Abstract
本发明公开了一种乌鸡肽铁螯合物粉制备方法,步骤包括:乌鸡肉浆水解后过滤得到乌鸡活性肽溶液;把乌鸡活性肽溶液转入真空反应釜中,与氯化亚铁反应得到乌鸡肽铁螯合物;然后经陶瓷滤膜过滤、乙醇洗涤、喷雾干燥,并在喷雾干燥时采用紫外光杀菌,最后真空包装得到乌鸡肽铁螯合物粉。本发明缩短了生产周期,提高了生产效率;在喷雾干燥时采用紫外光杀菌,所得产品无菌,并且产品为粉剂,真空包装,保存时间长。

技术领域
本发明涉及一种制备肽铁螯合物制备方法,尤其涉及一种乌鸡肽铁螯合物粉制备方法。
背景技术
铁是人体必需的化学元素之一,人体缺铁或铁利用不良时会影响血红蛋白的合成,引起缺铁性贫血。硫酸亚铁等非血红素铁价格低廉,但易与食物中植酸、多酚等成分形成难溶性化合物,吸收率较低,并易产生内源性自由基导致细胞膜脂质过氧化。血红素铁比非血红素铁的利用率高,但最高也只达25%左右并具有异味。活性肽铁是以蛋白质水解产生的分子活性肽为原料,与铁离子络合形成的螯合物。与氨基酸相比,肽的吸收效率更高且不易饱和。以活性肽为原料开发吸收利用率更高、生理作用更强的新型补铁剂。目前国内此类专利申请,如乳蛋白酶解液与亚铁盐螯合(200810240344.4)、酪蛋白肽螯合铁(201010040074.X)、椰子球蛋白亚铁络合物制备补铁剂(201010203183.9)、卵铁离子传递蛋白补铁剂(200610012762.9) 等,然而,这些物质多属于蛋白酶解物与铁的螯合,组成成分的分子量不清,而一般蛋白酶解物的分子量常高于5000Da,在人体吸收利用率和生理活性方面差强人意。乌鸡肽,又称乌鸡低聚肽,是以乌鸡蛋白为原料,酶解后获得的低分子量肽产物。低聚肽吸收速度快、耗能低、易于被人体消化、吸收和利用,具有降血压、抗氧化、抗疲劳、免疫调节等重要的生理活性。所以乌鸡肽铁螯合物是良好的补铁剂。
申请号201210380796.9的专利申请公开了一种乌鸡肽铁螯合物的制备方法,其特征在于该方法包括在乌鸡肽溶液中加入抗坏血酸,并且在pH3-6 的条件下,将乌鸡肽溶液与亚铁盐恒温下螯合,将螯合液用无水乙醇进行沉淀,得到乌鸡肽铁螯合物。这种方法沉淀时间长,占用场地大。
发明内容
本发明的目的在于提供一种乌鸡肽铁螯合物粉制备方法,该方法生产周期短,节约生产空间。
本发明采用的技术方案是: 乌鸡肽铁螯合物粉制备方法,其特征是包括如下步骤:1)把乌骨鸡宰杀后去毛,去内脏,去鸡头鸡爪,然后用绞肉机打成肉浆;2)加入风味蛋白酶水解肉浆,然后过滤得到乌鸡活性肽溶液;3)把乌鸡活性肽溶液转入真空反应釜中,调节PH为3-6,加入乌鸡肽质量的10%的氯化亚铁,加入0.5%的抗坏血酸,超声搅拌3-5分钟,使乌鸡活性肽与氯化亚铁混合均匀,然后加热到35-45℃,静置反应1-2h;然后用陶瓷滤膜过滤,得到乌鸡肽铁螯合物浓缩液,然后用乙醇洗涤浓缩液,再过滤得到高纯乌鸡肽铁螯合物,之后采用喷雾干燥器干燥高纯乌鸡肽铁螯合物得到乌鸡肽铁螯合物粉,并在喷雾干燥时采用紫外光杀菌,最后真空包装。
所述紫外光的波长为250-361nm。
本发明的有益效果是:1)乌鸡肽铁螯合物粉以小分子乌鸡肽作为新型补铁剂的原料,能够利用乌鸡的补血功效,并充分结合小分子活性肽的吸收利用率优势,以及乌鸡肽本身的生物活性,是具有增强生物吸收和补铁效果的新型补铁剂;2)采用陶瓷滤膜过减少浓缩沉淀时间,缩短了生产周期,提高了生产效率;3)在喷雾干燥时采用紫外光杀菌,所得产品无菌,并且产品为粉剂,真空包装,保存时间长。
具体实施方式
为了便于说明,下面结合实施例详细阐明本发明。
实施例1
把20kg乌骨鸡宰杀后去毛,去内脏,去鸡头鸡爪,然后用绞肉机打成肉浆;加入风味蛋白酶水解肉浆,然后过滤得到乌鸡活性肽溶液,测定乌鸡活性肽含量,乌鸡活性肽含量为1kg,把乌鸡活性肽溶液转入真空反应釜中,调节PH为4左右,加入100g氯化亚铁,加入50g的抗坏血酸,超声搅拌3分钟,使乌鸡活性肽与氯化亚铁混合均匀,然后加热到35℃,静置反应1h;然后用陶瓷滤膜过滤,得到乌鸡肽铁螯合物浓缩液,然后用乙醇洗涤浓缩液,再过滤得到高纯乌鸡肽铁螯合物,之后采用喷雾干燥器干燥高纯乌鸡肽铁螯合物得到乌鸡肽铁螯合物粉,在喷雾干燥时,干燥室内用250nm紫外光照射,最后真空包装。
实施例2
把100kg乌骨鸡宰杀后去毛,去内脏,去鸡头鸡爪,然后用绞肉机打成肉浆;加入风味蛋白酶水解肉浆,然后过滤得到乌鸡活性肽溶液,测定乌鸡活性肽含量,乌鸡活性肽含量为7kg,把乌鸡活性肽溶液转入真空反应釜中,调节PH为3,加入700g氯化亚铁,加入350g的抗坏血酸,超声搅拌5分钟,使乌鸡活性肽与氯化亚铁混合均匀,然后加热到45℃,静置反应2h;然后用陶瓷滤膜过滤,得到乌鸡肽铁螯合物浓缩液,然后用乙醇洗涤浓缩液,再过滤得到高纯乌鸡肽铁螯合物,之后采用喷雾干燥器干燥高纯乌鸡肽铁螯合物得到乌鸡肽铁螯合物粉,在喷雾干燥时,干燥室内用300nm紫外光照射,最后真空包装。
实施例3
把100kg乌骨鸡宰杀后去毛,去内脏,去鸡头鸡爪,然后用绞肉机打成肉浆;加入风味蛋白酶水解肉浆,然后过滤得到乌鸡活性肽溶液,测定乌鸡活性肽含量,乌鸡活性肽含量为6kg,把乌鸡活性肽溶液转入真空反应釜中,调节PH为6,加入600g氯化亚铁,加入300g的抗坏血酸,超声搅拌4分钟,使乌鸡活性肽与氯化亚铁混合均匀,然后加热到35℃,静置反应1-2h;然后用陶瓷滤膜过滤,得到乌鸡肽铁螯合物浓缩液,然后用乙醇洗涤浓缩液,再过滤得到高纯乌鸡肽铁螯合物,之后采用喷雾干燥器干燥高纯乌鸡肽铁螯合物得到乌鸡肽铁螯合物粉,在喷雾干燥时,干燥室内用361nm紫外光照射,最后真空包装。

1.一种乌鸡肽铁螯合物粉制备方法,其特征是:包括如下步骤:1)把乌骨鸡宰杀后去毛,去内脏,去鸡头鸡爪,然后用绞肉机打成肉浆;2)加入风味蛋白酶水解肉浆,然后过滤得到乌鸡活性肽溶液;3)把乌鸡活性肽溶液转入真空反应釜中,调节PH为3-6,加入乌鸡肽质量的10%的氯化亚铁,加入0.5%的抗坏血酸,超声搅拌3-5分钟,使乌鸡活性肽与氯化亚铁混合均匀,然后加热到35-45℃,静置反应1-2h;然后用陶瓷滤膜过滤,得到乌鸡肽铁螯合物浓缩液,然后用乙醇洗涤浓缩液,再过滤得到高纯乌鸡肽铁螯合物,之后采用喷雾干燥器干燥高纯乌鸡肽铁螯合物得到乌鸡肽铁螯合物粉,并在喷雾干燥时采用紫外光杀菌,最后真空包装。
