2.6 Effect of herbal wines on uric acid
The Uric Acid (UA) level of normal rats was significantly reduced (P <0.001) after taking the herbal alcohol at both low and medium concentrations, but was not significantly changed in the high concentration group. For rats with kidney-yang deficiency, the effect of herbal alcohol in reducing uric acid was not obvious (fig. 5).
2.7 Effect of herbal wines on thyroxine
In the normal group, the herbaceous plant wine has little influence on thyroxine T3 and T4, but the levels of T3 and T4 in the serum of rats after hydrocortisone molding are obviously reduced (P is less than 0.05), and the thyroxine level is increased after the rats are subjected to gastric lavage by the herbaceous plant wine. Thyroxine can generate heat through uncoupling of mitochondrial respiratory chain, promote metabolism of sugar, protein and the like, and has important effects on organism productivity and growth and development. The reduction of thyroxine in the model group indicates that the model accords with the general kidney-yang deficiency, the T3 and T4 levels can be improved after the model is fed with herbal wine, and the herbal wine has a certain treatment effect on rats with kidney-yang deficiency (figure 6).
2.8 Effect of herbal wines on sex hormones
The content of estradiol in each group of rats has no obvious change; the change of each group on the content of testosterone in serum is obvious, and the content of testosterone in the herb plant wine in the gavage is obviously increased (P is less than 0.05) in normal rats; in rats of a model group injected with hydrocortisone, the testosterone content is remarkably reduced (P is less than 0.05), which is consistent with the clinical symptoms of kidney-yang deficiency, and after the rats are infused with herbal wine, the testosterone level is remarkably improved, which indicates that the herbal wine can improve the androgen level and remarkably improve the symptoms of kidney-yang deficiency (figure 7).
2.9 Effect of herbaceous plant wine on pathological changes of liver tissue of rat with kidney-yang deficiency
The HE (400X) staining of the liver of each group of rats showed no significant change in liver morphological function after administration of herbal wine to normal and model rats (FIG. 8).
2.10 Effect of herbal wine on Kidney Yang deficiency in rat testis tissue
As can be seen from HE staining (100 ×), the rat convoluted tubule of the model group has obvious atrophy degeneration, and the number of interstitial cells and sperm cells is obviously reduced compared with the normal group. In the normal group, it can be seen that the sperm cells at all levels in the seminal tubules are well developed, the number of the sperm cells is large, the arrangement is compact, the cavities are filled with sperms, the boundaries of the tube walls are neat, the basement membrane is thin, and the development of the testicular interstitial cells is normal. After the herbaceous plant wine is infused, the normal rat does not have obvious change, but the model rat can obviously change, the number of seminal tubules and sperms is increased, and the arrangement is more compact (figure 9).
3. Conclusion of the experiment
1. After the glucocorticoid hydrocortisone model is made, the weight of a rat is obviously reduced, and a series of symptoms such as loose stool, reduced activity, shrugging of the back and the like appear, which indicates that the construction of the kidney-yang deficiency model is successful.
2. After the rats are fed with the herbal wine, the weight, the nutritional status and the liver function of the rats are not obviously changed, which indicates that the herbal wine has no obvious toxic or side effect on normal organisms, and does not change in a high-dose group, and indicates that the herbal wine is very safe in a dose range.
3. After the model is established, the two indexes of the kidney function, namely urea nitrogen and blood creatinine, are obviously increased, which is the specific embodiment of the kidney function degradation; meanwhile, the levels of thyroxine T3 and T4 which represent two indexes of energy metabolism are obviously reduced, which indicates that the metabolism of a model rat is reduced and the productivity is reduced, so that the cold intolerance symptom is caused, and the levels of blood creatinine and urea nitrogen are obviously reduced after the herbal wine is fed, which indicates that the herbal wine has the function of enhancing the kidney function, and simultaneously, the contents of thyroxine T3 and T4 are also increased, which indicates that the herbal wine can improve the metabolic rate of an organism.
4. It can be seen from the level of male hormone testosterone and HE staining of testis that the reproductive function of model group rat is obviously reduced compared with normal rat, and after 4 weeks of herbaceous plant liquor gavage, the content of serum testosterone is obviously improved, and reproductive function is obviously enhanced.
5. In addition, uric acid level is one of the indexes for measuring kidney function, and the increase of uric acid level is a high-risk factor of various diseases including gout.
6. Most indexes have obvious change in low concentration and medium concentration groups, and the dosage of the index is converted from the daily recommended drinking amount of normal people, so that proper drinking is recommended.

Claims (8)
1. The herbal wine with the effects of tonifying kidney and strengthening yang is characterized by being prepared from a herbal composition, wherein the herbal composition comprises the following traditional Chinese medicine components in parts by weight: 4-8 parts of clove, 2-4 parts of cinnamon, 20-40 parts of Chinese yam, 8-15 parts of donkey-hide gelatin, 10-20 parts of wolfberry fruit, 15-20 parts of mulberry, 10-20 parts of fructus alpiniae oxyphyllae, 12-18 parts of rhizoma polygonati and 15-25 parts of raspberry.
2. The herbal wine with the effects of tonifying kidney and strengthening yang as claimed in claim 1, wherein the herbal composition comprises the following traditional Chinese medicine components in parts by weight: 5-7 parts of clove, 2-4 parts of cinnamon, 25-35 parts of Chinese yam, 9-12 parts of donkey-hide gelatin, 15-18 parts of wolfberry fruit, 15-20 parts of mulberry, 12-17 parts of fructus alpiniae oxyphyllae, 12-18 parts of rhizoma polygonati and 15-20 parts of raspberry.
3. The herbal wine having the effects of invigorating kidney and strengthening yang as claimed in claim 1 or 2, wherein the alcohol content of the herbal wine is 35-37 vol%.
4. A method for preparing herbal wine having kidney-invigorating and yang-strengthening effects as claimed in claim 1, comprising the steps of:
(1) mixing flos Caryophylli, cortex Cinnamomi, rhizoma Dioscoreae, fructus Lycii, Mori fructus, fructus Alpinae Oxyphyllae, rhizoma Polygonati and Rubi fructus, pulverizing, soaking in Chinese liquor for 1-3 months, and filtering to obtain extractive solution;
(2) melting colla Corii Asini, mixing with the leaching solution, adding purified water, and reducing to obtain wine solution;
(3) standing the liquor for more than 30 days, sieving the supernatant with 200 mesh sieve, packaging the filtrate into wine bottles, testing under light, and packaging.
5. The preparation method according to claim 4, wherein in the step (1), the alcoholic strength of the white spirit is 37-42 vol%.
6. The method according to claim 5, wherein the liquor is Maotai-flavor liquor.
7. The method according to claim 4, wherein in the step (1), 400mL of distilled spirit is added to 100g of the pulverized material for soaking.
8. The method according to claim 4, wherein the alcohol content of the alcohol solution after the alcohol content is reduced by adding purified water in the step (2) is 35 to 37 vol%.
