本发明属于保健品领域,具体涉及以一种黄刺、沙棘、唐古特白刺果汁组合物和用途。
技术背景
随着现代生活节奏的加快,社会竞争的加剧,人们的体力、脑力、心理等疲劳长期积累,加之饮食不健康、环境辐射增多等因素,导致人体内自由基过多,使越来越多的人处于“亚健康”状态。自由基是一种因失去一个电子而成的不对称、不稳定的原子或分子,具有很大的能量。自由基总是企图夺取一个电子以达到新的平衡,为此它会攻击细胞组织中的脂质、蛋白质、糖类和DNA等物质,这就造成了脂质的过氧化、蛋白质的变性酶的失活、DNA的氧化损伤等。此外,当人们疲劳时,体内会产生了大量的代谢物,如乳酸、二氧化碳、血清尿素氮等,这类物质在体内积聚,刺激人体组织细胞和神经系统。身体长期处于疲劳状态,会造成体内激素代谢失调、神经系统调节功能异常、免疫力减低等不良后果。
白刺(Nitraria)为蒺藜科(Zygophyllceae)植物,是生长在海拔3000米以上的荒漠戈壁地区的主要建群植物之一。其抗逆性强,耐干旱盐碱,是优良的防风固沙灌木。白刺成熟浆果为紫红色,无毒可食,其果实中营养成分和医药保健活性成分种类多,含量丰富。种籽油主要成分为亚油酸、亚麻酸、花生四烯酸等不饱和脂肪酸,其中亚油酸的含量高达70%,在植物界中实属罕见。而其抗疲劳的作用已有文献报道,其具有降低运动后小鼠体内乳酸和尿素氮的功能。
沙棘为胡颓子科沙棘属植物(Hippophaerhamnoides),其又名酸刺、醋柳、黑刺。沙棘在我国北部及西南多个省区均有广泛分布,其生长海拔为1000-4500米之间,其植株亦具有很高的生态应用价值。其果实酸甜,营养丰富,药学应用价值极大,已开发应用至多个产品中。其果实的精华在于其籽油。已有多份报道探明,其籽油还有维生素类、多酚类、脂肪酸,等多种生物活性物质,其所含维生素种类之多,含量之丰富是任何果蔬不能比拟的,因此,被誉为“维生素宝库”。同时,亦有学者研究表明,和其他地区种植沙棘比,青藏高原沙棘果实中的维生素C含量比其他低海拔地区高2倍之多。
黄刺,小檗科植物直穗小檗BerberisdasystachyaMaxim.,是与白刺、沙棘(黑刺)一样为蒙、藏、维吾尔等少数民族药用与食用的传统野生浆果,统称为“青海三刺”。黄刺果实中含有丰富的维生素、氨基酸、多糖类等丰富的营养活性成分。《藏标》《藏本草》中对直穗小檗果实医药功能做了简要的记述,其具有治疗腹痛,消化不良,腹胀,痢疾等功能。黄刺种籽占黄刺成熟果实总质量的41-46%,其含油脂的主要成分为不饱和脂肪酸,如亚油酸和亚麻酸的含量就占了近60%,具有极高的营养和药用价值。
发明内容
本发明的目的在于提供一种基于黄刺、沙棘、唐古特白刺果汁组合物。本发明的另一目的在于提供该组合物或片剂的用途。
具体地,本发明提供了一种青海三刺果汁组合物,其特征在于:它是由如下体积配比的原料组成:
黄刺果汁1~5份、沙棘果汁1~2份、白刺果汁1~10份。
优选地,它是由如下体积配比的原料组成:
黄刺果汁4份、沙棘果汁1份、白刺果汁5份。
其中,所述黄刺果汁、沙棘果汁或白刺果汁是由黄刺、沙棘或白刺为原料,直接榨汁得到。
本发明还提供了上述组合物在制备具备抗氧化功效的食品、保健品或药品中的用途。
进一步地,所述食品、保健品或药品是清除ABTS自由基、超氧阴离子自由基或羟自由基的食品、保健品或药品。
优选地,所述食品、保健品或药品是清除超氧阴离子自由基的食品、保健品或药品。
本发明还提供了一种保健饮料或口服液,它是由上述青海三刺果汁组合物为活性成分,加上饮料或口服液中常用的辅料制备而成。当然,除饮料或口服液外,还可以将上述组合物用于制备其他产品。
本发明在实验中发现,三刺果汁组合物在多数配比下,其抗氧化作用是相似的,但是,在“黄刺果汁4份、沙棘果汁1份、白刺果汁5份”这一配比下,组合物的抗氧化活性明显优于其他各组,特别是在抗超氧阴离子自由基上的表现尤为突出,因此,上述配比作为本发明的优选配比,其对抗超氧阴离子自由基的作用可以作为本发明的优选用途;同时,上述最佳配比的抗氧化活性还显著优于各单一果汁。上述实验结论足以证明,本发明将3种果汁在特定的用量配比下,发挥了协同增效作用。
具体实施方式
实施例1本发明组合物的制备
分别取黄刺、沙棘或唐古特白刺果实,直接榨汁后,即得黄刺果汁、沙棘果汁或白刺果汁。
将各果汁以黄刺果汁4份、沙棘果汁1份、白刺果汁5份(v/v)进行配比混合,即得本发明组合物。
体外抗氧化活性实验
(1)材料:浆果果汁:为高压均质压榨所得果汁,由本发明实施例制备而成。各组合物配比见表1。
(2)实验仪器及试剂:TGL-16G-A型管式离心机,上海安亭科学仪器厂;UV-722N可见紫外分光光度计,上海尼柯有限公司;HD型恒温水浴锅,辽阳市恒温仪器厂。ABTS购于上海晶纯生化科技股份有限公司(阿拉丁);过硫酸钾、磷酸二氢钠、磷酸氢二钠、氯化钠、三氯乙酸等试剂均购于山东禹王实业有限公司;超氧阴离子试剂盒、羟自由基试剂盒均购于南京建成生物科技有限公司。
(3)ABTS清除能力:将ABTS溶解(+H2O)至终浓度为7mmol/L,过硫酸钾配制成2.45mmol/L,将二者溶液混合,放置于黑暗,室温条件下搅拌16h;而后将ABTS混合溶液用PBS溶液(PH=7.0)稀释到其吸光值(于734nm处测)为0.70±0.02;青海三刺浆果果汁按所需要的体积比量好后用蒸馏水配成终浓度为16mg/mL的混合果汁;最后将上述浓度的样品加入到ABTS混合溶液中,其样品与ABTS混合溶液比例为1∶20,混合后于室温下孵育6min,于734nm处测定吸光值。
计算公式:ABTS清除率(%)=(1-Asample/Acontrol)×100%
其中Acontrol为ABTS混合溶液的吸光值;Asample为ABTS混合溶液中加入样品后测定的吸光值。
(4)超氧阴离子自由基清除能力和羟自由基均采用南京建成试剂盒检测。青海三刺浆果果汁按所需要的体积比量好后用蒸馏水配成终浓度为16mg/mL的混合果汁。

本发明在前期实验中发现,按照表1中的前4种配比制备的果汁组合物,均具有良好的抗氧化活性,且各组活性较为相似,其中以第1组配比较好;但发明人意外地发现,当果汁组合物配比为“黄刺果汁4份、沙棘果汁1份、白刺果汁5份”时,组合物的抗氧化活性得到大幅的提高,特别是抗超氧阴离子自由基的能力提高尤为显著,因此本发明优选此配比;同时,上述最佳配比的抗氧化活性还显著优于各单一果汁。上述实验结论足以证明,本发明将3种果汁在特定的用量配比下,发挥了协同增效作用。
