Abstract

不溶性锌离子源诸如氧化锌也已公开用于洁齿剂中,例如参见美国专利中请公布2012/0207686(“‘686公布”)。然而,不溶性锌盐的使用通常由于可用于递送至牙齿表面的锌离子水平较低而受限,并且当使用较高水平的锌时有不希望的消费者涩味。
希望提供一种在生物膜、菌斑、口腔炎症等的预防和/或减少方面表现出比先前已知的组合物甚至更大的功效的口腔护理组合物。
简述
本发明涉及一种口腔护理组合物,例如洁齿剂组合物,所述组合物可有效地将亚锡离子源、氟离子源和不溶性锌离子源在低水分、优选单相的系统中合并,所述系统将所述离子有效地递送至口腔。已发现亚锡离子和锌离子有助于减少/预防口腔炎症诸如牙龈炎和牙周炎、菌斑形成、口腔生物膜形成、牙齿敏感性,并且提高呼吸益处。
本发明涉及一种口腔护理组合物,所述组合物包含:
(a)口腔可接受的载体;
(b)氟离子源;
(c)亚锡离子源;
(d)不溶性锌离子源;
(e)包含柠檬酸的缓冲系统,其中柠檬酸的量为所述组合物的至少1重量%,并且
其中所述组合物的总水含量小于15重量%。

不溶性锌离子源
在一个实施方案中,不溶性锌离子源为不溶性锌离子盐。如本文所用的,术语“不溶性”意指物质或化合物在25℃的水溶液中于pH 7下的溶解度小于0.1重量%(基于组合物的重量)。还将理解的是在较低或较高pH下,不溶性锌离子源可具有显著更大的溶解度。适合的不溶性锌离子源的实例包括氧化锌、磷酸锌、焦磷酸锌、以及其他锌化合物。不溶性锌离子源优选地为氧化锌。

锌离子源的量通常将以0.1重量%至2重量%存在于最终组合物中。在其他实施方案中,锌离子源以0.5%至1.5%或0.9%至1.1%的量存在。
亚锡离子源
本发明的组合物包含亚锡离子源。亚锡离子源可为具有无机或有机抗衡离子的可溶性或不溶性亚锡化合物。实例包括亚锡的氟化物、氯化物、氯氟化物、乙酸盐、六氟锆酸盐、硫酸盐、酒石酸盐、葡糖酸盐、柠檬酸盐、苹果酸盐、甘氨酸盐、焦磷酸盐、偏磷酸盐、草酸盐、磷酸盐、碳酸盐以及氧化物。
含有亚锡盐、尤其是氟化亚锡和氯化亚锡的口腔组合物描述于美国专利号5,004,597。亚锡盐洁齿剂的其他描述可见于美国专利号5,578,293。优选的亚锡盐为氟化亚锡和氯化亚锡二水合物。亚锡盐的其他实例包括乙酸亚锡、酒石酸亚锡和柠檬酸亚锡钠。
通常,可用于递送至口腔表面的可溶性亚锡离子水平越高,功效水平越高。本发明提供了存在于组合物中的来自不溶性亚锡离子源的高水平锌离子。通常,本发明的组合物具有大于50%可溶性亚锡离子,在其他实施方案中,在40℃下老化13周后为大于60%、大于70%、大于80%或大于90%。
亚锡离子源通常将以0.05重量%至5重量%的量存在于最终组合物中。在一个实施方案中,亚锡离子源以0.1%至1%或0.35%至0.55%的量存在。
亚锡离子和锌离子源自口腔护理组合物中的有效量的一种或多种离子源。有效量被定义为从至少1000ppm金属(锌和亚锡)离子、优选2,000ppm至15,000ppm。更优选地,金属离子以3,000ppm至13,000ppm以及甚至更优选地4,000ppm至10,000ppm的量存在。这是存在于组合物中以用于递送至牙齿表面的金属离子(亚锡和锌)的总量。
合并的金属离子源(亚锡和锌)通常将以0.15重量%至7重量%的量存在于最终组合物中。优选地,金属离子源以0.6%至2.5%、更优选1.25%至1.65%的量存在。
氟离子源
本文中氟离子源为能够提供游离氟离子的氟源。可溶性氟离子源包括氟化钠、氟化亚锡、氟化铟、氟化锌、以及单氟磷酸钠。氟化钠和氟化亚锡为优选的可溶性氟离子源。美国专利号2,946,725和美国专利号3,678,154公开了此类氟离子源以及其他氟离子源。
本发明组合物中的氟离子源在储存期间、在由消费者进行组合物的实际刷牙使用之前优选地作为组合物中的固体分散体存在。水在本发明组合物中的水平太低,不允许氟源在储存期间溶解在组合物中。因此,在储存期间使氟离子与其他成分诸如多磷酸盐、或二氧化硅(如果存在的话)之间的相互作用最小化,以提供在储存期间稳定的组合物。当组合物在刷牙时被唾液和/或水接触时,氟源优选地将分散并且活性离子将被递送到口腔。
本发明组合物可含有能够提供50ppm至3500ppm、以及优选500ppm至3000ppm的游离氟离子的可溶性氟离子源。为递送希望的量的氟离子,氟离子源可以总组合物的0.1重量%至5重量%、在一个实施方案中0.2重量%至1重量%、以及甚至0.3重量%至0.6重量%的量存在于总口腔护理组合物中。
缓冲系统
本文所述的组合物还含有包含柠檬酸的缓冲系统。令人意外地发现,在40℃下老化8或13周后,柠檬酸大于口腔护理组合物的0.6重量%的量将使得显著更多的锌离子和或亚锡离子存在于所述组合物中,它们来自组合物中的对应锌离子源和/或亚锡离子源。在一些实施方案中,柠檬酸的量为口腔护理组合物的0.5重量%-4重量%、或1重量%-3重量%、或1.1重量%-1.5重量%、或1.8重量%-2.2重量%、或1.3重量%、或2重量%。在一个实施方案中,缓冲系统包含柠檬酸和柠檬酸钠,尤其是柠檬酸三钠。如本文所用的缓冲剂是指可用于将组合物的pH调整至范围pH 3.0至pH 10、更典型地6至8的试剂。
除柠檬酸之外,可任选存在的其他缓冲剂包括碱金属氢氧化物、氢氧化铵、有机铵化合物、碳酸盐、倍半碳酸盐、硼酸盐、硅酸盐、磷酸盐、咪唑、以及混合物。特定缓冲剂包括磷酸一钠、磷酸三钠、苯甲酸钠、苯甲酸、氢氧化钠、氢氧化钾、碱金属碳酸盐、碳酸钠、咪唑、以及焦磷酸盐。缓冲剂的总量通常为本发明组合物的0.1重量%至30重量%、或0.1重量%至10重量%、或0.3重量%至3重量%。
总水含量
用于制备商业上适合的口腔组合物的水应优选地具有低离子含量并且不含有机杂质。在洁齿剂组合物中,水通常将构成本文的组合物的小于20重量%、或小于15重量%、或小于15重量%至大于10重量%。水的量包括添加的游离水和与其他物质诸如二氧化硅、表面活性剂溶液和/或颜色溶液一起引入的游离水。
口腔可接受的载体
在某些实施方案中,本发明提供了包含口腔可接受载体或赋形剂的组合物。如本文所用的,“口腔可接受载体”和“口腔可接受赋形剂”可互换使用,并且是指能安全用于本发明组合物中、与适合的益处/风险比率一致的物质或物质组合,它们可与其他成分相关同时保留显著的临床功效。此类载体材料应被选择为与所述组合物的其他成分相容,并且优选地基本上并不减小其他成分的功效。特定载体组分的选择依赖于所希望的产品形式,包括洁齿剂、清洗剂、凝胶以及涂剂。

如本文所用的,“粘附剂”为增强成分到它所应用的口腔表面的停留的物质或物质组合。此类粘附剂包括而不限于:粘合剂、成膜物和粘度增强剂;例如,亲水性有机聚合物、疏水性有机聚合物、聚硅氧烷胶、硅酮粘合剂、二氧化硅、以及其组合。在一个实施方案中,粘附剂是平均分子量(M.W.)为30,000至1,000,000,例如300,000至800,000的甲基乙烯基醚/马来酸酐共聚物。聚甲基乙烯基醚-马来酸酐共聚物的制备特别陈述于美国专利号5,047,490和4,627,977。聚甲基乙烯基醚马来酸酐共聚物也以商品名AN从ISP(Ashland),Wayne,N.J商购获得。
增稠剂
本发明的洁齿剂组合物可掺入一种或多种增稠剂,它们可用作粘合剂系统。增稠剂的总量通常为口腔护理组合物的0重量%至3重量%、或0.1重量%至1.5重量%、或0.2重量%%-0.5重量%。
一种此类增稠剂为聚乙烯吡咯烷酮(PVP),特别是交联PVP,任选地与树胶诸如黄原酸胶组合。其他增稠剂包括多糖、卡波姆、泊洛沙姆、改性纤维素(例如,羧甲基纤维素(CMC))以及其混合物。在一些实施方案中,这些增稠剂向低水分口腔护理组合物提供希望的稠度和胶凝。先前已知当水不存在来活化其胶凝结合特性时,与水和保湿剂一起提供希望的流变性的胶凝物质通常提供不太令人满意的流变性。这被认为对于甘油保湿剂而言尤其属实。增稠剂还可包括另外的无机增稠剂。
在本文中适用作增稠剂的多糖(包括树胶)包括角叉菜胶、结冷胶、刺槐豆胶、黄原酸胶、以及其混合物。角叉菜胶为源自海藻的多糖并且已知在牙膏中用作粘合剂或增稠剂,参见,例如,美国专利号6,187,293 B1和6,162,418。存在若干种角叉菜胶,它们可由其海藻来源和/或其硫酸化的程度和位置来区分。在本发明中适用的是κ角叉菜胶、改性κ角叉菜胶、i角叉菜胶、改性i角叉菜胶、以及其混合物。本文中适用的角叉菜胶包括从FMC公司以系列名称“Viscarin”商购的那些,包括但不限于Viscarin TP 329、Viscarin TP 388、以及Viscarin TP 389。
结冷胶为适用于本文中的另一种多糖。它是通过伊乐假单胞菌(pseudomonaselodea)有氧发酵的多糖。当它以0.1%至3%、优选0.4%至1.8%(w/w)的水平存在时,它也可形成可接受的低水分基质。
刺槐豆胶和黄原酸胶也为本文中适用的适合多糖。当组合物中水的水平低于10%时,作为增稠剂的刺槐豆胶或黄原酸胶可形成稳定且可接受的洁齿剂基质。
泊洛沙姆也适合用作在本文的低水分基质中的增稠剂。泊洛沙姆为合成的环氧乙烷和环氧丙烷的嵌段共聚物。它以多种类型可用。在本文中,泊洛沙姆407为优选的。它可部分溶解在水中。当温度高于65℃时,它可溶解在甘油中。POLOXAMER例如可获自BASFCORPORATION,New Jersey,USA。
卡波姆也适合用作在低水分基质,尤其是无水基质中的增稠剂。
改性纤维素诸如羟乙基纤维素和CMC也是低水分基质中的良好增稠剂。由于水的水平在本发明组合物中为有限的,具有疏水性链(C12-C20)的改性羟乙基纤维素为优选的,以增加此增稠剂在其他低极性溶剂诸如甘油、丙二醇和聚乙二醇(PEG)中的溶解度和水合作用。
增稠剂可包括无机增稠剂诸如胶状硅酸铝镁或细碎二氧化硅,以进一步改进纹理。另外的无机增稠剂(如果存在的话)可以口腔护理组合物的0.1重量%至15重量%、更优选地0.1重量%至5重量%的量使用。
保湿剂
保湿剂用于保持牙膏组合物以免于在暴露于空气后硬化并且某些保湿剂还可赋予牙膏组合物希望的甜味。用于本发明中的适合保湿剂包括甘油、山梨醇、聚乙二醇、丙二醇、木糖醇、以及其他可食用的多元醇类。优选的是甘油、聚乙二醇、聚丙二醇、以及其混合物,尤其是其混合物。保湿剂通常构成所述组合物的0.1重量%至70重量%、优选1重量%至60重量%、以及更优选15重量%至55重量%。
认为保湿剂对低水分基质的粘度具有显著影响。例如,当使用多糖作为组合物中的增稠剂时,当甘油或聚乙二醇的水平增加时,所述基质的粘度将增加。相反,当组合物中的丙二醇水平增加时,基质的粘度将减小。
抗齿垢剂
在本发明的不同实施方案中,口腔护理组合物可包含抗齿垢剂(牙垢控制剂)。通常,牙垢控制剂被归类为与一些增白剂不相容,但是本发明的实施方案将牙垢控制剂和增白剂并入在单相增白组合物。适合的抗齿垢剂包括但不限于,磷酸盐和多磷酸盐(例如,焦磷酸盐和三聚磷酸盐)、聚氨基丙磺酸(AMPS)、六偏磷酸盐、三水合柠檬酸锌、多肽(例如,聚天门冬氨酸和聚谷氨酸)、聚烯烃磺酸盐、聚烯烃磷酸盐、以及二磷酸盐(例如,EHDP;AHP)。口腔组合物可包含不同抗齿垢剂的混合物。在一个优选的实施方案中,使用焦磷酸四钠(TSPP)、三聚磷酸钠(STPP)、焦磷酸四钾(TKPP)、或其混合物。抗齿垢剂可构成0.1重量%至30%或0.1重量%至15重量%、或1重量%至10重量%、或2重量%至6重量%。
研磨剂
在牙膏组合物中还可包含研磨抛光物质。本发明的组合物中预期使用的研磨抛光物质可为并不过分磨损牙质的任何物质。典型的研磨抛光物质包括二氧化硅,包括凝胶和沉淀;氧化铝;磷酸盐,包括正磷酸盐、聚偏磷酸盐和焦磷酸盐;以及其混合物。特定实例包括正磷酸二钙二水合物、焦磷酸钙、磷酸三钙、聚偏磷酸钙、不溶性聚偏磷酸钠、水合氧化铝、β焦磷酸钙、碳酸钙、以及树脂性研磨物质(例如尿素和甲醛的颗粒状缩合产品)、以及其他实例,诸如在美国专利号3,070,510中公开的。还可使用研磨剂的混合物。如果洁齿剂组合物或特定相包含平均链长度为4或更大的多磷酸盐,则含钙研磨剂和氧化铝不是优选的研磨剂。最优选的研磨剂为二氧化硅。
各种类型的二氧化硅牙齿研磨剂为优选的,因为它们具有优异牙齿清洁和抛光性能的独特益处而不过度磨损牙釉或牙质。本文中的二氧化硅研磨抛光物质以及其他研磨剂通常具有范围在0.1至30微米之间并且优选在5至15微米之间的平均粒度。研磨剂可为沉淀的二氧化硅或二氧化硅凝胶,诸如美国专利号3,538,230和美国专利号3,862,307中所述的二氧化硅干凝胶,优选的是由W.R.Grace&Company,Davison Chemical Division以商品名销售的二氧化硅干凝胶。还优选的是沉淀的二氧化硅物质,诸如由Huber公司以商品名销售的那些,特别是携带标识“Zeodent 119”的二氧化硅。适用于本发明的牙膏中的二氧化硅牙齿研磨剂的类型更详细地描述于美国专利号4,340,583中。二氧化硅研磨剂也描述于美国专利号5,589,160;5,603,920;5,651,958;5,658,553;以及5,716,601。在本文描述的口腔护理组合物中的研磨剂通常以所述组合物的6重量%至70重量%的水平存在。优选地,牙膏含有所述组合物的10重量%至50重量%的研磨剂。
过氧化物源
本发明可在所述组合物中包含过氧化物源。过氧化物源选自由以下组成的组:过氧化氢、过氧化钙、过氧化脲、以及其混合物。优选的过氧化物源为过氧化钙。以下量表示过氧化物原料的量,尽管过氧化物源可含有除过氧化物原料之外的成分。本发明组合物可含有所述洁齿剂组合物的0.01重量%至10重量%、优选0.1重量%至5重量%、更优选0.2重量%至3重量%、以及最优选0.3重量%至0.8重量%的过氧化物源。
碱金属碳酸氢盐
所述组合物还可包含碱金属碳酸氢盐。碱金属碳酸氢盐可溶于水中并且除非加以稳定,否则倾向于在水性体系中释放二氧化碳。碳酸氢钠也称为小苏打,是优选的碱金属碳酸氢盐。碱金属碳酸氢盐也用作缓冲剂。本发明组合物可含有所述洁齿剂组合物的0.5重量%至50重量%、优选0.5重量%至30重量%、更优选2重量%至20重量%、以及最优选5重量%至18重量%的碱金属碳酸氢盐。
表面活性剂
所述组合物还可包含表面活性剂,通常也称为起泡剂。适合的表面活性剂为合理稳定且在整个宽pH范围内发泡。表面活性剂为阴离子型、非离子型、两性型、两性离子型、阳离子型、或其混合物。本文中有用的阴离子型表面活性剂包括烷基中具有8至20个碳原子的烷基硫酸盐的水溶性盐(例如,烷基硫酸钠)和具有8至20个碳原子的脂肪酸的磺化甘油单酯的水溶性盐。十二烷基硫酸钠和椰子基单甘油酯磺酸钠为这种类型的阴离子型表面活性剂的实例。其他适合的阴离子型表面活性剂为肌氨酸盐(例如月桂酰肌氨酸钠)、牛磺酸盐、月桂基磺基乙酸钠、月桂酰基羟乙磺酸钠、月桂基醚羧酸钠、以及十二烷基苯磺酸钠。还可使用阴离子型表面活性剂的混合物。许多适合的阴离子型表面活性剂公开于美国专利号3,959,458。
所述组合物中可用的非离子型表面活性剂可广泛地被定义为通过将环氧烷基(在自然界中为亲水性的)与在自然界中可为脂族或烷基芳基的有机疏水性化合物缩合产生的化合物。适合非离子型表面活性剂的实例包括泊洛沙姆(以商品名销售)、聚氧乙烯、聚氧乙烯山梨糖醇酐酯(以商品名销售)、聚乙二醇40氢化蓖麻油、脂肪醇乙氧基化物、烷基酚类的聚环氧乙烷缩合物、由环氧乙烷与环氧丙烷和乙烯二胺的反应产物缩合产生的产物、脂肪醇类的环氧乙烷缩合物、长链叔胺氧化物、长链叔膦氧化物、长链二烷基亚砜、以及此类物质的混合物。适用于本发明中的两性表面活性剂可广泛描述为脂肪族仲胺和叔胺的衍生物,其中脂肪族基团可为直链或支链的并且其中脂肪族取代基之一含有8至18个碳原子并且一个取代基含有阴离子水溶性基团,例如羧酸盐、磺酸盐、硫酸盐、磷酸盐、或膦酸盐。其他适合的两性表面活性剂为甜菜碱,确切的说为椰油酰胺丙基甜菜碱。还可使用两性表面活性剂的混合物。许多这些适合的非阴离子型表面活性剂和两性表面活性剂公开于美国专利号4,051,234。本发明组合物通常包含一种或多种表面活性剂,其各自的水平为所述组合物的0.25重量%至12重量%、优选0.5重量%至8重量%、以及最优选1重量%至6重量%。
在一个实施方案中,口腔护理组合物包含表面活性剂系统,其包含十二烷基硫酸钠(SLS)、椰油酰胺丙基甜菜碱或其混合物。
抗微生物剂
本发明组合物还可包含抗微生物剂。在此类试剂包括水不溶性非阳离子抗微生物剂,诸如卤化二苯醚、酚类化合物(包括酚及其同系物)、单烷基和多烷基以及芳族卤代酚、间苯二酚及其衍生物、双酚化合物和卤化N-水杨酰基苯胺、苯甲酸酯、以及卤化碳酰替苯胺、多酚和草药。水溶性抗微生物剂包括季铵盐和双-二胍盐、以及其他抗微生物剂。三氯生一磷酸盐为另一种优选的水溶性抗微生物剂。季铵试剂包括其中季氮上的一个或两个取代基具有8至20、通常10至18个碳原子的碳链长度(通常为烷基),而其余取代基(通常为烷基或苄基)具有较低数目的碳原子,诸如1至7个碳原子,通常为甲基或乙基。十二烷基三甲基溴化铵、十四烷基氯化吡啶鎓、溴化度米芬、N-十四烷基-4-乙基氯化吡啶鎓、十二烷基二甲基(2-苯氧基乙基)溴化铵、苄基二甲基硬脂基氯化铵、氯化十六烷基铵基吡啶、季铵化5-氨基-1,3-二(2-乙基-己基)-5-甲基六氢嘧啶、苯扎氯铵、苄索氯铵以及甲基苄索氯铵为典型季铵类抗菌剂的实例。其他化合物为双[4-(R-氨基)-1-吡啶嗡]链烷烃,如在1980年6月3日提交的Bailey的美国专利号4,206,215所公开的。
还可包括其他抗微生物剂,诸如双甘氨酸铜、甘氨酸铜、柠檬酸锌、以及乳酸锌。同样适用的是酶,包括内切糖苷酶、木瓜蛋白酶、葡聚糖酶、变异酶(mutanase)、以及其混合物。此类试剂公开于美国专利号2,946,725和美国专利号4,051,234中。特定抗微生物剂包括氯己定、三氯生、三氯生单磷酸盐、以及调味油(诸如百里酚)。三氯生为用于包含在本发明组合物中的优选抗微生物剂。三氯生和这种类型的其他试剂公开于美国专利号5,015,466和美国专利号4,894,220。这些试剂以所述组合物的0.01重量%至1.5重量%的水平存在。
二氧化钛也可加入到本发明组合物中。二氧化钛为增加组合物的不透明度的白色粉末。二氧化钛通常构成所述组合物的0.25重量%至5重量%。
着色剂也可加入到本发明组合物中。着色剂可处于水性溶液形式中,优选地水溶液中的1%着色剂。着色溶液通常构成所述组合物的0.01重量%至5重量%。
调味系统也可加入到所述组合物中。适合的调味组分包括冬青油、薄荷油、留兰香油、丁香花油、薄荷醇、茴香脑、水杨酸甲酯、桉叶油素、肉桂皮、1-乙酸酯、鼠尾草、丁子香酚、欧芹油、羟苯基丁酮、α-紫罗酮、甘牛至草、柠檬、橙、丙烯基乙基愈创木酚(propenylguaethol)、肉桂、香草醛、乙基香草醛、天芥菜精、4-顺式-庚烯醛、联乙酰、甲基-对-叔-丁基苯基乙酸酯、以及其混合物。着色剂也可为调味系统的一部分。本发明组合物中优选的着色剂为对薄荷烷羧基酰胺剂,例如N-乙基-对-薄荷烷-3-甲酰胺(在商业上称为“WS-3”)以及其混合物。调味系统通常以所述组合物的0.001重量%至5重量%的水平用于所述组合物中。
甜味剂可加入到所述组合物中。这些甜味剂包括糖精、葡萄糖、蔗糖、乳糖、木糖醇、麦芽糖、左旋糖、阿斯巴甜、环氨酸钠、D-色氨酸、二氢查耳酮、丁磺氨、以及其混合物。不同着色剂也可并入到本发明中。甜味剂和着色剂通常以所述组合物的0.005重量%至5重量%的水平用于牙膏中。
草药试剂包括但不限于黄连提取物、金银花提取物、以及其混合物,所述草药试剂也可以0.01%至0.05%的水平存在于本文中的组合物中。认为此类草药试剂提供抗菌功效。多酚类也可以0.01%至2%的水平包含在内。一种优选的多酚为茶多酚。
有效量的脱敏剂也可并入到本发明组合物中。脱敏剂包括选自碱金属盐与第II族金属或铝或阻塞小管的可聚合单体的氯化物、硝酸盐、硫酸盐或乙酸盐,碱金属硝酸盐或硝酸铵、草酸铵、柠檬酸以及柠檬酸钠的那些。优选的盐为硝酸钾、柠檬酸钾、以及其混合物。此类脱敏剂公开于例如美国专利号5,718,885。
减污剂诸如Plasdone S-630或氢氧化铝也可加入到组合物中。Plasdone为可通过将乙烯基吡咯烷酮聚合来合成的聚乙烯吡咯烷酮(PVP)。在商业上,它被生产为平均分子量范围10,000至700,000的一系列产品。在本文中,优选低分子量和中等分子量(从10,000至100,000)。为了有效去除污物,PVP的水平优选为0.5%至10%、更优选为1.0%至7.0%、以及甚至更优选为1.5%至5.0%。
本发明的组合物任选地包含营养素。适合的营养素包括维生素、矿物质、氨基酸(L-精氨酸)、以及其混合物。维生素包括维生素C和D、硫胺、核黄素、泛酸钙、烟酸、叶酸、烟酰胺、吡哆醇、氰钴素、对氨基苯甲酸、生物黄酮类、以及其混合物。营养补充剂包括氨基酸(例如,L-色氨酸、L-赖氨酸、蛋氨酸、苏氨酸、左卡尼汀以及L-肉碱)、抗脂肪肝剂(例如,胆碱、肌醇、甜菜碱、以及亚油酸)、以及其混合物。
应理解,虽然以上类别物质中每个的一般属性可能不同,但是可存在一些共有属性并且在两种或更多种这些类别的物质中任何给定物质可用于多种目的。所述组合物中的所有成分均可具有除了其主要功能之外的功能,并且可有助于所述组合物的总体特征,包括其稳定性、功效、稠度、口感、味道、气味等。优选地,载体成分被选择为与所述组合物的其他成分相容。
本发明的一种类型的产品形式为洁齿剂。术语“洁齿剂”通常是指用于清洁口腔表面的制剂。所述洁齿剂为一种口腔组合物,其不能故意吞咽用于全身施用治疗剂的目的,但是应用于口腔,以用于治疗口腔并且然后吐出。通常,洁齿剂与清洁工具诸如牙刷结合使用,通常通过将它施加于牙刷的刷毛并且然后刷洗口腔的可接近表面。优选地,洁齿剂为糊剂或凝胶(或其组合)的形式。
本发明的口腔护理组合物通常在单相中。然而,在一些实施方案中,组分为物理分离的,例如通过使用双隔室包装来分离。洁齿剂组合物可为糊剂、凝胶或任何构型或其组合。本发明的另一个实施方案包括双相或多相组合物,其包含本发明低水分组合物作为一相并且包含含有另外的洁齿剂组分以进一步增强洁齿剂产品的稳定性、性能和/或美学的至少一个另一单独相。例如,双相组合物可包含含有本发明低水分组合物与多磷酸盐和一种或多种离子活性剂的第一相和含有另外的活性剂的单独的第二相,所述另外的活性剂诸如漂白剂,优选为过氧化物源或牙齿表面调节剂,以提供改进的清洁、增白、抗污和口感益处。牙齿调节剂的实例为聚硅氧烷和改性聚硅氧烷,包括二有机基聚硅氧烷,诸如聚二甲基硅氧烷(PDMS);烷基-和烷氧基-聚二甲基硅氧烷共聚醇,诸如C12至C20烷基聚二甲基硅氧烷共聚醇;以及氨基烷基硅氧烷。这些硅氧烷聚合物描述于例如美国专利号5,759,523;6,024,891;6,123,950;6,019,962;以及6,139,823中。
洁齿剂组合物的分配器可为管、泵、或适用于分配牙膏的任何其他容器。在双相口腔组合物中,每种口腔组合物将包含分配器物理上分开的隔室中并且并排地分配。
使用方法
在实施方案的实践中,使用者仅需要将本文中的口腔护理组合物应用于人类或动物的牙齿表面希望的区域,以便获得希望的作用,例如减少或预防菌斑、增白、呼吸清新、预防龋病、减轻疼痛、维持牙龈健康、控制牙垢、控制侵蚀等等。洁齿剂控制牙齿表面侵蚀或预防矿物质脱去的用途为已知的并且描述于例如美国专利号6,685,920中。所述组合物也可应用于其他口腔表面,诸如齿龈和粘膜组织,尽管认为当洁齿剂应用于牙齿时获得最佳益处。洁齿剂组合物可直接或间接接触牙齿和/或口腔表面,然而,优选的是洁齿剂组合物直接应用。洁齿剂组合物可通过任何方式应用,但优选地使用牙刷或通过用洁齿剂浆液冲洗来应用。所述应用可为至少一天一次,尽管每天多至五次可为优选的,并且可持续一段持续时间来进行,所述持续时间诸如一周、多至一年、多至三年或持续一生
本发明的口腔组合物的制造可通过用于产生此类组合物的任何各种标准技术来实现。例如,用于洁齿剂的制备方法为已熟知的,例如,如美国专利号3,966,863;美国专利号3,980,767;美国专利号4,328,205;以及美国专利号4,358,437所述的。为了制备洁齿剂,可制备含有保湿剂(如甘油、丙三醇、山梨醇、以及丙二醇中的一种或多种)、增稠剂和抗菌剂(例如,三氯生)的媒介物,并且加入所述媒介物以及阴离子表面活性剂和两性表面活性剂的混合物,接着将抛光剂以及氟盐与预混合物共混。最后,掺混调味剂并将pH调节至6.8至7.0之间。

以下实施例进一步说明了优选实施方案,但是应理解本发明并不限于这些实施方案。
实施例
实施例1
程序
使用式A所指示的成分以及根据表1和表2中所指示地改变的量的CMC和柠檬酸、TKPP、SAPP和STPP来制备四种制剂。
测定所述制剂中的水溶性锡和水溶性锌的技术为通过原子吸收光谱法(FAAS)。可使用装备有空气乙炔燃烧器和氘背景校正的适合的双光束原子吸收光谱仪,例如,VarianSpectrAA 220FS。而且,可使用适合的锌空心阴极灯和AA级乙炔。组合物中的水溶性锌或锡可在将样品稀释在水中通过FAAS进行测量并与锌或锡标准物比较。将一部分制剂用水分散,将溶液转移到容量瓶中,将其用水稀释至体积并离心作用。通过原子吸收光谱法检测悬浮液中的锡或锌并通过将样品溶液的吸光度与已知锡或锌浓度的外部校准标准物的吸光度比较来进行定量。为了测定总锡或锌,不将样品分散在水中,而是在蒸汽浴上用适合的酸性混合物,例如盐酸、硝酸和/或氢氟酸消化样品。
将配制品在40℃下老化13周。在不同时间点测定总锡、总锌、可溶性锡和可溶性锌水平。

结果
比较性实施例-使用包含可溶性锌源(2重量%柠檬酸)和0.454重量%氟化亚锡与0.6重量%柠檬酸/3.0重量%柠檬酸三钠的配制品更早起作用。此配制品的可溶性锌的初始量仅0.39重量%,所述量在老化12周后轻微减少至0.34重量%。亚锡的量从其初始溶解度到12周老化时间段结束保持稳定(范围为0.31重量%-0.36重量%)。
令人意外的是,具有或不具有CMC且具有相同量(0.454重量%)的氟化亚锡和较少量(1重量%)的难溶性锌源(ZnO)的配制品当使用大于1重量%柠檬酸(范围为0.62重量%至0.69重量%)时显示更大初始溶解度,并且尽管老化13周仍维持此更高水平的溶解度(0.55重量%至0.63重量%)。
另外,当柠檬酸的量增加至2%时,可溶性锡的初始量和老化13周后的量(0.32重量%至0.35重量%)相当于比较性实施例中测量的范围。
本领域技术人员将了解的是,在不背离本发明的精神的情况下,可对本文所述的实施方案做出许多改变和修改。这意味着所有这些变化均属于所附权利要求书的范围内。
Claims (21)
1.一种口腔护理组合物,其在单相中包含:
(a)口腔可接受的媒介物;
(b)氟离子源;
(c)亚锡离子源;
(d)不溶性锌离子源;
(e)包含柠檬酸的缓冲系统,其中柠檬酸的量为所述组合物的至少1重量%,并且
其中所述组合物的总水含量基于所述组合物的重量小于15重量%。
2.如权利要求1所述的组合物,其中柠檬酸的量基于所述组合物的重量为0.5重量%-4重量%、或1重量%-3重量%、或1.1重量%-1.5重量%、或1.8重量%-2.2重量%。
3.如任一项前述权利要求所述的组合物,其中柠檬酸的量基于所述组合物的重量为1重量%-3重量%。
4.如任一项前述权利要求所述的组合物,其中柠檬酸的量基于所述组合物的重量为1.1重量%-1.5重量%。
5.如权利要求1-3中任一项所述的组合物,其中柠檬酸的量基于所述组合物的重量为1.8重量%-2.2重量%。
6.如任一项前述权利要求所述的组合物,其中所述不溶性锌离子源包括有机酸的锌盐。
7.如权利要求3所述的组合物,其中所述不溶性锌离子源包括氧化锌、磷酸锌、焦磷酸锌或其混合物。
8.如任一项前述权利要求所述的组合物,其中所述氟离子源和所述亚锡离子源包括氟化亚锡。
9.如任一项前述权利要求所述的组合物,其包含至少一种选自由以下组成的组的多磷酸盐:具有等于或小于三个磷原子的无机多磷酸盐。
10.如权利要求6所述的组合物,其中所述至少一种多磷酸盐选自由以下组成的组:焦磷酸四钠、三聚磷酸钠、以及其混合物;其中总的多磷酸盐构成所述组合物的1重量%至10重量%。
11.如任一项前述权利要求所述的组合物,其中所述缓冲系统包含柠檬酸和柠檬酸三钠的混合物。
12.如任一项前述权利要求所述的组合物,其包含聚乙烯吡咯烷酮、树胶或其混合物。
13.如任一项前述权利要求所述的组合物,其包含改性纤维素,诸如羧甲基纤维素。
14.如任一项前述权利要求所述的组合物,其还包含至少一种选自由以下组成的组的保湿剂:甘油、聚乙二醇、丙二醇、以及其混合物。
15.如任一项前述权利要求所述的组合物,其中所述组合物为一种包含以下中的一种或多种的洁齿剂:研磨剂、表面活性剂、发泡剂、维生素、聚合物、增白剂、酶、保湿剂、增稠剂、抗微生物剂、防腐剂、L-精氨酸、调味剂或着色剂。
16.在单相中的如任一项前述权利要求所述的组合物。
17.一种口腔护理组合物,其包含:
(a)0.5重量%-3重量%柠檬酸;
(b)0.5重量%-10重量%柠檬酸三钠;
(c)0.1重量%-5重量%氧化锌;
(d)0.1重量%-5重量%氟化亚锡;
(e)10重量%-50重量%甘油;
(f)1重量%-25重量%丙二醇;
任选地包含以下中的一种或多种:
(g)0.1重量%-12重量%黄原酸胶;
(h)0.25重量%-10重量%聚乙烯吡咯烷酮(PVP);
(i)0.2重量%-1.5重量%羧甲基纤维素(CMC);
(j)5重量%-60重量%二氧化硅(总计);
(k)0.1重量%-10重量%焦磷酸四钠(TSPP);
(l)0重量%-5重量%酸式焦磷酸钠(SAPP);
(m)0.1重量%-10重量%三聚磷酸钠(STPP);
(n)0.1重量%-10重量%十二烷基硫酸钠(SLS);
(o)0.1重量%-10重量%椰油酰胺丙基甜菜碱。
18.如权利要求17所述的口腔护理组合物,其包含:
(a)1.1重量%-1.5重量%或1.8重量%-2.2重量%柠檬酸;
(b)2重量%-4重量%柠檬酸三钠;
(c)0.5重量%-2重量%氧化锌;
(d)0.1重量%-2重量%氟化亚锡;
(e)10重量%-50重量%甘油;
(f)2重量%-10重量%丙二醇;
任选地包含以下中的一种或多种:
(g)0.1重量%-0.5重量%黄原酸胶;
(h)0.25重量%-10重量%聚乙烯吡咯烷酮(PVP);
(i)0.3重量%-0.9重量%羧甲基纤维素(CMC);
(j)10重量%-40重量%二氧化硅(总计);
(k)1重量%-4重量%焦磷酸四钠(TSPP);
(l)0.5重量%-1.5重量%酸式焦磷酸钠(SAPP);
(m)1重量%-4重量%三聚磷酸钠(STPP);

(d)0.4重量%-0.6重量%氟化亚锡;

(g)0.275重量%-0.325重量%黄原酸胶;
