In the present invention, the conditions for the culture activation of lactobacillus in the probiotic are: culturing at 25-40 deg.C for 22-26h under the following conditions: standing and culturing for 12-24h at 25-40 ℃, wherein the conditions of fermentation and culture are as follows: performing intermittent shaking culture at 25-40 deg.C for 48-96 hr at stirring speed of 100r/min and 5min/2 hr; the culture activation conditions of the bifidobacteria in the probiotics are as follows: culturing at 25-40 deg.C for 22-26h under the following conditions: anaerobic culture is carried out for 20-28h at the temperature of 32-40 ℃, and the culture conditions of the seed culture solution are as follows: standing and culturing for 16-30h at 32-40 ℃, wherein the fermentation culture conditions are as follows: 28-40 ℃, the initial pH value is 6.0-7.0, and the fermentation time is 48-96 h.
The culture medium for culture activation of lactobacillus in the probiotics is an MRS solid culture medium, and the primary seed culture medium and the fermentation culture medium are MRS liquid culture media without agar; the culture activation culture medium of the bifidobacteria in the probiotics is a TPY solid culture medium, and the primary seed culture medium and the fermentation culture medium are both TPY liquid culture media without agar.
The specific method of this example is as follows:
1. preparing each probiotic into lyophilized powder
(1) Preparation of culture Medium
The MRS solid culture medium comprises the following components in percentage by weight: 2.0 percent of glucose, 0.2 percent of ammonium citrate, 0.5 percent of sodium acetate, 0.5 percent of dipotassium phosphate, 0.02 percent of manganese sulfate, 0.05 percent of magnesium sulfate, 1.0 percent of peptone, 1.0 percent of beef extract, 0.5 percent of yeast extract, 800.1 percent of ground temperature and 1.5 percent of agar powder (imported), and the pH value is adjusted to 6.0. The liquid culture medium is a solid culture medium formula without agar.
The formula of the TPY solid culture medium is as follows: casein peptone 1%, soybean peptone 0.5%, yeast powder 0.25%, dipotassium hydrogen phosphate 0.2%, MgCl26H2O 0.05.05%, calcium chloride 0.015%, ZnSO 4.7H20.025% of O, 0.01% of ferric chloride, 0.005% of L-cysteine hydrochloride, 1.5% of glucose, 801 mL of tween-801 (1000mL plus 1mL), 1.5% of agar powder (inlet), adjusting the pH value to 6.3-6.6, and sterilizing at 115 ℃ for 20 min. The liquid culture medium is a solid culture medium formula without agar.
(2) Culture of probiotics
Respectively inoculating lactobacillus reuteri LY0032, lactobacillus plantarum ZW0045, lactobacillus gasseri GS0010, lactobacillus acidophilus and lactobacillus rhamnosus into an MRS solid slant culture medium, and culturing at 37 ℃ for 24 h; then transferring the seeds into an MRS liquid culture medium, and culturing for 24h at 37 ℃ to prepare first-grade seed liquid; then inoculating the strain into a fermentation culture medium by using an inoculation amount of 5% to perform high-density culture, wherein the fermentation condition is that the batch shaking is carried out for 72 hours at 37 ℃.
Inoculating Bifidobacterium lactis and Bifidobacterium longum into TPY solid slant culture medium, and culturing at 37 deg.C for 24 hr; then transferring the mixture into a TPY liquid culture medium, and performing static culture at 37 ℃ for 24 hours to prepare a first-grade seed solution; then inoculating the strain into a fermentation culture medium by the inoculation amount of 5 percent for high-density culture, and standing for 72 hours at the fermentation condition of 37 ℃.
(3) Preparation of lyophilized powder
When the density of the thallus in the fermentation liquid reaches 109cfu/mL, collecting the fermentation liquor. Centrifuging to obtain thallus, adding protective agent composed of 10% skimmed milk powder 91%, sucrose 8%, and glycerol 1%, and lyophilizing to obtain thallus powder.
2. Respectively pulverizing the Chinese medicinal materials, sieving to obtain Chinese medicinal powder with particle size less than 50um, weighing the Chinese medicinal powder in proportion, and mixing to obtain Chinese medicinal mixture;
3. taking Lactobacillus reuteri LY0032, Lactobacillus plantarum ZW0045 and Lactobacillus gasseri GS0010, and adding the active bacteria number of 1: 1: 2, adding the mixture into a traditional Chinese medicine mixture, adding auxiliary materials, uniformly mixing, wherein the viable count of three strains of lactobacillus reuteri LY0032, lactobacillus plantarum ZW0045 and lactobacillus gasseri GS0010 accounts for more than 80% of the total viable count, and then taking bifidobacterium lactis, lactobacillus acidophilus, bifidobacterium longum and lactobacillus rhamnosus according to the viable count of 1: 1: 1: 1 to obtain mixed bacterial powder, supplementing the mixed bacterial powder with bifidobacterium lactis, lactobacillus acidophilus, bifidobacterium longum and lactobacillus rhamnosus, and uniformly mixing to obtain a microecological preparation product, wherein the total viable count of the probiotics reaches 2.5 multiplied by 1010cfu/g。

Example 6 animal experiment verification of ethanol-induced mouse gastric mucosal injury model
(1) Procedure of experiment
40 male mice are taken and randomly divided into 4 groups, wherein the 4 groups are respectively named as CK (control group), MX (mice gastric mucosa injury model animal caused by ethanol), J123 (mixed with LY0032, ZW0045 and GS0010 in equal proportion) and J + Y (microecological preparation, J123+ traditional Chinese medicine), and each group of mice are subjected to intragastric administration for 1 time/d and 10 days by intragastric administration with corresponding medicines according to the table 1.

Model success judgment index:
the morphological observation indexes include general conditions such as stomach size, stomach wall thickness, number of folds, color of mucous membrane and bleeding. The normal group of stomach had a reddish mucous membrane with a smooth surface, a lot of mucus attached, thick full-thickness stomach wall, regular folds and high elasticity. The mucous membrane of the model group is pale, the elasticity of the gastric wall is poor, the plica is flat or has disordered trend, the mucous membrane layer becomes thin, a small amount of mucus is attached, and bleeding spots appear on partial gastric mucous membrane.
And (3) detecting the pathological histology: the mucosa is damaged and the tissue falls off in the model group. The control group had intact gastric mucosa and no significant detachment.
Fasting is carried out for 24h before the last administration, intragastric administration is carried out for 0.2 mL/treatment of absolute ethyl alcohol 1h after the administration, and the absolute ethyl alcohol is replaced by normal saline in a control group. Weighing after 3h, dislocating and killing cervical vertebra, picking the whole stomach, cutting along the greater curvature of the stomach, flushing with normal saline, observing the damage condition of the gastric mucosa of the mouse, and fixing with formaldehyde for observing the sliced tissues.
(2) Results of the experiment
The observation result of the gastric mucosa of the mouse is shown in fig. 1, compared with the control group (CK), the gastric mucosa of the model group (MX) is seriously damaged, and the protective effect of the composite bacterial powder group (J123) and the product group (J + Y) on the gastric mucosa is more obvious. The gastric mucosal sectioning results also fully validated this conclusion, as shown in figure 2. The microecological preparation product has a better protection effect on gastric mucosa injury caused by alcohol, namely the composition of the invention has a synergistic effect of the medicine-food homologous traditional Chinese medicine and the probiotics.
Example 7 animal experiment verification of antibiotic induced mouse gastrectasia model
(1) Procedure of experiment
40 male mice are randomly divided into 4 groups, wherein the 4 groups are respectively named as CK (control group), MX (mouse gastrectasia caused by antibiotics model animal), JF (equal proportion of LY0032, ZW0045 and GS0010 mixed), JY (microecological preparation product, JF + traditional Chinese medicine). Each group of mice was administered the corresponding drug for intragastric administration 1 time/day for 10 days. The specific grouping and operation is shown in table 2.

Except for the control group with normal saline, each group is drenched with lincomycin hydrochloride in 1-4 days, the beginning of day 5, the model group continues to drench the stomach with lincomycin hydrochloride, the experimental group is correspondingly intervened, weighing is carried out after 3 hours of last administration, cervical dislocation is killed, the abdominal cavity is opened to take out the whole stomach, and the gastrectasia condition of the mouse is observed.

(2) Results of the experiment
After the test, the bloating condition of the mice is shown in fig. 3, and the data of the bloating degree is shown in table 3. Compared with the model group, the probiotic group and the product group both have the function of relieving the mice gastrectasia caused by antibiotics.

The third part is the research on the application effect of the microecologics
EXAMPLE 8 use of the Microecological preparation (human trial)
In order to verify the efficacy of the microecological preparation in assisting the protection of gastric mucosa and relieving gastrectasia, 10 ginseng and products which are often uncomfortable in stomach are screened for trial administration, and 9 feedback tables are received. After 10 days of trial administration, feedback shows that the formula has good effects on relieving stomachache and gastrectasia, promoting digestion, increasing appetite and the like, and the result is shown in fig. 4.

6. The probiotic formulation according to claim 5, characterized in that it comprises 10 per gram of probiotic formulation9-1011Probiotic bacteria of CFU.
