Description
本発明の第1の態様によれば、磁気信号を発生するコア;ならびに前記磁気信号を発生するコアの表面上に少なくとも部分的に形成された被覆部分を含む、マルチモーダルPET(陽電子放射断層撮影)/MRI(核磁気共鳴画像化)造影剤であって、前記被覆部分には、機能化表面を有する内部層、および前記機能化表面上に形成された放射性核種で無電解めっきされた層の形態の外部層を含む複数の層が含まれる、前記造影剤が提供される。
磁気信号を発生するコアの表面を、機能化溶液と接触させて、前記磁気信号を発生するコア上に少なくとも部分的に内部層を形成させ、機能化表面を規定し;
前記機能化表面を、少なくとも二価スズイオンの供給源を含有する増感化(sensitizing)溶液と接触させて、スズ増感化表面を形成させ;
前記スズ増感化表面を、少なくとも貴金属イオンの供給源を含有する活性化溶液と接触させて、貴金属活性化表面を形成させ;次いで
前記貴金属活性化表面を、少なくとも放射性核種イオンの供給源ならびに前記放射性核種イオンを処理するための還元剤および/または加水分解剤を含有する無電解めっき溶液と接触させて、前記貴金属活性化表面上に前記放射性核種の無電解めっき層を形成させること
を含む方法が提供される。
前記TEOS前駆体を重合させて、前記内部層を、前記磁気信号を発生するコアを少なくとも部分的に封入するシリカ(SiO2)シェルとして形成させること
がさらに含まれる。
第4の態様による医薬製剤を患者に投与し;
前記製剤の投与中または投与後の前記目的の領域におけるPETおよび/またはMRIシグナルを比較することによって前記目的の領域における前記製剤中に存在するマルチモーダルPET/MRI造影剤の出現を検出し;
前記目的の領域のPETおよびMRI画像データを収集し;次いで
前記目的の領域のマルチモーダルPET/MRI画像を、PETおよびMRI画像データを用いて構築すること

本発明の範囲内でありうるいずれか他の方法にもかかわらず、本発明の好ましい実施態様は、添付の図面に関して例示としてのみ記載される。
下記の説明は、具体的な実施態様を記載することのみを目的とするものであって、上記記載について限定することを意図するものではないことが理解されるべきである。
本発明の好ましい実施態様によるマルチモーダルPET(陽電子放射断層撮影)/MRI(核磁気共鳴画像化)造影剤の合成方法が記載されている。
当業者によって評価されるように、MRI画像化目的のために用いられる造影剤のコアは、強磁性、常磁性または超常磁性信号を発生するコアのいずれかでありうる。強磁性物質は、一般に、鉄(Fe)、コバルト(Co)、またはニッケル(Ni)を含有する。このような物質は、外部磁場に置かれると大きな陽性磁化率を示し、外部磁場が除かれても磁性を維持し続ける能力を有し、これは、常磁性、超常磁性、および反磁性体と比較する際の識別因子である。常磁性体には、酸素、および、例えば、鉄(Fe)およびガドリニウム(Gd)のような様々な金属イオンが含まれる。これらのイオンは、陽性磁化率を生じる不対電子を有する。この磁化率の大きさは、強磁性体と比較して著しく低い。一方で、超常磁性体は、バルクで強磁性特性を示す元素の各ドメインからなる。それらの磁化率は、強磁性体と常磁性体との間である。
上記に示されるように、前記磁気信号を発生するコアを被覆するために用いられる被覆部分には、磁気信号を発生するコアの周囲に無電解めっきした放射性核種金属の第2の層の形成を促進することを活性化することができる複数の適切な機能化表面基を提示する内部層が含まれる。
一般的に、無電解めっきの方法は、電流を流すことなくめっきされる基質の表面上に金属を沈着させるために酸化還元反応を使用することが必要とされる。この反応は、典型的に、反応の還元プロセスを促進するのに適する触媒の使用に関連する。最も一般的に用いられる触媒系は、酸性(HCl)溶液中で塩化パラジウム(PdCl2)/塩化スズ(II)(SnCl2)触媒組成物を使用することである。具体的には、基質の表面は、まず、塩化スズ(II)溶液中に入れ、続いて塩化パラジウム溶液中で活性化させるによって増感化させる。次いで、活性化された表面は、めっきされる金属の溶液に曝露される。
本発明の目的のために、PET画像化を実現するために用いられる放射性核種金属は、ある程度のPET画像が生成可能である程度に十分に長い半減期を有する陽電子放射体でなければならないことが理解される。そのようなものとして、適する陽電子放射体は、銅(60Cu、61Cu、64Cu)、スカンジウム(44Sc)、チタン(45Ti)、鉄(52Fe)、マンガン(51Mn、52Mn)、コバルト(55Co)、ガリウム(66Ga、68Ga)、ヒ素(72As)、ルビジウム(82mRb)、ストロンチウム(83Sr)、テクネチウム(94mTc)、イットリウム(86Y)、ジルコニウム(89Zr)、およびインジウム(110In)からなる群から選択されてもよい。
本発明の1の特に好ましい実施態様を示す目的のために、マルチモーダルPET/MRI造影剤の合成方法は、無電解めっきされた放射性核種金属外部層の形成を容易にする使用のために、磁気信号を発生するコアとして酸化鉄ナノ粒子を、前記シェルの表面上に複数のシラノール基を提示するシリカシェル内部層を含む被覆部分とともに用いるコアシェルナノ粒子の配置について記載される。
前記方法の第1の工程(A)によれば、超常磁性酸化鉄コアを有するナノ粒子は、2段階方法により合成した。第1段階において、酸化鉄ナノ粒子は、出典明示により本明細書に取り込まれる文献方法(Parkら,2004)[2]による熱分解によって生成した。第2段階において、次いで、前記熱分解方法により得られた酸化鉄ナノ粒子は、本発明者らによって開発され、出典明示により本明細書に取り込まれる国際PCT特許出願番号PCT/AU2017/050981(Gammilonghiら)[4]の内容を構成する洗浄プロトコールを用いて精製した。これについて、前記磁気ナノ粒子は、ジエチルエーテルおよびメタノールを1:1(vol/vol)の割合で含む第1の溶媒組成物中で洗浄し、次いで、ヘキサンおよびエタノールを1:1(vol/vol)の割合で含む第2の溶媒組成物中で洗浄することによって精製し、最終的にシクロヘキサンから構成される第3の溶媒組成物中に分散させた。
前記方法の第2の工程(B)によれば、シリカ(SiO2)シェルは、精製した酸化鉄ナノ粒子を水などの極性溶媒中で分散可能にするために、前記ナノ粒子の周囲で成長させる。前記精製した酸化鉄ナノ粒子の表面上のSiO2シェルは、出典明示により本明細書に取り込まれる文献の方法(Hanら,2008)[5]であるが、記載されるプロトコールにより改変されている方法によるシクロヘキサン中水の逆ミクロエマルションを形成させることにより形成させた。具体的には、まず、精製した酸化鉄ナノ粒子をシクロヘキサン溶液(適する界面活性剤を含む)中で十分に分散させて、親水性に対する状態を改善させた。次いで、十分に分散させたナノ粒子の溶液をオルトケイ酸テトラエチル(TEOS)と1:1(vol/vol)の割合で数分間混合し、続いてアンモニア溶液(水中で28wt%)を加えて、逆ミクロエマルションを形成させた。室温で攪拌しながら6時間反応させ、約15nm(±1nm)の全体の粒径を有するシリカ被覆酸化鉄ナノ粒子を得た。反応が完了したら、メタノールを、逆ミクロエマルションを止めるために加え、シリカ被覆酸化鉄ナノ粒子を、室温で磁気分離または遠心分離を用いてメタノール層から抽出した。
水分散性シリカ被覆ナノ粒子コア(放射性核種を有する)を被覆するためには、出典明示により本明細書に取り込まれる出願人自身の実験(Andersonら,2016)[6]で用いた3工程「増感化-播種(seeding)-沈着(deposition)」プロセスが必要とされる。
前記方法の第4の工程(D)によれば、前記スズ増感化粒子は、続いて硝酸パラジウム溶液(Pd(NO3)2・2H2O)に5分間曝露され、スズ増感化シリカ表面上に金属Pd0核種の沈着を生じ、同時に以前に結合したSn2+イオンを置換する。この撒種反応の間、Pd0核種の自発形成または撒種は、Pd2+からPd0への還元中にSn4+イオンの形成により進行する。Pd2+/0(標準水素電極-SHEに対して+0.99V)およびSn4+/2+(SHEに対して+0.15V)の標準還元電位における有利な差異は、前記反応を自発的に進行することを可能にする(Andersonら,2016)[6]。Sn4+およびPd2+イオンの間の自発的な反応により酸化鉄ナノ粒子のシリカ表面上に形成されたPd0層は、触媒として作用し、すなわち、前記シリカ表面は、後の放射標識において活性化される。
銅めっき方法を、費用と放射性(ホット)廃棄物の発生を最小にしつつ最適化するために、モデル系として、銅めっき槽を、非放射性(コールド)銅を用いて開発した。この非放射性系と放射性64Cuとの間のバリエーションを最小にするために、酸性化した塩化銅[pH1.0,10-12M]を、本例で記載されるように最初に用いた。これは、64Cuが塩化銅の形態で生成され、pH1.0の塩酸溶液に送達されるためである。
銅槽プロトコールを非放射性銅について最適化させた後、前記無電解めっき方法を、64Cuを「ホット」銅供給源として用いて行った。これは、64Cuを非放射性銅の代わりに用いるわずかな変更を加えて、上記に詳細が記載されるプロトコールと同一のものに従って行った。市販品として購入した64Cuの銅イオン濃度は、フェムトモル濃度単位であることが知られている。しかしながら、この銅濃度はバッチ間で変動する。典型的には、受け取った市販の放射性薬剤の公知濃度は、メガベクレル(MBq)として定義される。それゆえ、この例では、64Cuの固定したMBq濃度を用いた。市販品として購入した64Cuを80MBq/mLまで希釈した。
具体的には、上記に記載されるマルチモーダルPET/MRI造影剤は、適する医薬的に許容される賦形剤と組み合わせて、ヒトまたは非ヒト対象への経口、筋肉内または静脈内注射による投与に適する製剤を供する。調製されると、前記製剤は、マルチモーダルPET/MRI画像化法を用いて対象における目的の領域を画像化するために用いることができる。
このように調製された製剤の画像化能力を調べるために、本発明者らは、生きているマウス対象に対して実験を行い、その方法は本明細書に記載される。
この考えを試験するために、本発明者らは、インジウムイオン(In3+)を+3の酸化状態の金属イオンの一例として用いて、パラジウム活性化シリカ被覆酸化鉄ナノ粒子を無電解めっきするための方法と同一の方法を用いた。本実施例における前記無電解めっき方法は、非放射性インジウム供給源を用いて行った。この非放射性系と放射性111Inとの間の変動を最小にするために、酸性化された塩化インジウム(InCl3)の保存溶液[pH1.0、10-6M]を金属イオンの供給源として用いた。これは、111Inが、市販の供給先から塩化インジウムの形で生成され、pH1.0の塩酸溶液中で送達されるためである。
この理論をさらに試験するために、本発明者らは、イットリウムイオン(Y3+)を、+3の酸化状態の金属イオンの別の例として用いて、パラジウム活性化シリカ被覆酸化鉄ナノ粒子を無電解めっきするための方法と同一の方法を用いた。この実施例における無電解めっき方法は、硝酸イットリウム六水和物(Y(NO3)3・6H2O)の形態中の非放射性イットリウム供給源を金属イオン源として用いて行った。
この理論をさらに試験するために、本発明者らは、ジルコニウムイオン(Zr4+)を+4の酸化状態の金属イオンの一例として用いて、パラジウム活性化シリカ被覆酸化鉄ナノ粒子を無電解めっきするための方法と同一の方法を用いた。この実施例における無電解沈着方法は、オキシ塩化ジルコニウム([Zr4(OH)8(H2O)16]Cl8(H2O)12)の形態の非放射性ジルコニウム供給源を金属イオン供給源として用いて行った。
本明細書に記載の本発明の実施態様は、マルチモーダルPET/MRI造影剤(一般に、造影剤と呼ばれる)の合成方法を提供する。これらのマルチモーダルPET/MRI造影剤は、独立して、核シンチグラフィーのためのガンマ放射体による診断用画像化、陽電子放射断層撮影(PET)画像化、核磁気共鳴画像化(MRI)、または独立して放射性核種治療応用、あるいは同時にマルチモーダル画像化応用(例えば、PET/MRI)または同時の画像化と放射性核種治療応用または治療診断応用のいずれかに用いることができる。
全ての化学物質は受け取った状態で使用した。全ての化学物質および溶媒は、Sigma-Aldrich Australiaから入手し、さらに精製することなく使用した。
超常磁性酸化鉄コアを有するナノ粒子は、2段階工程により合成した。
典型的には、0.5mLのポリオキシエチレン(5)ノニルフェニルエーテル(Igepal CO-520)を11mLのシクロヘキサンに加え、続いて20μLの上記で調製し十分に分散させたナノ粒子の2.5mg/mL(Fe当量濃度)のシクロヘキサン溶液を加えた。この溶液を300RPMで10分間混合し、20μLのオルトケイ酸テトラエチル(TEOS)を加えた。続いて、該溶液をさらに5分間混合させ、100μLのアンモニア溶液(水中で28wt%)を300RPMで攪拌しながら加えて、逆ミクロエマルションを生成した。15nm(±1nm)の全体の粒径を有するシリカ被覆酸化鉄ナノ粒子を、室温で攪拌しながら6時間の反応後に得た。反応が完了したら、10mLのメタノールを加えて、該逆ミクロエマルションを阻害し、前記シリカ被覆酸化鉄ナノ粒子を、磁気分離または遠心分離(10,000RPMで室温にて10分)を用いてメタノール層から抽出した。
酸化鉄ナノ粒子周囲のSiO2シェルの表面は、塩化スズ(SnCl2)の酸性溶液を用いてスズで増感化する。これを達成するために、水中に5mg当量の鉄を含む酸化鉄ナノ粒子を磁気的に沈殿させ、湿った沈殿物中において、酸性塩化スズ溶液[5mL、3mM、pH1.0]を加え、10分間反応させる。これにより、Sn2+イオンが陽性に荷電したシリカ表面に結合する。10分後、前記粒子を5mLの脱イオン化水で2回すすいで、磁気分離を用いて結合していないSn2+イオンを除く。この粒子の分離は、10,000RPMで5分間の遠心分離により行うこともできる。続いて、生じたナノ粒子は、TEMおよびエネルギー分散型X線分析(EDX)を用いて特徴付けした。
スズ増感化酸化鉄ナノ粒子上のパラジウムの撒種は、磁気的に分離された粒子を硝酸パラジウム水溶液(Pd(NO3)2・2H2O)[5mL、3mM]に5分間曝露することによって行い、以前に結合されたSn2+イオンを置換しつつ同時に、金属Pd0核種のシリカシェルの表面上への沈着を生じる。この反応の間、Pd0核種の自発的な形成がPd2+からPd0への還元中のSn4+イオンの形成により進行する。
金属沈着については、工程(D)からのパラジウム活性化シリカ被覆酸化鉄ナノ粒子を磁気分離にかけて、パラジウム活性化シリカ被覆酸化鉄ナノ粒子を含有する湿った沈殿物[湿った沈殿物中の200μg当量のFe]を上澄み物から単離する。生じた湿った沈殿物を、金属槽(水酸化ナトリウム水溶液[125μL,1M]、続いてすぐに酒石酸カリウムナトリウム水溶液[125μL,0.3M]、さらに酸性化された金属イオン源[250μL、10-6M、pH1.0]、続いてすぐにホルムアルデヒドム水溶液[500μL、1M]を還元剤として加えることを含む)に曝露する。反応が完了したら、得られた粒子を、磁気分離または遠心分離(10,000RPMで5分間)を用いて(1ml)脱イオン化水で2回すすぎ、残っている反応物または副生成物を取り除く。次いで、生じた沈殿物を、(0.1ml)の適切な溶媒(典型的には、水、生理食塩水、緩衝液または生物学的増殖媒体)中に2分間の超音波処理により分散させて、無電解金属めっきシリカ被覆酸化鉄ナノ粒子を得る。
マルチモーダルPET/MRI画像は、Siemens VB17を実行する300mm ボア7テスラ ClinScanを含むBruker ClinScan PET-MRスキャナー、およびSiemens Inveon Acquisition Workplace(IAW)ソフトウェアのもとで操作する磁石ボアの中心に1ブロックあたり15X15 LSO結晶(1.6X1.6X10mm)を備える3環の16検出器ブロックを含むリムーバブルPETインサートを用いて収集した。前記PET環内部に72mm ID ラット体MRI rfコイルを用いて、PET収集と同時に全体のマウス画像を収集した。(Bruker、ドイツ)。
本発明の好ましい実施態様で記載のマルチモーダルPET/MRI造影剤は、下記を含むが、これらに限定されないいくつかの応用で用いられる可能性を有する:
1.細胞治療への応用
2.診断用画像化
3.治療
4.治療診断(Theranostic)への応用。
他の実施態様において、マルチモーダルPET/MRI造影剤は、上記に記載されるものに限定されるものではないことは、関連技術の当業者によって評価される。例えば、上記に記載されるフルオロフォアは、磁気コア内に積載される量子ドットであることに限定されるものではない。例えば、前記フルオロフォアは、シリカ(SiO2)シェル層内に浸出した色素または放射性核種金属層に結合した色素もしくは実際の量子ドットでありうる。前記フルオロフォアのPET/MRI造影剤の表面への結合は、多くの結合技術のいずれか1つによって達成されうることが評価される。

Claims (16)
- 磁気信号を発生するコア;および
前記磁気信号を発生するコアの表面上に少なくとも部分的に形成された被覆部分
を含む、マルチモーダルPET(陽電子放射断層撮影)/MRI(核磁気共鳴画像化)造影剤であって、前記被覆部分には、機能化表面を有する内部層、および前記機能化表面上に形成された無電解沈着した放射性核種を含む外部層を含む複数の層が含まれ、前記機能化表面が、シリカである、前記造影剤。 - 前記磁気信号を発生するコアが、強磁性、常磁性、または超常磁性信号を発生するコアである、請求項1に記載のマルチモーダルPET/MRI造影剤。
- 前記磁気信号を発生するコアが、鉄(Fe)、コバルト(Co)、ニッケル(Ni)、ネオジム(Nd)、ガドリニウム(Gd)、およびマンガン(Mn)からなる群から選択される少なくとも1つの金属の酸化物、混合酸化物および/または水酸化物;鉄(Fe)、コバルト(Co)、ニッケル(Ni)、ネオジム(Nd)、ガドリニウム(Gd)、およびマンガン(Mn)からなる群から選択される少なくとも1つの金属イオンが積載された担体物質;鉄(Fe)、マグネタイト(Fe3O4)、マグヘマイト(γ-Fe2O3)、ヘマタイト(α-Fe2O3)、鉄合金、およびフェライト材料からなる群から選択される鉄系材料;鉄(Fe)、コバルト(Co)、ニッケル(Ni)、ネオジム(Nd)、ガドリニウム(Gd)、およびマンガン(Mn)からなる群から選択される少なくとも1つの金属の合金;ならびにこれらの組み合わせを含む、請求項1に記載のマルチモーダルPET/MRI造影剤。
- 前記放射性核種が、陽電子放射体であり、好ましくは、銅(60Cu、61Cu、64Cu)、スカンジウム(44Sc)、チタン(45Ti)、鉄(52Fe)、マンガン(51Mn、52Mn)、コバルト(55Co)、ガリウム(66Ga、68Ga)、ヒ素(72As)、ルビジウム(82mRb)、ストロンチウム(83Sr)、テクネチウム(94mTc)、イットリウム(86Y)、ジルコニウム(89Zr)、およびインジウム(110In、111In)からなる群から選択され、より好ましくは、64Cuである、請求項1に記載のマルチモーダルPET/MRI造影剤。
- 前記内部層が、金属、金属酸化物、金属カルコゲン化物、金属ニクトゲン化物、半金属、半金属酸化物、半金属カルコゲン化物、半金属ニクトゲン化物、自己組織化膜、高分子電解質、ポリマー、タンパク質、炭水化物、およびバイオポリマーからなる群から選択される種から形成され、好ましくは、シリカ(SiO2)である、請求項1に記載のマルチモーダルPET/MRI造影剤。
- 少なくとも1つのフルオロフォアをさらに含む、請求項1に記載のマルチモーダルPET/MRI造影剤。
- 少なくとも1つの量子ドットをさらに含む、請求項1に記載のマルチモーダルPET/MRI造影剤。
- マルチモーダルPET(陽電子放射断層撮影)/MRI(核磁気共鳴画像化)造影剤の合成方法であって:
磁気信号を発生するコアの表面を、機能化溶液と接触させて、前記磁気信号を発生するコア上に少なくとも部分的に内部層を形成させ、機能化表面を規定し;
前記機能化表面を、少なくとも二価スズイオンの供給源を含有する増感化溶液と接触させて、スズ増感化表面を形成させ;
前記スズ増感化表面を、少なくともパラジウムイオンの供給源を含有する活性化溶液と接触させて、パラジウム活性化表面を形成させ;次いで
前記パラジウム活性化表面を、少なくとも放射性核種イオンの供給源ならびに還元剤および/または加水分解剤を含有する無電解めっき溶液と接触させて、前記放射性核種イオンを、無電解沈着した放射性核種を含む層に前記パラジウム活性化表面上で変換させることを含む方法。 - 前記機能化溶液が、オルトケイ酸テトラエチル(TEOS)前駆体を含む、請求項8に記載の方法であって、前記TEOS前駆体を重合させて、前記内部層を、前記磁気信号を発生するコアを少なくとも部分的に封入するシリカ(SiO2)シェルとして形成させることをさらに含む方法。
- 前記増感化溶液が、塩化スズ(II)(SnCl2)を二価スズイオンの供給源として含む、請求項8に記載の方法。
- 前記活性化溶液が、硝酸パラジウムをパラジウムイオンの供給源として含む、請求項8に記載の方法。
- 前記放射性核種が、陽電子放射体、好ましくは、64Cuである、請求項8に記載の方法。
- 前記還元剤が、ホルムアルデヒドである、請求項8に記載の方法。
- 前記加水分解剤が、アンモニア、水酸化リチウム、水酸化ナトリウム、水酸化カリウム、および水酸化アンモニウムからなる群から選択されるアルカリである、請求項8に記載の方法。
- 前記無電解めっき溶液が、安定剤、錯化剤、および界面活性剤からなる群から選択される少なくとも1つの添加剤をさらに含む、請求項8に記載の方法。
- マルチモーダルPET/MRI画像化を用いる患者における目的の領域の画像化方法であって:
請求項1に記載のマルチモーダルPET(陽電子放射断層撮影)/MRI(核磁気共鳴画像化)造影剤を含む医薬製剤の患者への投与中または投与後の前記目的の領域におけるPETおよび/またはMRIシグナルを比較することによって前記目的の領域における前記製剤中に存在するマルチモーダルPET/MRI造影剤の出現を検出し;
前記目的の領域のPETおよびMRI画像データを収集し;次いで
前記目的の領域のマルチモーダルPET/MRI画像を、PETおよびMRI画像データを用いて構築すること
を含み、前記目的の領域は、バックグラウンド組織とは異なって見えるものである、方法。
