Abstract
本发明公开了一种从扁蓄草中提取扁蓄总黄酮的工业生产方法,制备工艺步骤如下:(一)将扁蓄药材粉碎20-40目,投到提取罐内,加入5-8倍原料重量的水,加温至80-90度,保温两小时,放出一次提取液,补水3-5倍,加温到80-90度,保温一小时,放出二次提取液,合并提取液,降温到60度。(二)提取液降温后经过大孔树脂吸附,直至水洗无糖色,再用50%-70%的酒精洗脱,收集酒精洗脱液直到流出液无色,洗脱液蒸馏回收酒精。(三)无酒精的水溶液用1倍到3倍的乙酸乙酯萃取2次,回收乙酸乙酯。(四)脱酯后的溶液,经喷雾、干燥、磨碎即得黄棕色粉体产品。本发明的有益效果:用水作提取溶剂,提取成本低,生产能耗低,溶剂残留无毒,工艺操作简单。

技术领域
本发明涉及中草药的提取方法,尤其是一种从扁蓄草中提取扁蓄总黄酮的工业生产方法。
背景技术
篇蓄为蓼科一年生或多年生草本植物,我国各地俗名有大萹蓄、鸟蓼、扁竹、竹节草、猪牙草、蓄辩、萹蔓、地萹蓄、粉节草、道生草 、萹蓄蓼、百节草、铁绵草、大蓄片、野铁扫把、路柳、斑鸠台、扁猪牙、乌要等,分布于全国各个省区,遍生在山坡、田野、路边。篇蓄全草含多种药用有效成分,主要为具有抗氧化性、清除体内过剩自由基、抗肿瘤等功效的扁蓄苷、槲皮苷、杨梅苷等黄酮类化合物,是常用利尿中草药之一。扁蓄的功能主要有利尿作用、抗菌作用、降压作用、提高子宫张力作用和止血作用以及降压、止泻、抗氧化和保肝作用。目前,提取扁蓄总黄酮的工业生产方法很少,在中国专利局专利授权公告号:CN102464686A,授权公告日2012年5月23日的专利文件中,公开了一种制备扁蓄苷的方法,其提取溶剂采用低碳醇,增加了提取成本,在萃取环节用水饱和正丁醇萃取,不是最理想的蒸馏方式,间接增加生产能耗,因此,研制一种提取成本低,生产效果最大化的提取扁蓄总黄酮的工业生产方法时目前急需解决的问题。
发明内容
为解决上述问题,本发明的目的是提供一种用水作提取溶剂,提取成本低,生产能耗低,溶剂残留无毒,工艺操作简单的从扁蓄草中提取扁蓄总黄酮的工业生产方法。
为实现上述目的,本发明的技术方案为:一种从扁蓄草中提取扁蓄总黄酮的工业生产方法,其中所述的总黄酮是指扁蓄苷、槲皮苷和杨梅苷化合物,其特征在于,制备工艺步骤如下:
(一)、将扁蓄药材粉碎20-40目,投到提取罐内,加入5-8倍原料重量的水,加温至80-90度,保温两小时,放出一次提取液,补水3-5倍,加温到80-90度,保温一小时,放出二次提取液,合并提取液,降温到60度。
(二)、提取液降温后经过大孔树脂吸附,直至水洗无糖色,再用50%-70%的酒精洗脱,收集酒精洗脱液直到流出液无色,洗脱液蒸馏回收酒精。
(三)、无酒精的水溶液用1倍到3倍的乙酸乙酯萃取2次,回收乙酸乙酯。
(四)、脱酯后的溶液,经喷雾干燥即得黄棕色粉体产品。
作为优选,所述步骤(二)中大孔树脂为LSA21、LSA10、PN700或DM-18。
作为优选,所述步骤(四)中所得黄棕色粉体产品中检测总黄酮含量采用分光光度法。
本发明的有益效果为用水作提取溶剂,经大孔树脂粗分离之后,用乙酸乙酯进一步提纯;水提取成本低,用乙酸乙酯萃取易于蒸馏、生产能耗低,并且溶剂残留无毒;适用的大孔树脂种类较多,大多数能吸附植物黄酮的树脂都可用来吸附篇蓄类黄酮;产品中重金属、农残、微生物等极低,符合纯天然要求,提取物可用作药品原料、保健品原料。
具体实施方式
1、实验室实施例:篇蓄药材全草50克,粉碎到20-40目,投入500毫升园底烧瓶中,加水300毫升,加温到80度,保温2小时,放出一次提取液;补水200毫升,加热到80度,维持1小时,放出二次提取液。
合并提取液得750毫升,冷却到60度,用PN700大孔树脂200克吸附,清水洗到流出液无色。
加入70%酒精500毫升洗脱,在旋转蒸发器上蒸发掉酒精。得到200毫升水溶液。
向水溶液加入乙酸乙酯300毫升萃取,取酯相;水相中再加乙酸乙酯100毫升萃取,合并酯相。
在旋转蒸发器上蒸发乙酸乙酯到彻底干燥,得干物质7.2克,总黄酮含量42.5%。
2、工业化生产实施例:篇蓄药材全草800公斤,粉碎到20-40目,投入6立方多功能提取罐中,加水4.5吨,加温到90度,保温2小时,放出一次提取液;补水3吨,加热到90度,维持1小时,放出二次提取液。
合并提取液得6.5吨水溶液,冷却到60度,用LSA21大孔树脂500公斤吸附,清水洗到流出液无色。在1000升/小时的真空浓缩器内蒸发酒精。得到300公斤水溶液。
向水溶液加入乙酸乙酯500公斤萃取,取酯相;再加乙酸乙酯300公斤萃取,合并酯相。
在1000升/小时的真空浓缩器上蒸发乙酸乙酯,当酯蒸尽时,补加水使浓缩液的密度为1.15克/毫升。
在喷雾干燥器中,进风温度280度、出风温度105度、离心喷雾机转速16000转/分的条件下,得到黄棕色粉体产品112公斤,检测其总黄酮含量40.8%。以上所述仅是本发明的优选实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱本发明技术原理的前提下,还可以做出若干改进和变型,这些改进和变型也应视为本发明的保护范围。

1.一种从扁蓄草中提取扁蓄总黄酮的工业生产方法,其中所述的总黄酮是指扁蓄苷、槲皮苷和杨梅苷化合物,其特征在于,制备工艺步骤如下:
(一)、将扁蓄药材粉碎20-40目,投入提取罐内,加入5-8倍原料重量的水,加温至80-90度,保温两小时,放出一次提取液;补水3-5倍,加温到80-90度,保温一小时,放出二次提取液,合并提取液,降温到60度;
(二)、提取液降温后经过大孔树脂吸附,直至水洗无糖色,再用50%-70%的酒精洗脱,收集酒精洗脱液直到流出液无色,洗脱液蒸馏回收酒精;
(三)、无酒精的水溶液用1倍到3倍的乙酸乙酯萃取2次,回收乙酸乙酯;
(四)、脱酯后的溶液,经喷雾干燥即得黄棕色粉体产品。
2.根据权利要求1所述的一种从扁蓄草中提取扁蓄总黄酮的工业生产方法,其特征在于,所述步骤(二)中大孔树脂为LSA21、LSA10、PN700或DM-18。
3.根据权利要求1所述的一种从扁蓄草中提取扁蓄总黄酮的工业生产方法,其特征在于,所述步骤(四)中所得棕色粉体产品中检测总黄酮含量采用分光光度法。
