本发明涉及一种由引入羧基的甘露聚糖涂层的磁共振成像造影剂,具体地说,是涉及一种为了高的体内稳定性且能够特异性的引入至抗原递呈细胞内而由引入羧基的甘露聚糖涂层的超顺磁性磁共振成像造影剂及其制备方法。本发明中提到的由引入羧基的甘露聚糖涂层的磁共振成像造影剂的表面带有大量的负电荷,从而能够提供较高的体内稳定性及生物相容性。同时,由于甘露聚糖中的甘露糖能够有效地引入至抗原递呈细胞内,从而能够使抗原递呈细胞自身和含有抗原递呈细胞的组织特异性地通过磁共振成像实现可视化。

技术领域
本发明涉及一种磁共振成像(magnetic resonance imaging;MRI)造影剂(contrast agent),特别地,涉及一种特异性的引入到抗原递呈细胞(antigenpresenting cell)中使抗原递呈细胞自身和含有抗原递呈细胞的组织特异性的通过磁共振成像实现可视化(visualization)的MRI造影剂,具体地说,就是涉及一种由引入羧基(carboxyl group)的甘露聚糖(mannan)涂层的(coating)超顺磁性(superparamagnetic)MRI造影剂。
背景技术
具有代表性的层析成像技术即磁共振成像,作为一种可以通过非创伤方式获得3维影像的方法,由于其对比度(contrast)和空间分辨率(spatialresolution)都很高,因此广泛应用于疾病的诊断。
MRI造影剂是一种注入(injection)到活体内后感知由强大的外部磁场和高频能量发生的T1,T2弛豫时间(relaxation time)而导致的差异并提高正常组织与非正常组织的对比度,因此MRI造影剂是能够针对局部解剖学或者功能性区域实现影像化处理的化学物质。一般情况下,MRI造影剂分为顺磁性(paramagnetic)造影剂和超顺磁性造影剂(Eur.Radiol.11:2319,2001)。
顺磁性造影剂的活体内毒性较强,且只能够生成针对mM标准的影像,因此人们更加关注能够生成μM标准的高敏感度影像的超顺磁性造影剂(Nano Lett.6:2427,2006;Nat.Med.13:95,2007)。
临床上广泛使用的超顺磁性造影剂都是在以如磁铁矿(magnetite,Fe3O4)或者磁赤铁矿(maghemite,Fe2O3)等超顺磁性氧化铁(superparamagnetic ironoxide,SPIO)为代表的超顺磁性纳米颗粒(nanoparticle)的基础上制备出来的。它们制备成铁磁流体(ferrofluid)注入到体内,该铁磁流体具有数十纳米以下的大小的均匀颗粒的稳定的胶体状。
纯超顺磁性氧化铁颗粒具有以下特点:①由于其具有疏水性,且体积与表面积之比较大,因此,颗粒间的疏水相互作用(hydrophobic interaction)较强,由此产生凝聚而形成簇(cluster);②其稳定性较低,原有的构造容易发生变化,因此其自身的磁性特性也很容易改变;③如果暴露在活体环境中,能够迅速生物降解;④纯氧化铁本身带有毒性,因此其对人体有害。所以,为了解决上述问题,提高含有上述超顺磁性纳米颗粒的氧化铁溶液的稳定性,就需要在颗粒表面进行修饰。
一般情况下,在表面修饰包括涂敷多种高分子,常用右旋糖酐(dextran)或者右旋糖酐的衍生物涂敷表面。现有的用于表面涂层的高分子除了右旋糖酐系之外,也可使用聚丙烯酸(Polyacrylic acid,PAA)、聚乙烯吡咯烷酮(Polyvinylpyrrolidone,PVP)、聚乙烯醇(Polyvinylalcohol,PVA)、聚乙二醇(Polyethylene glycol,PEG)等合成高分子(J.Mater.Chem.12:3654-3659,2002;J.Colloid Interface Sci.278:353-360,2004)或者明胶(Gelatin)、壳聚糖(Chitosan)、支链淀粉(Pullulan)等天然高分子(J.Colloid Interface Sci.283:446-451,2005;J.Magn.Magn.Mater.293:20-27,2005)。
另外,针对甘露聚糖而言,其虽然不是常用的,但是已公开也在表面修饰(涂层)上使用甘露聚糖(美国注册专利5,262,176及美国注册专利5,462,053)。本发明人也使用过甘露聚糖(韩国注册专利10-0949465)。据美国注册专利5,262,176中记载,使用甘露聚糖的目的与通常的高分子涂层的目的相同,即为了提高氧化铁胶体(colloid)在体内的稳定性。据美国注册专利5,462,053中记载,其目的也与通常的高分子涂层的目的相同,即为了提高造影剂溶液(suspension)的稳定性。据韩国注册专利10-0949465中记载,
高分子涂层不仅能够获得预期的一般效果,而且还具有通过甘露糖(mannose)特异性受体(receptor)的靶向目的。但是,上述甘露聚糖的使用还反映出由甘露聚糖导致的活体毒性的危险性,同时它在活体内循环系统(circulationsystem)中会逐渐衰减(trap)。因此,尤为重要的是研发出一种更加适合活体,即毒性显著降低且在活体内的半衰期延长的甘露聚糖及涂层上述甘露聚糖的MRI造影剂。
从另一个层面来说,如果能够利用MRI特异性地造影增强抗原递呈细胞或者含有大量抗原递呈细胞的组织,则可以提高其在临床上的应用率。通常情况下,在癌组织或者败血症(sepsis)等感染部位大量存在有抗原递呈细胞。在特定部位中大量存在有抗原递呈细胞就表明相应部位出现了异常症状。因此,考虑到临床应用,研发出一种能够检测上述抗原递呈细胞异常增加的诊断技术也非常重要。
在本发明中,整体上参照了多篇论文及专利文献,其引用部分都在括号内进行了标注。引用的论文及专利文献公开的全部内容作为参照引用到本说明书中,其目的在于对本发明所属技术领域及本发明的内容进行更加详细的说明。
发明内容
技术课题
因此,本发明的一个目的在于,解决上述现有技术中存在的问题及此前一直未解决的技术课题。
本发明的另一个目的在于,提供一种MRI造影剂及其制备方法,其体内稳定性及安全性都较高,能够特异增强针对抗原递呈细胞或者含有抗原递呈细胞的组织的造影。
本发明的另一个目的在于,提供一种能够用于诊断癌转移至淋巴结的MRI造影剂。
本发明的另一个目的在于,提供一种能够用于诊断败血症的MRI造影剂。
技术方案
为了解决上述课题,就需要克服现有的由甘露聚糖涂层的MRI造影剂的缺点。为此,本发明人预料,如果将羧基引入到甘露聚糖中并将其作为MRI造影剂的涂层材料,则不仅能够降低毒性而且还可以延长其在体内的半衰期。当研发出能够将羧基充分引入到甘露聚糖的技术之后,将由引入上述羧基的甘露聚糖涂层的MRI造影剂与由甘露聚糖涂层的MRI造影剂相比较,确认前者的使用更加安全,从而完成了本发明。
因此,本发明提供了一种由引入羧基的甘露聚糖涂层的作为超顺磁性纳米颗粒的MRI造影剂及其制备方法,上述羧基的体内稳定性及安全性都比较高,且具有能够特异增强针对抗原递呈细胞或者含有抗原递呈细胞的组织的造影的性能。
上述超顺磁性纳米颗粒的磁性物质虽然可以从氧化锰(MnO)、锰铁氧体(MnFe2O4)、铁-铂合金(Fe-Pt alloy)、钴-铂合金(Co-Pt alloy)及钴(Co)等物质中进行多种选择,但是,最优选使用磁铁矿(magnetite,Fe3O4)或者磁赤铁矿(maghemite,Fe2O3)等氧化铁(superparamagnetic iron oxide,SPIO)。
上述抗原递呈细胞虽然可以是树突细胞(dendritic cell)、巨噬细胞(macrophage)、B-细胞(B-cell)等多种类。但是,优选对象是带有甘露糖受体的细胞,特别优选的是巨噬细胞。
能够应用本发明的MRI造影剂的具有代表性的实例是癌症诊断。特别是优选用于诊断癌转移至淋巴结。另外,本发明的MRI造影剂也可以用于与炎症相关的诊断,特别是优选用于的败血症诊断。
另外,本发明还提供了一种增强针对由引入羧基的甘露聚糖涂层的抗原递呈细胞或者含有抗原递呈细胞的组织的造影增强的MRI造影剂的制备方法,包括以下几个步骤:
A)合成超顺磁性纳米颗粒的步骤;
B)向甘露聚糖中引入醛基合成醛-甘露聚糖的步骤;
C)利用上述醛-甘露聚糖使其氧化制备出引入羧基的羧酸-甘露聚糖的步骤;
D)利用上述制备的羧酸-甘露聚糖涂层上述合成的超顺磁性纳米颗粒的步骤。
如上所述,醛-甘露聚糖指的是引入醛基的甘露聚糖,羧酸-甘露聚糖指的是引入羧基的甘露聚糖。
虽然本发明人为了向甘露聚糖中引入羧基采用了多种方法,但是其他方法都很难将羧基充分地引入。通过本发明中记载的将醛-甘露聚糖作为中间体的方法能够充分引入羧基(整体甘露糖的10mol%得到羧化)。因此,本发明的特征在于,提供了由引入羧基的甘露聚糖涂层的MRI造影剂;以及对于引入上述羧基的甘露聚糖的制备而言,将醛-甘露聚糖用作中间体。
有益效果
由充分引入羧基(carboxyl group)的甘露聚糖(mannan)涂层(coating)的超顺磁性(superparamagnetic)MRI造影剂能够提供较高的体内稳定性(stability)及较低的活体毒性,并能够特异性的引入到抗原递呈细胞中,从而能够针对抗原递呈细胞及含有抗原递呈细胞的组织特异性的进行可视化处理。针对上述抗原递呈细胞及含有抗原递呈细胞的组织特异性的进行可视化处理,并具有较高的体内稳定性的MRI造影剂,可以用于有效检测乳腺癌、大肠癌、胃癌等的早期转移(metastasis)至淋巴结的情况,并且还可以用于败血症的早期诊断。
准确诊断癌是否转移至淋巴结对于癌症治疗和决定患者预后都非常重要,毫无疑问,它对败血症的早期诊断也是非常重要的。为了检测癌是否转移至淋巴结,目前广泛采用PET-CT(Positron Emission Tomography andComputed Tomography),但MRI与PET-CT相比,不存在放射性暴露的危险,并且其空间分辨率较高,因此很容易对早期的微转移做出准确的判定。
对于败血症,虽然目前主要通过ELISA(Enzyme-linked immunosorbentassay)等检查方法进行诊断,但是,诊断时间耗时较长。由于败血症的死亡率相当高,因此早期诊断非常关键。如果将本发明用于诊断败血症,就会使诊断变得更加快捷和简便。

具体实施方式
如上所述,本发明提供了一种由充分引入羧基的甘露聚糖涂层的体内稳定性较高且毒性显著降低的超顺磁性MRI造影剂。
下面,将对本发明的内容进行详细说明。但是,本领域技术人员完全可以在不偏离本发明技术思想及范围内对本发明进行多样的变化以及修改。
下面,将通过实施例对本发明进行具体说明。但是,这些实施例是为了对本发明进行详细说明而列举的示例,它并不限定本发明的范围。
实施例1:羧酸-甘露聚糖的制备
实施例1-1:醛-甘露聚糖的合成
将400mg的甘露聚糖溶于16ml的蒸馏水中,向其中缓慢添加0.1MNaIO4水溶液9ml(相对于甘露糖为0.4当量),然后在4℃避光条件下使其反应1小时。为了终结反应,向其中添加甘油(相对于甘露糖为0.8当量),并搅拌30分钟。将生成物转移到透析膜(MWCO1,000)上,在4℃的条件下对蒸馏水透析一天后进行冷冻干燥,获得393mg白色固体即醛-甘露聚糖。整体过程如图1所示。
实施例1-2:羧酸-甘露聚糖的合成
将370mg的醛-甘露聚糖溶于20ml的蒸馏水中,将溶液盛入装满冰块的水槽中,向其中加入过氧化氢(H2O2;相对于甘露糖为1.2当量)。然后,向其中缓慢加入3ml的0.5M NaClO2(相对于甘露糖为1.2当量),并将pH值调整到5。然后,去除水槽并继续搅拌,利用NaOH使反应物的pH值保持在5。当pH值不再变化时,将生成物的pH值调整到9后,利用透析膜(MWCO1,000)在4℃的条件下对蒸馏水透析一天,纯化后进行冷冻干燥,获得351mg白色固体即羧酸-甘露聚糖。以生成的羧酸-甘露聚糖为对象,利用1M HCl定量引入的羧基的量,结果确认甘露糖中引入了约13mol%的羧基,整个过程如图2所示。羧基的引入可以通过常用的方法即利用傅里叶变换红外光谱仪(Fourier transform infrared spectrometer;Nicolet Magna550series IIspectrometer,Midac,Atlanta,GA,USA)进行分析,其结果如图3所示。通过图3可以确定因引入羧基而出现了具有特征性的峰值(peak)。
实施例2:超顺磁性氧化铁纳米颗粒的制备
首先,为了制备超顺磁性氧化铁纳米颗粒(Superparamagnetic Iron OxideNanoparticles;SPIONs),利用碱性溶液(NH4OH)使Fe2+和Fe3+离子还原,并使SPIONs沉淀。具体地说,将氯化铁(III)(FeCl3·6H2O)2mol和氯化铁(II)(FeCl2·4H2O)1mol加入到70ml的蒸馏水中,制备混合水溶液。然后剧烈搅拌上述混合水溶液同时向其中加入7ml的氨水溶液。这时会形成黑色的沉淀物,将上述沉淀物利用永久磁铁固定,用蒸馏水清洗多次,直到pH值从10降到7为止,最后只获得pH值为7的沉淀物。
向上述沉淀物中加入2M的硝酸(HNO3)溶液20ml和0.35M的硝酸铁(Fe(NO3)3·9H2O)水溶液30ml并回流(reflux)1小时使其氧化。将氧化后形成的褐色悬浊液利用0.01M的硝酸透析2天,制备出SPIONs原液(stocksolution),并将其在4℃的条件下存放(参照图4)。
实施例3:由羧酸-甘露聚糖涂层的超顺磁性氧化铁纳米颗粒的制备
用蒸馏水将上述SPIONs原液稀释到6.25mg/ml,将上述稀释液在4℃条件下搅拌。另外,在此期间,将17.5mg的羧酸-甘露聚糖溶于200μl的蒸馏水中,制备出羧酸-甘露聚糖溶液。完成羧酸-甘露聚糖溶液的制备后,从此前制备的SPIONs原液的稀释液中取出800μl将羧酸-甘露聚糖溶液在4℃条件下搅拌的同时缓慢的逐滴地向上述稀释液中添加。添加完成后,将其在4℃条件下搅拌一夜。然后,加入少量NH4OH将pH值调整到7。最后,加入NaCl使NaCl比例调至0.9%,以调节离子强度。然后,就可以将上述溶液作为由羧酸-甘露聚糖涂层的超顺磁性氧化铁纳米颗粒(即,MRI造影剂)溶液使用。
作为参照物,还制备了由甘露聚糖涂层的SPIONs、由右旋糖酐涂层的SPIONs及由PVA(polyvinyl alcohol)涂层的SPIONs,其全部过程均与由羧酸-甘露聚糖涂层的SPIONs的制备过程相同。
实施例4:由羧酸-甘露聚糖涂层的SPIONs的特性分析
实施例4-1:测定由羧酸-甘露聚糖涂层的SPIONs的大小分布
为了测定依据本发明制备的由羧酸-甘露聚糖涂层的SPIONs的大小分布,利用将散射角调节到90度的光全散射测定仪(electrophoretic lightscattering spectrophotometer,ELS8000,Otsuka Electronics,Osaka,Japan)在25℃条件下测定。利用上述光全散射测定仪进行分析的样品是将由羧酸-甘露聚糖涂层的SPIONs放入蒸馏水后经超声波处理制备出来的。分析结果如图5所示,从图中可以看出生成了均匀分布的颗粒,并且颗粒的平均大小为34.1nm。
实施例4-2:测定Zeta电位(Zeta potential)
通过现有方法测定由羧酸-甘露聚糖涂层的SPIONs的Zeta电位(作为代表漂浮在液体中的胶体颗粒的表面电荷量程度的一个指标,指的是边界剪应力与本体溶液之间的电位差),测定结果为-31.69mV。根据上述Zeta电位数值能确定由羧酸-甘露聚糖涂层的SPIONs在现有的胶体标准下维持稳定的状态。
实施例5:与参照物进行比较
实施例5-1:与由甘露聚糖涂层的SPIONs进行毒性比较
将由羧酸-甘露聚糖涂层的SPIONs与由甘露聚糖涂层的SPIONs进行毒性比较。通过现有的单次静脉注射试验进行了比较。该试验在化学研究院附属安全性评估研究所进行的。具体地说,向7周龄ICR老鼠(每组雌雄各5只)以20、40及80mg Fe/kg的剂量单次静脉注射由羧酸-甘露聚糖涂层的SPIONs和由甘露聚糖涂层的SPIONs后,放置15天观察其一般症状,观察了死亡的动物,并测量了体重。第15天进行尸体解剖,观察其内部脏器有无异常。
对于注射由甘露聚糖涂层的SPIONs的情况,当其剂量达到20mg Fe/kg以上时,就会发现有部分动物死亡,还可以观察到动物出现活动性降低、不规则呼吸、腹部和胃及下腹部污染的一般症状。据此,可以计算出由甘露聚糖涂层的SPIONs导致的老鼠中的半数致死剂量在雌雄两种情况均为大约44mg Fe/kg。另外,对于注射由羧酸-甘露聚糖涂层的SPIONs,在所有的雌雄试验对象中均未观察到与注射试验物质相关的死亡,并且通过肉眼观察到的尸体解剖也没有发现什么特殊情况。因此,由羧酸-甘露聚糖涂层的SPIONs导致的老鼠半数致死剂量超过80mg Fe/kg。
通过上述结果可以确定,由羧酸-甘露聚糖涂层的SPIONs与由甘露聚糖涂层的SPIONs相比,前者是一种更加安全的MRI造影剂。
实施例5-2:测定引入至抗原递呈细胞内的情况
为了以抗原递呈细胞或者含有大量抗原递呈细胞的组织作为MRI造影的对象,所使用的MRI造影剂必须能够有效地引入至抗原递呈细胞内,并且优选的是,上述引入为特异性的。通过以下实验可以对此予以确认。
首先,检测由羧酸-甘露聚糖涂层的SPIONs引入至抗原递呈细胞内的效率。选定腹腔巨噬细胞(peritoneal macrophage)作为抗原递呈细胞的样本细胞,将从Balb/C老鼠中分离的腹腔巨噬细胞放入添加有由羧酸-甘露聚糖涂层的SPIONs的培养基中培养3小时。培养后,实施通常的普鲁士蓝染色(Prussian blue staining),观察由羧酸-甘露聚糖涂层的SPIONs引入至细胞内的情况。普鲁士蓝染色法是一种利用铁离子与亚铁氰化物反应生成铁氰化物(ferrocyan compound)原理的染色法,通过这种染色法可以确定铁离子的分布,其结果如图6所示。从图6中可以看出,由羧酸-甘露聚糖涂层的SPIONs有效地引入至细胞内。由此可知,由羧酸-甘露聚糖涂层的SPIONs能够以抗原递呈细胞或者含有大量抗原递呈细胞的组织为对象使用,其向细胞内的引入可称作内吞作用(endocytosis)。
做一个追加实验,将其与其它MRI造影剂的向抗原递呈细胞内的引入效率方面进行了比较。在这一实验中,排除前面实验已确认毒性的由甘露聚糖涂层的SPIONs,作为比较对象的其它MRI造影剂选择了由右旋糖酐(Dextran)涂层的SPION及由PVA涂层的SPION。在这一实验中,也选择了腹腔巨噬细胞作为抗原递呈细胞的样本细胞,将从Balb/C老鼠中分离的腹腔巨噬细胞放入分别添加有由羧酸-甘露聚糖涂层的SPIONs、由右旋糖酐涂层的SPION及由PVA涂层的SPION的培养基中培养3小时及24小时。培养后,将2×105的细胞固定在5%明胶(gelatin)上,然后将其放入微量离心管中并实施离心分离。进行离心分离后,通过MR幻影成像(phantomimaging)分析向细胞内引入的情况,其结果如图7所示。从图7中可以看出,SPIONs的引入越多颜色越黑,从而可以确定由羧酸-甘露聚糖涂层的SPIONs会更加迅速地引入到细胞内。从这一结果可以判定,由羧酸-甘露聚糖涂层的SPIONs与其它MRI造影剂相比,以抗原递呈细胞或者含有大量抗原递呈细胞的组织为对象,前者作为MRI造影剂更加优秀。
实施例5-3:测定向组织内的抗原递呈细胞内引入的情况
为了调查由羧酸-甘露聚糖涂层的SPIONs用作以含有抗原递呈细胞的组织为对象的MRI造影剂的可能性,将实际上向组织内的抗原递呈细胞内引入的效率与其它MRI造影剂进行了比较。选择巨噬细胞作为抗原递呈细胞的样本细胞,选择含有较多巨噬细胞的淋巴结组织作为样本组织。分别注射不同的MRI造影剂后通过普鲁士蓝染色法测定引入的情况。具体地,分别注射不同的MRI造影剂后,摘出淋巴结组织,然后将摘出的淋巴结浸入至1ml的4%福尔马林(formalin)溶液中。然后将其固定在石蜡(paraffin)内,为了染色而将其切开。将淋巴结切片用10%亚铁氰化钾及20%HCl染色20分钟,将染色的切片通过光学显微镜观察,可以观察到氧化铁颗粒呈现出蓝色的点,其结果如图8所示。从图8中可以看出,由羧酸-甘露聚糖涂层的SPIONs在淋巴结内的分布比其它MRI造影剂更好。由此可以确认,如果以由羧酸-甘露聚糖涂层的SPIONs作为造影剂引入时,能够有效地引入到组织内的抗原递呈细胞内,从而可以判定,由羧酸-甘露聚糖涂层的SPIONs可以用作以含有抗原递呈细胞的组织为对象的MRI造影剂。
实施例6:在癌是否转移至淋巴结的诊断中的用途
以诊断癌是否转移至淋巴结为目的,在MRI造影中对由羧酸-甘露聚糖涂层的SPIONs及现有的MRI造影剂即由右旋糖酐涂层的SPION及由PVA涂层的SPION的性能进行了比较。向癌转移至淋巴结的每只实验动物分别注射不同的造影剂使其获得1mg的铁离子,再对淋巴结实施MRI造影后进行比较。使用利用了韩国和顺全南大学医院的动物用线圈(4channel phasedarray rat head coil,Rapid Biomedical mdh,Rimpar,德国)的3T MR扫描仪(Siemens Medical System,Elegan,德国)实施了激发快速自旋回波(Turbo-spinecho;TSE)T2加权(repetition time ms/echo time ms of3,200/86,flip angle150°,echo train length of18,56mm field of view,2mm section thickness,0.2-mmintersection gap,320×320matrix,8NEX)MRI。其结果如图9及图10所示。
从图9中可以看出,与参照物的磁共振成像相比,注射了由羧酸-甘露聚糖涂层的SPIONs的磁共振成像随着时间的流逝颜色会变得更加暗淡。由此可以确认,由羧酸-甘露聚糖涂层的SPIONs在注射后相当短的时间内可提供组织间的对比度差异,因此它可以作为有效地诊断癌转移至淋巴结的MRI造影剂。图10中的信号强度(signal intensity)比表示针对相邻肌肉的淋巴结的相对信号强度比。与肌肉相比,由羧酸-甘露聚糖涂层的SPIONs能够更加有效地引入至淋巴结内。因此,可以确定其相对信号强度比更加优秀。
通过上述结果可以确定,依据本发明的由羧酸-甘露聚糖涂层的SPIONs不仅能够用作对含有抗原递呈细胞的组织进行可视化处理的MRI造影剂,还可以用作诊断癌转移至淋巴结与否的MRI造影剂。即,本发明的MRI造影剂可以有效地检测乳腺癌、大肠癌、胃癌等在早期转移至淋巴结。
实施例7:在败血症诊断中的用途
本发明的MRI造影剂可以用于败血症的早期诊断。败血症是一种因细菌感染而诱发的疾病。当被细菌感染时,感染部位就会对于细菌的脂多糖(lipopolysaccharide;LPS)引起反应产生大量的巨噬细胞。如果能容易地确认这样产生的巨噬细胞,就可以很方便地诊断败血症的病情,下面通过实验进行介绍。在下面的实验中,主要确认有多少由羧酸-甘露聚糖涂层的SPOINs有效地引入至因暴露在LPS中而被活化的巨噬细胞内。
将一部分腹腔巨噬细胞利用LPS进行处理,制备出具有如败血症状态的细胞。将上述经LPS处理的腹腔巨噬细胞和未经任何处理的腹腔巨噬细胞分别放入含有由羧酸-甘露聚糖涂层的SPOINs的培养基中培养3小时。培养后,分别针对每种细胞实施通常的普鲁士蓝染色,观察由羧酸-甘露聚糖涂层的SPOINs引入至细胞内的情况,其结果如图11所示。从图11中可以看出,具有如败血症状态的细胞即经LPS处理的细胞与未经任何处理的正常细胞相比,在经LPS处理的细胞中由羧酸-甘露聚糖涂层的SPOINs的引入显著增加。由此可以确定,本发明的MRI造影剂可以用于败血症的早期诊断。
在以上的说明中,对本发明的特定部分进行了详细阐述,对于具有本领域相关知识的技术人员来说,上述详细说明只不过是本发明的优选实施例,它并不限定本发明的范围。因此,本发明的保护范围并不局限于说明书的内容,必须要根据权利要求及等同于权利要求的范围来确定其保护范围。
Claims (15)
1.一种由羧酸-甘露聚糖涂层的超顺磁性纳米颗粒MRI 造影剂,其特征在于:
抗原递呈细胞自身及含有抗原递呈细胞的组织特异性的通过磁共振成像实现可视化。
2.如权利要求1所述的MRI 造影剂, 其特征在于:
上述抗原递呈细胞为树突细胞、巨噬细胞、B-细胞。
3.如权利要求1所述的MRI 造影剂, 其特征在于:
其用于癌转移至淋巴结的诊断。
4.如权利要求1所述的MRI 造影剂, 其特征在于:
其用于败血症的诊断。
5.如权利要求1所述的MRI 造影剂, 其特征在于:
上述羧酸-甘露聚糖是从醛-甘露聚糖制备。
6.如权利要求1所述的MRI 造影剂, 其特征在于:
构成上述羧酸-甘露聚糖的整体甘露糖的10 mol%以上被羧化了。
7.如权利要求1所述的MRI 造影剂, 其特征在于:
上述超顺磁性纳米颗粒为氧化铁。
8.如权利要求7所述的MRI造影剂,其特征在于:
上述氧化铁为磁铁矿(magnetite,Fe3O4)或者磁赤铁矿(maghemite, Fe2O3)。
9.一种MRI造影剂的制备方法,包括:
A) 合成超顺磁性纳米颗粒的步骤;
B) 向甘露聚糖中引入醛基以合成醛-甘露聚糖的步骤;
C)使醛-甘露聚糖氧化制备出引入羧基的羧酸-甘露聚糖的步骤;
D)由上述制备的羧酸-甘露聚糖涂层上述合成的超顺磁性纳米颗粒的步骤。
10.一种如权利要求9制备的由羧酸-甘露聚糖涂层的超顺磁性纳米颗粒MRI造影剂。
11.如权利要求10所述的由羧酸-甘露聚糖涂层的超顺磁性纳米颗粒MRI造影剂,其特征在于:
上述造影剂用于癌转移至淋巴结的诊断。
12.如权利要求10所述的由羧酸-甘露聚糖涂层的超顺磁性纳米颗粒MRI造影剂,其特征在于:
上述造影剂用于败血症的诊断。
13.如权利要求10所述的由羧酸-甘露聚糖涂层的超顺磁性纳米颗粒MRI造影剂,其特征在于:
构成上述羧酸-甘露聚糖的整体甘露糖的10 mol%以上被羧化了。
14.如权利要求10所述的由羧酸-甘露聚糖涂层的超顺磁性纳米颗粒MRI造影剂,其特征在于:
上述超顺磁性纳米颗粒为氧化铁。
15.如权利要求14所述的由羧酸-甘露聚糖涂层的超顺磁性纳米颗粒MRI造影剂,其特征在于:
上述氧化铁为磁铁矿(magnetite,Fe3O4)或者磁赤铁矿(maghemite, Fe2O3)。
