背景技术
从商业上而言,使工业规模的微生物发酵所产生的目的化合物的产率持续提高是十分重要的。
如果可以提高产率,这会解放对其它化合物的生产能力并降低对生产的新投入需要。
已知酸水解的淀粉可以作为经由里氏木霉(Trichoderma reesei)产生纤维素酶的诱导剂(Process Biochemistry,vol.27,no.6,1992,page327-334)。
发明内容
令人惊讶地发现,通过向培养基中添加褐化葡萄糖溶液,可以非常显著地提高目的化合物的产率,所以我们要求保护:
一种在培养基中发酵微生物、产生目的化合物的方法,该方法包括:
向培养基中添加褐化葡萄糖溶液,其中褐化葡萄糖溶液是经过酸处理并加热到至少90摄氏度的温度的葡萄糖溶液,并且其中葡萄糖溶液的浓度为至少500g/L。

微生物可以是可用于工业发酵的任何微生物,特别是细菌或真菌。
细菌
根据本发明的表达目的化合物的细菌可以是任何属的细菌。
在优选实施方式中,目的化合物可以得自革兰氏阳性细菌,特别是得自芽孢杆菌属(Bacillus)或链霉菌属(Streptomyces)的菌株。
在优选实施方式中,芽孢杆菌属的菌株选自嗜碱芽孢杆菌(Bacillusalkalophilus)、解淀粉芽孢杆菌(Bacillus amyloliquefaciens)、短芽孢杆菌(Bacillus brevis)、环状芽孢杆菌(Bacillus circulans)、凝结芽孢杆菌(Bacillus coagulans)、灿烂芽孢杆菌(Bacillus lautus)、迟缓芽孢杆菌(Bacillus lentus)、地衣芽孢杆菌(Bacillus licheniformis)、巨大芽孢杆菌(Bacillus megaterium)、嗜热脂肪芽孢杆菌(Bacillusstearothermophilus)、枯草芽孢杆菌(Bacillus subtilis)和苏云金芽孢杆菌(Bacillus thuringiensis)。

化合物可以是植物毒素,例如赛克斯托敏(thaxtomin)。目的化合物也可以是治疗性蛋白、或酶(例如,水解酶、转移酶、裂解酶、异构酶或连接酶;特别是纤维素酶)。
根据本发明的目的化合物可以是抗生素例如青霉素或头孢菌素或红霉素,或日用化学品例如柠檬酸。
赛克斯托敏
根据本发明的目的化合物可以是赛克斯托敏。赛克斯托敏是已知的植物毒素类。
赛克斯托敏包括环二肽家族的任何种类,例如含有4-硝基吲哚-3-基的2,5-二氧代哌嗪。合适的赛克斯托敏包括,描述为具有基础结构环-(L-4-硝基色氨酰基-L-苯丙氨酰基)的环二肽的试剂。在实施方式中,合适的二酮哌嗪部分可以是N-甲基化的,且包括携带苯丙氨酰基α-和环-碳羟基的同类物。根据本发明使用的合适赛克斯托敏的非限制性实例包括赛克斯托敏A、赛克斯托敏A邻位异构体、赛克斯托敏B、和C-14去氧赛克斯托敏B(赛克斯托敏D)、以及其中任一的衍生物。

蛋白
根据本发明的目的化合物可以是蛋白。

在特别优选的实施方式中,优选以下的水解酶:
淀粉葡糖苷酶:淀粉葡糖苷酶(也称为葡糖淀粉酶和葡聚糖1,4-α-葡糖苷酶,EC3.2.1.3)是催化从淀粉或相关寡糖和多糖分子的还原端释放D-葡萄糖的酶。
合适的淀粉葡糖苷酶包括真菌来源的那些,特别是得自丝状真菌或酵母,例如埃默森篮状菌(Talaromyces emersonii)、黑曲霉和泡盛曲霉的那些。包括化学改性突变体或蛋白工程突变体。

目的微生物的改性
产生目的化合物的微生物通常已经被包括与编码目的化合物的基因可操作地连接的启动子的表达构建体转化。在现有技术中使用并记载了多种启动子。
具有多于一个拷贝的对目的化合物进行编码的化合物通常也是有利的,以使产率尽可能高。
启动子优选为可诱导的启动子。
发酵
本发明可以用于工业规模的任何发酵,例如用于培养基为至少50升的任何发酵,优选为至少100升,更优选为至少500升,甚至更优选为至少1000升,特别是至少5000升。
微生物可以通过本领域已知的任何方法进行发酵。发酵培养基可以是包括复合氮和/或碳源的复合培养基,复合氮和/或碳源为例如大豆粉、大豆蛋白、大豆蛋白水解物、棉籽粉、玉米浆、酵母提取物、酪蛋白、酪蛋白水解物、马铃薯蛋白、马铃薯蛋白水解物、糖蜜等。发酵培养基可以是在化学上限定的培养基,例如WO98/37179中限定的。

有限碳条件
根据本发明,使用有限碳发酵可能是优势。
有限碳条件是指,微生物具有以特定生长速率生长的刚刚足够的碳,其中所述特定生长速率低于最大特定生长速率。
褐化葡萄糖
本发明公开,通过向培养基中添加褐化葡萄糖溶液,来代替仅向培养基中添加葡萄糖溶液,会达到令人惊讶的优势。

制作接种物以进行培养
从冷冻管接种菌株,且首先在28摄氏度在PDA琼脂上生长3天。从琼脂上切割出单个集落,用于对具有上述培养基以及作为碳水化合物来源的葡萄糖的摇瓶进行接种。摇瓶在28摄氏度、200RPM孵育2天;1天后,通过短时间的有力的手动摇晃,将壁生长摇晃到培养基/培养物中。该培养物用作上述摇瓶的接种物。
在2天的培养之后,测量生长和赛克斯托敏A的产生。
结论
基于与细胞干质密切相关的OD650,两种碳源引起几乎相同水平的生物质,而在褐化葡萄糖作为碳水化合物来源的培养物中的赛克斯托敏A水平令人惊讶地更高。
与葡萄糖作为碳水化合物来源的培养物相比,产率为170%。
Claims (15)
1.一种在培养基中发酵微生物、产生目的化合物的方法,包括:
向所述培养基中添加褐化葡萄糖溶液,其中所述褐化葡萄糖溶液是经过酸处理并加热到至少90摄氏度的温度的葡萄糖溶液,并且其中所述葡萄糖溶液的浓度为至少500g/l。
2.根据权利要求1所述的方法,其中所述微生物是细菌或真菌。
3.根据权利要求2所述的方法,其中所述细菌选自芽孢杆菌属、链霉菌属、埃希氏菌属和假单胞菌属。
4.根据权利要求2所述的方法,其中所述真菌选自绵霉属、曲霉属、头孢霉属、旋孢腔菌属、内座壳属、镰刀菌属、腐质霉属、毛霉属、脉孢菌属、青霉属、柄孢壳菌属、梨孢属和木霉属。
5.根据权利要求1所述的方法,其中所述目的化合物是次生代谢物。
6.根据权利要求1所述的方法,其中所述目的化合物是蛋白。
7.根据权利要求6所述的方法,其中所述蛋白是酶。
8.根据权利要求1所述的方法,其中所述发酵是分批、分批补料、反复分批补料或连续的发酵。
9.根据权利要求1所述的方法,其中所述酸处理使得葡萄糖溶液的pH小于pH4.5。
10.根据权利要求1所述的方法,其中所述葡萄糖溶液经过酸处理并在之后加热到至少90摄氏度的温度。
11.根据权利要求1所述的方法,其中所述葡萄糖溶液的浓度为500g/l至1200g/l。
12.根据权利要求1所述的方法,其中所述微生物已经被含有与编码所述目的化合物的基因可操作地连接的启动子的表达构建体转化。
13.根据权利要求12所述的方法,其中所述启动子是可诱导的启动子。
14.根据权利要求1所述的方法,其中在除去所述微生物后回收所述目的化合物。
15.根据权利要求5所述的方法,其中所述次生代谢物是赛克斯托敏。
