Abstract

背景技术
核酸常规上用于基因疗法用于替换非功能基因[1],并且经RNA干扰(RNAi)效应器分子比如miRNAs[2]、shRNAs[3]和siRNAs[4]用于抵消致病的突变。由于快速清除[5]、核酸酶[6]、缺乏器官特异性分布和低效的细胞摄取,裸露DNA和RNA难以在体内递送,通常专用的基因递送载体用于递送。
病毒载体和阳离子脂质体是最前沿的递送载体技术并且已经是相对成功的,大量的这些递送载体已经在临床试验中[7]。尽管这些成功,仍有明显的局限限制许多应用,最显著的局限是对于大部分病毒载体的免疫识别[8,9,10]和对于病毒比如慢病毒的诱变整合[11];和炎症毒性和对于脂质体的快速清除[12,13,14,15]。通过先天免疫系统的识别导致急性炎症反应,其可需要使用免疫抑制策略以克服现有策略潜在地将患者暴露于机会性感染的冒险的摄取和再施用问题[16,17,18]。针对递送媒介产生的抗体也使得在随后的施用中转基因表达急剧地减少[19]。
现有策略的固有风险和限制已经通常将它们限于治疗的益处明显超过风险的威胁人类生命的疾病,比如重症联合免疫缺陷[20],限于特定环境比如类似眼的免疫豁免部位中的疾病[1],或用于基因接种[21]。但是,对于慢性的和使人虚弱但不是危及生命的遗传疾病,比如肌强直性营养不良,需要相当更低的风险特征并且维持矫正基因治疗的能力几十年,不是几年,用于治疗介入。对于次类疾病潜在地不可接受的风险的例子是以上讨论的免疫抑制策略,突出的是由于试验管理员未知的对象服用的免疫抑制剂[22]引起的机会性感染,在最近的风湿性关节炎的AAV基因治疗试验中健康患者的死亡。随着逐渐增多数量的疾病显示具有遗传成分,包括肥胖、心脏疾病和精神疾病,有很大的可能性修饰易感基因用于预先的遗传方案,但是仅仅如果风险进一步下降并且实现长期的维持能力。因此,需要开发这样的技术,其能够避免免疫识别和炎症,同时保留好的递送效率,以便扩展基因疗法在致死疾病之外的用途。
方案之一可能在于使用特异性靶向待治疗的细胞类型或器官的外切体用于基因递送。外切体是内吞起源的小膜结合囊泡(30-100nm),其在多泡体与质膜融合之后释放进入细胞外环境。已经对许多免疫细胞,包括B细胞、T细胞和树突细胞(DC)描述了外切体产生。衍生自B淋巴细胞和成熟DC的外切体表达MHC-II、MHC-I、CD86和ICAM-1[23,24],并且在实验模型和临床试验中已经用于诱导特定的抗肿瘤T细胞毒素应答和抗肿瘤免疫[24,25]。外切体介导的基因递送的潜能已经由递送鼠mRNAs和miRNAs至人肥大细胞显示[26],并且神经胶质瘤衍生的外切体[27]已经表明将外源DNA质粒产生的mRNA转移至异源细胞。也已经公开了具有外源DNA、siRNA和其他修饰寡核苷酸的外切体[28]。
发明内容
本发明人也已成功地将靶向部分引入外切体,从而外切体可被靶向所选择的组织。尤其,本发明人已成功地将运铁蛋白(Tf)肽靶向部分引入外切体,从而外切体可被靶向表达运铁蛋白受体(TfR)的组织或细胞。本发明人已经显示表达Tf肽靶向部分的外切体具有与商业上可获得的常规的转染试剂比如lipofectamine和RNAi max相当的摄取效率。因此,本发明涉及在表面上包括Tf肽靶向部分的外切体、包括外切体跨膜蛋白和靶向部分的融合蛋白,并且涉及编码此类融合蛋白的核酸构建体。Tf靶向外切体可用货物比如外源遗传物质或生物治疗性蛋白和/或肽、和/或化疗试剂装载并且用于货物的递送,尤其用于核酸的递送用于基因治疗和基因沉默,以及生物治疗性蛋白和肽和化疗试剂的递送用于治疗。
根据本发明的一个方面,提供了包括外切体的组合物,其中外切体包括在外切体的表面上表达的靶向部分,并且所述靶向部分是运铁蛋白(Tf)多肽或肽。Tf多肽靶向部分或Tf肽靶向部分可以是保留与TfR结合的能力的任何Tf多肽或Tf肽。
在另一方面中,本发明提供包括外切体的组合物,其用于体内递送遗传物质、蛋白质和/或肽的方法中,该外切体包括在外切体的表面上表达的Tf多肽靶向部分或Tf肽靶向部分,其中该外切体衍生自不成熟的树突细胞。

具体实施方式
本发明涉及外切体,以及它们作为递送媒介的用途。外切体是内吞起源的小膜结合囊泡,其在多泡体与质膜融合之后被释放进入细胞外环境。因此,本申请涉及包括此类外切体的组合物。典型地,外切体的直径在30和100nm之间,但是可包括多达200nm的相似起源的膜颗粒。如本文所使用,外切体指由多泡体分泌的内体起源的纳米颗粒。
外切体由许多不同类型的细胞产生,包括免疫细胞比如B淋巴细胞、T淋巴细胞、树突细胞(DC)和肥大细胞。外切体也由以下产生,例如神经胶质瘤细胞、血小板、网织红细胞、神经元、小肠上皮细胞、肿瘤细胞、HELA细胞、人胚胎肾细胞(HEK细胞)、B2M17细胞、Bend3细胞、原代骨髓衍生的树突细胞、BV-2小胶质细胞和脑神经瘤(NEURO2A)细胞。根据本申请使用的外切体可衍生自任何适合的细胞,包括以上识别的细胞。外切体也已经从生理液体分离,比如血浆、尿、羊膜液和恶性积液。
在本发明的优选方面中,外切体衍生自DC,优选不成熟的DC。由不成熟的DC产生的外切体不表达MHC-II、MHC-I或CD86。如此,此类外切体不明显刺激幼稚T细胞并且不能在混合的淋巴细胞反应中诱导应答。因此,由不成熟的树突细胞产生的外切体是用于递送货物,比如遗传物质,尤其用于体内使用,例如基因疗法中的理想候选物。
因此,根据本发明的一个方面,提供衍生自树突细胞的外切体,用于货物——比如遗传物质和蛋白质和/或肽生物治疗剂或化疗剂——的体内靶向递送。在优选的实施方式中,本发明的外切体衍生自树突细胞并且提供用于遗传物质、蛋白质或肽生物治疗剂或化疗剂的体内靶向递送。
如上所概述,外切体由许多不同类型的细胞产生并且也已经从生理液体分离。因此,根据本发明,外切体可从以上讨论的任何合适的细胞类型获得,或通过从生理液体分离获得。典型地,本发明的方法包括从细胞培养基或组织上清液分离外切体。
可通过任何合适的方法从培养基收集由细胞产生的外切体。典型地,外切体的制品可通过离心、过滤或这些方法的组合从细胞培养物或组织上清液制备。例如,外切体可通过如下制备:差速离心,即低速(<20000g)离心以使较大颗粒成团,接着通过高速(>100000g)离心以使外切体成团、用合适的过滤器(例如,0.22μm过滤器)的粒度过滤、梯度超速离心(例如,蔗糖梯度)或这些方法的组合。
根据本发明,本发明的外切体可用外源遗传物质、蛋白质或肽或化疗剂装载。尤其,根据本发明,可制备外切体并且然后用期望的遗传物质、蛋白质或肽、或化疗剂装载,用于靶向递送。外切体也可用外源遗传物质或生物治疗性蛋白或肽装载,通过在细胞中过表达编码遗传物质或蛋白质或肽的合适的构建体,从而将物质装载进入外切体。
相比递送这种分子的常规手段,Tf-靶向的外切体递送外源遗传物质、蛋白质或肽的用途提供了许多优势。例如,当化合物使用外切体递送时,它们被保护避免降解并且更稳定;化合物可被递送至靶组织,比如特定类型的癌症,比如果没有封装在外切体内更有效和/或更特异;和/或该货物可能不太可能引起免疫应答,因为它包含在外切体中并且所以不能自由地被免疫细胞探测到,和/或结合抗体。Tf-靶向的外切体递送外源遗传物质、蛋白质或肽或化疗剂用途的其他潜在的优势包括在外切体的全身性递送之后绕过肝细胞和/或避免耐药性,比如药物转运体比如ABC-转运体的上调。
用于递送的适合的遗传物质包括外源DNA质粒和siRNA。合适的遗传物质也包括修饰的寡核苷酸和其他RNA干扰效应器部分比如miRNA和shRNA。根据本发明的一个方面,装载进入外切体的遗传物质不是通常与外切体相关的遗传物质,例如,核酸优选地不是当其由细胞产生时并入外切体的mRNA或miRNA。尤其,遗传物质可装载进入已经与细胞分离的外切体制品。可选地遗传物质可以在细胞中过表达,例如通过将合适的构建体引入细胞,并且随后装载进入外切体。因此,外源遗传物质指包括通常与外切体不相关的核酸的遗传物质。在尤其优选的实施方式中,核酸物质是通常不出现在外切体中的质粒DNA或其他核酸比如siRNA和修饰的寡核苷酸。
相似地,优选地装载进入外切体的蛋白质或肽不是通常与外切体相关的蛋白质或肽,例如,蛋白质或肽优选地不是当其由细胞——比如未修饰的细胞——产生时被并入外切体的蛋白质或肽。外源蛋白质或肽指包括通常与外切体不相关的蛋白质或肽的蛋白质或肽。此类外源蛋白质或肽可装载进入已经与细胞分离的外切体,或通过在宿主细胞中过表达合适的构建体,以便蛋白质或肽被装载进入外切体。在尤其优选的实施方式中,待装载进入外切体的蛋白质或肽是抗体或抗体片段,更优选地是单克隆抗体或其片段。

用于病况、疾病或不适的治疗和/或预防的任何生物治疗性蛋白和/或肽可并入根据本发明的外切体。在优选的实施方式中,并入外切体的蛋白质或肽是抗体或抗体片段。在更优选的实施方式中,抗体是单克隆抗体或其片段。
外切体也可用化疗剂——比如治疗药剂——装载,用于化疗剂的递送。合适的药剂的例子包括细胞毒素剂,例如用于癌症的治疗。
基于遗传物质、蛋白质或肽对其期望递送进入细胞的效果和进行该效果的机理选择待装载进入外切体的遗传物质、蛋白质或肽。例如,核酸可用于基因治疗,例如为了在细胞或细胞组中表达期望的基因。此类核酸通常以编码期望基因的质粒DNA或病毒载体的形式,并且其可操作地连接至合适的调节序列比如启动子、增强子等,使得一旦其被递送至待治疗的细胞,则表达质粒DNA。对基因治疗易感的疾病的例子包括血友病B(因子IX)、囊性纤维化(CTFR)和脊髓性肌萎缩(SMN-1)。
核酸可也用于例如免疫以表达期望针对其产生免疫应答的一种或多种抗原。因此,待装载进入外切体的核酸可编码期望针对其产生免疫应答的一种或多种抗原,包括但不限于肿瘤抗原,来自病原体比如病毒、细菌或真菌病原体的抗原。
核酸也可用于基因沉默。这种基因沉默可在治疗中用于关闭异常基因表达或在动物模型研究中用于产生单个或更多个遗传敲除。通常,此类核酸以siRNAs的形式提供。例如,包括siRNAs的RNAi分子可用于在肌细胞中敲低具有多个CUG重复的DMPK以治疗肌强直性营养不良。在其他实施例中,表达减少造成亨廷顿病的突变体亨廷顿基因(htt)的shRNA的质粒可用神经元特异外切体递送。其他靶基因包括用于治疗阿尔茨海默氏疾病的BACE-1。一些癌基因也可用siRNA或shRNAs比如ras、c-myc和VEGFR-2靶向。脑靶向siRNA装载的外切体可尤其用于阿尔茨海默氏疾病中BACE-1的沉默、帕金森疾病中α-突触核蛋白(synuclein)的沉默、亨廷顿疾病中htt的沉默和用于治疗中风以降低缺血损坏的神经元胱冬酶-3的沉默。
可使用包括2’-O-Me化合物和PNA的反义修饰的寡核苷酸。例如,此类寡核苷酸可设计为诱导外显子跳跃,例如突变体抗肌萎缩蛋白基因可被递送至肌细胞用于治疗杜兴氏肌营养不良症,例如抑制发夹环的反义寡核苷酸用于治疗肌强直性营养不良和例如反式剪接寡核苷酸用于治疗脊髓性肌萎缩。
作为用遗传物质装载本发明外切体的可选项,可装载蛋白质或肽。用于病况、疾病或不适的治疗和/或预防的任何生物治疗性蛋白和/或肽可并入根据本发明的外切体。在优选的实施方式中,并入外切体的蛋白质或肽是抗体或抗体片段。单个蛋白质或肽可并入外切体。可选地,多于一种的蛋白质和/或肽可并入外切体。多于一种的蛋白质和/或肽可作用在相同的或不同的靶上以带来其治疗和/或预防效果。
待并入外切体的蛋白质和/或肽可例如用于癌症的治疗和/或预防。可使用蛋白质或肽生物治疗剂治疗的癌症的例子包括白血病、结直肠癌、头颈癌、非霍奇金氏淋巴瘤、乳腺癌、卵巢癌、前列腺癌、胃癌、胰腺癌、腺癌和实体肿瘤。此类生物治疗性蛋白和/或肽典型地是抗体或其片段,尤其是单克隆抗体或其片段。
蛋白质和/或肽生物治疗也可用于自身免疫性疾病的治疗和/或预防。自身免疫性疾病起因于身体对通常存在于身体中的物质、组织和器官的过度活跃的免疫应答。
装载进入根据本发明的外切体的生物治疗性蛋白和/或肽也可用于治疗和/或预防其他病况,包括心血管疾病、血友病、脓毒、中风、包括杜兴氏肌营养不良(DMD)的肌营养不良、黄斑变性和阿尔茨海默氏疾病。
在优选的实施方式中,装载进入根据本发明的外切体的生物治疗剂是治疗性抗体。在更优选的实施方式中,治疗性抗体选自用于封闭Bace-1和b-淀粉样蛋白(b-amyloids)的活化位点的单克隆抗体;针对成胶质细胞瘤激酶的单克隆抗体和针对α4整联蛋白质用于治疗MS的抗体,例如那他珠单抗(Natalizumab)。
在优选的实施方式中,装载进入根据本发明的外切体的生物治疗剂是肽。在更优选的实施方式中,肽选自用于引起对病毒和/或肿瘤抗原的免疫应答的免疫显性(immune-dominant)肽;神经保护肽,比如δ-阿片样物质受体配体,例如Biphalin;用于神经炎症的免疫抑制肽,例如肾上腺髓质素和结合并且抑制NfKB信号传导的NBD肽。
外源遗传物质、蛋白质或肽或化疗剂可通过许多不同的技术引入外切体。例如,外切体可通过电穿孔或使用转染试剂装载。尽管外切体的小尺寸,仍可能使用电穿孔用外源遗传物质装载外切体[WO2010/119256]。考虑到与细胞相比外切体的小尺寸,这是令人吃惊的。考虑外切体的尺寸,用于细胞电穿孔的电压的外推将提示外切体的电穿孔将需要超高电压。但是,令人吃惊地,使用电穿孔用质粒DNA和siRNA装载外切体是可能的。电穿孔条件可根据货物的电荷和尺寸而改变。典型的电压范围为20V/cm至1000V/cm,比如20V/cm至100V/cm,电容通常在25μF和250μF之间,比如25μF和125μF之间。对于用抗体装载本发明的外切体,范围为150mV至250mV,具体地200mV的电压是优选的。

外切体也可通过用核酸构建体转化或转染宿主细胞装载,所述核酸构建体表达感兴趣的遗传物质或生物治疗性蛋白或肽,以便随着在细胞中产生外切体,遗传物质、生物治疗性蛋白或肽摄入外切体。
靶向
根据本发明,外切体靶向期望的细胞类型或组织。通过在外切体的表面上表达靶向部分实现该靶向,所述靶向部分结合在待靶向的细胞表面上表达的细胞表面部分。根据本发明,靶向部分是运铁蛋白(Tf)多肽或肽。在优选的实施方式中,靶向部分是Tf肽。典型地,Tf多肽靶向部分或Tf肽靶向部分表达为与通常在外切体的表面上表达的跨膜蛋白的融合蛋白。
Tf是糖蛋白,其以高亲和力可逆地结合铁。人运铁蛋白(Tf-h)由具有698个氨基酸残基的单条多肽链组成并且分子量为约80kDa。Tf-h的多肽序列显示在SEQ ID NO:1中。人Tf基因的多核苷酸序列显示在SEQ ID NO:2中。Tf-h以高亲和力结合两个Fe3+离子。四个不同的具有不同铁含量的同种型Tf共存在血浆中:(1)apo-运铁蛋白(APOTf,没有铁离子);(2)单铁运铁蛋白(铁在C-末端结构域);(3)单铁运铁蛋白(铁在N-末端结构域)和(4)二铁运铁蛋白(铁在两个结合位点)。在本发明的上下文中,Tf包括任何这四个同种型,单独或彼此组合。
Tf受体(TfR)是二硫化物-连接的同型二聚体糖蛋白,其在许多肿瘤的细胞表面高度表达。在本发明的优选实施方式中,存在于待被本发明的外切体靶向的细胞上的部分是TfR,其结合在外切体的表面上表达的Tf多肽或Tf肽靶向部分。
更详细地,本发明的外切体可通过在它们表面上表达Tf多肽靶向部分或Tf肽靶向部分,靶向具体的细胞类型或组织。合适的肽是结合在待靶向细胞的细胞表面上出现的细胞表面部分比如受体或它们的配体的那些。在优选的实施方式中,Tf多肽靶向部分或Tf肽靶向部分结合待靶向细胞的细胞表面上的TfR。合适的靶向部分的例子是短肽和完整的Tf蛋白,只要靶向部分可在外切体的表面上表达并且不干扰膜蛋白插入外切体。典型地,靶向肽相对于跨膜外切体蛋白是异源的。肽靶向部分的长度可通常小于100个氨基酸,例如小于50个氨基酸的长度,小于30个氨基酸的长度,至10、5或4个氨基酸的最小长度。
在优选的实施方式中,最小Tf靶向部分的氨基酸序列为SEQ IDNO:1的Tf蛋白的氨基酸50至56,即氨基酸序列PSDGPSV(SEQ IDNO:4)。
在尤其优选的实施方式中,Tf靶向多肽或肽由下述组成或包括下述:氨基酸序列CRTIGPSVC(SEQ ID NO:3)和/或SEQ ID NO:1的Tf蛋白的氨基酸50至56,即氨基酸序列PSDGPSV(SEQ ID NO:4)。
包括SEQ ID NOS:3或4的氨基酸序列的肽片段可用作本发明的靶向部分。这些片段的长度可为10个氨基酸或更短,20个氨基酸或更短的长度,30个氨基酸或更短的长度,50个氨基酸或更短的长度,100个氨基酸或更短的长度或更多。在优选的实施方式中,片段的长度为10个氨基酸或更短。片段优选地是SEQ ID NO:1的Tf蛋白片段。全长Tf蛋白可也用作本发明外切体中的靶向部分。
TfR在不成熟的红系细胞、胎盘组织和快速分裂的正常和恶性细胞中高度表达。因此,比如例如Tf蛋白、多肽和肽靶向部分可用于靶向具体的组织类型,或靶向患病的组织比如肿瘤。在本发明尤其优选的实施方式中,外切体靶向肿瘤。
本发明的Tf靶向外切体可特别有利地使货物能够递送至所有的细胞类型并且跨物种。这是因为Tf在跨物种中是高度保守的并且TfR在许多细胞类型的表面上普遍表达。因此,例如,本发明的小鼠外切体可被人细胞摄取并且本发明的人外切体可被小鼠细胞摄取。如此,Tf靶向外切体可认为是通用的体外转染试剂。另外,体内Tf靶向外切体理想地用于靶向具有提高的TfR表达的靶向组织,比如血脑屏障(BBB)的内皮细胞,肿瘤细胞和局部缺氧的位点(如在心脏的局部缺血或中风后的局部缺血)。
因此,本发明的Tf靶向外切体的潜在用途包括横跨BBB的遗传和非遗传货物的靶向;癌症疗法对肿瘤的靶向,因为相对于周围组织,肿瘤细胞表达显著提高水平的TfR;和疗法对涉及局部缺血的疾病的靶向,比如其中也具有提高水平的TfR表达的缺血性心脏病和中风。
与非靶向的外切体相比,在根据本发明的外切体的表面上Tf靶向部分的表达可增加由靶细胞对所述外切体和其货物的摄取至少两倍、至少三倍、至少四倍、至少五倍、至少十倍或至少二十倍。与使用常规商业上可获得的转染试剂比如lipofectamine或RNAi max递送相同的货物的摄取水平相比,在根据本发明的外切体的表面上Tf靶向部分的表达可产生的靶细胞对外切体货物的摄取水平相当。可选地,与使用常规商业上可获得的转染试剂递送相同的货物的摄取水平相比,在根据本发明的外切体的表面上Tf靶向部分的表达可增加通过靶细胞对外切体货物的摄取至少两倍、至少三倍、至少四倍、至少五倍、至少十倍或至少二十倍。
通过将其表达为与外切体跨膜蛋白的融合蛋白,Tf多肽靶向部分或Tf肽靶向部分在外切体的表面上表达。已知许多蛋白质与外切体相关;即随着其形成,它们被并入外切体。根据本发明使用的优选的蛋白质是跨膜蛋白的那些。例子包括但不限于Lamp-1、Lamp-2、CD13、CD86、Flotillin、Syntaxin-3、CD2、CD36、CD40、CD40L、CD41a、CD44、CD45、ICAM-1、整合蛋白α4、LiCAM、LFA-1、Mac-1α和β、Vti-1A和B、CD3ε和ζ、CD9、CD18、CD37、CD53、CD63、CD81、CD82、CXCR4、FcR、GluR2/3、HLA-DM(MHC II)、免疫球蛋白、MHC-I或MHC-II组分、TCRβ和四次跨膜蛋白。在本发明尤其优选的实施方式中,跨膜蛋白选自Lamp-1、Lamp-2、CD13、CD86、Flotillin、Syntaxin-3。在尤其优选的实施方式中,跨膜蛋白是Lamp-2。Lamp-2的序列在SEQ ID NO:5中示出。

外切体跨膜蛋白被修饰以并入Tf靶向部分。因此,外切体跨膜蛋白表达为包括Tf靶向部分的融合蛋白。Tf靶向部分被并入跨膜蛋白以便将其置于在外切体的表面存在的跨膜蛋白部分中。在本发明优选的方面中,外切体跨膜蛋白是Lamp-2并且Tf靶向部分表达为融合蛋白,其中Tf靶向部分存在于Lamp-2蛋白的N-末端附近,例如在Lamp-2N末端氨基酸的30个或20个氨基酸内,不包括信号序列。
例如,可在Tf靶向部分和跨膜蛋白的剩余部分之间提供间隔序列或接头序列,以避免在跨膜蛋白折叠时来自Tf靶向部分的干扰。

在本发明的实践中,通过在用于产生外切体的细胞中表达包括Tf靶向部分和外切体跨膜蛋白的融合蛋白,将Tf靶向部分引入外切体。该融合蛋白在细胞中的表达,允许融合蛋白随着其由细胞产生并入外切体。
例如,表达融合蛋白的多核苷酸构建体比如DNA质粒被转染进入细胞。任何适合的方法可用于将多核苷酸构建体引入细胞。多核苷酸构建体包括合适的启动子序列从而编码的融合蛋白在细胞中表达。也包括信号肽序列从而随着其产生,蛋白质被并入内质网膜。然后,膜蛋白随后被输出至外切体/溶酶体区室,之后并入外切体。信号序列通常是用于外切体跨膜蛋白的信号肽序列。例如信号肽序列优选衍生自Lamp-2。
以上详细讨论了用于产生外切体的优选细胞。通常,优选的细胞,比如不成熟的树突细胞用如上述的多核苷酸构建体转染,以便本发明的融合蛋白在细胞中表达。然后可收集由细胞产生的外切体。这种外切体具有插入膜的融合蛋白,以便外切体通过靶向部分靶向期望的组织或细胞类型。
可通过任何合适的方法从培养基收集由细胞产生的外切体。通常,外切体的制品可通过离心、过滤或这些方法的组合从细胞培养物或组织上清液制备。例如,外切体可通过如下制备:差速离心,即低速(<20000g)离心以使较大颗粒成团,接着通过高速(>100000g)离心以使外切体成团,用适合的过滤器(例如,0.22μm过滤器)的粒度过滤、梯度超速离心(例如,用蔗糖梯度)或这些方法的组合。
根据本发明的优选方面,靶向的外切体用外源遗传物质装载。尤其,根据本发明,如本文所述制备具有靶向部分的外切体,并且然后如上述用期望的用于递送遗传物质装载。
在本发明的一些实施方式中,选择外切体以便它们更容易靶向特定的组织类型。例如,衍生自不同细胞的外切体可对特定细胞亚型具有如它们的生理机能需要的天然亲和力,比如成熟的树突细胞衍生的外切体对T-细胞充分确立的亲和力。该亲和力可用于帮助将以上提到的货物特定地递送至组织。
递送/施用

本文提供的组合物可另外包含常规存在于药学组合物中的其他助剂组分。因此,例如,组合物可包含另外的兼容的药学活性材料,或可包含用于物理配制本发明组合物不同剂量形式的另外的材料,比如染料、增香剂、防腐剂、抗氧化剂、遮光剂、增稠剂和稳定剂。但是,当添加时,这种材料不应过分干扰本文提供组合物的组分的生物活性。
施用治疗上有效量的组合物。可根据不同的参数确定剂量,尤其根据待治疗患者的病况的严重性、年龄和体重;施用路径;和需要的用药法。医师将能够确定任何具体患者需要的施用路径和剂量。取决于单个构建体的相对功效,最佳剂量可以不同,并且通常可基于在体外和体外动物模型中发现有效的EC50s估计。一般而言,剂量从0.01mg/kg体重至100mg/kg体重。根据具体构建体的效力、待治疗对象的年龄、体重和病况、疾病的严重性以及施用频率和路径,典型的日剂量是从约0.1至50mg/kg体重,优选地从约0.1mg/kg至10mg/kg体重。可施用不同剂量的构建体,这取决于施用是否通过肌内注射或全身性(静脉内或皮下)注射。优选地,单个肌内注射的剂量范围是约5至20μg。优选地,单个或多个全身性注射的剂量范围为10至100mg/kg体重。
由于构建体清除(和任何靶向分子的分解),患者可能不得不反复治疗,例如每天、每周、每月或每年一次或多次。基于测量的构建体在体液或组织中残留的时间和浓度,本领域普通技术人员可容易评估给药的重复速度。成功的治疗之后,可能期望患者进行维持治疗,其中构建体以维持剂量施用,范围是0.01mg/kg至100mg/kg体重,每天一次或更多次至每20年一次。

实施例
实施例1–产生编码Tf靶向部分的构建体
Tf靶向配体附着至信号肽之后的N-末端,因为Lamp2b是I型膜蛋白,具有预测从外切体突出的N-末端。Tf靶向肽插入Lamp2b序列,以便其可位于外切体的外表面上,因此赋予外切体靶向能力。
由多核苷酸序列TGTCGTACCATCGGACCAAGTGTTTGT(SEQID NO:7)编码的Tf-衍生的肽CRTIGPSVC(SEQ ID NO:3)被插入之前国际公开号WO/2010/119256中描述的Lamp2b-表达载体。
表达载体基于pEGFP-C1载体(Clonetech),其中eGFP基因已经被去除。Lamp2b用来自C2C12细胞的cDNA克隆并且XhoI和BspEI限制酶切位点在信号肽序列之后与甘氨酸接头(Ala-Arg-{靶向肽}-Ser-Gly-Gly)一起插入。Lamp2b的信号肽对于膜插入是需要的,但是在成熟的蛋白质中被切除。全部的构建体然后在CMV启动子的下游用NheI和BamHI限制酶切位点克隆进入pEGFP-C1载体,在该过程中去除eGFP。

在Tf靶向肽侧翼的甘氨酸接头避免Tf靶向肽影响Lamp2b蛋白质的折叠。最后,Tf靶向肽应位于外切体的外表面上,从而赋予外切体靶向能力。
多核苷酸序列TGTCGTACCATCGGACCAAGTGTTTGT(SEQ IDNO:7)编码的Tf靶向肽CRTIGPSVC(SEQ ID NO:3)被克隆进入Lamp2b并且在外切体纯化之前4天转染进入树突细胞。在外切体的表面上表达靶向配体的能力的主要障碍是主要树突细胞难以转染并且可潜在地在转染之后分化。病毒载体的感染是不理想的,或者因为树突细胞可能被病毒活化[30],因此产生免疫刺激分子,其将并入至所得的外切体。选择Mirus Bio’s TransIT-LT1试剂,因为其好像有效地转导树突细胞而不显著地活化树突细胞。用5μg的pLamp2b-Tf肽表达载体和5μl的TransIT LT1转染试剂(Mirus Bio)在具有106细胞的6-孔板中在收获之后的第4天进行树突细胞的转染并且在第8天分离外切体。

细胞培养
在DC培养基(DMEM Glutamax,10%FBS,非必需氨基酸#和抗生物素)中培养树突细胞。在补充20%FBS和抗生物素的DMEMGlutamax中培养C2C12细胞。
实施例2–通过使用表达Tf靶向部分的外切体递送的siRNA的基因敲低
我们的目的是比较通过使用在它们的表面具有Tf靶向部分的外切体递送的siRNA的靶基因的敲低与通过使用非靶向外切体、由RVG肽靶向的外切体和使用常规的siRNA转染试剂递送的siRNA实现的敲低。
使用Tf靶向外切体、RVG靶向的外切体、非靶向外切体和两个商用siRNA转染试剂(lipofectamine(LIPO)和RNAi max)将抑制GAPDH的siRNA递送至人细胞系(HEK和B2M17)和小鼠细胞(N2A和骨髓树突细胞)。在使用外切体的情况下,这些衍生自树突细胞。对于每个外切体处理,除了电穿孔的外切体(E),还包括由与siRNA混合但是没有电穿孔的外切体(NE)组成的阴性对照。如在图1至4中可见,Tf-外切体比非靶向的外切体和RVG外切体在降低所有细胞类型中的GADPH表达方面更好,并且效率也与商用转染试剂lipofectamine和RNAi max相当。

Claims (25)

5.根据前述权利要求任一项所述的组合物,其中所述靶向部分包括与存在于待靶向的细胞上的部分结合的肽。
6.根据权利要求5所述的组合物,其中所述存在于待靶向的细胞上的部分是运铁蛋白受体(TfR)。
7.根据权利要求5或6所述的组合物,其中所述外切体包括外切体跨膜蛋白,其已经被修饰以并入所述肽靶向部分。
8.根据权利要求7所述的组合物,其中所述外切体跨膜蛋白选自Lamp-1、Lamp-2、CD13、CD86、Flotillin、Syntaxin-3。
9.根据权利要求8所述的组合物,其中所述外切体跨膜蛋白是Lamp-2b。
10.根据权利要求9所述的组合物,其中所述靶向部分存在于Lamp-2b的N-末端处或其附近并且用接头序列与Lamp-2b分开。
11.根据前述权利要求任一项所述的组合物,其中所述外切体衍生自树突细胞。
12.根据前述权利要求任一项所述的组合物,其中所述外切体用外源遗传物质、蛋白质和/或肽或化疗剂装载。
13.根据权利要求12所述的组合物,其中所述外源遗传物质是编码治疗性蛋白或免疫原的DNA质粒。
14.根据权利要求12所述的组合物,其中所述外源遗传物质是siRNA。
15.根据权利要求13所述的组合物,其中所述质粒编码用于基因治疗方法中的治疗性蛋白。
16.根据权利要求13所述的组合物,其中所述质粒编码免疫原,其用于对所述免疫原产生免疫应答的方法中。
17.根据权利要求12至16的任一项所述的组合物,其中所述外切体衍生自不成熟的树突细胞,其用于体内递送所述遗传物质、蛋白质和/或肽或化疗剂的方法中。
18.产生外切体的方法,所述外切体包括在所述外切体的表面上表达的Tf多肽靶向部分或Tf肽靶向部分,所述方法包括在用于产生外切体的细胞中表达包括所述靶向部分和外切体跨膜蛋白的融合蛋白,其中随着所述细胞产生所述融合蛋白,表达的所述融合蛋白并入所述外切体。
19.根据权利要求18所述的方法,其中编码所述融合蛋白的多核苷酸构建体转染进入所述细胞,所述多核苷酸构建体进一步包括启动子和信号肽序列。
20.根据权利要求18或19所述的方法,其中所述细胞是树突细胞。
21.包括外切体跨膜蛋白和异源靶向Tf多肽或Tf肽的多肽,其中所述靶向Tf多肽或Tf肽与待靶向的细胞表面上存在的Tf受体(TfR)结合,并且其中当所述多肽存在于外切体中时,所述靶向Tf多肽或Tf肽存在于所述外切体的表面上。
22.编码如权利要求22所限定的多肽的多核苷酸构建体,其中所述构建体包括编码5’外切体跨膜信号序列的多核苷酸,其可操作地连接至编码本发明所述多肽的多核苷酸。

24.将外切体靶向所选择的组织或细胞类型的方法,所述方法包括用如权利要求22或23所限定的多核苷酸构建体转染宿主细胞,在所述宿主细胞中表达所述构建体,和从其中已经表达所述构建体的所述宿主细胞中获得外切体。
