剂型为注射剂或口服制剂。
本发明还提供一种上述联合用药物在制备抗癌药物中的用途。
优选的,所述癌症为慢性粒细胞白血病。
本发明还提供一种绿原酸在制备抗癌药物毒副作用抑制剂中的用途。
优选的,所述抗癌药物为BCR-ABL小分子抑制剂。
目前,大量的实验均证明绿原酸具有抗诱变及抗癌的作用。绿原酸作为G6PT(葡萄糖-6-磷酸转移酶)的抑制剂,可以启动中性粒细胞及前髓细胞HL-60的程序性细胞死亡,抑制人Hep3B肝癌细胞的金属硫蛋白(MMP)分泌,抑制神经胶质瘤的迁移。。绿原酸对BCR-ABL小分子抑制剂的增敏作用可能与上述机理有关。其次绿原酸分子中含有一定量的活性羟基,能形成具有抗氧化作用的氢自由基,可以消除羟基自由基和超氧阴离子等自由基的活性,保护组织免受氧化作用的损伤。此外,绿原酸对上皮内淋巴细胞(IEL)上清液以及肠道固有层淋巴细胞(LPL)上清液中NF-γ和TNF-α水平影响均较大。体外研究显示绿原酸能诱导人淋巴细胞及人外周血白细胞生成IFN- γ和IFN-α,这些作用也可能与绿原酸在本申请中能降低BCR-ABL小分子抑制剂的毒副作用有关。
BCR-ABL耐药机制目前的研究表明主要分为与BCR-ABL相关与不相关两面,与BCR-ABL相关的耐药机制为:1,BCR-ABL基因扩增和过度表达在伊马替尼体外诱导耐药的CML细胞中18%,可以检测到BCR-ABL融合基因扩增和融合蛋白过表达。伊马替尼耐药倍数高的IM-R2细胞比对其耐药倍数低的IM-R1细胞有更高BCR-ABL 基因扩增和蛋白表达水平,并且与敏感细胞系比较,两者都有不同程度的BCR-ABL 蛋白高表达。2,BCR-ABL点突变,BCR-ABL点突变是伊马替尼耐药的主要机制,点突变加快CML疾病进程,降低患者生存率,导致疾病复发。
与BCR-ABL不相关的耐药机制为:1,膜转运蛋白介导的耐药机制,肿瘤多药耐药(MDR)与肿瘤细胞内化疗药物蓄积浓度的高低有关。说明膜表面转运蛋白的改变可能是耐药产生的原因。腺苷三磷酸结合转运蛋白(ABC转运蛋白)超家族是一组跨膜蛋白。目前研究最广泛的是,ABCB1(MDR-1/P-gp),ABCC1(多药耐药蛋白MRP1) 和ABCG2(乳腺癌耐药蛋白,BCRP),它们通过将药物排出细胞外使肿瘤细胞对化疗药物产生耐药。2,SFKs信号转导通路的激活,SFKs家族包括Src、Blk、Fgr、Fyn、 Hck、Lck、Lyn和Yes等成员,具有调节细胞增殖、分化和存活的作用,其表达具有组织特异性。研究表明CML的发病机制与SFKs有关,BCR-ABL激酶能够活化Lyn、Hck 和Fgr,从而活化信号传导子和转录激活子5(STAT5),上调细胞周期蛋白D1,促进细胞分裂,加速bcr-2转录。3,凋亡基因介导的耐药机制,c-FLIP((细胞型Fas相关死亡域样白介素-1β转换酶抑制蛋白)下调和DR5(DR5(肿瘤坏死因子相关诱导凋亡配体受体-5)上调可能会引发CML对伊马替尼耐药。4,克隆演变,克隆演变是遗传不稳定的体现,与CML疾病进程密切相关。克隆演变的患者在接受伊马替尼治疗后BCR-ABL 突变的概率增大,从而引发耐药。绿原酸逆转BCR-ABL小分子抑制剂的耐药性可能与对抗上述产生耐药的机制相关,具体的机理有待进一步的研究。
具体实施方式
为了使本领域的技术人员更好地理解发明的技术方案,下面结合具体实施方式对本发明作进一步的详细说明。
慢性粒细胞白血病(CML)以恶性髓系祖细胞克隆性增殖为特征,发病率占所有白血病的15%~20%,是人们发现的第一个与基因异常相关的血液系统恶性肿瘤。在受累细胞系中,可检测到Ph染色体和BCR-ABL融合基因。CML的病程可分为3期:CP(慢性期),AP(加速期),BP/BC(最终急性变期)。目前治疗CML主要着重于慢性期的治疗,并力争分子水平的缓解和治愈。
慢性粒细胞白血病发病率逐年增长,BCR-ABL小分子抑制剂的需求将会越来越大。从第1个治疗慢性粒细胞白血病的靶向药物伊马替尼上市以来,肿瘤的药物治疗迎来了靶向治疗的新时代。然而伴随着长期使用,部分患者出现了因BCR-ABL点突变而产生的耐药性。为了克服这一问题,第2代BCR-ABL抑制剂应运而生,包括尼洛替尼和达沙替尼,它们对于BCR-ABL的野生型及绝大多数突变型都有着超越伊马替尼的抑制作用,然而对于T315I突变型依然束手无策。以帕纳替尼为代表的第3代 BCR-ABL激酶抑制剂已经可以抑制T315I突变型,但其选择性却不尽如人意,在对各类突变型BCR-ABL络氨酸激酶激酶抑制效果提升的同时,第3代抑制剂作用的靶点也相对较多,从而可能导致了更多的不良反应。
BCR-ABL小分子抑制剂的毒副作用主要包括骨髓移植,表现为于治疗的12个月中出现各级中性粒细胞减少,以及血小板减少与贫血的发生;胃肠道副作用,轻到中度的恶心、呕吐、腹泻、便秘及腹部疼痛;肝功及胰腺损害,治疗的头几个月时,通常都会出现AST及ALT升高;液体潴留,通常表现为体重增加,眶周水肿或下肢水肿;还有肺毒性,心血管毒性等。
随着酪氨酸激酶抑制剂在临床上的使用,上述耐药问题逐渐严重,复发率逐渐增高,加之其毒副作用的问题,严重影响了CML的治疗效果及临床应用。为了解决该项问题,结合绿原酸在人体多个系统表现出的有益效果,本申请创造性的将二者结合起来,产生了意料不到的技术效果。
本发明公开一种一种抗肿瘤联合用药物及其在制备抗癌药物中的用途,所述联合用药物包括第一制剂和第二制剂,所述抗肿瘤联合用药物按重量份数计为第一制剂10-40份,优选10-30份,更为优选10-20份,第二制剂0.1-10份,优选1-8份,更为优选3-5份。本发明还公开了绿原酸在制备抗癌药物毒副作用抑制剂中的用途,所述抗癌药物为BCR-ABL小分子抑制剂。
本发明实验中,在MTT实验部分表明绿原酸联合伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼,可降低该系列药物对人正常造血干细胞HSPC-1的毒性,一定程度上提高该类药物的靶向性。在对慢性粒细胞白血病细胞株K56抑制作用试验中,绿原酸与伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼联用,在一定浓度范围内,达到了协同增效作用。
体外细胞实验表明,无细胞增殖抑制的绿原酸剂量能够逆转达沙替尼耐药细胞株的耐药性,体外动物实验进一步证实了绿原酸能够逆转厄洛替尼耐药细胞株的耐药性。
同时,体外动物实验还表明绿原酸与伊马替尼,尼洛替尼,达沙替尼,博舒替尼及帕纳替尼联用在小鼠身上有较好的抑瘤率,并且同时降低了此类药物的血液系统毒性,提高了小鼠的机能状态。
综上所述,本发明所述技术方案可有效提高BCR-ABL小分子抑制剂在慢性粒细胞白血病中的靶向性,降低其毒副作用,并可逆转BCR-ABL小分子抑制剂的耐药性。
以上是对发明内容的详细分析,下面为该发明的实施例。
实施例1 MTT测定联合用药的增殖抑制
1.1试验材料
受试药品:绿原酸、伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼。
细胞株:慢性粒细胞白血病细胞株K562、人正常造血干细胞HSPC-1
1.2试验方法
无菌条件下按100μl细胞接种于96孔培养板,接种细胞数均为5×l03/孔。细胞于37℃、5%CO2孵箱中分别孵育,细胞分为:空白对照组,阴性对照组、单独用药组、绿原酸组以及联合用药组的,空白对照组不接种细胞。每个孔培养液和药物总量为 200μl,每个浓度设3个复孔,并以等体积培养液代替药物作为空白对照。继续置5%CO2饱和湿度37℃温箱中继续培养。48h后,每孔加入浓度为5mg/ml MTT 20μl继续培养4h 后,小心吸弃孔内液体,加150μl二甲基亚枫溶解沉淀,充分振荡10min,溶解结晶物。选择570nm波长,在酶联免疫检测仪上测定各孔光吸收值,记录结果,计算抑制率及联合指数。以上实验至少重复3次。
1.3数据处理
(1)抑制率=(1-OD570(实验组-空白组)/OD570(对照组-空白组))×100%。
(2)联合用药以公式Q=E(a+b)/(Ea+Eb-Ea×Eb)计算有无协同作用。其中E(a+b)为两药合用的抑制率,即实测合并效应,Ea和Eb为两药单用时的抑制率,分母(Ea+Eb-Ea×Eb)为期望合并效应,Q为两者比值。Q值在0.85~1.15时,两药合并效应为相加(+),Q值在1.15~20时为协同(++),Q值>20为明显协同(+++),Q值在0.05~ 0.85时为拮抗,Q值<0.05为明显。

上述实验表明,绿原酸单独应用于人正常造血干细胞HSPC-1的IC50值为1367.3ug/ml,证明绿原酸本身对人正常造血干细胞无毒。绿原酸分别与伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼联用后,IC50值均高于伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼单独应用于人正常造血干细胞HSPC-1的IC50值,表明绿原酸联合伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼,可降低该系列药物对人正常造血干细胞HSPC-1的毒性,一定程度上提高该类药物的靶向性。
1.4.2单独用药及联合用药对肿瘤细胞株的影响

结果显示,在BCR-Abl小分子抑制药物与绿原酸联合用药对慢性粒细胞白血病细胞株K56抑制作用试验中,1umol伊马替尼与10-50μg/ml绿原酸联合用药的Q 值在1.15~20,两药合并效应为协同(++);12umol尼洛替尼与10-50μg/ml绿原酸联合用药的Q值在1.15~20,两药合并效应为协同(++);0.2umol达沙替尼与10-50 μg/ml绿原酸联合用药的Q值在1.15~20,两药合并效应为协同(++);5umol博舒替尼与10-50μg/ml绿原酸联合用药的Q值在1.15~20,两药合并效应为协同(++); 3umol帕纳替尼与10-50μg/ml绿原酸联合用药的Q值在1.15~20,两药合并效应为协同(++)。
实施例2绿原酸体外逆转人白血病细胞对达沙替尼的耐药性
2.1材料
受试药品:绿原酸,达沙替尼
细胞株:慢性粒细胞白血病细胞株K562为实验室常规培养细胞,临用前传代,取生长状态良好,处于对数期生长的细胞备用。
2.2试验方法
2.2.1耐药细胞株的培育
K562细胞株分别暴露于达沙替尼100nmol/L长达3个月,离心,培养出达沙替尼耐药的K562/NY细胞,每天观察细胞,3天左右1传3传代,保证细胞活力。
2.2.2检测达沙替尼对细胞株与耐药株的IC50,计算耐药倍数
取对数生长期K562细胞及耐药细胞株,调整细胞浓度为8×103个/孔接种于96孔板。实验分别分成3组:空白组、对照组及实验组。空白组只加入培养基,无需接种细胞;对照组加培养基并接种细胞;实验组在上述基础上再加入不同浓度的达沙替尼工作液,置于培养箱孵育48h后每孔加入5mg/ml MTT20μl,继续孵育4h,吸去上液,每孔加入DMSO150μl,静置30min,待结晶完全溶解。用酶标仪检测570nm处各孔吸光值OD值,计算肿瘤细胞生长抑制率。抑制率=(1-OD570(实验组-空白组)/ OD570(对照组-空白组))×100%。计算IC50。耐药倍数=耐药细胞IC50值/敏感细胞 IC50值。
2.2.3 MTT法测定无细胞毒的绿原酸浓度
K562细胞及耐药细胞培养及处理方法同上。实验分别分成3组:空白组、对照组及实验组。空白组只加入培养基,无需接种细胞;对照组加培养基并接种细胞;实验组在上述基础上再加入不同浓度的绿原酸工作液,使其最终浓度分别为1、2、4、8、 16、32、64、128μg/mL,置于培养箱孵育48h后每孔加入5mg/ml MTT20μl,继续孵育4h,吸去上液,每孔加入DMSO150μl,静置30min,待结晶完全溶解。用酶标仪检测570nm处各孔吸光值OD值,计算肿瘤细胞生长抑制率。抑制率= (1-OD570(实验组-空白组)/OD570(对照组-空白组))×100%,取抑制率10%以下浓度的绿原酸浓度作为无毒剂量的逆转浓度。
2.2.4绿原酸逆转耐药性K562/NY细胞株的作用
细胞培养以及实验方法同上。实验分组如下:K562/NY组、K562/NY+绿原酸 4ug/ml组、K562/NY+绿原酸8ug/ml组,实验组分别加入不同浓度的达沙替尼,每个浓度3复孔,测定各孔的OD值,观察无细胞毒性绿原酸(4μg/ml、8μg/ml) 联合达沙替尼作用于K562/NY与达沙替尼单用于对K562/NY组比较细胞杀伤作用的变化,计算达沙替尼对K562/NY的IC50以及用4ug/ml,8ug/ml逆转K562/NY后的IC50。逆转倍数=逆转前IC50值/逆转后IC50值。
2.3试验结果
2.3.1耐药倍数
达沙替尼对K562及K562/NY的IC50的IC50分别为0.12μmol/L和0.46μmol/L。按公式计算出耐药倍数为3.8。
2.3.2 MTT法测定对耐药细胞无细胞毒的绿原酸浓度结果
绿原酸对耐药细胞K562/NY均有抑制增殖作用,IC50为80.2ug/ml。当绿原酸浓度低于8μg/ml时,对K562/NY耐药细胞的抑制率均<10%,无显著细胞毒性。
2.3.3绿原酸对白血病耐药细胞的逆转作用
绿原酸无细胞毒浓度(4μg/ml、8μg/ml)作用于人白血病细胞耐药细胞K562/NY 之后,达沙替尼对耐药细胞株K562/NY的IC50降低。4μg/ml绿原酸作为逆转剂时,达沙替尼对K562/NY的IC50为0.3uM,逆转倍数为1.53,以8μg/ml绿原酸作为逆转剂时,达沙替尼对K562/NY的IC50为0.2uM,逆转倍数为2.3。

2.4小结
无细胞增殖抑制的绿原酸剂量能够逆转达沙替尼耐药细胞株的耐药性,其逆转倍数与剂量有一定相关性。
实施例3绿原酸与BCR-ABL小分子抑制剂的药物联合体内抑瘤的增效减毒作用
3.1材料与仪器
受试药品:绿原酸;伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼
受试细胞株:慢性粒细胞白血病细胞株K562
受试动物:BABL/c-nu小鼠,18-22g,雌
3.2试验方法
3.2.1实验动物肿瘤模型的建立
收集对数生长期细胞,1000rpm离心5min,细胞沉用PBS洗涤2次,计数后用无血清培养液调整细胞浓度为1x107/m1,无菌条件下送到动物室。在无菌实验条件下,小鼠右侧腋窝皮下注射1×107/m1的细胞,每只0.1ml,注射局部出现明显皮丘,1周后裸鼠左侧腋窝处出现米粒大结节,说明移植模型成功建立,此时按照随机分组法分成12组。
待肿瘤平均直径达100mm3后开始治疗,每天腹腔注射一次,生理盐水对照组给0.2ml无菌生理盐水。
3.2.3抗肿瘤作用评价
观察给药后每天测动物体重;待阴性组瘤体积约为0.5cm3时停止实验,眼球摘除取血测定WBC、HBC及HGB含量,脱颈椎处死小鼠并称重,剥取肿瘤并称重。
抑瘤率=(对照组瘤重-实验组瘤重)/对照组瘤重×00%。
两药合并Q=E(a+b)/(Ea+Eb-Ea×Eb),其中E(a+b)为两药合用的抑制率,即实测合并效应,Ea和Eb为两药单用时的抑制率,分母(Ea+Eb-Ea×Eb)为期望合并效应,Q为两者比值。Q值在0.85~1.15时,两药合并效应为相加(+),Q值在1.15~ 20时为协同(++),Q值>20为明显协同(+++),Q值在0.05~0.85时为拮抗,Q值<0.05为明显拮抗。
3.2.4统计学分析
实验数据用SPSS 17.0统计软件进行处理,所有数据均以均数士标准差(mean±SD)表示。组间比较采用单因素方差分析进行统计学处理,P<0.05认为有差异, P<0.01认为有显著性差异。
3.3试验结果
3.3.1联合用药对抑瘤率的影响
与模型组比较*p<0.05,**p<0.01;与阳性组比较△p<0.05,△△p<0.01
结果显示,绿原酸,伊马替尼,尼洛替尼,达沙替尼,博舒替尼及帕纳替尼使用较低剂量,对K562移植瘤有较弱的抑制作用,与模型空白组比较无显著性差异。但绿原酸与伊马替尼、绿原酸与尼洛替尼等联用有较好的抑瘤率,与单用相比有显著性差异。联合用药Q值计算结果表明,绿原酸与BCR-Abl小分子抑制联用有协同效应。
3.3.2联合用药对小鼠体重的影响
与阴性组比较*p<0.05,**p<0.01;与阳性组比较△p<0.05,△△p<0.01
结果显示,第15天绿原酸联合用药组的体重大于BCR-ABL靶向小分子抑制剂单独用药组,结果有差异,表明绿原酸联合用药组可提高小鼠体重,改善小鼠机能状态。
2.3.3联合用药的对小鼠血常规的影响
与阴性组比较*p<0.05,**p<0.01;与阳性组比较△p<0.05,△△p<0.01
结果显示绿原酸(20mg/kg)能够显著改善K562慢性粒细胞白血病小鼠使用伊马替尼、尼洛替尼、达沙替尼、博舒替尼、帕纳替尼后血常规中WBC、RBC、HGB的降低现象,说明绿原酸缓解BCR-ABL小分子抑制剂产生的骨髓抑制作用,血小板记数的增加可降低出血性不良反应的发生,同时血红蛋白增加可预防贫血及贫血相关性不良反应的发生,表明绿原酸从诸多方面降低了BCR-ABL小分子抑制剂的毒副作用。
实施例4绿原酸在制备抗多药耐药性的动物试验
4.1材料与方法
试药品:绿原酸,尼洛替尼
受试细胞株:慢性粒细胞白血病细胞株K562,系采用尼洛替尼浓度梯度递增法对K562细胞系诱导,并经克隆筛选建成,实验前脱药培养。
受试动物:BABL/C-nu小鼠,♀,体重18~22g;
4.2试验方法
4.2.1实验动物肿瘤模型的建立
将脱药后的耐药性细胞株,用培养液调整细胞浓度为1×107/m1,于小鼠右侧腋窝皮下注射1×107/m1的细胞,每只0.1ml。
4.2.2给药方法
待肿瘤平均直径达100mm3后开始分组,分别为尼洛替尼持续用药组、绿原酸+ 尼洛替尼组、绿原酸组、阴性组。
尼洛替尼持续用药组:灌胃给药,一天一次,10mg/kg,连续给药15天。
绿原酸+尼洛替尼组:先腹腔注射给绿原酸,一天一次,20mg/kg,连续给药5 天;停止绿原酸给药后,次日开始灌胃给尼洛替尼,一天一次,10mg/kg,连续给药 10天。
绿原酸组:腹腔注射给药,一天一次,20mg/kg,连续给药5天,后灌胃给生理盐水,一天一次,连续给药10天。
阴性组:腹腔注射给生理盐水,一天一次,连续给药5天,后灌胃给生理盐水,一天一次,连续给药10天。
4.2.3抗肿瘤作用评价
在给药结束后停止实验,脱颈椎处死小鼠并称重,剥取肿瘤并称重,计算抑瘤率。
4.2.4数据处理
抑瘤率%=[1-(给药组平均瘤重/阴性组平均瘤重)]×100%;
4.3试验结果
4.3.1各试验组对耐药性移植瘤抑瘤率的影响
与阴性组比较*p<0.05,**p<0.01;与尼洛替尼比较△p<0.05,△△p<0.01
结果显示,绿原酸组及尼洛替尼组对耐药性慢性粒细胞白血病K562小鼠移植瘤的抑瘤率较小,无明显抑瘤效果,而在绿原酸+尼洛替尼组对耐药性慢性粒细胞白血病K562小鼠移植瘤的抑瘤率显著,说明了绿原酸能够有效解决尼洛替尼引起的慢性粒细胞白血病K562耐药性。
以上仅是本发明的优选实施方式,应当指出的是,上述优选实施方式不应视为对本发明的限制,本发明的保护范围应当以权利要求所限定的范围为准。对于本技术领域的普通技术人员来说,在不脱离本发明的精神和范围内,还可以做出若干改进和润饰,这些改进和润饰也应视为本发明的保护范围。
Claims (4)
1.一种抗肿瘤联合用药物,其特征在于,包括绿原酸和药学上可接受的载体形成的第一制剂,及BCR-ABL小分子抑制剂和药学上可接受的载体形成的第二制剂,所述BCR-ABL小分子抑制剂为伊马替尼、尼洛替尼、博舒替尼、帕纳替尼,其中,伊马替尼与绿原酸质量比为1:20,尼洛替尼与绿原酸质量比为1:2,博舒替尼与绿原酸质量比为1:4,帕纳替尼与绿原酸质量比为3:20;其中以1mg/kg伊马替尼、10mg/kg尼洛替尼、5mg/kg博舒替尼或3mg/kg帕纳替尼分别与20mg/kg绿原酸的使用剂量联合用药。
2.根据权利要求1 所述的抗肿瘤联合用药物,其特征在于,所述药物的剂型为注射剂或口服制剂。
3.一种权利要求1 所述的联合用药物在制备抗癌药物中的用途,所述癌症为慢性粒细胞白血病。
4.一种绿原酸在制备抗癌药物BCR-ABL 小分子抑制剂毒副作用抑制剂中的用途,其特征在于,BCR-ABL 小分子抑制剂为伊马替尼、尼洛替尼、博舒替尼、帕纳替尼,其中,伊马替尼与绿原酸质量比为1:20,尼洛替尼与绿原酸质量比为1:2,博舒替尼与绿原酸质量比为1:4,帕纳替尼与绿原酸质量比为3:20;其中以1mg/kg伊马替尼、10mg/kg尼洛替尼、5mg/kg博舒替尼或3mg/kg帕纳替尼分别与20mg/kg绿原酸的使用剂量联合用药。
