Abstract
Description
A)超常磁性ナノ粒子を合成する工程と、
B)マンナンにアルデヒド基を導入してアルデヒド−マンナンを合成する工程と、
C)前記アルデヒド−マンナンを用いてそれを酸化させることによってカルボキシル基が導入されたカルボン酸−マンナンを製造する工程、及び
D)前記製造されたカルボン酸−マンナンで前記合成された超常磁性ナノ粒子をコーティングする工程とを含むカルボキシル基が導されたマンナンでコーティングされた抗原表示細胞または抗原表示細胞を含んだ組織の照映増強MRI造影剤の製造方法に関するものである。
実施例1−1:アルデヒド−マンナンの合成
400mgのマンナンを16mlの蒸留水に溶解した。そこに、0.1M NaIO4水溶液9ml(マンノ−スに対して0.4当量)をゆっくり添加して4℃の条件で光を遮断したまま一時間反応させた。反応を終結させるためにグリセロール(マンノ−スに対して0.8当量)を添加して、30分間撹拌した。生成物を透析膜(MWCO1,000)に移して蒸留水に対して4℃の条件で一日透析した後、凍結乾燥して393mgの白色固体であるアルデヒド−マンナンを得た。全体過程が図1に提示されている。
370mgのアルデヒド−マンナンを20mlの蒸留水に溶解した。この溶液を氷が満たされた水槽に入れてそこに過酸化水素(H2O2;マンノ−スに対して1.2当量)を添加した。そこに3mlの0.5M NaClO2(マンノ−スに対して1.2当量)をゆっくり添加して、pHを5に合わせた。以後、氷水槽を除去して継続して撹拌しながら、NaOHを用いて反応物のpHを5に維持させた。pHがこれ以上変わらない時、生成物のpHを9に合わせた後、透析膜(MWCO 1、000)を用いて蒸留水に対して4℃の条件で一日透析して精製した後、凍結乾燥させた。その結果、351mgの白色固体であるカルボン酸−マンナンを得た。生成されたカルボン酸−マンナンを対象に、1M HClを用いて導入したカルボキシル基の量を定量した結果、マンノ−スの約13モル%のカルボキシル基が導入されたことを確認することができた。全体過程が図2に提示されている。カルボキシル基の導入は、通常の方法であるフーリエ変換赤外線分光器(Fourier transform infrared spectrometer;Nicolet Magna 550 series II spectrometer,Midac,Atlanta,GA,米国)を用いて分析し、その結果を図3に示す。図3の結果から、カルボキシル基の導入による特徴的ピークが示されることを確認することができる。
まず、超常磁性酸化鉄ナノ粒子(Superparamagnetic Iron Oxide Nanoparticles;SPIONs)を製造するために、塩基性溶液(NH4OH)でFe2 +とFe3 +イオンを還元させてSPIONsを沈澱させた。具体的に、塩化鉄(III)(FeCl3・6H2O)2 モルと塩化鉄(II)(FeCl2・4H2O)1モルを蒸留水70mlに添加して混合水溶液を調製した。前記混合水溶液を激しく撹拌しながらアンモニア水溶液7mlを添加した。ここで、黒い沈殿物が形成されるが、前記沈殿物を永久磁石を用いて固定させながらpHが10から7に落ちるまで蒸留水で何回洗浄した。このようにして、pH7になった沈殿物のみを集めた。
前記SPIONs保存溶液を6.25mg/mlになるように蒸留水で希釈した。このように準備した希釈液を、4℃で撹拌する。一方、その間に17.5mgのカルボン酸−マンナンを200μlの蒸留水に溶解してカルボン酸−マンナン溶液を準備する。カルボン酸−マンナン溶液の準備が完了すると、前に準備したSPIONs保存溶液の希釈液から800μlを取ってそこにカルボン酸−マンナン溶液を4℃で撹拌しながらゆっくり一滴ずつ添加する。添加が完了すれば、それを4℃で一晩中撹拌する。最終的にNH4OHを少量入れてpHを7に合わせる。最後に、NaClを0.9%になるように添加してイオン強度を合わせる。このように調製された溶液をカルボン酸−マンナンがコーティングされた超常磁性酸化鉄ナノ粒子(すなわち、MRI造影剤)溶液に使用した。
実施例4−1:カルボン酸−マンナンがコーティングされたSPIONsの大きさ分布図測定
本発明によって製造されたカルボン酸−マンナンがコーティングされたSPIONsの大きさと分布を測定するため、散乱角が90度に調節されたトータル光散乱測定機(electrophoretic light scattering spectrophotometer,ELS8000,Otsuka Electronics,大阪、日本)を用いて25℃で測定した。前記トータル光散乱測定機分析のための試料は、カルボン酸−マンナンがコーティングされたSPIONsを蒸留水に入れた後、超音波処理をして準備した。図5の分析結果から分かるように均一に分布した粒子が生成されたことが分かり、粒子の大きさの平均は34.1nmだった。
カルボン酸−マンナンがコーティングされたSPIONsのゼータ電位(液体の中に浮遊するコロイド粒子の表面帯電量の程度を示す指標であり、shear boundaryとbulk solutionとの電位差)を通常の方法によって測定し、その結果、−31.69mVと測定された。このゼータ電位数値で、カルボン酸−マンナンがコーティングされたSPIONsが通常的コロイド基準で安定した状態を維持することを確認することができた。
実施例5−1:マンナンがコーティングされたSPIONsとの毒性学的比較
カルボン酸−マンナンでコーティングされたSPIONsとマンナンでコーティングされたSPIONsの毒性学的比較を実施した。通常の単回静脈投与試験を通じて比較した。試験は、化学研究院付設安全性評価研究所で実施した。具体的に、7週齢のICRマウス(群当り雄と雌各5匹使用)にカルボン酸−マンナンでコーティングされたSPIONsとマンナンでコーティングされたSPIONsを20、40及び80mg Fe/kgの用量で単回静脈投与した後15日間一般症状を観察、死亡動物の観察及び体重測定を実施して、15日目に剖検を実施して内部臓器の異常有無などを観察した。
抗原表示細胞または抗原表示細胞が多量に含まれた組織をMRI照映の対象にするためには、使用したMRI造影剤が抗原表示細胞に効果的に流入しなければならないしまたそれが特異的であるなら好ましい。これを確認するために、次の実験を実施した。
カルボン酸−マンナンでコーティングされたSPIONsの、抗原表示細胞を含んだ組織をターゲットにしたMRI造影剤としての活用可能性を調査するために、実際組織内の抗原表示細胞内への流入効率を、他のMRI造影剤と比較調査した。抗原表示細胞のモデル細胞では、大食細胞を選定し、モデル組織に大食細胞が多いリンパ節組織を選定した。各MRI造影剤を注入後、プルシアンブルー染色で流入程度を測定した。具体的に、各MRI造影剤を注入後、リンパ節組織を摘出して摘出したリンパ節を4%のホルマリン溶液1mlに浸しておいた後、それをパラフィン中に固定させて染色のために切開した。リンパ節のスライスを10%のフェロシアン化カリウム及び20%のHClで20分間染色し、染色したスライスは光学顕微鏡で観察した。酸化鉄粒子は、青い点で示される。その結果を、図8に示す。図8の結果から分かるように、カルボン酸−マンナンでコーティングされたSPIONsは、他のMRI造影剤に比べてリンパ節内によく分布していた。このことから、カルボン酸−マンナンでコーティングされたSPIONsを造影剤として注入する場合、組織内の抗原表示細胞にも効果的に流入し得ることを確認することができ、このことから抗原表示細胞が含まれた組織を対象にしたMRI造影剤として活用可能であることを確認することができた。
カルボン酸−マンナンでコーティングされたSPIONsを、既存に知られたMRI造影剤であるデキストラン-coated SPION及びPVA−coated SPIONとリンパ節への癌転移診断を目的としたMRI撮影において、その性能を比較してみた。リンパ節癌転移を有した実験動物当り1mgの鉄イオンになるように各造影剤を投与した後、リンパ節をMRIして比較した。和順全南大病院の動物用コイル(4 channel phased array rat head coil,Rapid Biomedical mdh,Rimpar,ドイツ)を用いた3T MRスキャナ(Siemens Medical System,Elegan,ドイツ)を使用してターボスピンエコー(Turbo-spin echo;TSE)T2-weighted(repetition time ms/echo time ms of 3,200/86, flip angle 150゜,echo train length of 18,56mm field of view,2mm section thickness,0.2-mm intersection gap,320×320 matrix,8 NEX)MRIを実施した。その結果は図9及び図10に提示されている。
本発明のMRI造影剤が、敗血症を早期診断する目的にも活用することができるかどうか調べた。敗血症は、バクテリアの感染によって誘発される疾患で、バクテリアに感染する場合感染部位はバクテリアのリポポリサッカライド(LPS)に反応して大食細胞を多量生成する。このように生成された大食細胞を容易に確認することができれば、敗血症の程度を簡便に確認することができる。実験は、次のように実施した。下記の実験では、カルボン酸−マンナンでコーティングされたSPOINsがLPSに露出して活性化した大食細胞内でどのくらい効率的に流入されるかを調べることにした。
Claims (15)
- 抗原表示細胞自体及び抗原表示細胞を含む組織を磁気共鳴映像で特異的に可視化させることができるカルボン酸−マンナンでコーティングされた超常磁性ナノ粒子MRI造影剤。
- 前記抗原表示細胞が、樹状細胞、大食細胞、B−細胞であることを特徴とする請求項1記載のMRI造影剤。
- リンパ節への癌転移診断に活用できることを特徴とする請求項1記載のMRI造影剤。
- 敗血症診断に活用できることを特徴とする請求項1記載のMRI造影剤。
- 前記カルボン酸−マンナンが、アルデヒド−マンナンから製造されることを特徴とする請求項1記載のMRI造影剤。
- 前記カルボン酸−マンナンが、全体を構成するマンノ−スの10モル%以上がカルボキシル化されていることを特徴とする請求項1記載のMRI造影剤。
- 前記超常磁性ナノ粒子が、酸化鉄であることを特徴とする請求項1記載のMRI造影剤。
- 前記酸化鉄が、マグネタイト(magnetite,Fe3O4)またはマグヘマイト(maghemite,Fe2O3)であることを特徴とする請求項7記載のMRI造影剤。
- A)超常磁性ナノ粒子を合成する工程と、
B)マンナンにアルデヒド基を導入してアルデヒド−マンナンを合成する工程と、
C)前記アルデヒド−マンナンを用いてそれを酸化させることでカルボキシル基が導入されたカルボン酸−マンナンを製造する工程、及び
D)前記製造されたカルボン酸−マンナンで前記合成された超常磁性ナノ粒子をコーティングする工程とを含む、MRI造影剤製造方法。 - 請求項9によって製造される、カルボン酸−マンナンでコーティングされた超常磁性ナノ粒子MRI造影剤。
- 造影剤が、リンパ節への癌転移診断に活用され得ることを特徴とする請求項10記載のカルボン酸−マンナンでコーティングされた超常磁性ナノ粒子MRI造影剤。
- 造影剤が、敗血症診断に活用され得ることを特徴とする請求項10記載のカルボン酸−マンナンでコーティングされた超常磁性ナノ粒子MRI造影剤。
- 前記カルボン酸−マンナンが、全体を構成するマンノ−スの10モル%以上がカルボキシル化されていることを特徴とする請求項10記載のカルボン酸−マンナンでコーティングされた超常磁性ナノ粒子MRI造影剤。
- 前記超常磁性ナノ粒子が、酸化鉄であることを特徴とする請求項10記載のカルボン酸−マンナンでコーティングされた超常磁性ナノ粒子MRI造影剤。
- 前記酸化鉄が、マグネタイト(magnetite,Fe3O4)またはマグヘマイト(maghemite,Fe2O3)であることを特徴とする請求項14記載のカルボン酸−マンナンにコーティングされた超常磁性ナノ粒子MRI造影剤。
