Abstract

本发明提供了一种基于CRISPR/Cas9载体系统获得番茄基因组定点敲除突变体的方法,将基因组编辑技术应用在园艺作物番茄上,主要根据目标基因设计靶向序列,将靶向序列插入CRISPR/Cas9的表达载体,并转入番茄植株体内,该载体系统利用靶向序列识别目标基因,Cas9核酸酶对番茄基因组目标基因剪切形成DNA双链断裂,继而在修复过程中产生突变。本发明所构建的番茄CRISPR/Cas9体系简单,能够快速、高效的进行番茄基因组定点修饰。

一种获得番茄基因组定点敲除突变体的方法
技术领域
本发明属于植物分子生物学领域,特别涉及一种番茄基因组定点敲除突变体的方法。
背景技术
近年来,基因组定点修饰技术受到广泛关注,发展迅速。最早实现定点修饰的技术是锌指核酸酶(ZFN)技术,但是该技术成本高、操作难度大。而转录激活因子样效应物核酸酶(TALEN)的出现一定程度上弥补了ZFN技术的缺陷,降低了成本,提高了效率。但是,TALEN技术载体构时要求针对每个碱基都要对应单一模块,载体构建过程繁琐。与TALEN技术相比,CRISPR定点修饰技术更为高效、简单,已经被广泛应用到动植物和微生物研究当中。
在物种基因组信息大量解析的背景下,基因组定点修饰技术CRISPR/Cas的出现,有力的推动了科研优势向产业优势的转化。该技术自2013年报道以来,目前已被成功应用于水稻、大豆、玉米、小麦、拟南芥甚至猴子等动植物的基因修饰,在基因功能研究、优良种质资源开发等领域表现出重要价值,但是目前CRISPR/Cas9体系在园艺作物上报道较少。
番茄是全球重要的蔬果作物,具有较高的营养价值和经济价值,也是研究果实发育的模式作物。研究番茄基因组定点编辑技术对于番茄科研、育种等领域都具有重要价值。因此开发一种获得番茄基因组定点敲除突变体的方法具有重要实际意义和应用价值。
发明内容
本发明的目的是提供一种获得番茄基因组定点敲除突变体的方法,为研究番茄基因功能和开发新种质资源奠定基础。
本发明通过以下技术方案实现:
(1)将Cas9序列通过酶切位点BamHⅠ和SacⅠ克隆到pC131-N1-YFP载体上,形成表达载体pC131-35S-Cas9;
(2)将番茄SlU6启动子和sgRNA通过融合PCR连接并克隆到pGEM-T载体上,SlU6启动子和sgRNA含有2个BsaⅠ酶切位点,形成pGEM-SlU6-sgRNA中间载体;
(3)根据番茄目标基因设计靶向序列,根据靶向序列合成带有与BsaⅠ酶切粘性末端互补序列的引物;
(4)将合成的引物通过BsaⅠ酶切位点克隆到中间载体pGEM-SlU6-sgRNA,并通过HindⅢ酶切位点将带有靶向序列的SlU6-sgRNA克隆到表达载体pC131-35S-Cas9,获得pC131-SlU6-sgRNA-35S-Cas9;
(5)将pC131- SlU6-sgRNA-35S-Cas9转化农杆菌GV 3101,并侵染番茄子叶;
(6)利用潮霉素对侵染后番茄子叶进行抗性筛选,分离抗性芽后进行生根培养;
(7)提取抗性苗基因组DNA,并通过对目标基因进行Sanger测序验证靶向位点突变情况。
采用上述技术方案的积极效果:本发明所建立的番茄基因组定点敲除突变体的方法克服了ZFN技术和TALEN技术体系的操作复杂、费时低效等缺点,能够快速、高效获得番茄基因组定点敲除突变体,非常有利于研究番茄基因功能和开发番茄新种质。

Claims (1)

1.一种获得番茄基因组定点敲除突变体的方法,其特征在于:该方法包括以下步骤:
(1)将Cas9序列通过酶切位点BamHⅠ 和SacⅠ克隆到pC131-N1-YFP载体上,形成表达载体pC131-35S-Cas9;
(2)将番茄U6启动子(SlU6)和sgRNA通过融合PCR连接并克隆到pGEM-T载体上,SlU6启动子和sgRNA含有2个BsaⅠ酶切位点,形成pGEM- SlU6-sgRNA中间载体;
(3)根据番茄目标基因设计靶向序列,根据靶向序列合成带有与 BbsⅠ酶切粘性末端互补序列的引物;
(4)将合成的引物通过BsaⅠ酶切位点克隆到中间载体pGEM-SlU6-sgRNA,并通过HindⅢ酶切位点将带有靶向序列的SlU6-sgRNA克隆到表达载体pC131-35S-Cas9,获得pC131-SlU6-sgRNA-35S-Cas9;
(5)将pC131- SlU6-sgRNA-35S-Cas9转化农杆菌GV 3101,并侵染番茄子叶;
(6)利用潮霉素对侵染后番茄子叶进行抗性筛选,分离抗性芽后进行生根培养;
(7)提取抗性苗基因组DNA,并通过对目标基因进行Sanger测序验证靶向位点突变情况。
