Abstract
本发明涉及一种新型环保固定液,包含质量分数为0.9‑1.4%的表面活性物质、质量分数为12‑20%的多元醇。本发明还公开了一种制备上述新型环保固定液的方法,将各组分混合后放入行星式重力搅拌机中搅拌得到。通过向甲醛固定液中添加表面活性物质和多元醇,使得所述新型环保固定液既降低了挥发性又保持了原有的固定功能和渗透性。使用磷酸缓冲剂可将新型环保固定液的pH稳定在中性的7.2‑7.4的范围,从而延长固定液的储存期限。
Description
技术领域
本发明涉及病理试剂领域,更特别地,涉及一种环保固定液。
背景技术
目前,甲醛固定液被广泛使用于医学病理学检测中,临床上使用的固定液为福尔马林稀释液,其甲醛含量在3.6%左右。
甲醛是一种常温下易挥发的有害化学物质,有资料显示,其为多种癌症及血液病诱因。甲醛在固定液的使用过程中极易逸失到空气中,被操作人员吸入而危害其健康。目前市场上的甲醛固定液在防止甲醛挥发上未作任何处理,对环境不友好,危害人身安全。
此外,作为固定液的主有效成分,甲醛容易聚合而失去效力。市售固定液多添加12%左右的甲醇作为甲醛聚合抑制剂。然而甲醇易使组织表面蛋白质固化,抑制甲醛的进一步深入作用,使标本组织松弛,影响后续的切片及显微镜观察。
因此,需要一种新的甲醛固定液,既能降低其中甲醛的挥发性,又不影响甲醛的渗透性。
发明内容
为解决以上问题,本发明提供了一种环保固定液,其包含质量分数为0.9-1.4%的表面活性物质和质量分数为12-20%的多元醇。通过向甲醛固定液中添加表面活性物质和多元醇,使得所述环保固定液既降低了挥发性又保持了原有的固定功能和渗透性。
优选地,所述表面活性物质为吐温80。
优选地,所述多元醇为异丙醇和/或丙三醇。
优选地,所述环保固定液还包含磷酸盐缓冲剂。甲醛在长期储存中会慢慢被氧化成甲酸,从而使得固定液的pH逐渐降低,从而影响固定效果。使用磷酸缓冲剂可将环保固定液的pH稳定在中性的7.2-7.4的范围,从而延长固定液的储存期限。
优选地,所述磷酸盐缓冲剂为质量分数为3.9-4.1%的磷酸二氢钠和6.3-6.6%的磷酸氢二钠。
优选地,所述环保固定液包含以下成分:3.6-4.0重量份的甲醛、9-15重量份的异丙醇、3-5重量份的丙三醇、3.9-4.1重量份的磷酸二氢钠、6.3-6.6重量份的磷酸氢二钠、0.9-1.4重量份的吐温80以及63.9-73.3重量份的水。
本发明还公开了一种制备上述环保固定液的方法,将各组分混合后放入行星式重力搅拌机中搅拌得到。
优选地,在所述行星式重力搅拌机中先以600转/分钟的转速搅拌30分钟,然后以200转/分钟的转速搅拌5分钟。
具体实施方式
以下结合实例对本发明的原理和特征进行描述,所举实例只用于解释本发明,并非用于限定本发明的范围。
1.环保固定液的制备
按表1中的组分配制环保固定液,然后将混合物放入行星式重力搅拌机,以600转/分钟的转速搅拌30分钟后再以200转/分钟的转速搅拌5分钟,得到的溶液密封保存即为本发明的一些实施例的环保固定液。经检测,这些环保固定液的pH都在7.2-7.4之间。

2.本发明实施例的环保固定液的挥发性测定
取市售10%福尔马林固定液(甲醛含量3.6%)及本例所述之环保固定液各500ML分置相同的口径为20cm的广口瓶内,敞口置于密闭空间(0.5m×0.5m×0.5m)中,静置3分钟后,按GBZ159-2004、GBZ/T160.54-2004《工作场所空气有毒物质测定脂肪族醛类化合物》进行采样分析。
市售10%福尔马林固定液样品甲醛挥发浓度为1.65mg/m3;实施例1-3的环保固定液样品甲醛挥发浓度分别为0.67mg/m3、0.47mg/m3和0.53mg/m3。可见,使用本发明的方法制备的环保固定液与市售的甲醛固定液相比挥发性降低为市售的28-40%,效果显著。
所述之环保固定液经临床使用验证,固定效果良好,性状稳定。
以上所述仅为本发明的较佳实施例,并不用以限制本发明,凡在本发明的精神和原则之内,所作的任何修改、等同替换、改进等,均应包含在本发明的保护范围之内。
Claims (6)
1.一种环保固定液,其特征在于,包含质量分数为0.9-1.4%的表面活性物质、质量分数为12-20%的醇、质量分数为3.6-4.0%的甲醛;
所述表面活性物质为吐温80;所述醇为异丙醇和丙三醇。
2.根据权利要求1所述的环保固定液,其特征在于,还包含磷酸盐缓冲剂。
3.根据权利要求2所述的环保固定液,其特征在于,所述磷酸盐缓冲剂为质量分数为3.9-4.1%的磷酸二氢钠和6.3-6.6%的磷酸氢二钠。
4.根据权利要求1-3中任一项所述的环保固定液,其特征在于,包含以下成分:3.6-4.0重量份的甲醛、9-15重量份的异丙醇、3-5重量份的丙三醇、3.9-4.1重量份的磷酸二氢钠、6.3-6.6重量份的磷酸氢二钠、0.9-1.4重量份的吐温80以及63.9-73.3重量份的水。
5.一种如权利要求1-4中任一项所述的环保固定液的制备方法,其特征在于,按比例将各组分混合后放入行星式重力搅拌机中搅拌得到所述环保固定液。
6.根据权利要求5所述的制备方法,其特征在于,在所述行星式重力搅拌机中先以600转/分钟的转速搅拌30分钟,然后以200转/分钟的转速搅拌5分钟。
