Abstract
本发明涉及食品加工技术领域,同时也涉及医药保健品技术领域,具体是一种海参番茄红素茶多酚片,原料按重量计的配比为:含量为6%的番茄红素粉14-17份,含量为80%的茶多酚粉5-7份,海参冻干粉45-55份,微晶纤维素粉30-35份,羧甲基淀粉钠5-8份,硬脂酸镁0-2份,包衣粉2-4份。与现有技术相比,本发明具有调节免疫、抗疲劳等功效,能提高机体防病抗病能力,快速恢复体力和精力,食用方便,是新一代高级海洋保健食品。

本发明涉及食品加工技术领域,同时也涉及医药保健品技术领域。
背景技术
海参是海洋生物中的极品,营养价值非常高,富含50多种对人体有益的成分,其中蛋白质含量高达55%且不含胆固醇,另外还含有海参素、海参皂苷、刺身粘多糖、硫酸软骨素等活性成分,具有补肾益精、滋阴健阳,补血润燥、调经祛劳、养胎利产等阴阳双补功效,可以提高记忆力、增强人体免疫力、抗病毒、抗衰老、降血脂、修复再生,是一种很典型的高蛋白、低脂肪、低胆固醇的健康食品,拥有海中“营养宝库”的美称。
传统海参干品的加工方法不仅粗糙而且工艺复杂繁琐,通常经过腌渍、烤参、拌灰、晒干制成,工序多、周期长、耗能大、操作成本高,大量有益成分随着废水液的流失而损失,并且食用非常不方便,水发时间较长,还容易出现结壳龟裂、脂肪氧化、产品表面变色等情况,导致海参失去原有风味,而冻干海参脱水彻底,产品无需任何防腐剂就可在常温下保存5年以上,并极大的保留了海参中的活性成分,附加值较高,越来越受到人们的青睐。
番茄红素是植物中所含的一种天然色素。主要存在于茄科植物西红柿的成熟果实中。它是目前在自然界的植物中被发现的最强抗氧化剂之一,拥有强大的清除自由基能力,对人体有优越的生理调节功能,可以有效的防治因衰老,免疫力下降引起的各种疾病,具有“植物黄金”之称。
茶多酚是茶叶中多酚类物质的总称。包括黄烷醇类、花色苷类、黄酮类、黄酮醇类和酚酸类等,主要为儿茶素类,占60-80%,具有解毒、抗辐射、抗氧化和清除自由基的作用,可以防治心脑血管疾病、提高综合免疫力、预防衰老、清除口臭、助消化、美容护肤,其抗氧化能力是BHA的4-6倍,VE的6-7倍,且用量很少即可发挥作用,并且没有合成物的潜在毒性,被健康及医学界誉为“辐射克星”。
目前海参营养成分的吸收主要以食补为主,加之烹饪方法的限制导致利用率不足,造成很多营养成分流失,公开号为CN103445168A的中国发明专利公开了一种《复合海参颗粒咀嚼片及其制备方法》它是由以下重量份数的原料制成的:海参50-80,杏鲍菇5-30,香菇5-20,海蓬子2-10,辅料5-30。服用、携带、运输都较方便,但其海参粉碎、过筛后得到2-10目粒状海参,人体吸收和利用率不高,与传统食用方法无异。
公开号为102302614B的中国发明专利公开了《一种复方海参茶多酚胶囊及其制备方法》,其具体步骤为:(1)将鲜活海参经过清洗、干燥、粉碎成海参粉,粉碎粒度为200μm-100nm;(2)取纯度≥90w%、粒度为通过80-100目筛孔的茶多酚粉;(3)将海参粉:茶多酚粉按照质量比1.0∶0.5-1.0进行充分混合;(4)将步骤(3)得到混合物进行精囊灌装,每囊0.2-0.6g。该发明虽然可能具有预防和治疗高血脂病症的作用,但由于其海参加工过程中经历了直接干燥过程,海参原有的有效成分可能部分流失,对人体的生理调节功能降低。将原料直接混合制胶囊容易导致内容物不匀、装量不准确,海参味道完全被掩盖难以感知具体成分,而且该剂型吞服不便,适口性差,单一抗氧化剂清除人体自由基能力不如复合抗氧化剂。
发明内容
本发明的目的是克服现有技术的不足,提供一种含有海参冻干粉、番茄红素、茶多酚,营养全面、功能突出的保健食品。
为了实现本发明的目的所采用的技术方案是:
一种海参番茄红素茶多酚片,原料按重量计的配比为:含量为6%的番茄红素粉14-17份,含量为80%的茶多酚粉5-7份,海参冻干粉45-55份,微晶纤维素粉30-35份,羧甲基淀粉钠5-8份,硬脂酸镁0-1份,包衣粉2-4份。
由于海参中DHA含量很高,DHA具有增强免疫力、促进儿童大脑发育以及预防癌症等作用,但由于容易被氧化而极大的限制了其功效的发挥。本发明采用将两种高抗氧化性成分加入冻干海参粉中,天然复合抗氧化剂的协同作用最大限度的保留了海参的营养成分,提高了海参的保健功效。冻干海参粉、番茄红素粉、茶多酚粉在本发明要求保护的配比范围内保健作用和产品质量最佳,超过或小于此范围则容易出现硬度欠佳、色泽较差、口感生涩等情况且番茄红素粉和茶多酚粉配合使用的抗氧化效果优于其中任何单一种类,辅之降血脂、提高免疫力的功能突出了产品的保健效果。
一种海参番茄红素茶多酚片的制备方法,包含如下步骤:
1)原料制备:新鲜海参去除内脏并用清水冲洗干净后进行高温漂烫,温度在65-70℃,时间不超过6分钟,然后进行泡发,24小时之后取出进行真空冷冻干燥,将新鲜刺海参在冻干舱内迅速冷冻到-40℃,在真空状态下,通过调节温度和压力,逐步升温使冰直接变为水蒸气,时间为50小时,得到冻干海参;冻干海参粗碎后过80目筛的万能粉碎机粉碎,得到海参冻干粉;番茄红素粉、茶多酚粉、微晶纤维素粉、羧甲基淀粉钠分别过80目筛;
本发明采用新鲜海参经真空冷冻干燥加工,有效的保护了海参有效成分的稳定性,并且干燥后形态松散,含水量低,不易受污染,延长了保质期,原料使用冻干海参粉加水后能够快速恢复生物活性,制成的海参片剂利用最先进的海参粉碎技术,将海参粉碎到80到120目的大小,直接穿过细胞膜,吸收率大大提高,并且溶出度也较好。
2)混合:将海参冻干粉、番茄红素粉、茶多酚粉、微晶纤维素粉、羧甲基淀粉钠置于三维运动混合机中混合20min,得到色泽均匀一致的混合粉;
3)制粒、干燥、整粒:将混合粉倒入制粒锅中,向混合粉中加入浓度20%的乙醇溶液利用湿法混合制粒机高速搅拌混合湿制软材,得湿颗粒;湿颗粒进入整粒机中整粒,使颗粒大小较均匀,再将湿颗粒放入高效沸腾干燥机干燥,温度为55-65℃,待物料干燥后取出得到干颗粒;
4)干颗粒分层:干颗粒倒入振荡筛中震荡分层,最上层为大于20目颗粒,中层为20-40目颗粒,最下层为小于40目的颗粒;收集20-40目颗粒备用;
5)压片:将20-40目干颗粒、硬脂酸镁置于三维运动混合机中混合5min,得混合颗粒;将混合颗粒放入旋转式压片机中压片,调整片重为0.6g/片,并去残、废片,得到素片;
在加工过程中,冻干海参粉和其他原料黏性较大,湿法制粒难度增大;比重较轻,成片困难。本发明采用加入20%的乙醇溶液可使粒度均匀分散,减少成球和原料浪费损失。压片的过程中在不减少冻干海参粉比例的前提下经过潜心试验研究设计合理的的原料配比,既保证了产品的质量要求,又增进了抗氧化能力,同时在该配方、颗粒大小和成片压力条件下的片剂溶出程度和吸收较好,在技术上取得质的飞跃。
6)包衣液的配制:取95%食用酒精,加纯化水配置成浓度为70%的乙醇溶液,倒入搅拌保温罐中,取包衣粉缓慢加入浓度为70%的乙醇溶液中,盖上盖子,搅拌约1小时,充分搅拌均匀,配置成8%的包衣液,备用;
7)薄膜包衣:启动高效包衣机,打开角座阀、排风和热风进行预热,使包衣滚筒中的温度达到45-50℃,待负压和温度稳定后将称好的素片倒入包衣滚筒;将配好的包衣液倒入保温罐中,预热5分钟,打开喷雾系统,调节雾化压力在0.15MPa,喷枪压力在0.8-1.0MPa,在启动蠕动泵的同时启动主机,滚筒不间断运转进行匀浆使片芯受热均匀;调整蠕动泵的转速和浆料调节器,使用喷枪均匀喷雾于转动的素片表面,55-65℃热风干燥;包衣完成后,停止喷浆,关闭热风,待外观光洁,色泽均匀,片芯冷却后,关闭匀浆和排风,得合格包衣片,是为海参番茄红素茶多酚片。
进一步地,所述的包衣粉为羟丙基甲基纤维素或三乙酸甘油酯。
进一步地,所述的番茄红素粉是通过天然微生物发酵法生产的。采用天然微生物发酵法生产的番茄红素,避免有机溶剂的残留且全程温度控制在60℃以内,较好的保证番茄红素的稳定性,具有质量安全、原料利用程度高的优点。
与现有技术相比,本发明结合使用抗氧化的番茄红素和茶多酚,更好的提高了易氧化物质的稳定性。制得的产品绿色、纯天然、低脂、低胆固醇,不含防腐剂,不含兴奋剂,具有调节免疫、抗疲劳等功效,能提高机体防病抗病能力,快速恢复体力和精力,食用方便,是新一代高级海洋绿色保健食品。
具体实施方式
一、以下结合具体事例对本发明作进一步详细说明。
实施例1:
备料:含量为6%的番茄红素粉17kg,含量为80%的茶多酚粉7kg,海参冻干粉55kg,微晶纤维素粉35kg,羧甲基淀粉钠8kg,硬脂酸镁1kg,包衣粉4kg。
原料的调配:新鲜海参去除内脏并用清水冲洗干净后进行高温漂烫,温度在65-70℃,时间不超过6分钟,然后进行泡发,24小时之后取出进行真空冷冻干燥,将新鲜刺海参在冻干舱内迅速冷冻到-40℃,在真空状态下,通过调节温度和压力,逐步升温使冰直接变为水蒸气,时间为50小时。
将符合标准的冻干海参粗碎后过80目筛的万能粉碎机粉碎,得到海参冻干粉。番茄红素粉、茶多酚粉、微晶纤维素、羧甲基淀粉钠去掉外包装进入洁净区,分别过80目筛。
混合:将海参冻干粉、番茄红素、茶多酚、微晶纤维素、羧甲基淀粉钠置于三维运动混合机中混合20min,得到色泽均匀一致的混合粉。
制粒、干燥、整粒:将混合粉倒入制粒锅中,向混合粉中加入浓度20%的乙醇溶液利用湿法混合制粒机高速搅拌混合两分钟湿制软材,得湿颗粒。湿颗粒进入整粒机中整粒,使颗粒大小较均匀,再将湿颗粒放入高效沸腾干燥机干燥,温度为55℃,干燥时间为30min,含水量低于5%时取出得干颗粒。
干颗粒分层:将干颗粒倒入振荡筛中震荡分层,最上层为大于20目颗粒,中层为20-40目颗粒,最下层为小于40目的颗粒。收集20-40目颗粒备用。
压片:将20-40目颗粒、硬脂酸镁分别置于三维运动混合机中混合5min,得混合颗粒;将各层混合颗粒分别放入旋转式压片机中压片,调整片重为0.6g/片,压片,并去残、废片,得到素片。
包衣液的配制:取95%食用酒精,加纯化水配置成浓度为70%的乙醇溶液,倒入电加热搅拌保温罐中,取包衣粉缓慢加入浓度为70%的乙醇溶液中,盖上盖子,搅拌约1小时,充分搅拌均匀,配置成8%的包衣液,备用。
薄膜包衣:启动高效包衣机,打开蒸汽阀、排风和热风,使包衣锅中的温度达到45-50℃,将称好的素片倒入包衣锅中进行匀浆使片芯受热均匀;调整蠕动泵的转速和浆料调节器,启动喷枪均匀喷雾于转动的素片表面,55-65℃热风干燥。包衣完成后,停止喷浆,关闭热风,待外观光洁,色泽均匀,片芯冷却后,关闭匀浆和排风,得合格包衣片,是为一种海参番茄红素茶多酚片。
实施例2:
其他同实施例1,只不过:番茄红素粉14kg,茶多酚粉5kg,海参冻干粉45kg,微晶纤维素粉30kg,羧甲基淀粉钠5kg,硬脂酸镁0kg,包衣粉2kg。
实施例3:
其他同实施例1,只不过:番茄红素粉15kg,茶多酚粉6kg,海参冻干粉50kg,微晶纤维素粉33kg,羧甲基淀粉钠7kg,硬脂酸镁0.5kg,包衣粉3kg。
实施例4:
其他同实施例1,只不过:番茄红素粉16kg,茶多酚粉5.5kg,海参冻干粉47kg,微晶纤维素粉32kg,羧甲基淀粉钠6kg,硬脂酸镁0.2kg,包衣粉3.3kg。
实施例5:
其他同实施例1,只不过:番茄红素粉16kg,茶多酚粉5.8kg,海参冻干粉49kg,微晶纤维素粉31kg,羧甲基淀粉钠6.8kg,硬脂酸镁0.8kg,包衣粉2.7kg。
对比例1,海参番茄红素片:
其他同实施例1,只不过:番茄红素粉34kg,茶多酚粉0kg。
对比例2,海参茶多酚片:
其他同实施例1,只不过:番茄红素粉0kg,茶多酚粉14kg。
二、小鼠功能试验。
1.番茄红素茶多酚片对小鼠体重的影响
样品:实施例1制备的海参番茄红素茶多酚片。
实验动物:选用北京华阜康生物科技股份有限公司繁殖的18-22g昆明种清洁级雌性小鼠48只,共分4组,每组12只。饲养于北京联合大学应用文理学院保健食品检测中心SPF级动物室。实验动物使用许可证号:SYXK(京)2012-0031。
基础饲料购于北京华阜康生物科技股份有限公司。
试验方法:按卫生部保健食品功能检测与评价技术规范:大鼠适应环境3d后,喂饲基础饲料5d,然后随机分为4组,即三个剂量组(0.1、0.2、0.6g/kgBW)、一个对照组。受试物以无菌水配制,每日一次经口给予,连续灌胃34天后进行测定,小鼠灌胃体积为0.1mL/10g鼠重。空白组用无菌水代替受试物,每日灌胃体积与受试物组相同,各剂量组均给予基础饲料。
由表1可见,各剂量组动物的初重、中期重、末重和增重与对照组比较无显著差异(P>0.05),说明在本实验条件下海参番茄红素茶多酚片对小鼠的体重无明显影响。

2.番茄红素茶多酚片清除自由基的能力
样品:实施例2制备的海参番茄红素茶多酚片。
实验动物:小白鼠48只,共分4组,每组12只,体重20-22g。
试验方法:按Smirnoff等的方法改进用FeSO4+H2O2产生·OH,以·OH氧化水杨酸所得产物测A510,以(对照组-加药组/对照组)×100%计算·OH的清除率。
试验结果:实验结果见表2

注:各剂量组与对照组比较,P<0.01。
由表2可见,各剂量的试验组与对照组均差异显著(P<0.01),说明在本实验条件下海参番茄红素茶多酚片对自由基的清除有显著影响;并且剂量在0.2g/kg对·OH就有一定的清除能力,说明番茄红素茶多酚片对·OH具有较强的清除作用。番茄红素片的剂量与清除率之间存在较好的量效关系。
3.番茄红素与茶多酚单独作用的体外抗氧化试验
样品:实施例1制备的海参番茄红素茶多酚片、对比例1制备的海参番茄红素片、对比例2制备的海参茶多酚片,对比例1、2制作的番茄红素或茶多酚片硬度较差,边缘破碎、掉渣。
实验动物:小白鼠30只,共分3组,每组10只。

由表3可见,在本实验条件下海参番茄红素结合茶多酚片清除自由基的能力与单独使用2倍比例的番茄红素或茶多酚清除率存在显著差异(P<0.01),相同剂量的条件下,番茄红素与茶多酚配合使用的抑制率是二者单独使用抗氧化能力的2-5倍,可见复合抗氧化剂的清除自由基能力明显优于单一抗氧化剂。
4.海参番茄红素茶多酚片对小鼠细胞免疫功能的影响
样品:实施例1制备的海参番茄红素茶多酚片。
实验动物:小白鼠48只,共分4组,每组12只,体重20-22g。
试验方法:取羊血,生理盐水洗涤3次,每只鼠经腹腔注射2%(v/v,用生理盐水配制)压积SRBC(2000r/min,10min)0.2mL,致敏后4天,测量左后足跖部厚度,同一部位测定三次,取平均值。然后在测定部位皮下注射20%(v/v,用生理盐水配制)压积SRBC20μL,于注射后24小时测量左后足跖部厚度,以攻击前后足跖厚度的差值来表示DTH的程度。受试样品组的差值显著高于对照组的差值,可判定该项试验结果阳性。

由表4可见,经口给予小鼠不同剂量的海参番茄红素茶多酚片34天后,0.2g/kgBW组与0g/kgBW组比较,小鼠足跖肿胀度有显著差异(P<0.05),0.6g/kgBW组与0g/kgBW组比较,小鼠足跖肿胀度有显著差异(P<0.05)即海参番茄红素茶多酚片在0.2g/kgBW、0.6g/kgBW组能提高小鼠足跖肿胀度。
Claims (3)
1.一种海参番茄红素茶多酚片的制备方法,由如下重量配比的原料,按照如下步骤制得:
含量为6%的番茄红素粉14-17份,含量为80%的茶多酚粉5-7份,海参冻干粉45-55份,微晶纤维素粉30-35份,羧甲基淀粉钠5-8份,硬脂酸镁0-1份,包衣粉2-4份;
1)原料制备:新鲜海参去除内脏并用清水冲洗干净后进行高温漂烫,温度在65~70℃,时间不超过6分钟,然后进行泡发,24小时之后取出进行真空冷冻干燥,将新鲜刺海参在冻干舱内迅速冷冻到-40℃,在真空状态下,通过调节温度和压力,逐步升温使冰直接变为水蒸气,时间为50小时,得到冻干海参;冻干海参粗碎后过80目筛的万能粉碎机粉碎,得到海参冻干粉;番茄红素粉、茶多酚粉、微晶纤维素粉、羧甲基淀粉钠分别过80目筛;
2)混合:将海参冻干粉、番茄红素粉、茶多酚粉、微晶纤维素粉、羧甲基淀粉钠置于三维运动混合机中混合20min,得到色泽均匀一致的混合粉;
3)制粒、干燥、整粒:将混合粉倒入搅拌锅中,向混合粉中加入浓度20%的乙醇溶液利用湿法混合制粒机高速搅拌混合湿制软材,得湿颗粒;湿颗粒进入整粒机中整粒,使颗粒大小较均匀,再将湿颗粒放入高效沸腾干燥机干燥,温度为55-65℃,待物料干燥后取出得到干颗粒;
4)干颗粒分层:干颗粒倒入振荡筛中震荡分层,最上层为大于20目颗粒,中层为20-40目颗粒,最下层为小于40目的颗粒;收集20-40目颗粒备用;
5)压片:将20-40目干颗粒、硬脂酸镁置于三维运动混合机中混合5min,得混合颗粒;将混合颗粒放入旋转式压片机中压片,调整片重为0.6g/片,压片,并去残、废片,得到素片;
6)包衣液的配制:取95%食用酒精,加纯化水配置成浓度为70%的乙醇溶液,倒入电加热搅拌保温罐中,取包衣粉缓慢加入浓度为70%的乙醇溶液中,盖上盖子,搅拌1小时,充分搅拌均匀,配置成8%的包衣液,备用;
7)薄膜包衣:启动高效包衣机,使包衣锅中的温度达到45-50℃,待负压和温度稳定后将称好的素片倒入包衣滚筒;将配好的包衣液倒入保温罐中,预热5分钟,打开喷雾系统,调节雾化压力在0.15MPa,喷枪压力在0.8~1.0MPa,在启动蠕动泵的同时启动主机,滚筒不间断运转进行匀浆使片芯受热均匀;调整蠕动泵的转速和浆料调节器,使用喷枪均匀喷雾于转动的素片表面,55-65℃热风干燥;包衣完成后,停止喷浆,关闭热风,待外观光洁,色泽均匀,片芯冷却后,关闭匀浆和排风,得合格包衣片,是为海参番茄红素茶多酚片。
2.根据权利要求1所述的一种海参番茄红素茶多酚片的制备方法,其特征在于所述的包衣粉为羟丙基甲基纤维素或三乙酸甘油酯。
3.根据权利要求1所述的一种海参番茄红素茶多酚片的制备方法,其特征在于所述的番茄红素粉是通过天然微生物发酵法生产的。
