背景技术
甜味剂是一种可以赋予食品或软饮料甜味的物质,非糖类甜味剂由于不会引起血糖的升高、脂肪堆积等健康问题,而受到了广泛的关注。
三叶苷作为一种非糖类甜味剂,其属于根皮素的糖苷化衍生物,普遍的研究认为根皮素类的糖苷化衍生物是一类钠葡萄糖协同转运因子(sodium/glucose cotransporterSGLT)的抑制剂,这类转运因子分布在人体的小肠、肾脏等器官内,通过对于这类转运因子的抑制可以达到降低血糖的目的。同时由于三叶苷具有很高的甜度,所以是一种很好的功能性甜味剂。
目前三叶苷的主要获得办法是使用壳斗科石柯属植物做为材料提取,例如多穗石柯(别名甜茶),但是石柯属植物分布面积小,种植量少,用于提取三叶苷会增加成本,限制产量。
本发明创造性地从苹果属植物中提取和分离出含有三叶苷的功能性甜味剂组合物,且工艺简便,产量可观,成本低廉,为进一步在食品、保健品、药品等领域应用三叶苷提供了源头保障。

具体地,本发明所述从苹果属植物中提取的功能性甜味剂组合物,其中应含有不少于80%的三叶苷,并且其中可能含有其他多酚类化合物,包含但不限于一种或多种成分,例如根皮苷、儿茶素、绿原酸、花色苷、芦丁及各种槲皮素的糖苷化衍生物和各种山萘酚的糖苷化衍生物等。
特别地,本发明所述的提取材料为苹果属植物,包括常见的栽培品种、观赏品种、原生种及变种以及用苹果属中任意种通过转基因技术获得的新品种。
本发明的实现方式包括浸提、萃取、初步分离、精细分离、浓缩和干燥结晶六个主要步骤。
浸提步骤通过使用醇的水溶液在一定条件下浸泡苹果属植物的材料,通过浸提步骤得到的粗提物再进一步纯化,例如通过大孔树脂或者聚酰胺柱层析的方法,这一步骤就是该发明所述的初步分离;再通过精细分离进一步去除其他杂质,提高三叶苷的含量,精细分离步骤结合具体需要可以通过柱层析法或高速逆流色谱法实现,浓缩步骤适用于任何需要缩小提取物体系体积的情况下,可以单一地或者组合地使用减压、加热、离心等手段;干燥结晶获得最终的甜味剂组合物。

本发明突破性的利用广泛栽种的苹果属植物来大量获取三叶苷,具有原料来源广泛、成本低廉的特点,且提取过程适用于主流的植物提取化工厂工艺,具有良好的兼容性,生产上易实现放大,适合于各种规模的工业化生产;最终的功能性甜味剂组合物产量大,纯度高,适合作为甜味剂进一步加工应用。

实施例
为了使本发明的目的、技术方案、优势及创新点更加清楚明白,以下结合附图及实施例对本发明进一步详细说明,但实施例并不限制本发明。并且,在本发明中可以做出多种改进和变型而不偏离本发明的精神或范围,这对于本领域技术人员是显而易见的。因此期望本发明涵盖在所附权利要求和它们的等同要求的范围内的这些改进和变形。
本发明的实施方式包括一种甜味剂组合物和获得这种甜味剂组合物的方法。
实施例1
将经过处理的苹果属植物的果实或果实残渣通过加热和/或冷冻减压的方法干燥后,用醇的水溶液浸泡,加热和/或加压,同时适度搅拌,以提高浸出率。浸提完毕后液体部分通过加热和/或减压浓缩后,再使用大孔树脂进行层析,使用醇的水溶液进行洗脱,收集含有目标化合物的组分,再次浓缩后使用高速逆流色谱仪进行精细分离,将配置好的溶剂体系依次注入仪器,在一定转速下,控制流速,并收集含有目标化合物的组分,充分干燥得到甜味剂组合物,使用高效液相色谱法检测甜味剂组合物种三叶苷的含量。
实施例2
将经过处理的苹果属植物的茎或叶通过加热和/或冷冻减压的方法干燥后,用醇的水溶液浸泡,热和/或加压,同时适度搅拌,以提高浸出率。浸提完毕后液体部分通过加热和/或减压浓缩后,再使用一定粒度的聚酰胺填装的层析柱进行初步分离,使用醇的水溶液进行洗脱,收集含有目标化合物的组分,浓缩后使用凝胶柱层析进行精细分离,得到含有甜味剂组合物的溶液,干燥结晶,得到甜味剂组合物,使用高效液相色谱法检测甜味剂组合物种三叶苷的含量。
适用于本文中实施例的苹果属植物的处理方式非限制性的实施例包括高温、低温、干旱、低氧、光照、辐射和转基因技术。适用于本文中实施例的干燥方法非限制性的实施例包括,烘干、阴干、冷冻干燥或其几种方法的组合。适用于本文中实施例的任何固液分离方式非限制性的实施例包括离心分离和过滤分离。
上述实施例的中对于甜味剂组合物的高效液相检测条件如下:使用InertsilODS-3色谱柱(5.0ml柱体积,4.6mm×250mm),Inertsil ODS-3保护柱(5.0μm,4.0mm×10mm),使用岛津公司(shimadzu)SPD-M20A二极管阵列DAD检测器,用于分析的具体条件在表1中。
两个实施例所获得的最终功能性甜味剂的组成没有实质差别,故仅用实施例1的高效液相检测结果来作说明结果如图1,此方法下三叶苷保留时间为20.30分钟,纯度为99.56%。
虽然已经就具体实施方式详细描述本发明,但是应当指出,本领域技术人员在理解了上述发明内容之后,可以容易地构思出这些实施方式的改变方案、变型方案和等同方案。因此,本发明的范围应当由所附权利要求及其任何等同要求的范围评价,同时,以上实施例并不限制本发明,在本发明的精神和原则之内所做出的任何修改、等同替换和改进等,均应包含在本发明的保护范围之内。
Claims (6)
1.一种甜味剂组合物的提取方法,其特征在于,所述甜味剂组合物的主要成分是三叶苷,其提取自苹果属植物(Malus),提取过程具体为:将经过处理的苹果属植物的果实或果实残渣通过加热和/或冷冻减压的方法干燥后,用醇的水溶液浸泡,加热和/或加压,同时适度搅拌,浸提完毕后液体部分通过加热和/或减压浓缩后,再使用大孔树脂进行层析,使用醇的水溶液进行洗脱,收集含有目标化合物的组分,再次浓缩后使用高速逆流色谱仪进行精细分离,将配置好的溶剂体系依次注入仪器,在一定转速下,控制流速,并收集含有目标化合物的组分,充分干燥得到甜味剂组合物;
所述醇的水溶液中的醇为乙醇;
所述精细分离的高速逆流色谱仪中使用的溶剂为正己烷、乙酸乙酯和水。
2.一种甜味剂组合物的提取方法,其特征在于,所述甜味剂组合物的主要成分是三叶苷,其提取自苹果属植物(Malus),提取过程具体为:将经过处理的苹果属植物的茎或叶通过加热和/或冷冻减压的方法干燥后,用醇的水溶液浸泡,加热和/或加压,同时适度搅拌,浸提完毕后液体部分通过加热和/或减压浓缩后,再使用聚酰胺粉末填装的层析柱进行初步分离,使用醇的水溶液进行洗脱,收集含有目标化合物的组分,浓缩后使用凝胶柱层析进行精细分离,得到含有甜味剂组合物的溶液,干燥结晶,得到甜味剂组合物;
所述醇的水溶液中的醇为乙醇;
所述聚酰胺粉末的粒度为200-400目。
3.根据权利要求1所述的提取方法,精细分离还可以使用高效液相色谱制备的方法,包括分析液相半制备和制备液相,其特征在于,所使用的色谱柱内填装填料应为各种规格的十八烷基硅烷键合硅胶(ODS)填料和/或亲水反相十八烷基硅烷键合硅胶填料,流动相为甲醇或乙腈。
4.根据权利要求2所述的提取方法,其特征在于,所述精细分离的凝胶柱选用Pharmadex LH-20或Sephadex LH-20。
5.一种由权利要求1或2所述的提取方法提取得到的甜味剂组合物,其特征在于,其中该组合物中三叶苷含量占该甜味剂组合物重量的80%以上。
6.权利要求1或2所述的甜味剂组合物的提取方法,其特征在于,进一步包括使用高效液相色谱检测三叶苷含量的步骤,具体为使用Inertsil ODS-3色谱柱,5.0ml柱体积,4.6mm×250mm,Inertsil ODS-3保护柱,5.0μm,4.0mm×10mm,使用岛津公司SPD-M20A二极管阵列DAD检测器,流动相A由甲酸10%,乙腈90%组合,流动相B由甲酸10%,水90%组合,流速1mL/min,注入体积10μL,UV-vis 280nm下检测,等度洗脱,流动相A 20%,流动相B 80%,其中所使用的色谱柱内填装填料应为烷基硅烷键合硅胶填料和/或亲水反相烷基键合硅胶填料。
