Step one, preparing a traditional Chinese medicine culture medium: respectively screening, cleaning and drying 4 parts of dandelion, 3 parts of honeysuckle, 3 parts of fructus forsythiae, 2 parts of dried orange peel, 2 parts of cortex magnoliae officinalis and 3 parts of liquorice, and mixing to obtain a traditional Chinese medicine mixture, crushing the traditional Chinese medicine mixture into 40-mesh medicinal material fine powder, and adding distilled water which is 10 times of the medicinal material fine powder in weight into the medicinal material fine powder to obtain a traditional Chinese medicine culture medium;
step two, activating microorganisms: inoculating microbial strains into a nutrient broth liquid culture medium, culturing at 37 ℃ for 24h, and carrying out passage twice; inoculating the bacterial liquid after two passages to a nutrient agar solid culture medium by using a leptospira loop, culturing for 24h at the temperature of 37 ℃, then selecting a single bacterial colony to a nutrient broth liquid culture medium, and culturing for 24h at the temperature of 37 ℃ to obtain an activated microorganism seed;
step three, preparing a seed solution: inoculating activated microorganisms into a nutrient broth liquid culture medium, wherein the inoculation amount of the activated microorganisms is 1% of the weight of the nutrient broth liquid culture medium, and then placing the activated microorganisms in a shaking incubator at the culture temperature of 37 ℃, the culture rotation speed of 180r/min and the culture time of 24h to obtain a seed solution;
step four, preparing the traditional Chinese medicine biological preparation: adjusting the pH value of the traditional Chinese medicine culture medium obtained in the step one to 7.0, sterilizing at 105 ℃ for 20min, cooling to room temperature, inoculating a seed solution with the weight being 2% of that of the traditional Chinese medicine culture medium, and then putting the seed solution into a shaking table shaking incubator, wherein the culture temperature is 37 ℃, the culture rotation speed is 150r/min, and the culture time is 48h, so as to obtain the traditional Chinese medicine biological preparation IV.
Secondly, respectively carrying out drug effect determination on the poultry Chinese medicinal biological preparations I to IV prepared by the invention, taking the poultry Chinese medicinal biological preparation III as an example:
(1) determination of total flavone content
a. Preparation of control solutions
Taking 9.7mg of rutin control, precisely weighing, placing in a 50mL measuring flask, adding appropriate amount of methanol for dissolving, adding distilled water to scale, and shaking to obtain control solution (containing 0.194mg of rutin per 1m L).
b. Drawing of standard curve
Precisely measuring reference substance solutions 0.0 mL, 0.2 mL, 0.5 mL, 1.0mL, 1.5 mL, 2.0 mL, 2.5 mL and 3mL, respectively placing in 10mL volumetric flasks, adding distilled water to 3mL, adding 5% sodium nitrite solution 0.4mL, mixing, and standing for 6 min; then 0.4mL of 10 percent aluminum nitrate solution is added, shaken up and placed for 6 min; adding 4.0mL of 4% sodium hydroxide solution, adding distilled water to the scale, shaking, standing for 15min, and taking 0.0 as blank. And (4) measuring the absorbance at 510nm by using an ultraviolet spectrophotometer, and calculating to obtain a regression equation by taking the absorbance as a vertical coordinate and the mass concentration as a horizontal coordinate.
c. 3 samples were tested in parallel and the arithmetic mean was taken.
(2) Total polysaccharide assay
a. Preparation of glucose standard solution and phenol solution
Taking a glucose standard product dried to constant weight at 105 ℃, precisely weighing 10.0mg in a 100mL volumetric flask, adding distilled water to scale, and preparing into a glucose standard solution with the concentration of 0.1 mg/mL.
40g of phenol, 0.5g of zinc powder and NaHC 0320 mg were weighed out and distilled (air-condensed), and the fraction at 182 ℃ was collected. 5.0g of the fraction was weighed into a 100mL volumetric flask, and distilled water was added to the scale to prepare a 5% phenol solution.
b. Drawing of glucose standard curve
0.2 mL, 0.4mL, 0.6 mL, 0.8 mL, 1.0mL, 1.2 mL, 1.4 mL, 1.6mL of glucose standard solution were respectively pipetted into a test tube, water was added to 1.6mL, then 6% phenol 1.0mL and concentrated sulfuric acid 5.0mL were added, shaken well, 2mL of distilled water was used as a reference, the absorbance value was measured at 490nm after standing at room temperature for 25min, a glucose standard curve was drawn, and a regression equation was calculated.
c. 3 samples were tested in parallel and the arithmetic mean was taken.

As can be seen from the above table, the content of total flavonoids in the biological traditional Chinese medicine preparation III is increased by 20.46% compared with the content of total flavonoids in the biological traditional Chinese medicine preparation IV, and the extraction of the total flavonoids as the active ingredients of the biological traditional Chinese medicine preparation III obtained under the fermentation conditions of the embodiment 4 is more sufficient than that of the biological traditional Chinese medicine preparation IV obtained under the fermentation conditions of the embodiment 5; the polysaccharide content of the traditional Chinese medicine biological preparation III measured by adopting a phenol-sulfuric acid method is increased by 25.77 percent compared with the polysaccharide content of the traditional Chinese medicine which is not fermented, the polysaccharide content of the traditional Chinese medicine composition is obviously increased by the fermentation treatment, and the total flavone of the effective components of the traditional Chinese medicine composition is more fully extracted.
(3) Clinical efficacy study
a. Study subjects: 180 healthy red feather broilers of 1 day old are taken and randomly divided into 6 groups, namely a blank control group (blank group), a bacterial liquid control group (bacterial liquid group), a traditional Chinese medicine biological agent test group (namely, non-sterilized group) I, II and III and a traditional Chinese medicine biological agent sterilization test group (IV), wherein each group is provided with 3 repetitions, and each repetition is 10. Cage culture is adopted, a trough and a water fountain are sufficient, food and water are taken freely, 24 hours of illumination are realized, the whole test time is 46 days, and the test time is divided into 2 stages (0-21 days old and 21-46 days old). Inoculating new-fluid dual vaccine at 8 days old, inoculating infectious bursal disease live vaccine at 21 days old, and inoculating by eye drop and nose drop.
The treatment of each group is as follows: the blank group is fed by adding 0.5% of distilled water into the word material, the bacteria liquid group is fed by adding 0.5% of bacteria liquid into the word material, the fermented traditional Chinese medicine sterilization group is fed by adding 0.5% of sterilized traditional Chinese medicine biological agent III into the word material, and the traditional Chinese medicine biological agent test groups I, II and III are respectively fed by adding 0.25%, 0.5% and 1.0% of traditional Chinese medicine biological agent II into the word material.
b. And (3) measuring growth performance indexes: and recording the feed amount, the residual feed amount and the scattered feed amount every day, and calculating the actual feed intake every day. The weight was measured on empty stomach in the morning of 1 day, 21 days and 46 days, respectively.
And calculating the average daily feed intake, average daily gain and material-weight ratio of each stage.

The results in the table show that the differences of the total feed intake, the average daily feed intake and the average daily gain of the broilers in each test group at the age of 0-21 days are extremely obvious (P is less than 0.01), the feed-weight ratios of the test groups are all lower than those of the blank group, and the differences are extremely obvious (P is less than 0.01); the average daily gain and feed weight of the broiler chickens in the test group II are extremely different from those of other test groups (P is less than 0.01), the average daily gain is increased by 27.3 percent compared with a blank group, and the feed weight ratio is reduced by 12.1 percent compared with the blank group. Compared with the bacterial liquid group, the total feed intake and the average daily feed intake of the test group II and the test group III have very obvious difference (P is less than 0.01). The average daily gain and the material-weight ratio of the test group II are very obvious in difference (P is less than 0.01) compared with the bacteria liquid group. In the age period of 21-46 days, the total feed intake and the average daily feed intake of the broilers of all groups except the group I are compared with those of the blank group, and the difference is very obvious (P is less than 0.01). Compared with the blank group, the average daily gain of the broilers in each test group is very obvious in difference (P is less than 0.01); compared with the blank group, the average daily gain of the broilers in the test I group is increased by 8.7 percent, the average daily gain of the broilers in the test II group is increased by 7.0 percent, the average daily gain of the broilers in the test III group is increased by 5.7 percent, and the average daily gain of the broilers in the test IV group is increased by 19.. The material weight ratio of each test group is obviously different (P is more than 0.01 and less than 0.05) compared with that of a blank group, wherein the material weight ratio of the test group II is minimum and is reduced by 18.2 percent compared with that of the blank group. Compared with the bacterial liquid group, the total feed intake, the average daily feed intake and the average daily gain of the groups I, II and III have very obvious difference (P is less than 0.01). The total feed intake, average daily feed intake, and average daily gain of the test groups I, II, and III were lower than those of the bacterial suspension group. Compared with the bacteria liquid group, the material weight ratio of the experiments II and III has obvious difference (P is more than 0.01 and less than 0.05), and the difference is lower than that of the bacteria liquid group.
c. Immune organ index: and (3) collecting blood of 46-day-old chickens, killing the chickens, weighing spleen and bursa of Fabricius, and calculating immune organ indexes.

The results in the table show that the spleen indexes of the broilers in the test group II and the test group IV are remarkably different (P is more than 0.01 and less than 0.05) compared with the blank group, and are not remarkably different (P is more than 0.05) compared with the bacteria liquid group; except group III, however, the spleen index of broilers in all test groups was higher than that of the blank group. The spleen indexes of the test groups I, II and IV are all higher than those of the bacteria liquid group. The bursa of Fabricius indexes of the broilers in each test group are not obvious in difference (P is more than 0.05) compared with those of a blank group and a bacteria liquid group, wherein the bursa of Fabricius indexes of the broilers in other test groups except the test II group are higher than those of the blank group, and the bursa of Fabricius indexes of the broilers in each test group are lower than those of the bacteria liquid group. The results are combined to obtain the traditional Chinese medicine biological preparation III which has certain promotion effect on the development of immune organs of the broiler chicken.

Claims (7)

1. A preparation method of a traditional Chinese medicine biological preparation for promoting growth and strengthening immunity of poultry is characterized by comprising the following steps:
step one, preparing a traditional Chinese medicine culture medium: respectively screening, cleaning and drying dandelion, honeysuckle, fructus forsythiae, dried orange peel, mangnolia officinalis and liquorice, and then mixing to obtain a traditional Chinese medicine mixture, crushing the traditional Chinese medicine mixture into medicinal material fine powder, and adding distilled water into the medicinal material fine powder to obtain a traditional Chinese medicine culture medium; the raw material medicines comprise, by mass, 2-6 parts of dandelion, 2-5 parts of honeysuckle, 2-5 parts of fructus forsythiae, 1-5 parts of dried orange peel, 1-5 parts of mangnolia officinalis and 1-5 parts of liquorice;
step two, activating microorganisms: inoculating Bacillus subtilis strain into nutrient broth liquid culture medium, culturing at 30-40 deg.C for 12-30 hr, and passaging twice; inoculating the bacterial liquid after two passages to a nutrient agar solid culture medium by using a leptospira, culturing for 12-30h at the temperature of 30-40 ℃, then selecting a single bacterial colony to a nutrient broth liquid culture medium, and culturing for 12-30h at the temperature of 30-40 ℃ to obtain an activated microorganism seed;
step three, preparing a seed solution: inoculating the activated microorganism obtained in the step two into a liquid culture medium, and performing shake culture to obtain the seed liquid;
step four, preparing the traditional Chinese medicine biological preparation: adjusting the pH value of the traditional Chinese medicine culture medium obtained in the step one, sterilizing at high temperature, cooling to room temperature, inoculating the seed solution, and placing into a shaking table shaking incubator for shaking culture to obtain the traditional Chinese medicine biological preparation.
2. The method for preparing a growth-promoting and immunity-enhancing poultry traditional Chinese medicine biological preparation according to claim 1, which is characterized in that the raw materials comprise, by mass: 4 parts of dandelion, 3 parts of honeysuckle, 3 parts of fructus forsythiae, 2 parts of dried orange peel, 2 parts of magnolia officinalis and 3 parts of liquorice.

The method for preparing a growth-promoting and immunity-enhancing poultry Chinese medicinal biological agent according to claim 1, wherein the third step is specifically: inoculating 0.5-5% of the activated microorganism obtained in the step two into a nutrient broth liquid culture medium, and then placing the culture medium in a shaking incubator at the culture temperature of 30-40 ℃, the culture rotation speed of 100-200r/min and the culture time of 12-30h to obtain the seed liquid.
