Abstract
本发明涉及一种治疗水貂钩端螺旋体病的中药组合物,由茵陈、栀子、大黄、甘草、桃仁、芒硝、猪苓、茯苓、滑石、泽泻制备而成。该中药组合物可以是颗粒剂,其制备步骤是:(1)将中药粉碎至60目,按配方量称取,三维混合得中药粉;(2)加水煮2次,每次煮30min,过滤,合并两次滤液。将滤液浓缩至原体积的10%~20%,得浓缩浸膏;(3)称取元明粉30份,白砂糖20份,淀粉40份,混合三粉;(4)将浓缩浸膏10份分3次均匀加入混合粉中。一边搅拌一边加入,搅拌时间为10-15min;(5)压粒,60-80℃烘干即得成品。本发明所提供的中药组合物可有效治疗水貂钩端螺旋体病,治愈率95%以上。本组方具有清热利湿,利胆退黄,利尿通淋的功效。
Description
技术领域
本发明属于中兽药制剂领域,尤其是一种治疗水貂钩端螺旋体病的中药组合物及其制备方法。
背景技术
钩端螺旋体病是由各种致病性钩端螺旋体引起的人兽共患的一种急性传染病,在世界各地均有发生。各种年龄动物均易感,其中以幼龄动物最易感,发病率和死亡率也最高。本病一年四季均有发生,但以夏秋季节高发。传播方式多种多样,经消化道感染是主要的感染途径。由于本病病原最终定位于肾脏,所以尿液在本病的蔓延扩散上有重要作用。如尿液接触破伤的皮肤和黏膜就可以感染,如尿液污染了饲料和水源也能造成本病的传播。钩端螺旋体经消化道或外伤进入机体,通过血液侵入器官组织内繁殖,主要在肝脏积聚和肾脏定位。菌体进入血液、组织,并在其中繁殖,形成菌血症。钩端螺旋体的代谢产物引起动物体温升高,血糖降低,红细胞被破坏崩解等病理过程,临床表现为发烧、体温升高、贫血、黄疸等。病原体在肝脏增殖,引起肝细胞变性坏死,导致胆红素蓄积,发生黄疸。在肾脏定位增殖造成肾脏细胞变性坏死,出血,出现血红蛋白尿。急性型表现为体温升高,采食减少,精神沉郁,粪便由病初的黄色逐渐变为淡绿色,最后呈煤焦油状。可视黏膜黄染,严重者皮肤也呈黄色。一般在停止采食后1-2天死亡。慢性型表现为食欲减退,喝水猛增,可视粘膜充血,淤血,并有出血斑。肾功能障碍者出现少尿、无尿或尿液粘稠呈黄色,亦可出现血尿,表现尿毒症症状。本病给水貂养殖业带来了巨大的经济损失。
现代兽医主要采用抗生素药物进行预防和治疗,如:青霉素、链霉素、强力霉素和阿莫西林。但不可避免的会产生药物残留。随着国际上对抗生素药物的控制标准越来越严格,选择纯天然的绿色中药替代抗生素药物,成为新的发展方向。
发明内容
本发明要解决的问题是:克服现有技术的不足,提供一种疗效好、无药残、安全可靠的中药组合物。
本发明要解决的另一技术问题是:提供一种治疗水貂钩端螺旋体病的中药配方。
本发明要解决的另一技术问题是:提供上述中药组合物的颗粒剂的制备方法。
为解决上述技术问题,本发明采用的技术方案是:一种治疗水貂钩端螺旋体病的中药组合物,其特征在于:由如下质量份数的原料制备而成:茵陈10~20份、栀子10~20份、大黄5~15份、甘草5~10份、桃仁5~15份、芒硝5~15份、猪苓5~20份、茯苓5~15份、滑石5~10份、泽泻5~10份。
进一步地,该中药组合物的最佳原料的质量份数如下:
茵陈15份、栀子15份、大黄10份、甘草5份、桃仁10份、芒硝5份、猪苓15份、茯苓10份、滑石10份、泽泻5份。
进一步地,该中药组合物是含有或不含有药用辅料的各种常规中药口服药物制剂形式。
进一步地,所述制剂可以是颗粒剂。
进一步地,该组合物的颗粒剂的制备方法是:
(1)将中药粉碎至60目,按配方量称取,三维混合得中药粉;(2)加水煮2次,每次煮30min,过滤,合并两次滤液。将滤液浓缩至原体积的10%~20%,得浓缩浸膏;(3)称取元明粉30份,白砂糖20份,淀粉40份,混合三粉;(4)将浓缩浸膏10份分3次均匀加入混合粉中。一边搅拌一边加入,搅拌时间为10-15min;(5)压粒,60-80℃烘干即得成品。
本发明具有的优点和积极效果是:
(1)本发明针对抗生素药物残留问题,发明了一种纯天然、无污染、无残留的绿色中兽药。本组方依据中药理论,选用清热利湿,利胆退黄的药物进行配伍,并采用简便易行的制备方法得到一种稳定的中药颗粒剂。将本发明所制得的产品用于临床试验,对水貂钩端螺旋体病的治愈率达到95%以上,其疗效明显优于抗生素治疗方案,值得推广。
(2)本发明各中药原料的药理是:
茵陈:清利湿热,利胆退黄。
栀子:泻火除烦,清热利湿,凉血解毒。
大黄:泻下攻积,清热泻火,凉血解毒。
甘草:补脾益气,祛痰止咳,清热解毒,调和诸药。
桃仁:活血祛瘀,润肠通便,止咳平喘。
芒硝:泻下攻积,润燥软坚,清热消肿。
猪苓:利水渗湿。
茯苓:利水渗湿,健脾,宁心。
滑石:利尿通淋,清热解暑,收湿敛疮。
泽泻:利水渗湿。
依据中医观点,钩端螺旋体病,即传染性黄疸病属湿热黄疸。配方之中,茵陈苦泄下降,性寒清热,善清利脾胃肝胆湿热,使之从小便而出,为治黄疸之要药。栀子亦有清利下焦湿热之功效,可用治肝胆湿热郁结之黄疸。大黄能泻下通便,导湿热外出,故可治湿热蕴结之症。钩端螺旋体病的另一症状为喝水增加但却尿少、无尿。茯苓、猪苓、泽泻同为利水渗湿之药,茯苓味甘而淡,甘则能补,淡则能渗,药性平和,即可祛邪,又可扶正,利水而不伤正气,猪苓、泽泻甘淡渗泄,利水作用较强,滑石利尿通淋,四者合用,治疗水热互结,小便不利。桃仁富含油脂,能润燥滑肠,治疗肠燥便秘证。芒硝能泻下攻积,且性寒能清热,味咸润燥软坚,对湿热积滞,大便燥结者尤为适宜。芒硝与大黄相须为用,可增强泻下通便作用。甘草调和诸药,可解毒,亦可缓解大黄对肠胃的刺激,还可矫正方中药物滋味。诸药合用,既可清热利湿,利胆退黄,又可利水渗湿,润燥通便。

下面通过具体实施例对本发明作进一步详述,以下实施例只是描述性的,不是限定性的,不能以此限定本发明的保护范围。
实施例1
一种治疗水貂钩端螺旋体病的中药颗粒剂,各原料的重量份数如下:茵陈15份、栀子15份、大黄10份、甘草5份、桃仁10份、芒硝5份、猪苓15份、茯苓10份、滑石10份、泽泻5份。其制备方法是:(1)将中药粉碎至60目,按配方量称取,三维混合得中药粉;(2)加水煮2次,每次煮30min,过滤,合并两次滤液。将滤液浓缩至原体积的10%~20%,得浓缩浸膏;(3)称取元明粉30份,白砂糖20份,淀粉40份,混合三粉;(4)将浓缩浸膏10份分3次均匀加入混合粉中。一边搅拌一边加入,搅拌时间为10-15min;(5)压粒,60-80℃烘干即得成品。

实施例4

实施例6
一种治疗水貂钩端螺旋体病的中药颗粒剂,各原料的重量份数如下:茵陈10份、栀子10份、大黄13份、甘草8份、桃仁7份、芒硝7份、猪苓15份、茯苓15份、滑石7份、泽泻8份。其制备方法是:(1)将中药粉碎至60目,按配方量称取,三维混合得中药粉;(2)加水煮2次,每次煮30min,过滤,合并两次滤液。将滤液浓缩至原体积的10%~20%,得浓缩浸膏;(3)称取元明粉30份,白砂糖20份,淀粉40份,混合三粉;(4)将浓缩浸膏10份分3次均匀加入混合粉中。一边搅拌一边加入,搅拌时间为10-15min;(5)压粒,60-80℃烘干即得成品。
临床试验
实验1:
山东省某水貂养殖场10月龄左右的水貂陆续发病,出现粪便黄稀下痢,重者如煤焦油样,粘膜贫血、饮水迅猛,食欲减退、精神沉郁、眼部疾患、后肢瘫痪,脉搏增快。初步怀疑为钩端螺旋体病。经实验室病原检查确定为钩端螺旋体引起。遂将已发病的病貂隔离,并分组进行治疗。其中一组30头给予实施例1所制得的产品,每5g产品兑水1L,给病貂自由饮用,连续用药5d后,与治疗前相比,病貂精神恢复,烦渴多饮症状缓解,粪便成形颜色正常,基本康复,治愈率达到96.3%。另一组28头肌肉注射青霉素,每日2次,连续注射5天,大群治愈,治愈率88.2%。还有一组28头肌肉注射链霉素,每日2次,连续注射5天,大群治愈,治愈率86.4%。其他试验结果见表1。
实验2:
重庆某水貂养殖场从外地购入45日龄左右的水貂50只。饲养至第3日部分水貂发病,至第5日全群均有不同程度感染。病貂最初精神不振,不喜活动,体温升高,食欲不振,但尚有饮欲,排尿呈黄红色;严重者神志低落,目滞耳耸,食欲降低甚至废绝,粪便时有溏稀,眼结膜潮红,耳部皮肤渐有褐色痂块。对3只急性死亡病例进行剖检,均见皮肤、眼结膜等呈不同程度的黄染,下颌、头部轻度水肿,胸腹腔内有少量淡黄色积液,肝脏肿大呈黄褐色,肾脏稍肿,肾盂肾盏结构模糊不清,胃壁和肠系膜有小点出血,其中2只膀胱内积有少量尿液,色较黄。经临床和剖检诊断为钩端螺旋体病。遂给予全群实施例1所制得产品。每5g颗粒剂兑水1L,让病貂自由饮用,连续饮用7天,病情基本得到控制,各项指征恢复正常。

实验证明实施例1所制得产品疗效显著,明显优于抗生素药物,且具有统计学差异,P>0.05。

Claims (4)
1.一种治疗水貂钩端螺旋体病的中药组合物,其特征在于:由如下质量份数的原料制备而成:茵陈10~20份、栀子10~20份、大黄5~15份、甘草5~10份、桃仁5~15份、芒硝5~15份、猪苓5~20份、茯苓5~15份、滑石5~10份、泽泻5~10份。
2.根据权利要求1所述的治疗水貂钩端螺旋体病的中药组合物,其特征在于:最佳原料的质量份数如下:
茵陈15份、栀子15份、大黄10份、甘草5份、桃仁10份、芒硝5份、猪苓15份、茯苓10份、滑石10份、泽泻5份。
3.根据权利要求1所述的治疗水貂钩端螺旋体病的中药组合物,其特征在于:所述制剂可以是颗粒剂。
4.一种制备如权利要求3所述的治疗水貂钩端螺旋体病的中药组合物的方法,其特征在于,制备方法是:(1)将中药粉碎至60目,按配方量称取,三维混合得中药粉;(2)加水煮2次,每次煮30min,过滤,合并两次滤液。将滤液浓缩至原体积的10%~20%,得浓缩浸膏;(3)称取元明粉30份,白砂糖20份,淀粉40份,混合三粉;(4)将浓缩浸膏10份分3次均匀加入混合粉中。一边搅拌一边加入,搅拌时间为10-15min;(5)压粒,60-80℃烘干即得成品。
