Abstract

一种止血粉的制备方法,属于外用医药技术领域。本发明的制备方法包含以下步骤:(1)介孔硅二氧化硅颗粒制备;(2)止血浸渍液的制备;(3)浸渍制得止血粉。本发明制得的止血粉主要成分为壳聚糖、凝血酶和鞣酸。凝血酶与鞣酸的氢键结合,使凝血酶的存储稳定性大大提高,负载大量凝血酶和壳聚糖的介孔硅二氧化硅颗粒,止血性能大大提高,弥补了纯壳聚糖止血效果有限的缺陷。本发明止血粉能快速止血,特别适用于严重出血伤员的院前紧急救治,制备方法和使用方法方便,制得的止血粉稳定性好。

技术领域
本发明涉及一种止血粉的制备方法,属于外用医药技术领域。
背景技术
壳聚糖(chitosan)又称脱乙酰甲壳素,是由自然界广泛存在的几丁质(chitin)经过脱乙酰作用得到的,化学名称为聚葡萄糖胺(1-4)-2-氨基-B-D葡萄糖。自1859年,法国人Rouget首先得到壳聚糖后,这种天然高分子的生物官能性和相容性、血液相容性、安全性、微生物降解性等优良性能被各行各业广泛关注,在医药、食品、化工、化妆品、水处理、金属提取及回收、生化和生物医学工程等诸多领域的应用研究取得了重大进展。
壳聚糖在弱酸溶剂中易于溶解,特别值得指出的是溶解后的溶液中含有氨基(NH2+),这些氨基通过结合负电子来抑制细菌。壳聚糖的抑制细菌活性,使其在医药、纺织和食品等领域有着广泛的应用。壳聚糖及其衍生物有较好的抗菌活性,能抑制一些真菌、细菌、和病毒的生长繁殖。截止到2013年认为其可能的机制有三:一是由于壳聚糖的多聚阳离子,易于真菌细胞表面带负电荷的基团作用,从而改变病原菌细胞膜的流动性和通透性;二是干扰DNA的复制与转录;三是阻断病原菌代谢。
纯壳聚糖的止血性能十分有限,对于广泛出血创面的止血效果不很理想,常采用复合其他止血剂如凝血因子、氯化钙等方法提高止血性能。其中凝血酶是一种很常用的局部速效止血药,已经FDA认证,可以在临床上直接使用。通过将凝血酶与壳聚糖进行复合,可以制备出新型止血材料。但由于凝血酶是一种蛋白质基止血因子,存储稳定性较差,需冻干后在低温下保存,这大大制约了凝血酶在急救敷料上的应用。凝血酶和鞣酸被证实在生理pH下存在氢键相互作用,通过将凝血酶/鞣酸复合在壳聚糖中有望制得一种止血效率高、生物安全性好、易存储的止血敷料。
发明内容
本发明的目的是提供一种止血粉的制备方法。
本发明的技术方案包括如下:
本发明止血粉的制备方法包括以下步骤:
(1)介孔硅二氧化硅颗粒制备:将20ml正硅酸乙酯、10ml乙醇、2-2.5ml平均分子量为5800的聚醚多元醇P123、1ml2mol/L盐酸在45℃混和搅拌3-7小时,在450-650℃煅烧5-10h,球磨1-4h,150目过筛后得到介孔硅二氧化硅颗粒;
(2)将4g壳聚糖溶于200ml体积浓度为0.5-2%的乙酸水溶液中,加入6-9g聚乙二醇20000,得到壳聚糖溶液,称为A液;将树枝状聚醚酰亚胺溶解在磷酸缓冲液中,配置质量浓度为1-3mg/mL的聚醚酰亚胺溶液,称为B液;将鞣酸溶解在磷酸缓冲液中,配置质量浓度为1-3mg/mL的鞣酸溶液,称为C液;将凝血酶溶解在磷酸缓冲液中,配置质量浓度为1-3mg/mL的凝血酶溶液,称为D液;将上述A、B、C、D液混合均匀,得到止血浸渍液;
(3)向步骤(2)制备的止血浸渍液中加入10-30g步骤(1)制备的介孔硅二氧化硅颗粒,室温搅拌24h,将沉淀物冷冻干燥后悬浮于300ml二甲基亚砜中,用氢氧化钠水溶液调pH值=7,离心、用去离子水洗涤3-5次后冷冻干燥,得到止血粉。
凝血酶与鞣酸的氢键结合,使凝血酶的存储稳定性大大提高,负载大量凝血酶和壳聚糖的介孔硅二氧化硅颗粒,止血性能大大提高,弥补了纯壳聚糖止血效果有限的缺陷。
本发明的有益效果是:
本发明止血粉能快速止血,特别适用于严重出血伤员的院前紧急救治,制备方法和使用方法方便,制得的止血粉稳定性好。
具体实施方式
以下结合技术方案详细叙述本发明的具体实施方式。
本发明止血粉的制备方法包括以下步骤:
(1)介孔硅二氧化硅颗粒制备:将20ml正硅酸乙酯、10ml乙醇、2ml平均分子量为5800的聚醚多元醇P123、1ml2mol/L盐酸在45℃混和搅拌5小时,在500℃煅烧7h,球磨3h,150目过筛后得到介孔硅二氧化硅颗粒;
(2)将4g壳聚糖溶于200ml体积浓度为1.2%的乙酸水溶液中,加入7g聚乙二醇20000,得到壳聚糖溶液,称为A液;将树枝状聚醚酰亚胺溶解在磷酸缓冲液中,配置质量浓度为1mg/mL的聚醚酰亚胺溶液,称为B液;将鞣酸溶解在磷酸缓冲液中,配置质量浓度为3mg/mL的鞣酸溶液,称为C液;将凝血酶溶解在磷酸缓冲液中,配置质量浓度为3mg/mL的凝血酶溶液,称为D液;将上述A、B、C、D液混合均匀,得到止血浸渍液;
(3)向步骤(2)制备的止血浸渍液中加入30g步骤(1)制备的介孔硅二氧化硅颗粒,室温搅拌24h,将沉淀物冷冻干燥后悬浮于300ml二甲基亚砜中,用氢氧化钠水溶液调pH值=7,离心、用去离子水洗涤3次后冷冻干燥,得到止血粉。

Claims (1)

1.一种止血粉的制备方法,其特征在于,所述的止血粉的制备方法包括以下步骤:
(1)介孔硅二氧化硅颗粒制备:将20ml正硅酸乙酯、10ml乙醇、2-2.5ml平均分子量为5800的聚醚多元醇P123、1ml2mol/L盐酸在45℃混和搅拌3-7小时,在450-650℃煅烧5-10h,球磨1-4h,150目过筛后得到介孔硅二氧化硅颗粒;
(2)将4g壳聚糖溶于200ml体积浓度为0.5-2%的乙酸水溶液中,加入6-9g聚乙二醇20000,得到壳聚糖溶液,称为A液;将树枝状聚醚酰亚胺溶解在磷酸缓冲液中,配置质量浓度为1-3mg/mL的聚醚酰亚胺溶液,称为B液;将鞣酸溶解在磷酸缓冲液中,配置质量浓度为1-3mg/mL的鞣酸溶液,称为C液;将凝血酶溶解在磷酸缓冲液中,配置质量浓度为1-3mg/mL的凝血酶溶液,称为D液;将所述的A、B、C、D液混合均匀,得到止血浸渍液;
(3)向步骤(2)制备的止血浸渍液中加入10-30g步骤(1)制备的介孔硅二氧化硅颗粒,室温搅拌24h,将沉淀物冷冻干燥后悬浮于300ml二甲基亚砜中,用氢氧化钠水溶液调pH值=7,离心、用去离子水洗涤3-5次后冷冻干燥,得到止血粉。
