本发明涉及一种猪繁殖与呼吸综合征糖玻璃活疫苗的备方法,其特征在于包括以下步骤:将PRRS病毒液与耐热配方T28C‑5等体积混匀后,分装于西林瓶中,1ml/瓶,进行梯度真空泡沫干燥;本发明所述方法制备的猪繁殖与呼吸综合征糖玻璃活疫苗,活性强,玻璃态的疫苗水分含量低,分子运动受限制,大大减缓了病毒的衰变。

技术领域
本发明涉及一种猪繁殖与呼吸综合征糖玻璃活疫苗的制备方法。
背景技术
兽用活疫苗通常是疫病防控的有力手段之一,但由于我国地区差异大,“冷链”的缺失往往会造成疫苗免疫失败。目前国内真空步冻干燥目前国内减毒活疫苗主要采用真空冷冻干燥工艺制备,其在预冻阶段形成的冰晶颗粒对病毒有一定的损伤,并且疫苗的保存与运输均需要冷链支持。
早在上世纪90年代,国外研究学者发现用真空梯度干燥工艺制备的泡沫干燥疫苗可以显著提高耐热性能,此种方式的原理是将疫苗包埋于黏度较高的非还原糖中,随着水分的失去而逐步固相化,形成一种固态的糖玻璃疫苗,由于非还原糖非常稳定,因此包裹疫苗的糖玻璃不会参与化学反应,也不会参与蛋白质反应,制备的疫苗呈现出致密的干燥泡沫状。
1999年,英国Worrall选择海藻糖作为保护剂分别对副粘病毒、支原体等进行梯度真空干燥,通过测定病毒滴度下降1.0g,牛瘟病毒能够在45℃保存3d、37℃保存14 d反刍兽疫疫苗也可以在37℃保存7 d、25℃保存14 d M12006年,美国Vu Truong—Le采用40%蔗糖溶液对流感病毒进行了冷冻泡沫干燥,可在25℃保存2年、37℃保存146d。11年SatoshiOhtake用含25%海藻糖的保护剂溶液对重组肠炎沙门氏菌进行了泡沫干燥,37℃和25℃保存12周,细菌CFU基本无损失;Worrall使用含2.5%海藻糖的保护剂制备牛反刍兽疫疫苗,在45℃下保存14d效价损失很小。
发明内容
本发明提出一种猪繁殖与呼吸综合征糖玻璃活疫苗的制备方法。
一种猪繁殖与呼吸综合征糖玻璃活疫苗的制备方法,包括以下步骤:
(1)将PRRS病毒液与耐热配方T28C-5等体积混匀后,分装于西林瓶中,1ml/瓶,进行梯度真空泡沫干燥;
(2)设置隔板15-35℃,冷确至零下20-60℃;
(3)样品入舱后温度平衡至25℃后开启真空,初始真空60 000 Pa,之后每2 min调节次真空,最终调至300 Pa维持约1 h,
(4)待样品温度升至25℃后,真空度再降至10 Pa,维持12~18 h,之后真空降至0.1Pa,维持12~18 h整个干燥过程共计30~42 h;
(5)干燥结束,样品出舱轧盖、保存。
所述隔板温度为25℃,冷却至零下40℃。
所述真空度维持16h。
所述真空降至0.1 Pa后维持18h。
本发明所述方法制备的猪繁殖与呼吸综合征糖玻璃活疫苗,活性强,玻璃态的疫苗水分含量低,分子运动受限制,大大减缓了病毒的衰变。
