Abstract

本发明是一种木耳多糖提取方法,所用原料为木耳,经除杂、粉碎、过筛,用稀的盐酸溶液浸泡,并加入纤维素酶进行酸解,酸解结束后,采用短时高温灭酶,灭酶后将滤液减压浓缩,加入乙醇,在冰水浴中静置结晶,过滤出结晶物即木耳多糖。

本发明涉及活性物质提取分离技术领域,具体涉及一种木耳多糖提取方法。
背景技术
木耳是我国珍贵的药用和食用胶质真菌,多糖为木耳的重要活性成分,它具有抗凝血、抗肿瘤、抗炎症等细胞保护作用,还具有降低血脂、胆固醇、血液粘度、抗血栓形成降低血糖、抗糖尿病、抗衰老等作用。传统木耳多糖提取采用水提取,木耳细胞粗大、壁厚,多糖难以从胞内扩散出来,需要多次浸提,操作时间长、收率低。
发明内容
本发明所要解决的技术问题是针对现有技术的不足,提供一种新的木耳多糖提取方法。
本发明所要解决的技术问题是通过以下的技术方案来实现的。原料为木耳,经除杂、粉碎、过筛,用稀的盐酸溶液浸泡,并加入纤维素酶进行酸解,酸解结束后,采用短时高温灭酶,灭酶后将滤液减压浓缩,加入乙醇,在冰水浴中静置结晶,过滤出结晶物即木耳多糖。
与现有技术相比,本发明工艺简便、省时,提取率高,对于木耳的综合开发利用及提高其经济价值具有实际意义。
具体实施方式
原料。剔除发霉变质以及杂质,粉碎,过50目筛,备用。
酸解。配制PH值为5的盐酸溶液10升,将木耳粉1000克用盐酸溶液浸泡,并加入纤维素酶10克,搅拌均匀,溶液温度55℃,浸泡时间90分钟。
灭酶。将浸泡液温度迅速升至90℃,并维持2分钟,降至室温、过滤,保留滤液。
浓缩。将滤液在50℃下减压浓缩至三分之一体积。
沉淀。向溶液中加入溶液体积的3倍乙醇,并不断搅拌,在冰水浴中静置12小时,过滤出结晶物即木耳多糖。

1.一种木耳多糖提取方法,其特征在于:所用原料为木耳,经除杂、粉碎、过筛、酸解、灭酶、浓缩、沉淀、干燥而成。
2.根据权利要求1所述的一种木耳多糖提取方法,其特征在于:所述过筛是过40-80目筛。
3.根据权利要求1所述的一种木耳多糖提取方法,其特征在于:所述酸解是用PH值为2-7的盐酸溶液浸泡木耳,并加入木耳质量0.5-1.5%的纤维素酶,浸泡液温度为40-80℃,酸解时间为60-120分钟。
4.根据权利要求1所述的一种木耳多糖提取方法,其特征在于:所述浓缩是在40-70℃温度下减压浓缩。
5.根据权利要求1所述的一种木耳多糖提取方法,其特征在于:所述沉淀是向浓缩液中加入乙醇,在4℃左右静置8-16小时。
