背景技术
多种吸入器可用于雾化药物。例如,美国专利号5,586,550(通过引用并入本文)描述了一种包含分配装置的吸入器,其中具有锥形孔的膜被振动使得与膜的背面接触的液体从膜的正面作为气雾分配。另一单剂量吸入系统以及可被雾化的药物的示范例在美国专利申请号2011/0168172和2011/0168170中被描述,上述申请的公开通过引用并入本文。
上述吸入器的一个方面是需要向分配装置提供计量的液体药物,使得该计量可被雾化并输送至患者的肺。然而,输送已知量的液体药物(其通常必须存储在无菌环境中)被证明是具有挑战性的,尤其是当需要以受控的和可重复的方式分配该计量时。因此,本发明涉及将上述药物输送至吸入器用于后续雾化的方法。
发明内容
一实施例提供一种以可重复的方式向雾化装置提供计量液体药物的方法。该方法使用一种雾化装置,该雾化装置包括具有衔口的外壳、外壳内设置用于通过衔口喷射液体微滴的振动元件、容纳液体药物直至液体药物被振动元件雾化的存储器以及外壳上与腔室相连通的开口。尽管结合一种特定类型的吸入器进行说明,将理解其他类型的雾化装置可通过本文所描述的方法进行使用。
为提供计量的液体药物,使用到一种分配器,该分配器包括细长的分配器主体,其具有近端和远端的尖头,液体药物通过该尖头进行分配。分配器还包括分配机构,其在操作时每次压缩分配器主体从尖头分配计量的液体药物。

因为处方可能要求比分配器单次喷射所提供的药物更多的药物,该过程可能按照需要被重复多次以向腔室内提供处方所要求的量。这点可通过“泵送动作”完成,使用者“泵送”分配器(使之在每次泵送下压缩),直至提供正确的泵送次数。例如,如果处方要求一定量的液体药物,分配器可被泵送5次,每次泵送的微滴具有总量五分之一的计量体积。
在某些情况下,尖头可包括至少一条对准结构,其在分配液体药物时协助保持分配器大体上与外壳垂直。当计量被分配至腔室内时,振动元件被操作以将已分配的液体药物喷射成喷雾。
在另一实施例中,本发明提供一种示范性雾化系统。该系统包括雾化装置,该雾化装置包括具有衔口的外壳、外壳内设置用于通过衔口喷射液滴的振动元件、容纳液体药物直至液体药物被振动元件雾化的存储器以及外壳上与腔室液体连通的开口。

具体实施方式
本发明的某些方面涉及向雾化装置(也称吸入器)分配计量液体药物的技术。尽管对于大量的雾化装置有用,在某些情况下液体将被分配进入雾化装置,该装置包括外壳(其界定分配出口或衔口)、振动膜或网(其正面暴露在出口处且背面用于容纳待分配的液体)以及振动机构(其与外壳相连并可操作以振动膜并通过膜分配液体的雾。
多种容器或分配器可用于存储液体药物,然后将计量的液体药物分配至存储器内,在存储器内液体药物与膜的背面接触。以此方式,通过操作振动机构一段时间以完全雾化背面处的计量的液体,计量的液体可在出口或衔口处被分配。容器或分配器通常具有用于存储液体的封闭区和用于在每次操作所述振动机构时分配计量液体的机构。例如,容器可被压缩或泵送以喷射已知体积的液滴。
可被用于上述吸入器的示范气雾生成器也被描述在美国专利号5164740、6629646、6926208、7108197、5938117、6540153、6540154、7040549、6921020、7083112、7628339、5586550、5758637、6085740、6467476、6640804、7174888、6014970、6205999、6755189、6427682、6814071、7066398、6978941、7100600、7032590、7195011以及美国专利申请公开号2011/0168172和2001/0168170中,上述所有专利和申请通过引用并入本文。上述引用文件描述示范气雾生成器和制备上述气雾生成器的方法。每一申请或专利的至少这些特征通过引用被并入本文。气雾生成器可包括具有锥形孔的振动膜,锥形孔的大小在约3μm-8μm之间,优选地为约3μm-6μm之间,且在某些情况下为约4μm。膜具有半球形外形,且随围绕孔的环形压电元件振动。膜的直径可在约5mm-8mm之间。膜的厚度也可在约50微米-70微米之间。一般来说,膜将以约50kHz-150kHz的频率振动。
多种液体药物可从容器中被分配。例如,液体药物可包括胰岛素制剂,如无防腐胰岛素,包括之前通过引用并入的美国专利申请公开号2011/0168170中所述的任何胰岛素制剂。例如,可被分配的无防腐胰岛素制剂可以不含任何防腐剂,包括苯酚、间甲酚、氯化甲酚、麝香草酚和它们的混合物等。由于不含上述防腐剂,使得制剂可使用高频运作的振动网或孔板被雾化成液体喷雾。由于不含上述防腐剂,便允许一定剂量的制剂与振动网相接触而不至于在制剂中形成大量发泡。相应地,该制剂可被更快地雾化。此外,大体上所有的液体均可被雾化。上述制剂包含大量的水和少量的人胰岛素。该制剂也可包括多种浓度的人胰岛素。例如,制剂的浓度可以在约100IU胰岛素/毫升-1200IU胰岛素/毫升之间,且更优选地在约200IU胰岛素/毫升-800IU胰岛素/毫升之间。除了水和人胰岛素之外,制剂还可包括锌、醋酸酯、氯化物和钠。锌离子和醋酸离子来自药剂,如胰岛素。氯离子和钠离子在胰岛素溶解和调整pH值时被加入。比方说,氯化钠NaCl的浓度在800IU胰岛素/毫升制剂中可以是约20mM,在400IU胰岛素/毫升制剂中可以是10mM,而在200IU胰岛素/毫升制剂中可以是5mM。
其他液体药物也可以被分配。例如,上述药物可包括其他蛋白制剂、哮喘和慢性阻塞性肺病的治疗、疫苗以及镇痛治疗。
现参照附图,对一种向吸入器中分配计量药物的示范方法进行说明。图1中显示的是吸入器10,其包括具有衔口14的外壳12。外壳内设置有具有振动网的气雾生成器以及用于控制气雾生成器运作的电子件,气雾生成器与本文所述的哪些类似。网被定位成成在网振动时通过衔口16喷射气雾。外壳12包括上表面13,其具有被定位在开口20上的可滑动的盖子18(见图2)。盖子18向后滑动会露出开口20。此外,开口20通向存储器22或漏斗(见图3和4),存储器22或漏斗朝向振动网24逐渐变细,振动网24是与本文中所述的任何气雾生成器相似的气雾生成器的一部分。更确切地,网24具有暴露于存储器22的背面,使得当液体被提供至存储器22内时液体开始与网24的背面相接触。当网24被振动时(通过操作向气雾生成器提供电力的“开始(on)”按钮26),液体从网24的正面喷射成雾化的喷雾,且可通过衔口14吸入。
图5显示的是与前述吸入器10相似的吸入器100的内部组件。吸入器100具有外壳112,外壳112包括通向存储器122的开口120,存储器122具有开放底端150,其将已分配的液体汇集输送至振动网(未示出)的背面,振动网是气雾生成器154的一部分,该气雾生成器154具有用于振动网的振动元件155(如环形压电换能器)。气雾生成器154具有圆形开口156,网被设置在开口156上方。当振动时,网将液体以雾化喷雾的形式喷射至俘获腔室内和衔口处。以此方式,喷雾可通过衔口处的吸入动作被吸入。
在一实施例中,待雾化的液体被存储在由图3和4充分显示的分配器30中。为方便起见分配器30可被描述成近端32和在末尾处具有尖头36的远端34。分配器30还包括用于将液体药物存储在无菌环境下的筒38。分配器30包括内阀,以使远端34可相对于近端32移动,由此压缩分配器30。通过此方式,每次分配器被压缩(或“泵送”)时计量体积的液体被喷射进入存储器22。例如,图3显示分配器30处于未被压缩状态。在图4中,压力被施加以压缩分配器30并将液滴喷射进入存储器22。相似的过程可被用于吸入器100,其中液体被注入至存储器122内。该过程可按需要被重复多次,以便于将适当体积的液体药物分配至存储器22内。在分配所需量的液体药物后,分配器30可被移除并存储以便于后续使用。每次泵送可被分配的示例单位体积可在约5-100微升之间。
尖头36包括形如环形或圆形台阶的肩部40,其用作防止尖头36被进一步插入开口20中的阻挡部。肩部40被插入一定的距离,以足以使其不仅用作阻挡部还允许尖头36坐定在开口20中处于大体垂直于外壳12的上表面13的稳定位置。如此,一旦分配器30被放置在开口20中,其将自立于大体垂直的方向。该位置允许使用者在准备向气雾生成器中分配单元体积的液体时方便抓握分配器30。尽管显示为具有肩部,也可使用其他坐落机制,如与开口20的锥形相匹配的锥形体、凸起的凸起部或翼、棘爪等。
操作分配器30的一个示范技术显示在图1和2中。应理解,相似的过程可用于吸入器100。在图1和2中,分配器30被单手抓握,使得四根手指环绕分配器30,基本上围绕着筒38。通过四根手指抓握分配器30,达到了牢固的抓握,使得分配器30可被容易地泵送以喷射液体。此外,人的拇指可位于近端32处以施加额外的压力。通过适当的抓握,盖子18可向后滑动且尖头34可被置于开口20中。如前所述,尖头34包括比开口20宽的肩部40,以防止尖头34与网24接触。此外,肩部40可被设计成尖头34与网24充分隔开,使得当足量的计量液体被分配至存储器22内时,尖头34不会与已分配的液体相接触。以此方式,当分配器30从吸入器上移除时,其将不会移除部分已分配的液体。通常,尖头34将与网24相分离的距离在约5mm-20mm之间。尖头34可为锥形,以便于与存储器22的锥形体相匹配。存储器22的锥形体便于将所有已分配的液体输送至网24的背面,且尖头34的锥形防止其与存储器22的壁接触。
如图2中清楚示出,当尖头34在适当位置时,使用者将筒38压向尖头34。同时,吸入器10也在适当位置。这致使分配器30压缩。然后,内阀被打开以允许计量的液体从尖头34被分配并进入存储器22。筒38每次被压下或泵送时,又一计量液体被喷射。该动作按所需被进行多次,以便于向存储器22内提供规定剂量。
通过以所示方式抓持分配器30,泵送动作可以很轻易发生。这与鼻喷雾分配器不同,鼻喷雾分配器通常以直立方式被致动,通过小心并同时用中指和食指(尖头在指尖延伸)将近端压向处于拇指的同等压力下的分配器容器的远端。对于这种类型的鼻喷雾,当足够的压力被等同施加至两端时产生喷雾。相反,分配器30可以在尖头与吸入装置相接合时通过仅向分配器的远端施加压力而被轻易致动。吸入装置及匹配结构被构建使得计量的药物持续地从分配器被输送至装置中,且使用者可通过不规律的按压来完成,只要压力大于或等于全程压缩分配器所需的力。如果吸入装置被装载且被放置在桌上或任何其他自由支撑表面上,与同时拿着装置和分配器(不借助于支撑表面)时分配定量液体所需的力量相比,将分配器按压进装置至致动点所需的力减少50%。
图6显示的是可用在吸入器10或吸入器100上的分配器80的替代实施例。分配器80具有在其远端的尖头82,该尖头82上具有协助将尖头82对准并定位在上表面13中的开口12中的多个对准结构84。对准结构84可具有多种形状、大小和结构。尽管显示为四个均等间隔的结构84,应理解可以使用其他数量,如仅有单个对准结构。结构84用作限制尖头82插入开口20中的阻挡部。结构84也提供一种触感,让使用者知道尖头82已被适当插入。当所有四个结构84与吸入器10的表面相连接时,分配器80已被适当插入并准备好向吸入器10中分配计量。此外,结构84用来将分配器80支撑在大体上与上表面13垂直或呈直角的方向。以此方式,当被插入开口20中时分配器80可自立,使得使用者易于按照图1和2中所述的方式抓握并分配。

Claims (11)
1.一种向雾化装置提供计量液体药物的方法,所述方法包括:
提供雾化装置,所述雾化装置包括具有衔口的外壳、所述外壳内被设置成通过所述衔口喷射液滴的振动元件、容纳所述液体药物直至被所述振动元件雾化的存储器以及所述外壳上的与所述存储器液体连通的开口;
提供分配器,所述分配器包括细长的分配器主体,所述分配器主体包括延伸于其近端(32)和在其远端(34)处的尖头(36)之间的纵向轴线,液体药物通过所述尖头被分配,其中所述分配器还包括包含液体药物的容器(38)以及分配机构,所述分配机构在所述分配器主体每次被压缩时从所述尖头(36)分配计量的所述液体药物,其中所述分配机构包括定位在所述尖头(36)与所述容器(38)之间的分配机构壳体,所述分配机构壳体能沿所述纵向轴线扩张和收缩从而分配计量的所述液体药物;

5.一种雾化系统,包括:

向所述远端方向施加轴向力至所述细长的分配器主体以轴向压缩所述分配机构壳体,且致使所述分配机构被压缩时计量的所述液体药物被喷射进入所述存储器内。
9.根据权利要求8中所述的方法,还包括重复将所述细长的分配器主体压向所述尖头,以向所述存储器内分配多个计量的所述液体药物。
10.根据权利要求8所述的方法,其中所述多个对准结构配置成在分配所述液体药物时所述对准结构协助保持所述分配器与所述雾化装置外壳大体垂直。
11.根据权利要求8所述的方法,还包括操作振动元件以将已分配的液体药物以雾化喷液形式喷射。
