Abstract
本发明涉及一种杀菌组合物,具体地讲是一种含有枯草芽孢杆菌与醚菌酯的杀菌剂配方,以及该杀菌剂的制备方法。该杀菌组合物,由下述重量百分含量的组分组成:枯草芽孢杆菌母粉1~25%,醚菌酯10%~30%,余量为助剂。本发明提供了一种含有枯草芽孢杆菌与醚菌酯的杀菌剂,可提高药效、延长药剂使用时间、降低使用成本、协同增效作用显著、对病害的防治效果好,主要用于防治番茄枯萎病,番茄青枯病,小麦赤霉病、白粉病、番茄炭疽病,葡萄白粉病、灰霉病,大豆根腐病,苹果霉心病,棉花立枯病,水稻稻瘟病等病害。

技术领域
本发明涉及一种杀菌组合物,具体地讲是一种含有枯草芽孢杆菌与醚菌酯的杀菌剂配方,以及该杀菌剂的制备方法。
背景技术
枯草芽孢杆菌,英文名(Bacillussubtilis),是芽孢杆菌属的一种,革兰氏阳性菌,菌落表面粗糙不透明,污白色或微黄色。研究证实枯草芽孢杆菌能够产生枯草菌素、多粘菌素、制霉菌素、短杆菌肽等多种具有不同结构和功能的抗菌物质,这些抗菌物质对致病菌或内源性感染的条件致病菌有明显的抑制作用,抗菌谱广,可防治多种植物病害。枯草芽孢杆菌还有以下优点:有较强的竞争和定殖能力,抢占病原菌的生活空间,改善土壤微生物环境,阻止和干扰病原菌对植物叶面和其他器官的侵染;能分泌溶解酶腐蚀线虫表皮;可分解有机质为作物提供养分,促进作物生长;对人畜无毒无害,不污染环境。由于枯草芽孢杆菌具有生长快、营养简单以及产生耐热、抗逆芽孢等突出特征,使其有利于生防菌剂在生产、剂型加工及在环境中的存活、定殖与繁殖,并且批量生产工艺简单,成本较低,施用方便,储存期长,现已成为种理想的生防细菌。
醚菌酯(kresoxim-methyl)又称(E)-2-甲氧亚氨基-[2-(邻甲基苯氧基甲基)苯基]乙酸甲酯,是一种高效、广谱、新型杀菌剂。具有保护、治疗、铲除、渗透和内吸活性,与其它常用的杀菌剂无交互抗性,且比常规杀菌剂持效期长。具有高度的选择性,对作物、人畜及有益生物安全,对环境基本无污染。醚菌酯对半知菌、子囊菌、担子菌、卵菌纲等真菌引起的多种病害具有很好的活性。
然而,经验告诉我们农药如果长期单一使用,都不可避免地产生抗性,杀菌剂也不例外。据最近有关杂志报到醚菌酯已在德国产生抗性。因此,通过复配渠道,采用醚菌酯与枯草芽孢杆菌复配的杀菌组合物将是抵御抗性、延缓抗性发展的有效途径。醚菌酯与枯草芽孢杆菌复配的另一作用是通过有目的地引进枯草芽孢杆菌,恶化病菌生存环境,形成优势互补,既能发挥化学药剂高效、快速的抑菌作用,又能发挥草芽孢杆菌在诱导植物抗病性、持久地控制作物土传病害流行的目的。实验证明醚菌酯对枯草芽孢杆菌有促进生长的作用,并不影响枯草芽孢杆菌发挥其有效的作用机制。且有关醚菌酯与枯草芽孢杆菌复配国内外均无报道。
发明内容
本发明提供了一种可提高药效、延长药剂使用时间、降低使用成本、协同增效作用显著、对病害的防治效果好的含有枯草芽孢杆菌与醚菌酯的杀菌剂。
本发明提供的枯草芽孢杆菌,分类命名:枯草芽孢杆菌XM16(BacillussubtilisXM16)从保藏单位:中国微生物菌种保藏管理委员会普通微生物中心(英文简称CGMCC)购买,保藏编号:CGMCCNO.0372,保藏日期:1998年12月2日。
本发明提供了所述含有枯草芽孢杆菌与醚菌酯的杀菌剂的可湿性粉剂、水悬浮剂及其制备方法。
本发明提供了含有枯草芽孢杆菌与醚菌酯的杀菌剂,其中先把枯草芽孢杆菌制成母粉,方法如下:将枯草芽孢杆菌XM16(Bacillus.subtilisXM16)经过斜面培养、摇床扩大培养、种子罐发酵和发酵罐发酵;将菌种的发酵菌液在吸附搅拌设备中用麦饭石进行吸附,吸附比例为按菌液∶麦饭石=1:6(L/kg),然后搅拌均匀,得到枯草芽孢杆菌粉剂,其中含菌量10-1000亿CFU/g。
本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂,由下述重量百分含量的组分组成:
枯草芽孢杆菌母粉1~25%,
醚菌酯10%~30%,
载体20%~81%,
分散剂3%~15%,
润湿剂1%~5%,
渗透剂2%~8%。
其中载体为高岭土、硅藻土、绢云母粉、膨润土、白炭黑、轻质碳酸钙中的一种或两种以上任意混合。分散剂为木质素磺酸钠、木质素磺酸钙、烷基酚聚氧乙烯基磷酸酯、三聚磷酸钠中的一种或两种以上任意混合。润湿剂为十二烷基硫酸钠、十二烷基苯磺酸、二丁基萘磺酸钠、琥珀酸二辛酯磺酸钠中的一种或两种以上任意混合;渗透剂选自脂肪醇聚氧乙烯醚、酯磺酸盐、有机硅中的一种或两种以上任意混合。所述载体优选为高岭土、白炭黑。所述分散剂优选为木质素磺酸钠。所述润湿剂优选为十二烷基苯磺酸。所述渗透剂优选为脂肪醇聚氧乙烯醚。所述枯草芽孢杆菌母粉的重量百分含量优选为10%~22%,所述醚菌酯的重量百分含量优选为12%~25%,所述载体的重量百分含量优选为30%~70%,所述分散剂的重量百分含量优选为3%~10%,所述润湿剂的重量百分含量优选为2%~4%,所述渗透剂的重量百分含量优选为3%~5%。

其中分散剂为木质素磺酸钠、木质素磺酸钙、烷基酚聚氧乙烯基磷酸酯、三聚磷酸钠中的一种或两种以上任意混合。润湿剂为十二烷基硫酸钠、十二烷基苯磺酸、二丁基萘磺酸钠、琥珀酸二辛酯磺酸钠中的一种或两种以上任意混合;增稠剂为阿拉伯胶、黄原胶、羟甲基纤维素聚乙烯醇、聚乙烯醇吡咯烷酮、硅酸镁铝和膨润土的一种或两种以上任意混合;增效剂为油酸甲酯、有机硅、渗透剂中的一种或两种以上任意混合。稳定剂为环氧氯丙烷、环氧大豆油、多烷基取代芳烃、二甘醇、三甘醇、邻二氯苯的的一种或两种以上任意混合。防冻剂为乙二醇或丙三醇中的一种或两种混合。水为自来水、蒸馏水或去离子水中的一种。所述分散剂优选为木质素磺酸钙,所述润湿剂优选为十二烷基硫酸钠,所述增稠剂优选为黄原胶,所述增效剂优选为有机硅,所述稳定剂优选为环氧大豆油,所述防冻剂优选为乙二醇。所述枯草芽孢杆菌母粉的重量百分含量优选为10%~22%,所述醚菌酯的重量百分含量优选为12%~25%,所述分散剂的重量百分含量优选为3%~8%,所述润湿剂的重量百分含量优选为2%~4%,所述增稠剂的重量百分含量优选为3%~8%,所述增效剂的重量百分含量优选为3%~6%,所述稳定剂的重量百分含量优选为1%~4%,所述防冻剂的重量百分含量优选为2%~5%。
本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂的制备方法,是将枯草芽孢杆菌母粉、醚菌酯、高岭土、木质素磺酸钠和脂肪醇聚氧乙烯醚投入螺旋搅拌机中搅拌均匀,再放入粉碎机中粉碎。即为枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂。
本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂的制备方法,是将枯草芽孢杆菌母粉、醚菌酯、分散剂、润湿剂、增稠剂、增效剂、稳定剂、防冻剂和水等按投料比装入带高剪切搅拌的反应釜中,粉碎混合,直到混合体成均匀粘稠液是停止搅拌;经搅拌粉碎的物料通入砂磨机中,通冷却水进行砂磨超微粉碎,终点粒径<3μm,即为枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂。
通过实验发现醚菌酯对枯草芽孢杆菌有促进生长的作用,枯草芽孢杆菌增长率达30%以上,并不影响枯草芽孢杆菌发挥其有效的作用机制。
本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌剂主要用于防治番茄枯萎病,番茄青枯病,小麦赤霉病、白粉病、番茄炭疽病,葡萄白粉病、灰霉病,大豆根腐病,苹果霉心病,棉花立枯病,水稻稻瘟病等病害。
具体实施方式
实施例一把枯草芽孢杆菌XM16(BacillussubtilisXM16)用NA培养基(牛肉浸膏3.0g,酵母膏1g,蛋白胨5g,葡萄糖10g,琼脂20g,蒸馏水1000mL)28℃振荡(150r/m)培养2d,备用。把20%醚菌酯稀释500×、1000×、2000×置于NA培养基中,把培养好的枯草芽孢杆菌XM16稀释109倍涂布于配药的NA培养基中,设清水为对照,分别做三个重复,实验结果见表1。

其中增生率为增加生长作用的一个指数,增生率=(处理菌生长菌株数-对照菌生长菌株数)/对照菌生长菌株数×100%。实验结果表明醚菌酯有促进枯草芽孢杆菌XM16生长的作用。
实施例二枯草芽孢杆菌母粉的制备
将枯草芽孢杆菌XM16(BacillussubtilisXM16)经过斜面培养、摇床扩大培养、种子罐发酵和发酵罐发酵;将菌种的发酵菌液20L在吸附搅拌设备中用麦饭石120kg进行吸附,然后搅拌均匀,进行粉碎,得到枯草芽孢杆菌母粉,其中要求含菌量10-1000亿CFU/g。
实施例三本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂,由下述重量百分含量的组分组成:
枯草芽孢杆菌母粉15%,
醚菌酯15%,
载体60.5%,
分散剂4.5%,
润湿剂2%,
渗透剂3%。
在反应釜中加入15千克枯草芽孢杆菌母粉、15千克醚菌酯、60.5千克高岭土、4.5千克木质素磺酸钠、2千克十二烷基苯磺酸和3千克脂肪醇聚氧乙烯醚高速搅拌均匀,30分钟后取出反应物放入粉碎机中粉碎,过筛。所得物即是枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂。
实施例四本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂,由下述重量百分含量的组分组成:
枯草芽孢杆菌母粉15%,
醚菌酯15%,
分散剂5%,
润湿剂2%,
增稠剂4%,
增效剂3%,
稳定剂1%,
防冻剂3%,
余量加水至100%。
在反应釜中加入15千克枯草芽孢杆菌母粉、15千克醚菌酯、5千克木质素磺酸钙、2千克十二烷基硫酸钠、4千克黄原胶、3千克有机硅、1千克环氧大豆油、3千克乙二醇和30千克自来水装入带高剪切搅拌的反应釜中,粉碎混合,直到混合体成均匀粘稠液是停止搅拌1小时后取出的反应物,经搅拌粉碎的物料通入砂磨机中,通冷却水进行砂磨超微粉碎,终点粒径<3μm。即是枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂。
实施例五本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂,由下述重量百分含量的组分组成:
枯草芽孢杆菌母粉20%,
醚菌酯20%,
载体49%,
分散剂5%,
润湿剂2.5%,
渗透剂3.5%。
在反应釜中加入20千克枯草芽孢杆菌母粉、20千克醚菌酯、49千克高岭土、5千克木质素磺酸钠、2.5千克十二烷基苯磺酸和3.5千克脂肪醇聚氧乙烯醚高速搅拌均匀,30分钟后取出反应物放入粉碎机中粉碎,过筛。所得物即是枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂。
实施例六本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂,由下述重量百分含量的组分组成:
枯草芽孢杆菌母粉20%,
醚菌酯20%,
分散剂6%,
润湿剂3.5%,
增稠剂4.6%,
增效剂4%,
稳定剂2%,
防冻剂4%,
余量加水至100%。
在反应釜中加入20千克枯草芽孢杆菌母粉、20千克醚菌酯、6千克木质素磺酸钙、3.5千克十二烷基硫酸钠、4.6千克黄原胶、4千克有机硅、2千克环氧大豆油、4千克乙二醇和35千克自来水装入带高剪切搅拌的反应釜中,粉碎混合,直到混合体成均匀粘稠液是停止搅拌1小时后取出的反应物,经搅拌粉碎的物料通入砂磨机中,通冷却水进行砂磨超微粉碎,终点粒径<3μm。即是枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂。
实施例七用实施例三至六所配制的枯草芽孢杆菌与醚菌酯的杀菌剂对番茄枯萎病菌和青枯病菌进行抑菌圈试验。设实施例三所配制的30%枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂为A;实施例四所配制的30%枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂为B;实施例五所配制的400%枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂为C;实施例六所配制的40%枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂为D。
供试病原菌菌株:
番茄枯萎病菌由海南省农科院植保所提供。用PDA培养基(200g马铃薯、20g葡萄糖、20g琼脂和1000mL蒸馏水,pH5.6~6.6)28℃培养5d,备用。
番茄青枯病菌由海南省农科院植保所提供。用NA培养基(牛肉浸膏3.0g,酵母膏1g,蛋白胨5g,葡萄糖10g,琼脂20g,蒸馏水1000mL)28℃振荡(150r/m)培养2d,备用。
在PDA平板中央放置枯萎病菌菌块,直径为7mm;培养皿四周呈十字架型放置4张滤纸片(离培养皿中心距离为22.5mm),每张滤纸片上加入不同稀释倍数的A、B、C;设清水对照。每处理设3个重复。28℃恒温培养,待对照平板上病原菌菌落长满时,测量处理菌落生长直径。计算抑制率:抑制率(%)=[(对照菌落直径-处理菌落直径)/(对照菌落直径-7)]×100%。
吸取青枯菌液涂布NA平板(0.3mL菌液/平板)。培养皿中央放置1张灭菌滤纸片,滤纸片上加入不同稀释倍数的的A、B、C、D;设清水对照。每处理设3个重复。28℃恒温培养2d,测量抑菌圈直径,试验结果见表2。

从结果得知本发明的枯草芽孢杆菌与醚菌酯的杀菌剂有较好的杀菌效果,对番茄枯萎病抑制率达90%以上,对番茄青枯病也有较好的抑制效果。
实施例八用实施例三至六所配制的枯草芽孢杆菌与醚菌酯的杀菌剂防治番茄枯萎病和青枯病的田间小区效果。
试验时间:于2014年11月5日左右在海南省澄迈县金江大塘种植基地内进行。
试验品种:金丰1号。
试验药剂为实施例三至六所配制的枯草芽孢杆菌与醚菌酯的杀菌剂(依次设为A、B、C、D)及20%枯草芽孢杆菌母粉、20%醚菌酯,清水为对照。
田间药效试验:番茄苗移栽时(12月底)灌根处理,移栽后每隔两周左右(15d)灌根一次。整个生长期共需要用药8-9次。将实验药剂稀释1000倍、2000倍;每个处理设2次重复,设保护行。
3月上旬调查各个小区的番茄枯萎病和青枯病发病指数,分级标准如下:0级为茎内维管束正常,外部无症状;1级为茎内维管束1/4以下变色;3级为茎内维管束1/4~1/2变色;5级为茎内维管束1/2~3/4变色;7级为茎内维管束3/4以上变色,部分叶片萎蔫;9级为整株枯死。

防治效果(%)=[(对照的病情指数-防治的病情指数)/对照的病情指数]×100%
田间试验结果见表3

从表3可以看出,本发明的枯草芽孢杆菌与醚菌酯的杀菌剂,按不同的用量进行大田试验,药后其对番茄枯萎病和番茄青枯病的防治效果均优于20%枯草芽孢杆菌母粉和20%醚菌酯的防治效果,防治效果可达88%以上。且各用药处理对番茄均无药害。

1.本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂,由下述重量百分含量的组分组成:
枯草芽孢杆菌母粉1~25%,
醚菌酯10%~30%,
载体20%~81%,
分散剂3%~15%,
润湿剂1%~5%,
渗透剂2%~8%,
其特征是所述载体优选为高岭土、白炭黑,所述分散剂优选为木质素磺酸钠,所述润湿剂优选为十二烷基苯磺酸,所述渗透剂优选为脂肪醇聚氧乙烯醚;所述枯草芽孢杆菌母粉的重量百分含量优选为10%~22%,所述醚菌酯的重量百分含量优选为12%~25%,所述载体的重量百分含量优选为30%~70%,所述分散剂的重量百分含量优选为3%~10%,所述润湿剂的重量百分含量优选为2%~4%,所述渗透剂的重量百分含量优选为3%~5%。
2.根据权利要求1所述的一种含有枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂,其特征是将枯草芽孢杆菌母粉、醚菌酯、高岭土、木质素磺酸钠和脂肪醇聚氧乙烯醚投入螺旋搅拌机中搅拌均匀,再放入粉碎机中粉碎,即为枯草芽孢杆菌与醚菌酯的杀菌可湿性粉剂。
3.本发明所提供的枯草芽孢杆菌与醚菌酯的杀菌水悬浮剂,由下述重量百分含量的组分组成:
枯草芽孢杆菌母粉1~25%,
醚菌酯10%~30%,
分散剂2%~12%,
润湿剂1%~10%,
增稠剂2%~15%,
增效剂1%~10%,
稳定剂1%~8%,
防冻剂1%~7%,
余量加水至100%,
其特征是所述分散剂优选为木质素磺酸钙,所述润湿剂优选为十二烷基硫酸钠,所述增稠剂优选为黄原胶,所述增效剂优选为有机硅,所述稳定剂优选为环氧大豆油,所述防冻剂优选为乙二醇;所述水优选为为自来水、蒸馏水或去离子水中的一种,所述枯草芽孢杆菌母粉的重量百分含量优选为10%~22%,所述醚菌酯的重量百分含量优选为12%~25%,所述分散剂的重量百分含量优选为3%~8%,所述润湿剂的重量百分含量优选为2%~4%,所述增稠剂的重量百分含量优选为3%~8%,所述增效剂的重量百分含量优选为3%~6%,所述稳定剂的重量百分含量优选为1%~4%,所述防冻剂的重量百分含量优选为2%~5%。
