Abstract

本发明涉及一种黑果枸杞中多糖、色素、蛋白质、籽油和籽饼的联合提取方法。主体工艺包括研磨、水提、醇沉、水溶、浓缩等步骤。将黑果枸杞挑选后研磨,先后经50℃水提(pH3)和90℃水提,合并两次提取液,离心,从渣中分离出黑果枸杞籽,经烘干、压榨获得黑果枸杞籽油和黑果枸杞籽饼产品;将上清液醇沉后过滤得清液和沉淀物,将清液浓缩后冷冻干燥得黑果枸杞色素产品;将沉淀加水溶解,不溶物即为蛋白质,将水溶液再次醇沉、干燥即得黑果枸杞多糖产品。本发明工艺简便,可同时得到黑果枸杞中的色素、多糖、蛋白质、籽油和籽饼产品,用于抗氧化、预防心血管疾病等保健品的原料,从而高值化综合利用黑果枸杞。

黑果枸杞中多种有效成分的联合提取方法
技术领域:
本发明涉及一种黑果枸杞高值化综合利用的方法,将黑果枸杞中色素、枸杞多糖、蛋白质和枸杞籽联合提取。
背景技术:
黑果枸杞中含有丰富的原花青素、多糖、蛋白质等各种营养成分。其中,原花青素是最有效的天然自由基清除剂,其功效是维生素C的20倍、VE的50倍;枸杞多糖具有抗疲劳、降血糖、增强免疫力等生理功能,黑果枸杞蛋白质氨基酸组成符合WHO/FAO的参考蛋白模式,属于优质蛋白。其药用保健价值远远高于普通红枸杞,被誉为“软黄金”。
目前,有多种方法用于提取黑果枸杞中的色素或多糖,如乌鲁木齐益广康源生物科技有限公司申报的“黑果枸杞果实提取花青素及其制备方法”专利(申请号:201510128567.1申请公布号:CN 104789612 A),青海康元药用资源科技利用有限公司申报的“一种黑果枸杞固液隔离提取多糖的方法”专利(申请号:201610100244.6申请公布号:CN105542029 A)。现有方法只能对其中的一种物质进行单独的提取,而且在提取工艺过程对其它成分有一定程度破坏,从而造成了黑果枸杞的浪费。
本发明对黑果枸杞中多种有效成分进行联合提取,产品可用于制备高附加值的食品和保健品,可大大提高黑果枸杞的资源利用率。
发明内容:
本发明所要解决的技术问题是提供一种联合提取黑果枸杞中的色素、多糖、蛋白质和枸杞籽的方法。本发明解决上述技术问题所采用的技术方案为:将黑果枸杞干果充分研磨后按1∶50的固液比于50℃ pH3的盐酸溶液浸提150分钟。离心后收集沉淀按1∶50的固液比于90℃水浸提30分钟。再次离心使固液分离,从沉淀中收集得黑果枸杞籽,经烘干、压榨可获得黑果枸杞籽油和黑果枸杞籽饼;合并两次提取液,真空浓缩后加入95%乙醇,使乙醇浓度达到80~85%,4℃下静置过夜后过滤得上清液和沉淀,将上清液进行真空浓缩回收乙醇后冷冻干燥得黑果枸杞色素产品;将沉淀用水溶解,过滤收集不溶物、干燥即得黑果枸杞蛋白质,水溶液进行二次醇沉(乙醇浓度达到80~85%)、过滤、干燥即得黑果枸杞多糖产品。
与现有技术相比,本发明的优点在于:工艺简便、得率高,实现了黑果枸杞中各种有效成分的联合提取,使原料得到了最大限度的利用,减少了黑果枸杞资源浪费。

具体实施方法:
实例:取5kg黑果枸杞干果充分研磨后按1∶50的固液比加入pH3的盐酸溶液于50℃浸提150分钟。离心使固液分离,收集沉淀按1∶50的固液比加入蒸馏水250L于90℃浸提30分钟,再次离心使固液分离。收集沉淀中的枸杞籽1kg,烘干后压榨获得黑果枸杞籽油和黑果枸杞籽饼;将两次浸提上清液合并、真空浓缩,加入95%的乙醇调至80%,4℃下静置过夜醇沉后过滤,将上清液进行真空浓缩后进行冷冻干燥即得黑果枸杞色素产品2.92kg。滤渣用水溶解后,离心得到不溶性物质即为蛋白质(干重42g),上清液加95%的乙醇调至80%进行二次醇沉,过滤,收集沉淀、干燥即得黑果枸杞多糖116g。

Claims (2)

1.黑果枸杞中多种有效成分的联合提取方法,其特征在于,将黑果枸杞中黑果枸杞色素、黑果枸杞多糖、黑果枸杞蛋白质、黑果枸杞籽油、黑果枸杞籽饼依次提取出来,实现了黑果枸杞的综合利用,且工艺简便易行。
2.黑果枸杞中多种有效成分的联合提取方法,其步骤为:
(1)将黑果枸杞干果充分研磨,用pH=3的盐酸溶液按照1∶50的固液比,50℃下浸提150min;
(2)将步骤(1)所得混合溶液离心,分别得到上清液和沉淀;
(3)将步骤(2)所得沉淀用蒸馏水按1∶50固液比,90℃浸提30min;
(4)将步骤(3)所得混合液离心得上清液和沉淀,收集沉淀再经干燥即得黑果枸杞籽;
(5)将步骤(4)所得黑枸杞籽压榨,即得黑枸杞籽油和黑枸杞籽饼;
(6)将步骤(2)和步骤(4)所述的上清液合并,真空浓缩;
(7)向步骤(6)所得浓缩液中加入95%的乙醇使乙醇浓度达到80~85%,4℃下静置过夜;
(8)将步骤(7)所得混合物过滤得上清液和沉淀;
(9)将步骤(8)所得上清液经真空浓缩回收乙醇后冷冻干燥即得黑果枸杞色素产品;
(10)将步骤(8)所得沉淀以蒸馏水溶解后离心,得上清液和沉淀;
(11)将步骤(10)中上清液加入95%的乙醇使乙醇浓度达到80~85%,4℃下静置过夜;
(12)将步骤(11)所得混合物过滤得上清液和沉淀;
(13)将步骤(12)中所得沉淀经干燥即得黑果枸杞多糖产品;
(14)将步骤(10)中所得沉淀干燥即得黑果枸杞蛋白质产品。
