技术领域
本发明涉及一种速溶茶复方制品及其制备方法,特别涉及含蛹虫草、人参、罗汉果、茶叶等中草药植物,运用生物工程技术制成一种速溶茶的制备方法。
背景技术
中药材是中医药事业传承和发展的物质基础,是关系国计民生的战略性资源。中国具有得天独厚的丰富药物资源,保护中药材资源、发展中药材产业,开发中药材产品对于发展中国特色医药健康产业、促进人类健康事业和生态环境建设的都具有十分重要的意义。
本发明是以蛹虫草、人参、阿胶、肉桂、玉竹、砂仁、葛根、黄精、枸杞、甘草、罗汉果、海藻、苦丁茶、乌龙茶为原料,经粉粹、酶解、膜过滤、浓缩、低温喷雾干燥、配制、制粒、灭菌、包装加工制成的可直接冲泡的速溶茶。富含多种天然物质活性成分,对于提高人体机能具有显著的作用,是一种药食同源的新型健康产品。
蛹虫草(Cordyceps militaris ) 别名:北冬虫夏草,北虫草。蛹虫草含有虫草菌素、虫草多糖、腺苷和N6-[β-(乙酰胺甲酰);虫草环肽A;核苷;麦角甾醇过氧化物。具有刺激免疫球蛋白和抗体的产生,增强机体免疫和抗氧化能力,抗病毒、抗菌、明显抑制肿瘤生长,延缓衰老,抗衰防皱。扶正固本,保护心脏、肝脏,抗痉的作用。其独特药理作用已日益引起药学界的重视。
人参(Panax ginseng C. A. Mey.) 别名: 黄参、血参、人衔、神草。《神农本草经》中就认为,人参有“补五脏、安精神、定魂魄、止惊悸、除邪气、明目开心益智”的功效,“久服轻身延年”。人参含总皂苷约 4%,氨基酸和肽类、含氮有机化合物约占 12%-15%,以及胆碱、烟酸、维生素、微量元素等。
罗汉果(Siraitia grosvenorii)是我国特有的珍贵葫芦科植物,素有良药佳果之称。营养成分:蛋白质7.2%,粗蛋白质9%。葡萄糖、果糖约14%。D-甘露糖(鲜果含量)0.08%。罗汉果甙Ⅳ无甜味。功能主治:清热润肺,止咳,利咽,滑肠通便。用于肺火燥咳,咽痛失音,肠燥便秘。对肠管运动机能有双向调节作用。
阿胶(SHAN DONG DONG-E E-JIAO CO)。含有明胶原、骨胶原、蛋白质及钙、钾、钠、镁、锌等17种元素,所含蛋白质水解后能产生多种必需氨基酸。阿胶善于治疗血虚引起的各种病症,并通过补血起到滋润皮肤的作用,还能调经保胎,增强体质,改善睡眠,健脑益智,延缓衰老,有助于防治包括癌症在内的多种疾病。
肉桂(Cinnamomum cassia)中含有桂皮醛、丁香醛等挥发油,它们具有很强的杀菌作用,可以消灭身体中的某些致病菌,有用于肾阳不足的畏寒、肢冷、腰膝冷痛,亦可用于肾不纳气的虚喘、气逆的防病治病功效。
玉竹(Polygonatum odoratum (Mill.)Druce)含有的维生素A,改善干裂、粗糙的皮肤状况,使之柔软润滑。美容护肤。玉竹多糖体对离体蛙心有强心作用,提高血清溶血素抗体水平,改善脾淋巴细胞对ConA的增殖反应。
砂仁(Fructus Amomi),主要作用于人体的胃、肾和脾,能够行气调味,和胃醒脾,治疗胸脘胀满、腹胀食少等病症。
葛根(Pueraria lobata)淀粉含量在8~10%,每 100克葛根含异黄酮14.82毫克,促进新陈代谢,加强肝脏解毒功能,防止酒精对肝脏的损伤,促进胆汁分泌具有延缓衰老、调节血脂血糖、促进造血功能等方面的作用止脂肪在肝脏堆积。
黄精(Polygonatum sibiricum),又名:鸡头黄精。具有补气养阴,健脾,润肺,益肾功能。用于治疗脾胃虚弱,体倦乏力,口干食少,肺虚燥咳,精血不足,内热消渴等症。对于糖尿病很有疗效。
枸杞(Lycium chincnse Mill. )富含枸杞多糖,枸杞多糖是一种水溶性多糖,由阿拉伯糖、葡萄糖、半乳糖、甘露糖、木糖、鼠李糖这6种单糖成分组成,具有生理活性,能够增强非特异性免疫功能,提高抗病能力,抑制肿瘤生长和细胞突变。
甘草(Glycyrrhiza uralensis Fisch)味甘甜,性平和,入心、脾、肺、胃四经。生用偏凉,可泻火解毒、缓急止痛;炙用偏温,能散表寒、补中益气。此外,甘草还善于调和药性,解百药之毒。
海藻(seaweed)海藻中具有增强免疫力及抗癌活性的物质,属特殊多糖类、蛋白质、脂质、色素及低分子物质,海藻富有食物纤维,这些纤维属硫酸多糖或酸性多糖之类的物质,具有增强免疫力及抗癌的活性;
苦丁茶(Ilex latifolia Thunb.)现代科学临床应用表明,苦丁茶具有消暑解毒、消炎杀菌、化痰止咳、健胃消积、提神醒脑、减肥抗癌和抗辐射、降血压、降血脂、降胆固醇等功效。被国内外消费者誉为保健茶、益寿茶、美容茶,是一种应用极为广泛的天然多功能植物饮料。
乌龙茶(Oolong tea)。茶叶含多酚类、植物碱、蛋白质、氨基酸、维生素、果胶素、有机酸、脂多糖、糖类、酶类、色素等。乌龙茶中的主成分--单宁酸,可以降低血液中的胆固醇含量,有抑制胆固醇上升的效果。具有消除危害美容与健康的活性氧,能改善皮肤过敏,经常饮用能瘦身,抗肿瘤、预防老化功效。
以上述原料,传统做法系采用直接粉碎配伍、或采用溶剂进行简单提取等工艺制成成品、半成品等。大部分中草药的有效成分都被包裹在植物细胞壁内,用一般的溶剂提取难以破坏细胞壁,提取率低,而且影响环境保护。
发明内容
本发明的目的在于提供一种虫草人参复方速溶茶及其制备方法,本发明产品选用天然植物进行配伍制成,无毒副作用,长期服用具有提高免疫力、滋养肝肾等功效;本方法集酶解、膜过滤、浓缩、低温喷雾干燥于一体,比传统的溶剂提取方法提取率更高,比单一的提取方法成分更全面,制成的虫草人参复方速溶茶溶解时间快,营养成分高,是一种高效环保的中草药复方制品生产技术。
为实现上述目的,本发明采用如下技术方案:
一种虫草人参复方速溶茶,主要原料按重量份数计:蛹虫草1500份、人参1500份、阿胶500份、肉桂500份、玉竹500份、砂仁500份、葛根500份、黄精500份、枸杞500份、甘草500份、罗汉果1000份、海藻500份、苦丁茶500份、乌龙茶1000份。
以蛹虫草、人参、阿胶、肉桂、玉竹、砂仁、葛根、黄精、枸杞、甘草、罗汉果、海藻、苦丁茶、乌龙茶为主要原料,经粉粹、酶解、高压均质处理、膜过滤、浓缩、低温喷雾干燥、制粒、灭菌、包装得到速溶茶。包括如下步骤:
(1)将合格原料按配比称量,粉碎,过10-30目筛;
(2)将过筛后的粉料用饮用水浸泡半小时,料液比为1kg:10-15kg,然后加入原料重量0.5-1.0wt%纤维素酶、0.5-1.0wt%果胶酶和0.1-0.5wt%植酸酶,用1-3mol/L的盐酸调节pH为4.5-6.5,50-60℃酶解45-60min;所述的纤维素酶活力为20000U/g,果胶酶活力为30000U/g,植酸酶活力为100000U/g;酶解结束将料液升温至100℃,灭酶3-5min,然后将料液采用80目板框过滤,得酶解滤液备用;
(3)将酶解滤液先经胶体磨均质,旋转磨转速800-1000r/min,再经高压均质机均质,压力为40-60MPa下过200目膜过滤;
(4)将步骤(3)得到的滤液在真空度为0.09MPa下减压浓缩至浓度Brix=20.0-25.0%;
(5)将浓缩液采用低温喷雾干燥制成中草药活性物质干浸膏粉;低温喷雾干燥设备控制参数:水份蒸发率1500-2000mL/h,进风温度控制100-130℃,出风温度控制60-80℃,空气压缩机产气量4.2m3/h,工作压力2-5bar;
(6)将中草药活性物质干浸膏粉采用干法制粒;使用含10wt%羟丙基纤维素的80wt%乙醇溶液作为黏合剂,按干浸膏粉和羟丙基纤维素的重量比为10∶2计量,将干浸膏粉和黏合剂混合均匀,经搅拌、60-80℃烘干、整粒,得到颗粒剂,其含水量为4-5%;
(7)采用颗粒包装机无菌充填包装,包装材料为医用铝膜袋;包装工艺车间温度为18℃-22℃,湿度 50-60%。
本发明的显著优点在于:本发明利用纤维素酶、果胶酶、植酸酶将中草药进行酶解,同时利用低温提取工艺、强化传质等作用,促进酶分子在体系中的扩散,加速酶解进行,也加速植物有效成份的浸出提取。该方法不但提高了提取率,而且将纤维素和活性物质转化为人体易吸收的糖类物质,提高原料的利用价值,使资源充分得到利用。本发明是以传统中草药为原料,以低温酶解、高压均质、真空干燥等集成的制备方法。比传统的提取方法提取率更高,比单一的水提或醇提所得成分更全面,制成的中草药速溶茶营养更充分、食用更便捷。本方法工艺先进,提取得率高,是一种高效环保的生产技术。
具体实施方式
实施例一:一种虫草人参速溶茶复方制品
1、原料组成为:蛹虫草1500g、人参1500g、阿胶500g、肉桂500g、玉竹500g、砂仁500g、葛根500g、黄精500g、枸杞500g、甘草500g、罗汉果1000g、海藻500g、苦丁茶500g、乌龙茶1000g。
2、制备方法:
(1) 将合格原料按配方称量,然后粉碎,过筛,目数为10-30目。
(2) 将过筛后的粉碎粉用饮用水浸泡半小时,料液比为1:10(即每1kg粉碎粉用10kg的水),然后加入原料重量0.5%的纤维素酶,0.5%的果胶酶,0.1%的植酸酶,酶用2mol/L的盐酸调节pH为4.5-6.50,温度为50-60℃条件下,酶解45-60min。所述的纤维素酶活力为20000U/g,果胶酶活力为30000U/g,植酸酶活力为100000U/g。酶解反应后将料液升温至100℃,灭酶3-5分钟,然后将料液采用80目板框过滤,得酶解滤液备用。
(3)高压均质处理:经酶解处理的酶解液先经胶体磨均质,旋转磨转速800-1000r/min。使菌丝体等物料颗粒崩解,达到均质,再经高压均质机(40MPa-60MPa),使酶解液在高压作用下通过200目膜过滤。
(4) 将步骤3所得的酶解高压均质膜过滤滤液进行浓缩,注入低温真空浓缩罐中浓缩,在真空度为0.09MPa下,减压浓缩,浓度Brix=20.0-25.0%,
(5) 采用低温喷雾干燥,获得中草药活性物质干浸膏粉1650 g。提取得率为16.65%。低温喷雾干燥设备控制参数:水份蒸发率1500-2000ml/hour;进风温度控制100℃-130℃;出风温度控制60℃-80℃;空气压缩机 产气量4.2 m3/h,工作压力2-5bar。
(6) 将中草药活性物质干浸膏粉采用干法制粒工艺,使用含10%羟丙基纤维素(L-HPC)和80%乙醇液作为黏合剂,以浸膏粉∶羟丙基纤维素为10∶2的比例置于制粒机搅拌,并经干燥箱烘干,整粒机整粒,即可得到粒度均匀的颗粒,获得高浓度、低辅料量、均匀颗粒剂。 颗粒剂含水量为4-5%。干燥温度60℃-80℃。
(7)包装。采用颗粒包装机无菌充填包装,包装材料为医用铝膜袋。包装工艺车间温度为18℃-22℃,湿度 50-60%。
实施例二:一种虫草人参速溶茶复方制品
1、原料组成为:蛹虫草1500 g、人参1500 g、阿胶500 g、肉桂500 g、玉竹500 g、砂仁500 g、葛根500 g、黄精500 g、枸杞500 g、甘草500 g、罗汉果1000 g、海藻500 g、苦丁茶500 g、乌龙茶1000 g。
2、制备方法:
(1)将合格原料按配方称量,然后粉碎,过筛,目数为10-30目。
(2) 将过筛后的粉碎粉用饮用水浸泡半小时,料液比为1:12.5(即每1kg粉碎粉用12.5kg的水),然后加入原料重量0.75%的纤维素酶,0.75%的果胶酶,0.25%的植酸酶,酶用1.5mol/L的盐酸调节pH为4.5-6.5,温度为50-60℃条件下,酶解45-60min。所述的纤维素酶活力为20000U/g,果胶酶活力为30000U/g,植酸酶活力为100000U/g。酶解反应后将料液升温至100℃,灭酶3-5分钟,然后将料液采用80目板框过滤,得酶解滤液备用。
(3)高压均质处理:经酶解处理的酶解液先经胶体磨均质,旋转磨转速800-1000r/min。使物料颗粒崩解,达到均质,再经高压均质机(40MPa-60MPa),使酶解液在高压作用下通过200目膜过滤。
(4) 将步骤3所得的酶解高压均质膜过滤滤液进行浓缩,注入低温真空浓缩罐中浓缩,在真空度为0.09MPa下,减压浓缩,浓度Brix=20.0-25.0%,
(5) 采用低温喷雾干燥,获得中草药活性物质干浸膏粉1720g。提取得率为17.20%。低温喷雾干燥设备控制参数:水份蒸发率1500-2000ml/hour;进风温度控制100℃-130℃;出风温度控制60℃-80℃;空气压缩机 产气量4.2 m3/h,工作压力2-5bar。
(6) 将中草药活性物质干浸膏粉采用干法制粒工艺,使用含10%羟丙基纤维素(L-HPC)和80%乙醇液作为黏合剂,以浸膏粉∶羟丙基纤维素为10∶2的比例置于制粒机搅拌,并经干燥箱烘干,整粒机整粒,即可得到粒度均匀的颗粒,获得高浓度、低辅料量、均匀颗粒剂。 颗粒剂含水量为4-5%。干燥温度60℃-80℃。
(7)包装。采用颗粒包装机无菌充填包装,包装材料为医用铝膜袋。包装工艺车间温度为18℃-22℃,湿度 50-60%。
实施例三:一种虫草人参速溶茶复方制品
1、原料组成为:蛹虫草1500 g、人参1500 g、阿胶500 g、肉桂500 g、玉竹500 g、砂仁500 g、葛根500 g、黄精500 g、枸杞500 g、甘草500 g、罗汉果1000 g、海藻500 g、苦丁茶500 g、乌龙茶1000 g。
2、制备方法:
(1) 将合格原料按配方称量,然后粉碎,过筛,目数为10-30目。
(2) 将过筛后的粉碎粉用饮用水浸泡半小时,料液比为1:15(即每1kg粉碎粉用15kg的水),然后加入原料重量1.0%的纤维素酶,1.0%的果胶酶,0.5%的植酸酶,酶用1.5mol/L的盐酸调节pH为4.5-6.5,温度为50-60℃条件下,酶解45~60min。所述的纤维素酶活力为20000U/g,果胶酶活力为30000U/g,植酸酶活力为100000U/g。酶解反应后将料液升温至100℃,灭酶3-5分钟,然后将料液采用80目板框过滤,得酶解滤液备用。

实施例1-3制得的虫草人参速溶茶主要成分含量表
上述主要成分检测标准与方法:
1、腺苷含量:采用NY/T2116-2012《虫草制品中虫草素和腺苷的测定高效液相色谱法》。
2、人参皂甙:采用 GB/T 22996-2008《人参中多种人参皂甙含量的测定 液相色谱-紫外检测法》。
3、碳水化合物:采用 GB/28050-2011《食品安全国家标准,预包装食品营养标签通则》。
以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。
Claims (3)
1.一种虫草人参复方速溶茶,其特征在于:主要原料按重量份数计:蛹虫草1500份、人参1500份、阿胶500份、肉桂500份、玉竹500份、砂仁500份、葛根500份、黄精500份、枸杞500份、甘草500份、罗汉果1000份、海藻500份、苦丁茶500份、乌龙茶1000份。
2.一种制备如权利要求1所述的虫草人参复方速溶茶的方法,其特征在于:以蛹虫草、人参、阿胶、肉桂、玉竹、砂仁、葛根、黄精、枸杞、甘草、罗汉果、海藻、苦丁茶、乌龙茶为主要原料,经粉粹、酶解、高压均质处理、膜过滤、浓缩、低温喷雾干燥、制粒、灭菌、包装得到速溶茶。
3.根据权利要求2所述的方法,其特征在于:包括如下步骤:
(1)将合格原料按配比称量,粉碎,过10-30目筛;
(2)将过筛后的粉料用饮用水浸泡半小时,料液比为1kg:10-15kg,然后加入原料重量0.5-1.0wt%纤维素酶、0.5-1.0wt%果胶酶和0.1-0.5wt%植酸酶,用1-3mol/L的盐酸调节pH为4.5-6.5,50-60℃酶解45-60min;所述的纤维素酶活力为20000U/g,果胶酶活力为30000U/g,植酸酶活力为100000U/g;酶解结束将料液升温至100℃,灭酶3-5min,然后将料液采用80目板框过滤,得酶解滤液备用;
(3)将酶解滤液先经胶体磨均质,旋转磨转速800-1000r/min,再经高压均质机均质,压力为40-60MPa下过200目膜过滤;
(4)将步骤(3)得到的滤液在真空度为0.09MPa下减压浓缩至浓度Brix=20.0-25.0%;
(5)将浓缩液采用低温喷雾干燥制成中草药活性物质干浸膏粉;低温喷雾干燥设备控制参数:水份蒸发率1500-2000mL/h,进风温度控制100-130℃,出风温度控制60-80℃,空气压缩机产气量4.2m3/h,工作压力2-5bar;
(6)将中草药活性物质干浸膏粉采用干法制粒;使用含10wt%羟丙基纤维素的80wt%乙醇溶液作为黏合剂,按干浸膏粉和羟丙基纤维素的重量比为10∶2计量,将干浸膏粉和黏合剂混合均匀,经搅拌、60-80℃烘干、整粒,得到颗粒剂,其含水量为4-5%;
(7)采用颗粒包装机无菌充填包装,包装材料为医用铝膜袋;包装工艺车间温度为18℃-22℃,湿度 50-60%。
