本发明涉及一种番茄红素环糊精包合物的制备方法,属于食品添加剂及其制备方法领域。所述的番茄红素环糊精包合物的制备方法,该方法包括以下步骤:(1)精确称取精制的番茄红素和β-环糊精,将番茄红素用有机溶剂溶解;(2)将β-环糊精加入热水中,制成β-环糊精饱和溶液,在保持搅拌的情况下,将番茄红素溶液缓慢滴加至β-环糊精水溶液中,密封后,保持温度持续搅拌包合;(3)包合后打开容器,使丙酮挥发,继续搅拌,冷藏,过滤,并先后用蒸馏水和无水乙醚快速洗涤。(4)低温真空干燥,研碎即得番茄红素环糊精包合物。本发明所述的番茄红素环糊精包合物的制备方法,其包合率达到79.6%,产率达到86.3%。经实验证明,该番茄红素经β-环糊精包合后,大大提高了番茄红素的水溶性和稳定性。

技术领域
本发明涉及一种番茄红素的制备方法,属于食品添加剂及其制备方法领域。
背景技术
番茄红素是一类非常重要的类胡萝卜素,具有优越的生理功能。它具有较强的清除自由基、猝灭单线态氧的能力。其清除单线态氧的速率常数是目前常用抗氧化剂维生素E的100倍,是β-胡萝卜素的2倍。近来的流行病学研究表明,多食用番茄红素含量丰富的食物可大大降低人体患某些癌症的危险,番茄红素越来越多地被确认为同其他类胡萝卜素一起与癌危险呈负相关,甚至是唯一与某些癌症呈负相关的类胡萝卜素。现在有越来越多的研究表明,番茄红素可以防止前列腺癌、乳腺癌及消化道(包括结肠、直肠及胃)癌的发生。此外,番茄红素对预防和治疗心血管疾病、动脉硬化和肿瘤等各种成人病以及增强人体免疫等方面具有重要意义。最近几年来,番茄红素在人体健康及疾病防治方面的潜在作用引起了人们越来越浓厚的兴趣。
番茄红素是由11个共轭及两个非共轭碳-碳双键组成的直链型碳氢化合物,它的这一结构特点导致了它极不稳定,在光、热和氧的作用下很容易被氧化降解。再者,番茄红素是一种脂溶性的类胡萝卜素,在水中是不溶解的,这些特点都大大限制了它的应用。
近年来,环糊精包合技术在食品及医药中应用日益广泛,用环糊精做成的包和物能有效改善客体物质的理化性质,增加客体物质的溶解度和稳定性,提高药物的生物利用率,减少不良反应,掩盖不良气味等。此法具有很大的潜在应用价值,许多物质,如香料、色素、脂溶性维生素和许多药品都可以与环糊精形成包结络合物。
本发明提供一种用环糊精包合番茄红素,以期提高其对光、热、氧的稳定性,及其在水中的溶解能力的制备方法。
发明内容
本发明旨在提供一种能明显改善了对光、氧和热的稳定性的番茄红素环糊精包合物的制备方法。
本发明所述的番茄红素环糊精包合物的制备方法,该方法包括以下步骤:
(1)精确称取精制的番茄红素和β-环糊精,将番茄红素用有机溶剂溶解;
(2)将β-环糊精加入热水中,制成β-环糊精饱和溶液,在保持搅拌的情况下,将番茄红素溶液缓慢滴加至β-环糊精水溶液中,密封后,保持温度持续搅拌包合;
(3)包合后打开容器,使丙酮挥发,继续搅拌,冷藏,过滤,并先后用蒸馏水和无水乙醚快速洗涤。
(4)低温真空干燥,研碎即得番茄红素环糊精包合物。
优选的,本发明所述的步骤(1)中的番茄红素与β-环糊精的重量比1:200,有机溶剂为丙酮;
更优选的,本发明所述的步骤(2)中的热水温度为45-55℃,络合温度为50-55℃,络合时间为1-2h;
进一步优选的,本发明所述的步骤(3)中的搅拌时间为2h,冷藏温度为4-6℃, 冷藏时间为24h;
更进一步优选的,本发明所述的步骤(4)中的干燥温度为45-50℃,干燥时间为24h。
本发明所述的番茄红素环糊精包合物的制备方法,其包合率达到79.6%,产率达到86.3%。经实验证明,该番茄红素经β-环糊精包合后,大大提高了番茄红素的水溶性和稳定性。包合物可以在水中形成稳定的溶液,体系呈淡红色,并且明显改善了对光、氧和热的稳定性。
具体实施方式
实施例一:
精确称取精制的番茄红素20g和β-环糊精400g,然后将番茄红素用丙酮溶剂完全溶解;同时将β-环糊精加入50℃热水中,搅拌至溶解,制成β-环糊精饱和溶液,在保持搅拌的情况下,将番茄红素溶液缓慢滴加至β-环糊精水溶液中,密封后,保持50℃的温度下持续搅拌1h进行包合,然后打开容器,使丙酮挥发,继续搅拌2h,后在5℃下冷藏24h,过滤,并先后用蒸馏水和无水乙醚快速洗涤,在45℃下真空干燥24h,研碎即得番茄红素环糊精包合物。
经测定,本发明所述的番茄红素环糊精包合物的制备方法其包合率达到79.6%,产率达到86.3%。
并经实验证明,该番茄红素经β-环糊精包合后,大大提高了番茄红素的水溶性和稳定性。包合物可以在水中形成稳定的溶液,体系呈淡红色,并且明显改善了对光、氧和热的稳定性。
Claims (5)
1.番茄红素环糊精包合物的制备方法,该方法包括以下步骤:
(1)精确称取精制的番茄红素和β-环糊精,将番茄红素用有机溶剂溶解;
(2)将β-环糊精加入热水中,制成β-环糊精饱和溶液,在保持搅拌的情况下,将番茄红素溶液缓慢滴加至β-环糊精水溶液中,密封后,保持温度持续搅拌包合;
(3)包合后打开容器,使丙酮挥发,继续搅拌,冷藏,过滤,并先后用蒸馏水和无水乙醚快速洗涤;
(4)低温真空干燥,研碎即得番茄红素环糊精包合物。
2.如权利要求1所述的番茄红素环糊精包合物的制备方法,其特征在于所述的步骤(1)中的番茄红素与β-环糊精的重量比1:200,有机溶剂为丙酮。
3.如权利要求1所述的番茄红素环糊精包合物的制备方法,其特征在于所述的步骤(2)中的热水温度为45-55℃,络合温度为50-55℃,络合时间为1-2h。
4.如权利要求1所述的番茄红素环糊精包合物的制备方法,其特征在于所述的步骤(3)中的搅拌时间为2h,冷藏温度为4-6℃, 冷藏时间为24h。
5.如权利要求1所述的番茄红素环糊精包合物的制备方法,其特征在于所述的步骤(4)中的干燥温度为45-50℃,干燥时间为24h。
