骨质疏松是以骨量减少、骨组织微观结构退化为特征,致使骨骼脆性增加及骨折危险性增加的一种代谢性疾病,常见于老年人,目前我国有骨质疏松患者 (包括骨量减少)8400万,占总人口的6.6%,到2050年将增加1倍以上,达21千万人,占总人口的13.2%。随着中国人口老龄化日趋明显,骨质疏松症已成为一个较大的社会问题,严重影响高龄人群的健康和寿命。骨质疏松症常用的治疗药物主要有雌激素类药物,其副作用较明显,和乳腺癌、子宫内膜癌的发病存在着联系,并能够引发心肌梗塞、中风、深静脉血栓形成、肺栓塞的发病。另一类主要来源于植物的异黄酮属于植物雌激素,有雌激素样效应而无明显雌激素样副作用,被认为是雌激素的天然替代品。因此来源天然且富含异黄酮的食物或药物由于其具有增加骨密度作用,无毒副作用等,被广泛用于骨质疏松的预防与治疗,而且随着人口高龄化的进展,人们对具有增加骨密度功能的天然产品需求将不断增长。
鹰嘴豆( Chickpea)属豆科野豌豆族鹰嘴豆属(Cicer arietinum L ) ,一年生或越年生草本植物,鹰嘴豆在《维吾尔族药志》、《维吾尔民族医常用药材》、《药物宝库》、《草本遗》、《医典》等传统典籍中均有记载。中医认为,鹰嘴豆味甘、性平、无毒,具有补中益气、温肾壮阳功效,现代研究表明,鹰嘴豆主要活性成分为异黄酮,包括鹰嘴豆芽素A、芒柄花素等异黄酮类成分,具有一定的雌激素样作用,但目前尚未见有关于鹰嘴豆增加骨密度的报道。另鹰嘴豆还含有大量蛋白质、优质氨基酸和矿物质,也为治疗和预防骨质疏松提供必要的营养支持。
目前公开的鹰嘴豆食品药品组合主要有:专利《一种包含鹰嘴豆、山药的食品及其制作方法》(申请号:201310730140.X)公开的鹰嘴豆、山药和VC以及调味品等制成果蔬泥产品,专利《一种具有抗疲劳功效的鹰嘴豆保健营养粉》(申请号:201310420729.X )公开了鹰嘴豆、山药、红景天、人参叶、龙眼药、酸枣仁、芡实、枸杞和薄荷等中药制成的具有抗疲劳功效的营养粉,专利《一种助消化鹰嘴豆奶茶粉及其制备方法》(申请号:201410291927.5)公开了由鹰嘴豆、大腹皮、淡竹叶、黄花蒿、马鞭草、苦瓜根、野艾蒿、葫芦茶、羊栖菜、富国菜、云雾果、枇杷汁、燕麦片和杏仁粉等制成的营养粉,专利《一种具有调节血脂作用的鹰嘴豆保健营养粉》(申请号:201310417872.3)鹰嘴豆和银耳、荷叶、水蛭、人参、陈皮、干姜、百合等制成具有其降血脂效果的营养粉,鹰嘴豆和茯苓、芡实、山药、党参、枸杞、龙眼肉、百合、酸枣仁、红枣等制成的有改善营养性贫血作用营养粉。上述已有的鹰嘴豆与中药组方,组方大,药味多,药物作用成分复杂,未见增加骨密度作用的配方。因此,充分利用鹰嘴豆异黄酮具有雌激素样作用,同时配以食品营养素钙剂和维生素D3,可以从补充雌激素样品成分、促进钙吸收及补充钙等途径发挥增加骨密度作用,起到预防骨质疏松的作用。
钙剂治疗骨质疏松已经有多年的历史,无机钙剂因碱性过强且能中和胃酸而导致消化不良、便秘或腹泻等问题,柠檬酸钙是一种源自柠檬酸的钙盐,作为食品钙强化剂,吸收效果比无机钙好,同时柠檬酸钙能促进脂肪分解,保持身体酸碱平衡。
肠道Ca2+吸收不良是骨质疏松的重要发病原因之一,维生素 D3可以促进肠道对Ca2+的吸收,而并可促进肾脏对钙的重吸收,研究认为钙或维生素D单独服用在预防原发性老年性骨质疏松骨折的效果往往不理想,因此足够的钙和维生素D对于保持对骨质疏松症最佳治疗效果是非常必要的。
发明内容
本发明的首要目的在于提供一种鹰嘴豆组合物,该化合物具有增加骨密度作用。
本发明的另一目的在于提供上述鹰嘴豆组合物在制备增加骨密度食品、保健品或药品中的应用。
本发明通过以下技术手段实现:
一种具有增加骨密度作用的鹰嘴豆组合物。
该组合物由如下重量份的原料制成:鹰嘴豆970-980重量份,柠檬酸钙20-30重量份,维生素D3 10-20×10-6重量份。
优选地,所述组合物由如下重量份的原料制成:鹰嘴豆974重量份,柠檬酸钙26重量份,维生素D3 15×10-6重量份。
鹰嘴豆主要活性成分之一为异黄酮,具有一定的雌激素样作用,而无明显雌激素样副作用,可诱导破骨细胞的凋亡,抑制骨吸收并刺激成骨细胞形成。钙代谢异常是骨质疏松症的重要病理生理改变,钙剂长期广泛用于骨质疏松症治疗,但大量研究提示单纯补钙效果不明显,需同时辅以补充维生素D,促进肠道对Ca2+的吸收及肾对Ca2+的重吸收。
因此,本组合物通过将鹰嘴豆、柠檬酸钙和维生素D3混合,从诱导破骨细胞调亡、刺激成骨细胞形成、促进钙吸收及直接补充钙营养的多重途径,协同发挥增加骨密度作用。
本发明上述具有增加骨密度作用的鹰嘴豆组合物的制备方法,可以采用如下方法:
(1)粉碎过筛:取鹰嘴豆粉碎过80 -300目筛,将柠檬酸钙和维生素D3过80-120目筛,备用。
(2)混合:将鹰嘴豆粉、柠檬酸钙和维生素D3混合均匀,即得。
本发明提供上述组合物在制备用于增加骨密度的食品、保健品或药品的应用。
优选地,所述食品、保健品为固体饮料、饼干或糕点,所述药品为胶囊或片剂。
优选地, 所述食品、 保健品或药品还包括食品、保健品或药品可接受的辅助药物或辅料。
本发明具有以下优点:
1)本组合物通过将鹰嘴豆、柠檬酸钙和维生素D3混合,具雌激素样作用刺激成骨细胞形成和诱导破骨细胞调亡,促进钙吸收及直接补充钙营养的多重途径,协同发挥增加骨密度作用,组份取材均为普通食品及有机钙剂,组方简单,安全性高。
2)本发明可将该组合物制成固体饮料、饼干和糕点等食品,或者加入辅助药物和药学上可接受的载体,制成制剂,适用于骨质疏松症人群,口感适宜,方便携带和食用。

下面通过具体实施方式来进一步说明本发明,以下实施例为本发明具体的实施方式,但本发明的实施方式并不受下述实施例的限制,其他的任何未背离本发明的精神实质与原理下所作的改变、修饰、替代、组合、简化,均应为等效的置换方式,都包含在本发明的保护范围之内。
实施例1
一种具有增加骨密度作用的鹰嘴豆组合物,该组合物由如下重量份的原料制成:鹰嘴豆974重量份,柠檬酸钙26重量份,维生素D3 15×10-6重量份。
粉碎过筛:取鹰嘴豆粉碎过200目筛,将柠檬酸钙和维生素D3过120目筛,备用。
混合:将鹰嘴豆粉、柠檬酸钙和维生素D3混合均匀,即得。
实施例2 增加骨密度作用的药理实验
采用实施例1的组合物为受试样品。
试验对象及饲养管理:SD雌性大鼠,250-290g,饲养温度与湿度:20-25 ℃,40-70%,采用12 h:12 h昼夜间断照明;自由进食饮水。
分组与剂量设置:手术造模后适应性饲养一周,将大鼠按照体重随机分为模型组,组合物高、中、低剂量组,每组各10只。另取10只正常大鼠作为空白对照组。另外分别设定一个中剂量组(0.4 g/kg)和一个低剂量组(0.2 g/kg),连续灌胃给药3个月。
数据分析:本实验所有数据均采用均值±标准差()表示,组间差异性分析采用SPSS 17.0软件进行单因素方差分析,以P<0.05作为统计学差异参考标准。
实验方法:除空白组外,其它组雌性大鼠麻醉,切除卵巢,待伤口愈合(7天后)开始分组给药。各组动物给药3个月后,麻醉,取血检测血钙。解剖,分离股骨进行钙密度扫描。取大鼠股骨,一般灰化后用原子吸收的方法进行骨钙含量检测。取血清检测血钙、碱性磷酸酶等指标含量。
血清相关指标结果:由表1可知,给予组合物3个月后,该两项指标虽有上升的趋势,其中血钙浓度上升4.3%,AKP活力最高上升13.72%。

骨密度及骨钙含量
由表2可见,与空白组比较,模型组股骨骨密度、股骨干重及骨钙含量均明显下降(P<0.05),给予组合物3个月后,各组骨密度、股骨干重及骨钙含量均有增加的趋势,但只有高、中剂量组的股骨干重差异具有统计学意义(P<0.05)。

通过本实施例研究发现,组合物增加加骨密度组合物对机体血钙水平、碱性磷酸酶等指标具有一定的增加作用,对骨密度和骨钙含量等均有一定的改善作用、其中对增加股骨干重有显著性作用,长期食用有利于增加骨密度。
实施例3增加骨密度组合物饼干制备
一种具有增加骨密度作用的鹰嘴豆组合物饼干,该组合物由如下重量份的原料制成:
该组合物由如下重量份的原料制成:鹰嘴豆970重量份,柠檬酸钙30重量份,维生素D310×10-6重量份。
其制备工艺包括如下步骤:
粉碎过筛:取鹰嘴豆粉碎过300目筛,将柠檬酸钙和维生素D3过100目筛,备用。
混合:将鹰嘴豆粉、柠檬酸钙和维生素D3混合均匀,即得鹰嘴豆增加骨密度组合物混合粉。
制饼:取鹰嘴豆增加骨密度组合物混合粉300g、面粉153.8g、玉米淀粉23.5g、鸡蛋40g、麦芽糖醇200g、食用油200g、碳酸氢铵6.72g、泡打粉3.36g、小苏打1.34g、食盐0.84g、钙粉0.34g、甜蜜素0.10g,混合均匀,加水制成团,压成圆形生饼,以面温140℃,底温180℃进行烘烤,时间8min,得熟饼,包装即得增加鹰嘴豆增强骨密度组合物饼干。
实施例4 增加骨密度组合物胶囊制备
一种具有增加骨密度作用的鹰嘴豆组合物饼干,该组合物由如下重量份的原料制成:
该组合物由如下重量份的原料制成:鹰嘴豆980重量份,柠檬酸钙20重量份,维生素D320×10-6重量份。
其制备工艺包括如下步骤:
粉碎过筛:取鹰嘴豆粉碎过80目筛,将柠檬酸钙和维生素D3过80目筛,备用。
混合:将鹰嘴豆粉、柠檬酸钙和维生素D3混合均匀,即得组合物混合粉。
取上述混合粉,加入硬脂酸镁混匀, 填充胶囊,即得。
Claims (6)
1.一种具有增加骨密度作用的鹰嘴豆组合物,其特征在于,该组合物由如下重量份的原料制成:鹰嘴豆970-980重量份,柠檬酸钙20-30重量份,维生素D3 10-20×10-6重量份。
2.根据权利要求1所述的具有增加骨密度作用的鹰嘴豆组合物,其特征在于,所述组合物由如下重量份的原料制成:鹰嘴豆974重量份,柠檬酸钙26重量份,维生素D3 15×10-6重量份。
3.根据权利要求1或2所述的具有增加骨密度作用的鹰嘴豆组合物的制备方法,其特征在于,包括如下方法:
(1)粉碎过筛:取鹰嘴豆粉碎过80 -300目筛,将柠檬酸钙和维生素D3过80-120目筛,备用;
(2)混合:将鹰嘴豆粉、柠檬酸钙和维生素D3混合均匀,即得。
4.根据权利要求1或2所述的具有增加骨密度作用的鹰嘴豆组合物在制备具有增加骨密度的食品、保健品或药品中的应用。
5.根据权利要求4 所述的具有增加骨密度作用的鹰嘴豆组合物在制备增加骨密度的食品、保健品或药品中的应用,其特征在于,所述食品、保健品为固体饮料、饼干或糕点,所述药品为胶囊或片剂。
6.根据权利要求5所述的具有增加骨密度作用的鹰嘴豆组合物在制备增加骨密度的食品、保健品或药品中的应用,其特征在于,所述食品、保健品或药品还包括食品、保健品或药品可接受的其它辅助药物或辅料。
