背景技术
感冒发烧是指感冒常常能引起发烧症状,感冒的发生主要由于体虚,抗病能力减弱而引起的。当气候剧变时,人体内外功能不能适应,邪气乘虚由皮毛、口鼻而入,引起一系列发热症状,四季均可发生,是当今发病率非常高的一种疾病。目前,感冒发烧的治疗手段主要以化学药为主,化学药起效快,但药效持续时间较短,病情容易反复。
岭南中草药,多属植物性草药,且多为一年生的草本植物,其性味多苦寒,其功效大多具有清热利湿或祛湿的作用,多用于感冒,高热烦渴,扁桃体炎,咽喉炎,气管炎,百日咳,肠炎,痢疾,传染性肝炎,野蕈、砒霜中毒等。
因此,根据感冒发烧治疗手段的优缺点,结合岭南中草药的特点,研究开发一种具有解热作用且药效持续时间较长的岭南中草药制剂,具有重要的意义,同时也具有很大的市场开发潜力。

本发明是一种解热中药组合物,配方由以下组分组成:草珊瑚:垂叶榕:石仙桃的重量比为2:0.8-1.2:0.8-1.2。优选的,配方由以下组分组成:草珊瑚:垂叶榕:石仙桃的重量比为2:0.9-1.1:0.9-1.1。最优选的,配方由以下组分组成:草珊瑚:垂叶榕:石仙桃的重量比为2:1:1。
解热中药组合物可以为汤剂。或者,为了患者服用的方便和根据提取物的特性,可以制成制剂,以所述解热中药组合物配方中组分的提取物为有效成分,添加相应药用辅料,按照中药常规制剂工艺制备成任何一种口服制剂。制剂的剂型可以为片剂、胶囊剂、颗粒剂、口服液或滴丸剂等。其中提取物优选为水提物,可按常规的水提工艺制备,例如:取配方的药材按照君臣佐使处方比例混合,每次用6~12倍药材份量的水,加热回流提取1~3小时,提取1~3次,过滤,合并滤液,浓缩到每毫升含生药1.0~5.0g,即得,再添加相应药用辅料可制备成任何一种口服制剂。
配方中,草珊瑚,又名九节茶,是金粟兰科草珊瑚属草珊瑚Sarcandra glabra(Thunb.)Nakai的全草。苦,辛,平。清热解毒、凉血消斑、祛风除湿、通络止痛。用于感冒高热、疮疡脓肿等,主治流行性感冒、流行性乙型脑炎、肺炎、阑尾炎、盆腔炎、跌打损伤、风湿关节痛、闭经、创口感染、菌痢等。《岭南采药录》记载“取叶煎水服,能祛风退热,又能祛风湿”。《生草药性备要》记载“煲水饮,退热”。
垂叶榕,又名细叶榕,是桑科榕属垂叶榕Ficus benjamina L.的气根、树皮、叶芽、果实等。微苦,涩,微寒。清热解毒,祛风,凉血,滋阴润肺,发表透疹,催乳,用于感冒发热、湿热泻痢等。《岭南采药录》记载“煎汤饮,能退热”。
石仙桃,是兰科石仙桃属石仙桃Pholidota chinensis Lindl.的全草。甘,淡,微寒。养阴清热、润肺止咳,用于热病津伤口渴,阴虚燥咳,潮热盗汗。
本方以九节茶和垂叶榕作为处方中的君臣药物,发挥其解热作用,而石仙桃作为处方中的使药,辅助治疗高热后津伤燥咳,滋阴养肺,符合中医整体治疗观念。

实施例1 取九节茶162g,小叶榕81g,石仙桃81g,剪碎,加8倍药材重量的水,回流提取1.5小时,提取2次,合并提取液,过滤,合并滤液,浓缩到250ml,得到汤剂。

实施例3 取九节茶162g,小叶榕81g,石仙桃81g,剪碎,加8倍药材重量的水,回流提取1小时,提取1次,合并提取液,过滤,合并滤液,浓缩到250ml,得到汤剂。
实施例4 取九节茶162g,小叶榕81g,石仙桃81g,剪碎,加10倍药材重量的水,回流提取1.5小时,提取3次,合并提取液,过滤,合并滤液,浓缩到250ml,得到汤剂。
实施例5 取九节茶162g,小叶榕81g,石仙桃81g,剪碎,加12倍药材重量的水,回流提取1小时,提取2次,合并提取液,过滤,合并滤液,浓缩到250ml,得到汤剂。
实施例6 取九节茶162g,小叶榕81g,石仙桃81g,剪碎,加10倍药材重量的水,回流提取2小时,提取1次,合并提取液,过滤,合并滤液,浓缩到250ml,得到汤剂。
实施例7 取九节茶162g,小叶榕64.8g,石仙桃64.8g。制备方法同实施例1。
实施例8 取九节茶162g,小叶榕97.2g,石仙桃97.2g。制备方法同实施例1。

实施例10 取九节茶162g,小叶榕72.9g,石仙桃81g。制备方法同实施例1。
实施例11 取九节茶162g,小叶榕89.1g,石仙桃81g。制备方法同实施例1。
实施例12 取上述实施例1所得汤剂,加入适量纯净水,搅拌加热煮沸,煮沸20分钟,待冷却到30~40℃时,加入适量壳聚糖和山梨酸钾,搅拌使溶解均匀,过滤,最后加纯净水调整至1000ml,过滤,分装,灭菌,即得口服液。每支口服液10ml。
实施例13 取上述实施例1所得汤剂,浓缩,干燥,得到干浸膏;取干浸膏12.0g,研成细粉,与适量的磷酸氢钙、微晶纤维素、三氯蔗糖、交联聚维酮和羧甲基淀粉钠,混匀,3%聚维酮溶液为粘合剂,过14目筛进行制粒,65℃干燥,再过14目筛进行整粒,然后加入适量的硬脂酸镁,压制成100片,即得分散片。分散片每片0.5g。
实施例14 取上述实施例1所得汤剂,浓缩,干燥,得到干浸膏;取干浸膏15.4g,研成细粉,加入适量的药用淀粉和硬脂酸镁,混合均匀,过筛,装入胶囊,制成100粒,即得硬胶囊。每粒内容物重0.2g。
实施例15 取上述实施例1所得汤剂,浓缩,干燥,得到干浸膏;取干浸膏10.5g,加入适量的丙二醇、卵磷脂和大豆油,振动磨超微20分钟,使混匀,制成软胶囊100粒,即得软胶囊。每粒含内容物0.3g。
实施例16 取上述实施例1所得汤剂,浓缩,干燥,得到干浸膏;取干浸膏17.6g,研成细粉,加入适量的药用淀粉和微粉硅胶,混合,过14目筛进行制粒,65℃干燥,再过14目筛进行整粒,然后加入适量的硬脂酸镁,混匀,压制成100片,即得片剂。每片重0.3g。
实施例17 取上述实施例1所得汤剂,浓缩,干燥,得到干浸膏;取干浸膏16.8g,研成细粉,加入适量的糊精和乳糖,混匀,制粒,65℃干燥,过14目筛进行整粒,分装,制得100包,即得颗粒剂。每包重5g。
实施例18 取上述实施例1所得汤剂,浓缩,干燥,得到干浸膏;取干浸膏15.4g,研成细粉;另取适量的聚乙二醇4000加热熔融,加入上述干浸膏,混匀,滴入冷却的液体石蜡中,制成滴丸,即得滴丸剂。每丸重0.4g。
实验例1
取上述实施例1所得汤剂,浓缩成100ml,再依次配制成高、中、低剂量,作为受试药物进行解热实验,以此来证明本发明的解热疗效。
取大鼠70只,体重180~220g,每日测肛温1次,连续2日,使大鼠适应此测温操作。实验当日早晨先测动物基础体温,然后以干酵母悬液按1ml/100g背部皮下注射,4h后测体温,根据造模后体温与基础体温的差值均衡分组,分别为空白对照组、模型组、阳性药组、药物提取液高、中、低剂量组。空白对照组:灌胃生理盐水(10ml/kg);模型对照组:灌胃生理盐水(10ml/kg);药物高、中、低剂量按10ml/kg进行灌胃;阳性对照组:灌胃阿司匹林0.2g/kg;中成药组:灌胃连花清瘟胶囊溶液0.375g/kg。分别测定给药后0.5h、1h、2h、3h、4h的肛温,以不同时间所测肛温与各基础肛温的差值为体温变化的指标。实验结束后,所有实验数据均用SPSS23.0统计软件处理。计数指标采用表示,其中结果P<0.05具有统计学意义。分析结果如表1和图1。

注:与模型组比较:*P<0.05,表示具有统计学意义。
根据实验分析结果,可以看出:与模型组相比,空白对照组每个时间段均具有显著性差异(P<0.05),表明酵母悬液致大鼠发热模型成功;与模型组相比,阿司匹林组在给药后0.5h~1h之间体温下降明显,从给药后2h开始机体体温变化出现反复;与模型组相比,中药处方制备的汤剂高、中剂量组均有显著性差异(P<0.05),表明从给药后2h开始,该处方高、中剂量组对酵母致热的大鼠具有解热作用,提示该中药处方具有解热作用,且药效较为平缓,持续时间较长。

1.一种解热中药组合物,其特征在于:配方由以下组分组成:草珊瑚:垂叶榕:石仙桃的重量比为2:0.8-1.2:0.8-1.2。
2.根据权利要求1所述的解热中药组合物,其特征在于:所述的配方由以下组分组成:草珊瑚:垂叶榕:石仙桃的重量比为2:0.9-1.1:0.9-1.1。
3.根据权利要求1所述的解热中药组合物,其特征在于:所述的配方由以下组分组成:草珊瑚:垂叶榕:石仙桃的重量比为2:1:1。
4.根据权利要求1所述的解热中药组合物,其特征在于:所述的解热中药组合物为汤剂。
5.一种解热中药制剂,是以权利要求1、2或3所述解热中药组合物配方中组分的提取物为有效成分,添加药用辅料制得的制剂。
6.根据权利要求5所述的解热中药制剂,其特征在于:所述制剂的剂型为片剂、胶囊剂、颗粒剂、口服液或滴丸剂。
7.根据权利要求5所述的解热中药制剂,其特征在于:所述提取物为水提物。
