Abstract
本发明公布了银杏慢消化淀粉负载GBE的技术方法,以我国特有资源银杏淀粉及银杏叶提取物(GBE)为原料。首先,将银杏淀粉脱脂后,经过普鲁兰酶和α-淀粉酶对糊化后的淀粉进行复合脱支处理,灭酶;然后,加入用酒精溶液溶解的GBE,震荡,重结晶一次;最后,在真空干燥箱中干燥一段时间后,将样品置于冷冻干燥机中,冷冻干燥处理,将样品粉碎,过筛网,即得银杏慢消化淀粉包埋的GBE颗粒。此颗粒用银杏慢消化淀粉作药物壁材,将GBE药物包埋,可以起到抗氧化作用。且对包埋后的GBE可以起到缓释的效果。此银杏慢消化淀粉负载的GBE颗粒特别适于三高患者使用。

技术领域
本发明属于天热产物的加工领域,涉及的是银杏慢消化淀粉做药物载体的方法。
背景技术
慢消化淀粉(slowlydigestiblestarch,SDS),指那些能在小肠中被完全消化吸收但速度较慢的淀粉(20-120min);其中,慢消化淀粉呈部分结晶的网状结构,具有低的溶胀程度,并在消化过程中水解速率近似恒定。近年来临床研究发现,慢消化淀粉具有缓慢消化吸收、持续释放能量、维持餐后血糖稳态、改善葡萄糖耐量,还可降低餐后胰岛素分泌,提高机体对胰岛素的敏感性、预防和治疗各种与饮食相关慢性疾病的特殊生理学功能。慢消化淀粉作为一种新型的功能性产品,恰恰兼顾了糖尿病营养与治愈功能两大需求,不仅顺应了当代食品及药品科技发展的趋势,还满足了消费者的健康需求,这对合理利用碳水化合物预防胰岛素抵抗相关的代谢综合症发生有着重要意义。因此,众多科研工作者都着眼于慢消化淀粉的研究。
申请号为200710131771.4和200710195581.9的专利,两者都是采用普鲁兰酶与α-淀粉酶对糊化后的淀粉进行复合脱支处理,重结晶,热液处理制备了慢消化淀粉。申请号为200780050404.X的专利公开了颗粒粒径D90小于50μm,缓慢消化淀粉含量为15-75%的慢消化淀粉制作方法;申请号为200910071744.1的专利采用普鲁兰酶酶解,然后在结晶的方法制备慢消化淀粉;申请号为200910200212.3的专利采用转葡糖苷酶以及淀粉酶酶解对淀粉进行酶解,在进行结晶处理等方式制备慢消化淀粉;申请号为201010142423.9的专利以新型水解-转苷淀粉酶协同修饰重组淀粉分子链,从而获得具有慢消化特性的耐温树枝状淀粉。申请号为201010228265.9的专利采用高功率超声波降解大米淀粉形成短链直链淀粉,经过高温脂质反应和短时结晶形成单螺旋V-型微晶淀粉-脂质复合物,制备慢消化淀粉。申请号为201110112582.9的专利将醇溶蛋白放于烧杯中,加入乙醇,水浴搅拌至醇溶蛋白完全溶解,加入油酸,搅拌均匀,加入玉米淀粉,继续搅拌。在高压均质机中均质3次,离心喷雾干燥得到慢消化淀粉产品。申请号为201310058866.3的专利采用普鲁兰酶和葡萄糖苷转移对小麦淀粉进行处理,在通过反复重结晶的方法制备慢消化淀粉。
上述众多研究方法都属于慢消化淀粉制作技术。没有对慢消化淀粉的应用进行研究。本发明中将叙述一种慢消化淀粉的应用技术,以银杏淀粉为原料,制备慢消化淀粉,负载银杏叶提取物(GBE)的技术。
发明内容
本发明的技术方案是,将银杏淀粉脱脂后,经过普鲁兰酶和α-淀粉酶对糊化后的淀粉进行复合脱支处理,灭酶后,加入用酒精溶液溶解的GBE,震荡,重结晶一次后,在真空干燥箱中干燥一段时间后,将样品置于冷冻干燥机中,冷冻干燥处理,即得银杏慢消化淀粉包埋的GBE颗粒。
具体制备工艺为:
1)将银杏淀粉用石油醚脱脂处理;
2)将脱脂后的银杏淀粉糊化,然后先添加普鲁兰酶,后加入α-淀粉酶,酶解温度保持在40-70℃,酶解时间2-8小时,然后在高压灭菌锅中进行灭酶处理30分钟;
3)灭酶处理后,加入用50-80%酒精溶液溶解的GBE溶液,放入振荡器内震荡6-10小时,0-10℃重结晶处理24-36小时;
4)放入真空干燥箱中在40-80℃干燥4-10小时;
5)将干燥后的样品置于冷冻干燥机中,降温到-40℃至0℃开始冷冻干燥,最后获得银杏慢消化淀粉包埋的GBE的干燥样品,粉碎过40-120目筛网,获得银杏慢消化淀粉负载的GBE颗粒。
有益效果
本发明所用原料为我国特有林业资源之一银杏淀粉及其叶子的提取物,所得产品为银杏慢消化淀粉负载的GBE颗粒。本颗粒不仅可以起到减缓淀粉消化,还可以对GBE包埋,抗氧化及缓释作用。此加工方法的特点为:
1)充分利用我国林业资源;
2)银杏慢消化淀粉可以用作药物壁材,将GBE药物包埋,起到抗氧化作用;
3)包埋后的GBE可以起到缓释的效果;
4)此银杏慢消化淀粉负载的GBE适于糖尿病患者使用。
具体实施方式
实例一
将银杏淀粉用石油醚脱脂处理。将脱脂后的银杏淀粉糊化4公斤,然后先添加普鲁兰酶,后加入α-淀粉酶,酶解温度保持在40-70℃,酶解时间6小时。然后在高压灭菌锅中进行灭酶处理30分钟。灭酶处理后,加入用50-80%酒精溶液溶解的GBE溶液,放入振荡器内震荡7小时。0-10℃重结晶处理24小时。放入真空干燥箱中在60℃干燥8小时。将干燥后的样品置于冷冻干燥机中,降温到-40℃至0℃开始冷冻干燥,最后获得银杏慢消化淀粉包埋的GBE的干燥样品,粉碎过100目筛网,获得银杏慢消化淀粉负载的GBE颗粒。
实例二
将银杏淀粉用石油醚脱脂处理。将脱脂后的银杏淀粉糊化10公斤,然后先添加普鲁兰酶,后加入α-淀粉酶,酶解温度保持在40-70℃,酶解时间8小时。然后在高压灭菌锅中进行灭酶处理30分钟。灭酶处理后,加入用50-80%酒精溶液溶解的GBE溶液,放入振荡器内震荡8小时。0-10℃重结晶处理30小时。放入真空干燥箱中在50℃干燥10小时。将干燥后的样品置于冷冻干燥机中,降温到-40℃至0℃开始冷冻干燥,最后获得银杏慢消化淀粉包埋的GBE的干燥样品,粉碎过80目筛网,获得银杏慢消化淀粉负载的GBE颗粒。
实例三
将银杏淀粉用石油醚脱脂处理。将脱脂后的银杏淀粉糊化15公斤,然后先添加普鲁兰酶,后加入α-淀粉酶,酶解温度保持在40-70℃,酶解时间4小时。然后在高压灭菌锅中进行灭酶处理30分钟。灭酶处理后,加入用50-80%酒精溶液溶解的GBE溶液,放入振荡器内震荡10小时。0-10℃重结晶处理36小时。放入真空干燥箱中在70℃干燥9小时。将干燥后的样品置于冷冻干燥机中,降温到-40℃至0℃开始冷冻干燥,最后获得银杏慢消化淀粉包埋的GBE的干燥样品,粉碎过60目筛网,获得银杏慢消化淀粉负载的GBE颗粒。
Claims (2)
1.银杏慢消化淀粉负载GBE的方法,其特征在于:
加工方法包含下列步骤:
(1)将银杏淀粉用石油醚脱脂处理;
(2)将脱脂后的银杏淀粉糊化,然后先添加普鲁兰酶,后加入α-淀粉酶,酶解温度保持在40-70℃,酶解时间2-8小时,然后在高压灭菌锅中进行灭酶处理30分钟;
(3)灭酶处理后,加入用50-80%酒精溶液溶解的GBE溶液,放入振荡器内震荡6-10小时,
0-10℃重结晶处理24-36小时;
(4)放入真空干燥箱中在40-80℃干燥4-10小时;
(5)将干燥后的样品置于冷冻干燥机中,降温到-40℃至0℃开始冷冻干燥,最后获得银杏慢消化淀粉包埋的GBE的干燥样品,粉碎过40-120目筛网,获得银杏慢消化淀粉负载的GBE颗粒。
2.根据权利要求1所述的降温到-40℃至0℃开始冷冻干燥,最后获得银杏慢消化淀粉包埋的GBE的干燥样品,其特征在于,冷冻干燥过程是阶梯式升温过程,使干燥真空度保持恒定。
