Abstract
本发明公开了一种提升机体对外邪抵抗力的组合物及其制备方法。该组合物是由鸡血藤、牛尾菜、黑老虎根、香附、牛膝、穿破石、乌药、山香、羊耳菊、地耳草为原料制备而成的。动物药效学结果显示:在调节小鼠免疫功能实验中,能显著提高加Con?A和不加Con?A脾细胞培养液之间的吸光度差值;提高小鼠胸腺和脾脏器指数。提示本组合物可能是通过促进胸腺和脾脏的发育,促进T淋巴细胞的分化和B淋巴细胞的产生而提高免疫功能。明显优于模型组。可以认为本发明能促进小鼠T淋巴细胞的分化和B淋巴细胞的产生,从而提高小鼠的免疫功能。

本发明涉及医药卫生领域,具体涉及一种提升机体对外邪抵抗力的组合物及其制备方法。
背景技术
中医理论认为一切致病因素为邪气,一切抗病能力为正气。正气,又分为卫气、元气和主要组织器官的脏腑之气等。所谓卫气是能起防御外邪侵犯人体的正气。如果正气旺盛,机体就能祛邪外出,免于生病,即所谓“正气内存,邪不可干”。如果正气虚衰,则邪气容易侵入机体而引起疾病,即所谓致病是由于“邪之所揍,其气必虚”。因此中医在防病治病过程中提出的扶正祛邪以固本的基本原则,与增强机体免疫力的保健方法是一致的。
免疫系统是覆盖全身的防卫网络,保护身体的第一道防线。免疫系统(immunologicfunction)是生物机体在种系进化和个体发育过程中逐步获得的防卫能力。主要包括三个方面:机体抵抗外界传染性因子的免疫防护功能;机体清除损伤和死亡细胞,维持自身生理平衡的自身稳定功能;对机体监视,发现并清除突变细胞的免疫监视功能。在正常生理条件下,机体的免疫系统依靠其先天具有的免疫能力及后天获得的免疫能力发挥共同免疫作用,保持机体的生理功能相对稳定。若免疫功能发生异常,必然会导致机体生理功能的失调,从而出现病理性变化。

中国专利申请CN200710114135.0公开一种强力提高免疫力膏药及其制备方法,药物组成:主药包括黄连、黄芩、干姜、黄柏、附子、胡椒各50~75g等,辅药包括三部分,第一部分为丁香、冰片各30g~50g等,第二部分为乳香、没药、丁香、制川椒、砂仁、枳实、青皮各70~100g等,第三部分为硇砂、阿魏各100g~130g等。其制备方法为:先将主药用熟油浸泡、油榨、再自然冷却,然后滤除药渣,与黄丹充分化合均匀后倒入水内浸泡;将辅药第一部分研粉待用;将辅药第二部分研粉待用;将辅药第三部分放入水中慢火煎药,将获得的药汤浓缩后倒入辅药第二部分研粉中搅拌均匀,晒干后粉碎为粉末备用;将药膏加热,再与第一部分研粉、晒干后粉碎的粉末按比例混合均匀,即为免疫膏药。

现有技术方案组方太复杂,服用群体限制较大,需要提供一种顺应市场需要,方便较大人群服用的药物。

本发明所述的提升机体对外邪抵抗力的组合物,是由以下重量份的原料制备而成的:鸡血藤70-82份、牛尾菜70-82份、黑老虎根135-170份、香附135-170份、牛膝70-82份、穿破石70-82份、乌药70-82份、山香70-82份、羊耳菊70-82份、地耳草70-82份。
优选地,是由以下重量份的原料制备而成的:鸡血藤76份、牛尾菜76份、黑老虎根153份、香附153份、牛膝76份、穿破石76份、乌药76份、山香76份、羊耳菊76份、地耳草76份。
以下是本发明的药材来源:
鸡血藤:为豆科植物密花豆SpatholobussuberectusDunn的干燥藤茎。秋、冬二季采收,除去枝叶,切片,晒干。【性味与归经】苦、甘,温。归肝、肾经。【功能与主治】活血补血,调经止痛,舒筋活络。用于月经不调,痛经,经闭,风湿痹痛,麻木瘫痪,血虚萎黄。
牛尾菜:菝葜科植物牛尾菜SmilaxripariaA.DC.的干燥根机根茎。夏、秋季采挖,除去藤茎及泥沙,晒干。【性味】甘、苦、平。【功能与主治】舒筋通络,补气活血,祛痰止咳。用于筋骨疼痛,气虚浮肿,跌打损伤,咳嗽吐血。
黑老虎根:为木兰科植物厚叶五味子Kadsuracoccinea(Lem.)A.C.Smith的干燥根。收载于《中国药典》1977年版。或海风藤,即异型南五味子Kadsuraheteroclita(Roxb.)Craib的干燥藤茎,收载于《广西中药材标准》。【性味】甘、微辛,温。【功能与主治】祛风散寒,行气止痛,舒筋活络。用于风湿性关节炎,腰肌劳损,感冒,产后风瘫。
香附:本品为莎草科植物莎草CyperusrotundusL.的干燥根茎。秋季采挖,燎去毛须,置沸水中略煮或蒸透后晒干,或燎后直接晒干。【性味与归经】辛、微苦、微甘,平。归肝、脾、三焦经。【功能与主治】疏肝解郁,理气宽中,调经止痛。用于肝郁气滞,胸胁胀痛,疝气疼痛,乳房胀痛,脾胃气滞,脘腹痞闷,胀满疼痛,月经不调,经闭痛经。
牛膝:本品为苋科植物牛膝AchyranthesbidentataBL.的干燥根。冬季茎叶枯萎时采挖,除去须根及泥沙,捆成小把,晒至干皱后,将顶端切齐,晒干。【性味与归经】苦、甘、酸,平。归肝、肾经。【功能与主治】逐瘀通经,补肝肾,强筋骨,利尿通淋,引血下行。用于经闭,痛经,腰膝酸痛,筋骨无力,淋证,水肿,头痛,眩晕,牙痛,口疮,吐血,衄血。
穿破石:本品为桑科植物构棘Cudraniacochinchinensis(Lour.)KudoetMasam.的干燥根。全年均可采挖,除去须根,洗净,切片或段、晒干。【性味】微苦,凉。【功能与主治】止咳,退黄,活血,通络。用于肺结核,湿热黄疸,胁肋疼痛,跌扑瘀痛,风湿痹痛。
乌药:本品为樟科植物乌药Linderaaggregata(Sims)Kosterm.的干燥块根。全年均可采挖,除去细根,洗净,趁鲜切片,晒干,或直接晒干。【性味与归经】辛,温。归肺、脾、肾、膀胱经。【功能与主治】行气止痛,温肾散寒。用于寒凝气滞,胸腹胀痛,气逆喘急,膀胱虚冷,遗尿尿频,疝气疼痛,经寒腹痛。
山香:本品为唇形科植物山香Hyptissuaveolens(L.)Poit.的干燥全草。夏秋季采收,除去杂质,阴干。【性味】辛、苦,平。【功能与主治】疏风散瘀,行气利湿,解毒,止痛。用于感冒头痛,胃肠胀气,风湿骨痛,跌打肿痛,创伤出血,痈肿疮毒,虫蛇咬伤,湿疹,皮炎。
羊耳菊:本品为菊科植物羊耳菊Inulacappa(Buch.-Ham.)DC.的干燥地上部分。夏、秋季采割,除去杂质,干燥。【性味】辛、微苦,温。归肺、肝、胃经。【功能与主治】祛风,利湿,行气化滞。用于风湿关节痛,胸膈痞闷,疟疾,痢疾,泄泻,产后感冒,肝炎,痔疮,疥廯。
地耳草:本品为藤黄科植物地耳草HypericumjaponicumThunb.的干燥全草。春、夏二季花开时采挖,除去杂质,晒干。【性味与归经】苦、辛、平。归肝、脾经。【功能与主治】清利湿热,散瘀消肿。用于急、慢性肝炎,疮疖痈肿。
本发明的另一方面是提供了本发明中药组合物的活性成分的制备方法,该方法是采用水提取。具体的制备方法如下:
取鸡血藤、牛尾菜、黑老虎根、香附、牛膝、穿破石、乌药、山香、羊耳菊、地耳草,加水提取2-3次,每次加水量相当于药材总重量的6-12倍,每次提取时间为1-3小时,合并提取液,滤过,滤液浓缩至60-80℃时相对密度为1.05-1.20的清膏,即得。
本发明所述的提升机体对外邪抵抗力的组合物,可以与医药学上可接受的载体配合,制成各种常用制剂,如片剂、颗粒剂、胶囊剂、咀嚼片、丸剂等。
本发明的药学上可接受的载体包括但不限于以下:
稀释剂:淀粉、糖粉、乳糖、糊精、微晶纤维素、无机盐、糖醇类等。
润湿剂与粘合剂:纯化水、乙醇、明胶、聚乙二醇、纤维素衍生物等。
崩解剂:淀粉、羧甲基淀粉钠、纤维素衍生物、交联聚维酮等。
润滑剂:硬脂酸镁、微粉硅胶、滑石粉、聚乙二醇等。
助溶剂:水、乙醇、甘油、丙二醇、液状石蜡、植物油等。
矫味剂:蔗糖、单糖、芳香剂等。
防腐剂:苯甲酸、山梨酸、甲酯、乙酯、丙酯等。
方解:本方证由六淫之邪侵袭人体,卫外不固;或起居无常,熬夜伤津,致气血逆乱等。以上原因,均可耗伤气血,使气滞血瘀,致体质渐衰,抵抗力低下。本方主要以活血化瘀,益气通络为调理之则,提升机体免疫力。方中重用鸡血藤为君,鸡血藤入肝经,其性苦而不燥,温而不烈,为强补气血之品,且兼行血散瘀、舒经活络之功,可助行气血,两擅其功。牛尾菜性味甘,平,功能补气活血,舒经活络,可助君药行药势;黑老虎根味辛,性温,善行经络气血及温养经脉,助君药畅行气血;香附味辛、苦,能散能收,尤善调理气机,使气血运行有常;牛膝归肝、肾经,其活血祛瘀之力较强,又可补益肝肾,以旺正气。牛膝性善下行,善引气血下注,荣养腰膝;穿破石能舒筋活血,助君药药势,又能清热解毒,抵御外邪;以上五味药共为臣药。方中佐以乌药、山香、羊耳菊和地耳草。乌药气雄性温,能行气止痛,以其气雄而善可携诸药奏活血化瘀,益气通络之效,本品外解表而理肌,内宽中而顺气,以之祛邪气则天行疫瘴即却,故于机体存正气,御邪气大有裨益;山香散瘀之效佳,又能祛风解表、清热,御风热之邪于外;羊耳菊功能散寒解表、解毒,实腠理,用治本证可防外感风寒之邪;地耳草苦凉,入肝、胆经,即能清热解毒,又兼活血之功,用治本证尤为适宜。以上诸药合用,标本兼顾,共奏活血化瘀,益气通络之功,提升机体对外邪的抵抗力。
本发明提供的一种提升机体对外邪抵抗力的组合物具有以下优点:
1、本发明处方中各药合用,共同起到活血化瘀,益气通络之功,之功的作用,能够起到较好的提升机体对外邪抵抗力的作用。
2、在小鼠免疫功能调节实验中,与模型组相比,给予本发明后的小鼠,胸腺及脾脏的脏器指数明显提高,免疫调节功能显著增强,且效果优于现有技术。
3、本发明组合物配方合理创新,生产成本低、工艺简单。
具体实施方式
下面通过实施例进一步说明本发明。应该理解的是,本发明的实施例是用于说明本发明而不是对本发明的限制。根据本发明的实质对本发明进行的简单改进都属于本发明要求保护的范围。
实施例1
取鸡血藤82g、牛尾菜82g、黑老虎根170g、香附170g、牛膝82g、穿破石82g、乌药82g、山香82g、羊耳菊82g、地耳草82,加水提取3次,每次加水量相当于药材总重量的6倍,每次提取时间为1小时,合并提取液,滤过,滤液浓缩至60℃时相对密度为1.05的清膏,干燥成干膏粉,粉碎,加入辅料淀粉、蔗糖粉、滑石粉、硬脂酸镁,制得颗粒剂。
实施例2
取鸡血藤70g、牛尾菜70g、黑老虎根135g、香附135g、牛膝70g、穿破石70g、乌药70g、山香70g、羊耳菊70g、地耳草70,加水提取2次,每次加水量相当于药材总重量的8倍,每次提取时间为2小时,合并提取液,滤过,滤液浓缩至70℃时相对密度为1.10的清膏,干燥成干膏粉,粉碎,加入辅料淀粉、蔗糖粉、滑石粉、硬脂酸镁,制得颗粒剂。
实施例3
取鸡血藤76g、牛尾菜76g、黑老虎根153g、香附153g、牛膝76g、穿破石76g、乌药76g、山香76g、羊耳菊76g、地耳草76g,加水提取2次,每次加水量相当于药材总重量的12倍,每次提取时间为1.5小时,合并提取液,滤过,滤液浓缩至80℃时相对密度为1.05的清膏,干燥成干膏粉,粉碎,加入辅料淀粉、乙醇、滑石粉,制得片剂。
药效学试验:
一、对小鼠免疫功能的影响
1材料
1.1药物
本发明实施例1-3、胸腺肽肠溶片,批号:141230,哈高科白天鹅药业集团有限公司。
1.2动物
ICR小鼠,雄性,18-22g,由广东省医学实验动物中心提供,动物生产许可证SCXK(粤)2003-0002。
1.3试剂与仪器
RPMI1640完全培养基,批号150128,南京凯基生物科技发展有限公司。Hank’s液,批号141209,sigma。刀豆蛋白A(ConA),批号141108,sigma。酶标仪,型号MultiskanMK3,Thermo。CO2培养箱,型号INC108,memmert。
2方法
2.1对ConA诱导的小鼠淋巴细胞转化试验
ICR小鼠50只,雄性,随机分为对照组、胸腺肽阳性组(0.5mg/kg)和实施例组(5.0g/kg),每组10只。对照组小鼠给予等容积生理盐水,给药容积均为20mL/kg。每天灌胃给药1次,连续给药30d。
末次给药30min后脱颈椎处死,称体质量。无菌取脾和胸腺,称湿重,计算脏器指数。无菌取脾和胸腺。将脾脏置于盛有适量无菌Hank’s液的小平皿中,用镊子轻轻撕碎,制成单细胞悬液,200目筛网过滤,洗涤,计数,最后用RPMI1640完全培养液调整细胞浓度为3×106个/mL。将细胞悬液分2孔加入24孔培养板中,每孔1mL。一孔加75μLConA液,另一孔不加ConA液作为对照,置于37℃、5%CO2培养箱中培养72h。培养结束前4h,每孔轻轻吸去上清液0.7mL,加入0.7mL不含小牛血清的RPMI1640培养液,同时加入MTT50μL。培养结束后,每孔加入1mL酸性异丙醇,吹打混匀。在酶标仪上测OD值。酶标仪的参数设定:波长为570nm,带宽为9nm,闪光次数为5,设定时间为5ms。计算加ConA孔吸光度与不加ConA孔吸光度的差值。
2.3统计方法
所有实验数据均以表示。计量数据采用SPSS13.0统计软件进行方差分析及组间检验。P<0.05有统计学意义。
3结果
各组脾和胸腺的脏器指数、吸光度差值的原始数据均符合方差齐性要求。与对照组比较,经LSD检验,实施例组均能显著增加吸光度差值及胸腺指数、脾脏指数(P<0.01或P<0.05);与阳性组比较,实施例3组能显著增加吸光度差值及脾脏指数,差异显著(P<0.05),实施例组均能显著增加胸腺指数(P<0.01)。结果见表1。

4讨论
脾脏和胸腺为机体重要淋巴器官。脾内淋巴细胞总数的55%为B淋巴细胞,还拥有25%的全身循环T淋巴细胞。胸腺则是T淋巴细胞分化成熟的地方。当T淋巴细胞受ConA刺激后母细胞发生增殖反应,活细胞特别是增殖细胞中的线粒体水解反应细胞的增殖情况。结果表明,本发明能显著提高加ConA和不加ConA脾细胞培养液之间的吸光度差值;提高小鼠胸腺和脾脏器指数。提示本发明可能是通过促进胸腺和脾脏的发育,促进T淋巴细胞的分化和B淋巴细胞的产生而提高免疫功能。
结论:本发明中药组合物可显著提高加ConA和不加ConA脾细胞培养液之间的吸光度差值;提高小鼠胸腺和脾脏器指数。提示本发明可能是通过促进胸腺和脾脏的发育,促进T淋巴细胞的分化和B淋巴细胞的产生而提高免疫功能。
虽然,上文中已经用一般性说明、具体实施方式及试验,对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
Claims (5)
1.一种提升机体对外邪抵抗力的组合物,其特征在于它是由如下重量份的原料制备而成的:鸡血藤70-82份、牛尾菜70-82份、黑老虎根135-170份、香附135-170份、牛膝70-82份、穿破石70-82份、乌药70-82份、山香70-82份、羊耳菊70-82份、地耳草70-82份。
2.如权利要求1所述的提升机体对外邪抵抗力的组合物,其特征在于它是由如下重量份的原料制备而成的:鸡血藤76份、牛尾菜76份、黑老虎根153份、香附153份、牛膝76份、穿破石76份、乌药76份、山香76份、羊耳菊76份、地耳草76份。
3.如权利要求1或2所述的提升机体对外邪抵抗力的组合物,其特征在于其活性成分是采用水提或40—80%体积百分浓度的乙醇提取制备而成的。
4.如权利要求3所述的提升机体对外邪抵抗力的组合物,其特征在于其活性成分是通过以下方法制备而成的:取鸡血藤、牛尾菜、黑老虎根、香附、牛膝、穿破石、乌药、山香、羊耳菊、地耳草,加水提取2-3次,每次加水量相当于药材总重量的6-12倍,每次提取时间为1-3小时,合并提取液,滤过,滤液浓缩至60-80℃时相对密度为1.05-1.20的清膏,即得。
5.一种提升机体对外邪抵抗力的中药制剂,由权利要求1或2所述的提升机体对外邪抵抗力的组合物与医药行业上可接受的载体配合制备而成。
