Abstract

本发明涉及一种吡啶酮生物碱类化合物的制备方法和用途。本发明用采自南极长城站附近苔藓样品中的绳状青霉GWT2‑24(Penicillium funiculosum GWT2‑24)生产出结构新颖的吡啶酮生物碱类化合物。本发明的吡啶酮生物碱可有效预防或治疗高血脂及相关并发症。

Description

一种吡啶酮生物碱类化合物及其制备方法和用途
技术领域:
本发明涉及用绳状青霉GWT2-24(Penicillium funiculosum GWT2-24)(保藏编号是:CGMCC 8986保藏日期:2014年4月1日保藏单位:中国微生物菌种保藏管理委员会普通微生物中心保藏地址:北京市朝阳区北辰西路1号院3号中国科学院微生物研究所,100101)生产包含吡啶酮生物碱类化合物的方法;本发明还涉及该类化合物在预防和治疗高血脂及相关并发症中的用途。
背景技术:
本发明人从绳状青霉GWT2-24(Penicillium funiculosum GWT2-24)(保藏编号是:CGMCC 8986)液体发酵产物中分离出多个以吡啶酮为基本骨架的生物碱类化合物。研究发现所示吡啶酮生物碱化合物具有显著的降脂活性,表明上述吡啶酮生物碱进一步有可能开发成新型降血脂药物。
发明内容:
本发明旨在提供一种结构新颖的具有抑制脂肪代谢活性的新化合物。其结构式是
本发明的式I化合物可通过微生物发酵培养来获取含有该类化合物的发酵物,然后从发酵物中采用Sephadex LH20凝胶柱层析,中压MPLC,半制备HPLC等方法分离纯化得到。
本发明的下述实施例中列举了利用绳状青霉GWT2-24(Penicillium funiculosumGWT2-24)(保藏编号是:CGMCC 8986)制备本发明式I化合物的实例。
附图说明:
图1是化合物I(吡啶酮生物碱)对肝癌HepG2细胞脂质调节活性图,图2是化合物I(吡啶酮生物碱)对肝癌HepG2细胞总胆固醇(TC)调节活性图,图3是化合物I(吡啶酮生物碱)对肝癌HepG2细胞总甘油三酯(TG)调节活性图。其中数值平均值以±标准差表示,***P<0.001,脂质堆积模型组vs正常对照组;P<0.05,P<0.01,P<0.001,给药组vs脂质堆积模型组。

在如下的实施例中所指的化合物I的化学结构(结构式中的阿拉伯数字是化学结构中碳原子的标位)是:
实施例1化合物I的发酵生产及分离精制
1发酵生产
生产菌的发酵培养:按培养微生物的常规方法,取绳状青霉GWT2-24(Penicilliumfuniculosum GWT2-24)(保藏编号是:CGMCC 8986)适量,接种到PDA固体斜面培养基上,在28摄氏度培养箱中培养5天。
取斜面培养5天的绳状青霉GWT2-24(Penicillium funiculosum GWT2-24)适量,接种到装有300mL培养基[培养基组成(克/升):麦芽糖20.0,葡萄糖10.0,甘露醇20.0,味精10.0,酵母膏3.0,玉米浆1.0,KH2PO40.5,MgSO4·7H2O 0.3,pH调节至6.5]的1000mL锥型瓶中,在28摄氏度条件下静置培养30天,获得发酵产物。
2浸膏的获得
用纱布将发酵液和菌丝体分离。将菌丝体用丙酮浸提三次,减压浓缩至无丙酮,所得水相用等量乙酸乙酯萃取三次。发酵液直接用等量乙酸乙酯萃取三次,合并乙酸乙酯相,减压浓缩,得粗浸膏,共4.5克。
3化合物的分离精制
浸膏(4.5克)用甲醇溶解后,以甲醇为溶剂进行LH20柱层析,分为4个流份。组分2先以C-18 ODS中压柱层析,以甲醇-水为溶剂进行梯度洗脱,最后经反相半制备高效液相色谱(甲醇∶水=50∶50)得化合物I(6mg)。
化合物I白色固体,分子式C14H13NO4,HR-ESI-MS m/z:260.0923[M+H]+,计算值260.0921。IR(KBr)vmax 3353,1715,1658,1533,1350,1207,935,734,698cm-11H和13C-NMR数据见表1。

a)本表信号归属基于DEPT、HMQC及HMBC图谱解析结果。碳信号的多重度利用DEPT方法确定并分别用s(单重峰)、d(二重峰)、t(三重峰)和q(四重峰)表示。

c)此栏中的数字和代号分别代表在HMBC谱中与相应行中的1H给出偶合相关信号的13C核。
实施例2化合物的脂质调节作用活性测试
1实验样品及实验方法
被测样品溶液的配制 测试样品为上述实施例1中分离精制的化合物I纯品。精密称取适量样品,用DMSO配制成所需浓度的溶液,供测活性。
细胞系及细胞的继代培养 细胞系及细胞的继代培养,采用肝癌HepG2细胞株。用含10%胎牛血清(FBS)的DMEM培养基,并加入100μg/mL的青霉素和链霉素,在37℃于通入5%二氧化碳的培养箱中继代培养。
脂质调节作用活性测试方法
取对数生长期细胞,消化、计数,以适宜浓度接种到已放入无菌盖玻片的6孔板中,培养24小时至细胞完全贴壁。细胞生长汇合至70%-80%时,用100μM不饱和脂肪酸-油酸(OA)刺激细胞制成脂质堆积模型;给药组同时给予不同浓度的样品(1μmol/L);阳性药组给予辛伐他汀(1μmol/L)。除空白组外,其它组需添加油酸100mmol/L共孵育24小时,24小时后,弃去培养基,用PBS洗2遍,加4%多聚甲醛在4℃固定12小时。每孔加油红O工作液20微升,室温染色15分钟,PBS洗净油红工作液,每孔加DMSO 150微升溶解附着脂质上染料,酶标仪358nm波长下测定OD值。
1)细胞内总胆固醇(TC)测量
弃去培养液,用PBS洗2遍,收集细胞于管中;加200微升氯仿∶异丙醇∶NP40(7∶11∶0.1)混合溶液,超声裂解细胞;13000转/分钟离心15分钟,取上清液于新管中50℃烘1小时,以除去氯仿;将样品放于真空浓缩离心机中抽干,除去剩余有机溶剂。试剂盒测定样品中TC含量,样品加200微升Assay Buffer溶解,具体操作见Cholesterol/Cholesteryl EsterQuantitation Kit(BioVision)说明书。BCA试剂盒测定样品中蛋白含量。
2)细胞内总甘油三酯(TG)含量测定
收集细胞,加300微升5%NP-40超声破碎细胞;将样品放到80-100℃水浴中缓慢加热2至5分钟至样品变浑浊,冷却至室温,重复3次。12000转/分钟离心2分钟,取上清液于新管中,加适量去离子水稀释样品。试剂盒测定样品中TG含量,具体操作按TriglycerideQuantification Kit(BioVision)说明书进行。BCA试剂盒测定样品蛋白含量。
2实验结果
用不饱和脂肪酸建立脂质堆积模型对所示化合物I降脂活性进行了研究,以辛伐他汀为阳性对照药,结果发现所述化合物I(吡啶酮生物碱)对细胞内的总胆固醇(TC)和总甘油三酯(TG)均具有明显降低作用。活性结果见附图。
3结论
化合物I具有较好的降脂活性,可作为新型降血脂剂,用于预防和治疗高血脂及相关并发症。

Claims (3)

1.如下式结构所示的化合物
2.权利要求1所述化合物的制备方法,其特征在于包括以下步骤:先以PDA固体斜面培养基对保藏编号是CGMCC 8986的绳状青霉GWT2-24(Penicillium funiculosum GWT2-24)在28℃培养箱中培养5天,再在发酵培养基中对该真菌进行静置发酵培养30天,将所得菌丝体用丙酮浸提三次,减压浓缩后用乙酸乙酯萃取三次,发酵液直接用等量乙酸乙酯萃取三次,合并乙酸乙酯相,减压浓缩,得粗浸膏;将所述粗浸膏通过Sephadex LH20凝胶柱层析,以甲醇为溶剂;再经C-18 ODS中压柱层析,以甲醇-水为溶剂进行梯度洗脱;最后经反相半制备高效液相色谱分离纯化得权利要求1所述化合物,制备梯度为甲醇∶水=50∶50,其中所述发酵培养基成分为麦芽糖20.0克/升,葡萄糖10.0克/升,甘露醇20.0克/升,味精10.0克/升,酵母膏3.0克/升,玉米浆1.0克/升,KH2PO4 0.5克/升,MgSO4·7H2O 0.3克/升,pH调节至6.5。
