Abstract

本发明公开了一种糜蛋白酶冻干粉的冷冻干燥技术。本发明的糜蛋白酶冻干粉针剂外观饱满均一、孔隙率高、复溶性好、稳定性高,并且处方简单,药用辅料添加比例小,对人体的副作用小,安全可靠。本发明对糜蛋白酶冻干粉的冷冻干燥工艺做了进一步的改进,并进行两次除菌。

Description

一种糜蛋白酶无菌冻干粉的冷冻干燥技术
技术领域
本发明涉及化学制药工艺领域,具体地说涉及一种糜蛋白酶冻干粉的冷冻干燥技术。
背景技术
糜蛋白酶又称胰凝乳蛋白酶,分子量为42000道尔顿,用于肽链的酶解,专门水解肽键,属于肽链内切酶,水解产率很高,但其专一性小于胰蛋白酶。其固体状态较稳定,呈白色棒状结晶;而水溶液极不稳定,pH在5~8时,其活力最强,也最易失去活性。
糜蛋白酶为胰腺分泌的一种蛋白水解酶,能迅速分解变性蛋白质,作用、用途与胰蛋白酶相似,比胰蛋白酶分解能力强、毒性低、不良反应小。一般用于手术后创口或创伤愈合、抗炎及防止局部积血、水肿、扭伤血肿、乳房手术后局部肿胀、鼻炎、中耳炎等。也可用于白内障摘除、松解睫状肌韧带,以减少膜破裂和视网膜损伤。此外,糜蛋白酶与胰蛋白酶或其他化学药剂共同治疗各种炎症具有协同作用,故常用于外科创伤及眼睛消肿治疗。
由于糜蛋白酶是生物大分子,经冻干后的蓬松度较差,溶解速度较慢。为此,本发明提出了一种使糜蛋白酶冻干粉蓬松度高、稳定性高的冷冻干燥技术。
发明内容
本发明提供了一种糜蛋白酶冻干粉的冷冻干燥技术,能够制备蓬松度高、稳定性高的糜蛋白酶冻干粉。所述冷冻干燥包括以下步骤:
1.预冻:搁板温度在5-15℃进箱,并将冻干箱温度以2-6℃/min的速度降至-20~-15℃,保温1-3h;再将冻干箱温度以0.7-1.2℃/min的速度降至-40~-50℃,等制品温度达-30~-40℃时,计时保温1-3小时。
2.一次干燥:将冷凝温度降至-48~-58℃,抽真空至10pa以下,将冻干箱温度以0.45-0.55℃/min的速度升至4-8℃,保温至水晶消失,然后继续保温1-3小时。
3.二次干燥:冻干箱以0.5-0.8℃/min升温至25-35℃,待制品温度达20-28℃时,计时保温2-5小时,检查真空度无变化,结束冻干过程,充氮全压塞,出箱。
具体实施方式
下面结合具体实施例对本发明作进一步说明。
实施例1
1.配制药液;
2.药液经两次除菌过滤;
3.滤液分装,半压塞;
4.冷冻干燥:
(1)预冻:搁板温度在5℃进箱,并将冻干箱温度以2℃/min的速度降至-20℃,保温1小时;再将冻干箱温度以0.7℃/min升温至25℃,待制品温度达20℃时,计时保温5小时;
(2)一次干燥:将冷凝温度降至-48℃,抽真空至10pa以下,将冻干箱温度以0.45-0.55℃/min的温度升至4-8℃,保温至水晶消失,然后继续保温1-3小时;
(3)二次干燥:冻干箱以0.8℃/min升温至35℃,待制品温度达28℃时,计时保温2小时;检查真空度无变化,结束冻干过程,充氮全压塞,出箱。
实施例2
1.配制药液;

(2)一次干燥:将冷凝温度降至-58℃,抽真空至10pa以下,将冻干箱温度以0.5℃/min的速度升至8℃,保温至水晶消失,然后继续保温2小时;
(3)二次干燥:冻干箱温度以0.7℃/min的速度升温至28℃,待制品温度达24℃时,计时保温2小时;检查真空度无变化,结束冻干过程。充氮全压塞,出箱。

1.一种糜蛋白酶冻干粉的冷冻干燥技术,其特征在于,所述的冷冻干燥包括以下步骤:
(1)、预冻:搁板温度在5-15℃进箱,并将冻干箱温度以2-6℃/min的速度降至-20~-15℃,保温1-3h;再将冻干箱温度以0.7-1.2℃/min的速度降至-40~-50℃,等制品温度达-30~-40℃时,计时保温1-3小时;
(2)、一次干燥:将冷凝温度降至-48~-58℃,抽真空至10pa以下,将冻干箱温度以0.45-0.55℃/min的速度升至4-8℃,保温至水晶消失,然后继续保温1-3小时;
(3)、二次干燥:冻干箱以0.5-0.8℃/min升温至25-35℃,待制品温度达20-28℃时,计时保温2-5小时,检查真空度无变化,结束冻干过程,充氮全压塞,出箱。
