Abstract

本发明公开了一种雷公藤多苷片,由下述重量份的原料制备而成:雷公藤85‑90份、中药辅料6‑8份、硬脂酸镁1‑3份、麦芽糊精6‑8份、矫味剂0.01‑0.05份。本发明雷公藤多苷片,具有低毒高效的显著效果,并且通过添加矫味剂,改善了雷公藤多苷片的口感。

技术领域
本发明涉及中药制剂技术领域,具体涉及一种雷公藤多苷片。
背景技术
雷公藤,为卫矛科植物雷公藤的根,又叫黄藤、黄腊藤、菜虫药雷公藤、红药、水莽草,味苦、辛,性凉,大毒。归肝、肾经。功效祛风除湿、通络止痛、消肿止痛、解毒杀虫。可用于类风湿性关节炎、原发性肾小球肾病、肾病综合征、紫瘢性、狼疮性肾炎、红斑狼疮、亚急性、慢性重症肝炎、慢性活动性肝炎;亦可用于过敏性皮肤脉管炎、皮炎、湿疹、银屑病性关节炎、麻风反应、白噻氏病、复发性口疮、强直性脊柱炎等。
现代研究发现,雷公藤药材中含有雷公藤甲素和内酯酮。其中,雷公藤甲素具有良好的抗炎和免疫抑制活性,是雷公藤的重要活性成分;而雷公藤内酯酮的雄性抗生育作用非常显著,是雷公藤抗生育副作用的主要成分。在抗炎作用方面,它能拮抗和抑制炎症介质的释放及实验性炎症及关节炎的反应程度。在抑制免疫作用方面,它能抑制T细胞功能、延迟型变态反应、白介素的分泌、分裂源及抗原刺激的T细胞分裂与繁殖。
但是,长期服用雷公藤多苷片,可能会对患者的消化系统、血液系统、泌尿系统、心血管系统等产生一定的影响,严重者甚至出现急性中毒肝损伤、全血细胞减少、急性肾功能衰竭等症状。
因此,在对雷公藤药材进行制剂时,需要对雷公藤的毒性进行控制,使雷公藤多苷片在使用时尽可能低毒释放,产生疗效。
发明内容
针对现有技术中存在的上述不足,本发明所要解决的技术问题是提供一种雷公藤多苷片。
本发明目的是通过如下技术方案实现的:
一种雷公藤多苷片,由下述重量份的原料制备而成:雷公藤85-90份、中药辅料6-8份、硬脂酸镁1-3份、麦芽糊精6-8份,矫味剂0.01-0.05份。
优选地,所述中药辅料为皂角、生姜、甘草中一种或多种的混合物。
更优选地,所述中药辅料为皂角、生姜、甘草的混合物,所述皂角、生姜、甘草的质量比为(1-3):(1-3):(1-3)。
优选地,所述矫味剂为山梨醇、甜蜜素、阿斯巴甜中一种或多种的混合物。
更优选地,所述矫味剂为山梨醇、甜蜜素、阿斯巴甜的混合物,所述山梨醇、甜蜜素、阿斯巴甜的质量比为(1-3):(1-3):(1-3)。
具体的,在本发明中:
皂角,拉丁学名:Gleditsia sinensis Lam.,又名鸡栖子、长皂荚,内含黄酮甙、酚类、氨基酸。可祛痰止咳、开窍通闭、杀虫散结。用于中风口禁、中暑不醒、咽喉肿痛、咳逆上气、痰喘咳嗽、腹部肿痛、风热牙痛、脱肛。
生姜,拉丁学名:Zingiber officinale Roscoe,又名姜根、百辣云、勾装指、因地辛、炎凉小子、鲜生姜、蜜炙姜。用于脾胃虚寒、食欲减退、恶心呕吐、痰饮呕吐、胃气不和的呕吐、风寒、寒痰咳嗽、感冒风寒、恶风发热、鼻塞头痛。
甘草,拉丁学名:Boehmeria gracilis C.H.Wright。气微,味甜。功能主治:清热解毒、祛痰止咳、脘腹等。
山梨醇,CAS号:50-70-4。
甜蜜素,CAS号:139-05-9。
阿斯巴甜,CAS号:22839-47-0。
氧化铝,即Al2O3,CAS号:1344-28-1。
硬脂酸镁,CAS号:557-04-0,采用青岛郎顺弘盛有限公司提供的食品级硬脂酸镁。
麦芽糊精,CAS号:9050-36-6,采用苏州华航化工科技有限公司公司提供的食品级麦芽糊精。
本发明雷公藤多苷片,具有低毒高效的显著效果,并且通过添加矫味剂,改善了雷公藤多苷片的口感。
具体实施方式
实施例1
雷公藤多苷片原料(重量份):雷公藤85份、中药辅料6份、硬脂酸镁2份、麦芽糊精6份、矫味剂0.03份。
雷公藤多苷片的制备方法,包括以下步骤:
(1)将中药辅料粉碎过120目筛,用中药辅料重量15倍的质量浓度为80%的乙醇浸渍24小时,再加热回流提取4次,采用180目滤布过滤,得到滤液,滤液减压浓缩至药材重量的3倍,得到中药辅料浸膏;
(2)将雷公藤和中性氧化铝按质量比1:2混合均匀,粉碎过140目筛,得到混合物粉末,将混合物粉末装上硅胶柱,混合物粉末与硅胶的质量比为1:2,再加入混合物粉末与硅胶总体积2.5倍的氯仿,浸渍32h,然后用氯仿洗脱至洗脱液为无色;再用有机溶剂洗脱至洗脱液为无色,收集有机溶剂洗脱液,其中有机溶剂由二氯甲烷和甲醇按体积比为10:1混合均匀得到;减压加热浓缩除去有机溶剂,即得雷公藤多苷固体;
(3)将中药辅料浸膏、雷公藤多苷固体、硬脂酸镁、麦芽糊精和矫味剂混合均匀,60℃干燥8小时,压片,即得。
所述中药辅料由皂角、生姜、甘草按质量比为1:1:1混合均匀得到。
所述矫味剂由山梨醇、甜蜜素、阿斯巴甜按质量比为1:1:1混合均匀得到。
上述份,均为重量份。
实施例2
与实施例1基本相同,区别仅仅在于:所述中药辅料辅料由皂角、生姜按质量比为1:1混合均匀得到。得到实施例2的雷公藤多苷片。
实施例3
与实施例1基本相同,区别仅仅在于:所述中药辅料辅料由生姜、甘草按质量比为1:1混合均匀得到。得到实施例3的雷公藤多苷片。
实施例4
与实施例1基本相同,区别仅仅在于:所述中药辅料辅料由皂角、甘草按质量比为1:1混合均匀得到。得到实施例4的雷公藤多苷片。
实施例5
与实施例1基本相同,区别仅仅在于:所述矫味剂由山梨醇、甜蜜素按质量比为1:1混合均匀得到。得到实施例5的雷公藤多苷片。
实施例6
与实施例1基本相同,区别仅仅在于:所述矫味剂由甜蜜素、阿斯巴甜按质量比为1:1混合均匀得到。得到实施例6的雷公藤多苷片。
实施例7
与实施例1基本相同,区别仅仅在于:所述矫味剂由山梨醇、阿斯巴甜按质量比为1:1混合均匀得到。得到实施例7的雷公藤多苷片。
测试例1
对实施例1-7制备的雷公藤多苷片的稳定性进行测试。
分别取实施例1-7制得的雷公藤多苷片用铝塑复合膜袋密封包装,将它们置于38℃下放置3个月,测定各试样中3月时的黄芪甲苷相对于该试样0月时的残余含量。测试方法参照刘法千等《一种自雷公藤前段浸膏中提取雷公藤氯内酯醇的方法》,具体结果见表1。
上述实施例中雷公藤氯内酯醇的高效液相检测方法为:高效液相色谱仪Agilent1260;流动相:甲醇:水(50:50);色谱柱:C18柱(4.6×250mm);检测波长选择219nm;流速为:1.0ml/min,室温操作。

比较实施例1与实施例2-4,实施例1(皂角、生姜、甘草复配)稳定性明显优于2-4(皂角、生姜、甘草任意二者复配)。比较实施例1与实施例5-7,实施例1(山梨醇、甜蜜素、阿斯巴甜复配)稳定性明显优于实施例5-7(山梨醇、甜蜜素、阿斯巴甜任意二者复配)。
测试例2
将实施例1-7制备的雷公藤多苷片,置于25℃,相对湿度85%环境下保藏半年,然后进行大肠杆菌(ATYCC 25922)菌落总数测试,参照《GBT 4789.2-2008食品卫生微生物学检验菌落总数测定》。具体测试结果见表2。

比较实施例1与实施例2-4,实施例1(皂角、生姜、甘草复配)防腐性能明显优于实施例2-4(皂角、生姜、甘草任意二者复配)。比较实施例1与实施例5-7,实施例1(山梨醇、甜蜜素、阿斯巴甜复配)防腐性能明显优于实施例5-7(山梨醇、甜蜜素、阿斯巴甜任意二者复配)。

1.一种雷公藤多苷片,其特征在于,由下述重量份的原料制备而成:雷公藤85-90份、中药辅料6-8份、硬脂酸镁1-3份、麦芽糊精6-8份、矫味剂0.01-0.05份;
所述中药辅料为皂角、生姜、甘草的混合物,所述皂角、生姜、甘草的质量比为(1-3):(1-3):(1-3);
所述矫味剂为山梨醇、甜蜜素、阿斯巴甜的混合物,所述山梨醇、甜蜜素、阿斯巴甜的质量比为(1-3):(1-3):(1-3)。
