Abstract
本发明属于食品加工技术领域,涉及一种富含大豆多肽的膳食补充剂及其制备方法。包括以下步骤:A、制备发酵基料,将大豆多肽溶于水中,加入糖,匀浆得到匀浆液,杀菌后冷却,B、制备母发酵剂,取乳品溶液,杀菌后冷却,在乳品溶液中加入复合乳酸菌悬液,厌氧培养即得母发酵剂,C、制备发酵液,发酵基料置于无菌条件下,调节温度至35-42℃,随后加入适量母发酵剂,培养3-6小时,D、水解,发酵液升高温度至45-55℃,在45-55℃下水解,得水解液,E、干燥,向水解液中加入适量环糊精,干燥脱除水分,得到干燥粉末。本发明营养丰富、口感好,易于被人体吸收。
Description
技术领域
本发明属于食品加工技术领域,涉及一种大豆多肽,尤其涉及一种富含大豆多肽的膳食补充剂及其制备方法。
背景技术
与普通的大豆蛋白相比,大豆多肽具有溶解性好、营养全面、易消化吸收、无豆腥味、受热不变性等特点。作为一种优良的膳食补充剂和保健食品原料,在膳食中添加大豆多肽或食用富含大豆多肽的保健食品,具有提高人体免疫力、抗疲劳、补充微量元素、改善骨质疏松症状和预防心血管系统疾病等功效。
大豆多肽是以大豆榨油后得到的豆粕或大豆分离蛋白为原料,经蛋白酶水解以后的混合物,其主要成分是平均分子量1000道尔顿以下的寡肽。在未经提纯的大豆多肽中,通常还含有少量的大豆异黄酮、大豆低聚糖和矿物质等。然而,大豆蛋白在水解过程中产生的疏水性氨基酸低分子量多肽具有苦味,影响了大豆多肽的整体口感;大豆异黄酮主要以糖苷形式存在,无法被人体直接吸收利用;以水苏糖、棉籽糖为主的大豆低聚糖,食用后容易导致胀气,上述这些因素限制了大豆多肽功能特性的发挥。
中国专利文献公开了一种大豆多肽的制备方法[申请号:201310487428.9],包括以下步骤:(1)制备大豆蛋白浆液,(2)调节温度和pH值进行第一次酶解,用粗胰酶进行酶解,(3)升温将粗胰酶灭活,(4)调节温度和pH值进行第二次酶解,用中性蛋白酶进行酶解,(5)升温将中性蛋白酶灭活,(6)调节pH值,过滤、浓缩、干燥、包装的成品大豆多肽。该方案采用复合酶多级定向水解大豆蛋白,针对不同性质的大分子蛋白质分阶段用不同性能的酶进行水解,水解专指度大为增强,从而极大地提高了酶的利用效率,得到的大豆多肽质量高,活性强。但是该方案的营养成分单一,口感略苦,有待进一步改进。
发明内容
本发明的目的是针对上述问题,提供一种口感好,营养丰富,易于被人体吸收的富含大豆多肽的膳食补充剂。
本发明的另一目的是提供一种加工时间短、反应效率高的富含大豆多肽的膳食补充剂的制备方法。
为达到上述目的,本发明采用了下列技术方案:一种富含大豆多肽的膳食补充剂的制备方法,包括以下步骤:
A、制备发酵基料,将大豆多肽溶于水中,在其水溶液中加入0-10%(w/w)的糖,待溶胀后匀浆得到匀浆液,匀浆液杀菌后冷却即为发酵基料,
B、制备母发酵剂,取乳品溶液,杀菌后冷却至42℃以下,以无菌操作向杀菌后的乳品溶液中加入1-5%(v/v)的复合乳酸菌悬液,在35-42℃下厌氧培养3-5小时,即得母发酵剂,乳品溶液为鲜奶、纯牛奶、复原乳中的任意一种或几种,
C、制备发酵液,将上述步骤A中得到的发酵基料置于无菌条件下,调节温度至35-42℃,随后加入适量步骤B中得到的母发酵剂,培养3-6小时,
D、水解,将步骤C得到的发酵液升高温度至45-55℃,加入适量防腐剂,加酸度调节剂调节pH值至5.5-6.5,并在45-55℃下水解,得水解液,
E、干燥,向水解液中加入适量环糊精,混合均匀,得到混合物,将上述混合物经干燥脱除水分,得到干燥粉末,所得粉末即为富含大豆多肽的膳食补充剂。
在上述的富含大豆多肽的膳食补充剂的制备方法中,在步骤A中,所述的糖为单糖、双糖或多糖中的任意一种或几种的组合,匀浆液在100-121℃下杀菌10-60min。
在上述的富含大豆多肽的膳食补充剂的制备方法中,在步骤B中,乳品溶液加热至68-70℃,并保持该温度杀菌10-30min,杀菌完成后,乳品溶液在10min内冷却至42℃以下。
在上述的富含大豆多肽的膳食补充剂的制备方法中,在步骤B中,所述复合乳酸菌悬液的制备方法为:取保加利亚乳酸杆菌、干酪乳杆菌、瑞士乳杆菌、嗜酸乳杆菌、双歧杆菌、嗜热链球菌、鼠李糖乳杆菌和植物乳杆菌菌种中的任意两种或两种以上,分别接种于MRS液体培养基中,在37℃下厌氧培养6-24小时后混合得到所述复合乳酸菌悬液。
在上述的富含大豆多肽的膳食补充剂的制备方法中,在步骤C中,母发酵剂的加入量为发酵基料体积的5-20%(v/v)。
在上述的富含大豆多肽的膳食补充剂的制备方法中,在步骤D中,所述的防腐剂为山梨酸钾、丙酸钙、双乙酸钠和乳链球菌素的任意一种或几种的组合,防腐剂在发酵液中的添加量为0-0.25%(w/w),水解时间为1-3小时且为连续水解。
在上述的富含大豆多肽的膳食补充剂的制备方法中,在步骤D中,所述的酸度调节剂为氢氧化钙、氢氧化钾、氢氧化钠、碳酸氢三钠、碳酸钠、碳酸钾、碳酸氢钠和碳酸氢钾的任意一种或几种的组合。
在上述的富含大豆多肽的膳食补充剂的制备方法中,在步骤E中,混合物中还加入适量调味剂和香精并混合均匀,干燥采用喷雾干燥,并使干燥粉末的含水量在4-6%之间,水解液中环糊精的质量达到水解液总固形物质量的5-10%,所述的环糊精为α-环糊精、β-环糊精或γ-环糊精的任意一种或几种的组合。
在上述的富含大豆多肽的膳食补充剂的制备方法中,所述的环糊精为α-环糊精或其改性衍生物、β-环糊精或其改性衍生物、γ-环糊精或其改性衍生物中的任意一种或几种的组合。
根据上述的制备方法制得的富含大豆多肽的膳食补充剂。
与现有的技术相比,本发明的优点在于:
1、本发明使得大豆多肽作为膳食补充剂的营养组成更加全面,其中所含有的维生素类营养成分,为乳酸菌发酵所得,非人工合成后添加,具有更高的安全性。
2、本发明采用发酵与水解分阶段进行的方法,使乳酸菌的发酵和糖苷酶水解作用分别在各自适宜的条件下进行,提高了反应效率,缩短了加工时间。
3、本发明利用乳酸菌发酵产生的糖苷酶将大豆异黄酮苷转化为异黄酮苷元,更利于人体吸收。
4、本发明利用乳酸菌发酵产生的糖苷酶将大豆低聚糖水解,避免了食用后的胀气问题。
5、本发明利用乳酸菌发酵产生的氨肽酶将苦味肽水解,提升了产品的口感。
6、本发明使用糊精掩蔽了异黄酮苷元的苦味,提升了产品的口感。
具体实施方式
实施例1
一种富含大豆多肽的膳食补充剂的制备方法,包括以下步骤:
以蛋白酶水解脱脂豆粕制备得到的大豆多肽粉为原料,将大豆多肽粉溶于水至溶液总固形物含量为10%,待溶胀后匀浆处理;匀浆液在121℃下杀菌10min,冷却后作为发酵基料。
取保加利亚乳酸杆菌、嗜热链球菌、鼠李糖乳酸杆菌冻干粉,分别接种于MRS液体培养基中,震荡分散均匀后在37℃下厌氧培养12小时;
将上述保加利亚乳酸杆菌、嗜热链球菌、鼠李糖乳酸杆菌的菌悬液按照体积比1:1:1混合,得到复合乳酸菌悬液;
取新鲜生乳,加热至70℃,并保持该温度30min,随后迅速冷却至42℃;
以无菌操作向杀菌后的乳中加入以体积计5%(v/v)的复合乳酸菌悬液,在42℃下厌氧培养5小时,即得母发酵剂。
将发酵基料置于无菌条件下,调节温度至42℃,按照体积比20%(v/v)的比例加入母发酵剂,在42℃下厌氧培养6小时,得发酵液。
将发酵液升高温度至55℃,加入丙酸钙,使丙酸钙的质量百分比浓度达到0.25%,用0.1mol/L的氢氧化钠溶液调节pH至6.5,并在55℃下继续水解3小时,随后加入β-环糊精,使β-环糊精的质量达到发酵液总固形物量的10%,按照发酵液质量的0.1%加入香精,按照发酵液质量的6%加入蔗糖,溶解并混合均匀。
将上述混合物过筛,除去会堵塞离心喷雾机中旋转分离器的不溶物,随后进行喷雾干燥,进风温度为180℃,出风温度为105℃,调节进料流量以控制喷雾干燥后的粉末含水量在4-6%之间,用铝塑复合真空包装袋将粉末包装后即为成品。
所得产品中水溶性维生素含量0.05%,多肽含量61%,大豆苷元、染料木素和黄豆黄素三种苷元含量合计1.9%。
实施例2
一种富含大豆多肽的膳食补充剂的制备方法,包括以下步骤:
以蛋白酶水解大豆分离蛋白制备得到的大豆多肽粉为原料,将大豆多肽粉、水、20%的葡萄糖溶液按照质量比以1:1:1的比例混合,待吸水溶胀后,匀浆处理至糊状;
匀浆液在110℃下杀菌45min,冷却后作为发酵基料。
取植物乳杆菌、双歧杆菌和嗜酸乳杆菌菌种,分别接种于MRS液体培养基中,震荡分散均匀后在37℃下厌氧培养12小时;
将上述菌悬液按照体积比2:1:1混合,得到复合乳酸菌悬液;
将脱脂乳粉与69℃热水按照质量比2:9混合均匀得到复原乳,调节复原乳温度为69℃,并保持该温度杀菌20min,随后10min内迅速冷却至37℃;
以无菌操作向杀菌后的复原乳中加入以体积计2.5%的复合乳酸菌悬液,在37℃下厌氧培养4小时,即得母发酵剂。
将发酵基料置于无菌条件下,调节温度至37℃,按照体积比10%的比例加入母发酵剂,在37℃下厌氧培养2小时,得到发酵液。
将发酵液升高温度至50℃,加入占发酵液质量0.01%的山梨酸钾,用0.1mol/L的碳酸钠溶液调节pH至6.0,并在50℃下继续水解2小时,随后加入重量比为1:1的α-环糊精和β-环糊精,使α-环糊精和β-环糊精的总质量达到发酵液总固形物量的7%,按照发酵液质量的0.15%加入香精,按照0.12g/kg发酵液的用量加入复合甜味剂,复合甜味剂的配比为一份糖精钠加十份甜蜜素(质量比),搅拌发酵液使香精、复合甜味剂溶解并混合均匀。
将上述混合物直接喷雾干燥,进风温度为180℃,出风温度为105℃,调节进料流量以控制喷雾干燥后的粉末含水量在4-6%之间。用纸塑复合袋将粉末包装后即为成品。
所得产品中水溶性维生素含量0.03%,多肽含量80%,大豆苷元、染料木素和黄豆黄素含量合计1.1%。
实施例3
一种富含大豆多肽的膳食补充剂的制备方法,包括以下步骤:
以蛋白酶水解大豆分离蛋白制备得到的大豆多肽粉为原料,将大豆多肽粉、水、20%的蔗糖溶液按照质量比以3:3:2的比例混合,待吸水溶胀后,匀浆处理至糊状;
匀浆液在100℃下杀菌60min,冷却后作为发酵基料。
取植物乳杆菌和嗜酸乳杆菌菌种,分别接种于MRS液体培养基中,震荡分散均匀后在37℃下厌氧培养6小时;
将上述菌悬液按照体积比2:1混合,得到复合乳酸菌悬液;
将脱脂乳粉与68℃热水按照质量比2:9混合均匀得到复原乳,调节复原乳温度为68℃,并保持该温度杀菌10min,随后10min内迅速冷却至40℃;
以无菌操作向杀菌后的复原乳中加入以体积计1%的复合乳酸菌悬液,在35℃下厌氧培养5小时,即得母发酵剂。
将发酵基料置于无菌条件下,调节温度至35℃,按照体积比20%的比例加入母发酵剂,在35℃下厌氧培养3小时,得到发酵液。
将发酵液升高温度至45℃,加入占发酵液质量0.01%的山梨酸钾,用0.1mol/L的碳酸钠溶液调节pH至5.5,并在45℃下继续水解1小时,随后加入β-环糊精,使β-环糊精的质量达到发酵液总固形物量的5%,按照发酵液质量的0.1%加入香精,按照0.12g/kg发酵液的用量加入复合甜味剂,复合甜味剂的配比为一份糖精钠加十份甜蜜素(质量比),搅拌发酵液使香精、复合甜味剂溶解并混合均匀。
将上述混合物直接喷雾干燥,进风温度为180℃,出风温度为105℃,调节进料流量以控制喷雾干燥后的粉末含水量在4-6%之间。用纸塑复合袋将粉末包装后即为成品。
所得产品中水溶性维生素含量0.02%,多肽含量85%,大豆苷元、染料木素和黄豆黄素含量合计0.9%。

Claims (10)
1.一种富含大豆多肽的膳食补充剂的制备方法,其特征在于,包括以下步骤:
A、制备发酵基料,将大豆多肽溶于水中,在其水溶液中加入0-10%(w/w)的糖,待溶胀后匀浆得到匀浆液,匀浆液杀菌后冷却即为发酵基料,
B、制备母发酵剂,取乳品溶液,杀菌后冷却至42℃以下,以无菌操作向杀菌后的乳品溶液中加入1-5%(v/v)的复合乳酸菌悬液,在35-42℃下厌氧培养3-5小时,即得母发酵剂,
C、制备发酵液,将上述步骤A中得到的发酵基料置于无菌条件下,调节温度至35-42℃,随后加入适量步骤B中得到的母发酵剂,培养3-6小时,
D、水解,将步骤C得到的发酵液升高温度至45-55℃,加入适量防腐剂,加酸度调节剂调节pH值至5.5-6.5,并在45-55℃下水解,得水解液,
E、干燥,向水解液中加入适量环糊精,混合均匀,得到混合物,将上述混合物经干燥脱除水分,得到干燥粉末,所得粉末即为富含大豆多肽的膳食补充剂。
2.根据权利要求1所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,在步骤A中,所述的糖为单糖、双糖或多糖中的任意一种或几种的组合,匀浆液在100-121℃下杀菌10-60min。
3.根据权利要求1所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,在步骤B中,乳品溶液加热至68-70℃,并保持该温度杀菌10-30min,杀菌完成后,乳品溶液在10min内冷却至42℃以下。
4.根据权利要求1所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,在步骤B中,所述复合乳酸菌悬液的制备方法为:取保加利亚乳酸杆菌、干酪乳杆菌、瑞士乳杆菌、嗜酸乳杆菌、双歧杆菌、嗜热链球菌、鼠李糖乳杆菌和植物乳杆菌菌种中的任意两种或两种以上,分别接种于MRS液体培养基中,在37℃下厌氧培养6-24小时后混合得到所述复合乳酸菌悬液。
5.根据权利要求1所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,在步骤C中,母发酵剂的加入量为发酵基料体积的5-20%(v/v)。
6.根据权利要求1所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,在步骤D中,所述的防腐剂为山梨酸钾、丙酸钙、双乙酸钠和乳链球菌素的任意一种或几种的组合,防腐剂在发酵液中的添加量为0-0.25%(w/w),水解时间为1-3小时且为连续水解。
7.根据权利要求1所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,在步骤D中,所述的酸度调节剂为氢氧化钙、氢氧化钾、氢氧化钠、碳酸氢三钠、碳酸钠、碳酸钾、碳酸氢钠和碳酸氢钾的任意一种或几种的组合。
8.根据权利要求1所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,在步骤E中,混合物中还加入适量调味剂和香精并混合均匀,干燥采用喷雾干燥,并使干燥粉末的含水量在4-6%之间,水解液中环糊精的质量达到水解液总固形物质量的5-10%,所述的环糊精为α-环糊精、β-环糊精或γ-环糊精的任意一种或几种的组合。
9.根据权利要求8所述的富含大豆多肽的膳食补充剂的制备方法,其特征在于,所述的环糊精为α-环糊精或其改性衍生物、β-环糊精或其改性衍生物、γ-环糊精或其改性衍生物中的任意一种或几种的组合。
10.根据权利要求1-9任意一项所述的制备方法制得的富含大豆多肽的膳食补充剂。
