Abstract
本发明涉及一种人工授精液及其制备方法,具体为达氏鲟人工授精液及其制备方法,NaCl、KCl、MgCl2·6H2O、CaCl2、葡萄糖、Tris-base溶解在去离子水中形成混合溶液,用Hcl调节混合溶液的pH至7.8形成达氏鲟人工授精液;每500毫升的达氏鲟人工授精液中,含有0.585gNaCl、0.0019gKCl、0.5083gMgCl2·6H2O、0.2775gCaCl2、5.4g葡萄糖、1.2114g Tris-base。本发明提供的达氏鲟人工授精液,完全针对达氏鲟精子激活所需物质配制,由不同离子和大分子物质组成的,这些离子和大分子物质对精子的运动可以起到一定的优化作用,精子的激活率超过90%,精子的活力和寿命显著延长,精子的活力可从103s-123s提高到172s-185s,受精率从13%-29%提高到54%-58%。

技术领域
本发明涉及一种人工授精液及其制备方法,具体为达氏鲟人工授精液及其制备方法。
背景技术
达氏鲟又称长江鲟和沙腊子,是我国特有的淡水定居性鲟科鲟属鱼类,分布于长江上游和金沙江下游,国家I级重点保护动物和长江上游一级急切保护鱼类,近年来,由于过度捕捞、水利工程、航运及水体污染等诸多人类活动的影响,达氏鲟野生种群数量逐日减少,种群资源衰退严重,长江流域中的达氏鲟几近灭绝,为了保护这一珍稀鱼类资源,对达氏鲟进行人工繁殖研究,最终实现规模化人工养殖,势在必行。
达氏鲟生长较快,抗病力强,能适应于溪河、池塘、水库等环境,性周期短,口感较好,是引种、驯化养殖的优良品种,但是其受精率偏低,无法获得批量受精卵。现有技术是性成熟达氏鲟经注射催产激素后,采用两种人工授精方法授精,1.干法授精,采用的人工授精液为江水;2.半干法授精,采用的人工授精液为生理盐水。
达氏鲟在自然环境中繁殖受精以江水为介质,现有人工繁殖技术是将江水或生理盐水作为人工授精液使用,由于人工授精过程与达氏鲟自然繁殖过程存在一定的区别,因此,江水或生理盐水的理化指标无法完全满足人工授精要求,其受精率仅为自然条件下的10-30%左右。
同时达氏鲟卵属于强粘性卵类型,人工授精过程中卵遇水后极易粘成团或成块,而达氏鲟精液粘稠,精子密度高,精卵不易混合均匀,若精子活力不高,寿命不长,其受精率将受极大影响,很难获得批量受精卵。
并且江水中的离子成分、渗透压、pH值不确定,其用量控制不准确,存在随意性,且江水易受到各种污染,产生对精子的毒害作用,影响受精率;生理盐水中的钠离子虽然起调节授精液渗透压的作用,但精子的激活还与多种相关离子及pH等因素有关,此外,0.65%的生理盐水也并非达氏鲟精子适应的最适渗透压浓度,因此,采用生理盐水作为授精介质,效果也不理想。
发明内容
针对上述技术问题,本发明提供一种适用于达氏鲟的人工授精液,解决现阶段达氏鲟人工繁殖过程中受精率低的问题。
具体技术方案为:
达氏鲟人工授精液,由以下方法制得:
NaCl、KCl、MgCl2·6H2O、CaCl2、葡萄糖、三羟甲基氨基甲烷溶解在去离子水中形成混合溶液,用Hcl调节混合溶液的pH至7.8形成达氏鲟人工授精液;每500毫升的达氏鲟人工授精液中,含有0.585gNaCl、0.0019gKCl、0.5083gMgCl2·6H2O、0.2775gCaCl2、5.4g葡萄糖、1.2114g三羟甲基氨基甲烷即Tris-base。
达氏鲟人工授精液所用药品均为分析纯,人工授精前2-3小时配制,常温保存。
本发明提供的达氏鲟人工授精液,完全针对达氏鲟精子激活所需物质配制,由不同离子和大分子物质组成的,这些离子和大分子物质对精子的运动可以起到一定的优化作用,精子的激活率超过90%,精子的活力和寿命显著延长,精子的活力可从103s-123s提高到172s-185s,受精率从13%-29%提高到54%-58%。
具体实施方式
结合实施例,说明本发明具体实施方式。
选择性腺发育较成熟的达氏鲟雌雄鱼,雌鱼用LHRH-A2+鲤脑垂体,2针注射,2针间隔12h,雄鱼为1针注射。注射后鲟鱼放在流水池中待其性腺成熟。
达氏鲟精子的采集:选择性腺发育成熟的达氏鲟雄鱼,用干毛巾擦干生殖孔周围及腹部的水,用软管进行导精,精液充氧后在4℃进行保存待用。
达氏鲟人工采卵:雌鱼第2针注射后应检查池中雌鱼排卵情况,对已开始排卵的达氏鲟雌鱼用干毛巾揩干其生殖孔周围以及腹部,轻轻挤压腹部,将雌鱼的卵全部挤入干燥的容器中,常温备用。
NaCl、KCl、MgCl2·6H2O、CaCl2、葡萄糖、Tris-base溶解在去离子水中形成混合溶液,用Hcl调节混合溶液的pH至7.8形成达氏鲟人工授精液;每500毫升的达氏鲟人工授精液中,含有0.585gNaCl、0.0019gKCl、0.5083gMgCl2·6H2O、0.2775gCaCl2、5.4g葡萄糖、1.2114g Tris-base。
应用本发明提供的达氏鲟人工授精液和现有技术作为授精液,分3次对达氏鲟人工授精进行了对比试验,操作采用干法和半干法,同在气温23℃、水温21℃条件下进行。
结果显示,采用干法授精,江水作为授精液3次受精率分别为17.5%、12.3%、11.4%,平均受精率为13.73%;利用本发明提供的达氏鲟人工授精液,其受精率分别为55.3%、51.7%、57.8%,平均受精率为54.93%,较江水作为授精液提高了41.2%。
采用半干法授精,生理盐水作为授精液3次受精率分别为27.6%、28.3%、31.4%,平均受精率为29.1%;采用半干法利用本发明的达氏鲟人工授精液,其受精率分别为56.4%、60.2%、59.5%,平均受精率为58.7%,较生理盐水作为授精液提高了29.6%。

1.达氏鲟人工授精液,其特征在于,由以下方法制得:NaCl、KCl、MgCl2·6H2O、CaCl2、葡萄糖、三羟甲基氨基甲烷溶解在去离子水中形成混合溶液,用Hcl调节混合溶液的pH至7.8形成达氏鲟人工授精液;每500毫升的达氏鲟人工授精液中,含有0.585gNaCl、0.0019gKCl、0.5083gMgCl2·6H2O、0.2775gCaCl2、5.4g葡萄糖、1.2114g三羟甲基氨基甲烷。
