发明背景
核酸酶活性在自然界中普遍存在,并通常存在于任何发酵液中。已知某些核酸酶非常稳定。
有时,获得自发酵液的枯草杆菌蛋白酶产物无核酸酶活性是非常重要的;例如,在将DNA和RNA从组织或细胞系中分离时枯草杆菌蛋白酶产物用于去除核酸酶的情况,或在枯草杆菌蛋白酶产物用于分离PCR或RT-PCR模板的情况。

核酸酶
核酸酶可以是产生感兴趣的枯草杆菌蛋白酶的微生物还产生的任何核酸酶,或者可以是在枯草杆菌蛋白酶产物制备过程中已污染枯草杆菌蛋白酶产物的外源性核酸酶。核酸酶可以是核糖核酸酶(RNase)或脱氧核糖核酸酶(DNase),特别是核糖核酸酶(RNase)。

根据本发明,感兴趣的枯草杆菌蛋白酶优选在芽孢杆菌种中产生。优选地,芽孢杆菌种选自:嗜碱芽孢杆菌(Bacillus alkalophilus)、解淀粉芽孢杆菌(Bacillus amyloliquefa)、短芽孢杆菌(Bacillus brevis)、环状芽孢杆菌(Bacillus circulans)、克劳氏芽孢杆菌(Bacillus clausii)、凝结芽孢杆菌(Bacilluscoagulans)、耐盐嗜碱芽孢杆菌(Bacillus halodurans)、灿烂芽孢杆菌(Bacilluslautus)、迟缓芽孢杆菌(Bacillus lentus)、地衣芽孢杆菌(Bacillus licheniformis)、巨大芽孢杆菌(Bacillus megaterium)、短小芽孢杆菌(Bacillus pumilus)、嗜热脂肪芽孢杆菌(Bacillus stearotherophilus)、枯草芽孢杆菌(Bacillus subtilis)、以及苏芸金芽孢杆菌(Bacillus thuringiensis)。优选地,感兴趣的枯草杆菌蛋白酶在克劳氏芽孢杆菌、迟缓芽孢杆菌、地衣芽孢杆菌或枯草芽孢杆菌中产生。
公众可从多个保藏中心容易地获得这些种类的菌株,诸如,美国典型培养物保藏中心(ATCC)、德国微生物菌种保藏中心(DSM)、荷兰微生物菌种保藏中心(CBS)、美国农业研究菌种保藏中心(NRRL)。
发酵液
产生感兴趣的枯草杆菌蛋白酶的微生物可通过本领域已知的任何方法来发酵。发酵培养基可为在例如WO 98/37179中描述的基本培养基,或者发酵培养基可为包含复合氮和碳源的复合培养基,其中复合氮源可如WO2004/003216中描述的被部分水解。
发酵可以分批、重复分批、补料分批、重复补料-分批或连续发酵工艺来进行。
在补料分批工艺中,在发酵之前,不将包含一种或多种结构和/或催化元素的化合物添加至培养基中,或者,将部分包含一种或多种结构和/或催化元素的化合物添加至培养基中,在发酵工艺过程中分别地补入所有或其余部分的包含一种或多种结构和/或催化元素的化合物。可以将选择用于补料的化合物一起或彼此分开补入发酵工艺中。
在重复补料分批或连续发酵工艺中,在发酵过程中,另外补入完全起始培养基。所述起始培养基可与结构元素补料一起或分开补入。在重复补料-分批工艺中,以规律的时间间隔去除部分包含生物质的发酵液,而在连续工艺中,部分发酵液的去除连续进行。由此,发酵工艺补充有对应于去除的发酵液量的一部分新鲜培养基。
发酵/回收步骤
感兴趣的枯草杆菌蛋白酶可以通过本领域已知的方法发酵,例如US3,723,250中描述的。
感兴趣的枯草杆菌蛋白酶不限于,但通常将通过使用下列一些或所有步骤来回收:
1)预处理发酵液;
2)从发酵液去除细胞和其他固体物质(初步分离);
3)过滤;
4)浓缩(例如,超滤、结晶、蒸发);
4b)新步骤:核酸酶灭活步骤;
5)过滤;
6)进一步纯化(例如,盐沉淀、色谱法);
7)稳定和标准化。
在预处理步骤中,将细胞和固体物质絮凝,且沉淀某些可溶组分。该步骤可如WO 96/38469中所描述进行。细胞和其他固体物质的去除可以通过离心、过滤或微滤进行。
可以包括过滤步骤,以改进枯草杆菌蛋白酶溶液的澄清度和/或去除微生物细胞。浓缩步骤通常涉及超滤步骤或在减压下蒸发水。在浓缩过程中,可形成固体物质,其可通过进一步过滤去除。最后,可如本领域已知将枯草杆菌蛋白酶产物进一步纯化、稳定和标准化至所需浓度。
除了上述列出的单元操作以外,还可以应用一些其他的回收工序和步骤,例如,使用活性碳和/或使用不同的吸收剂处理枯草杆菌蛋白酶溶液,以去除例如不期望的化合物,诸如负面活性(side-activity)和有色化合物。
根据本发明的核酸酶灭活步骤通常在浓缩步骤后进行。
核酸酶灭活工艺
然后,将获得自发酵液的枯草杆菌蛋白酶溶液的pH(通常在絮凝、离心和超滤之后)调节到pH 8.5至pH 10.5范围的pH值,特别地调节到pH 9.0至pH 10.0范围的pH值。
然后,添加一种或多种多元醇。多元醇可以在pH调节之前、同时或之后添加。
然后,将获得自发酵液的枯草杆菌蛋白酶溶液的温度调节到50℃至80℃范围的温度,优选调节到55℃至75℃范围的温度,更优选调节到60℃至70℃范围的温度,特别地,调节到63℃至67℃范围的温度。
通常,可以在高pH和高温下温育枯草杆菌蛋白酶溶液持续达30分钟的一段时间,优选持续达15分钟的一段时间,更优选持续达10分钟的一段时间,特别地,持续达5分钟的一段时间。
通过使用多元醇、高pH以及高温的组合,核酸酶活性被降低至低于起始值的5%,优选核酸酶活性被降低至低于起始值的4%,更优选核酸酶活性被降低至低于起始值的3%,甚至更优选核酸酶活性被降低至低于起始值的2%,特别地,核酸酶活性被降低至低于起始值的1%,尤其,核酸酶活性被降低至低于起始值的0.5%。
通过使用多元醇、高pH以及高温的组合,枯草杆菌蛋白酶活性被维持在高于起始值的80%,优选枯草杆菌蛋白酶活性被维持在高于起始值的90%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的91%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的92%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的93%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的94%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的95%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的96%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的97%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的98%,更优选枯草杆菌蛋白酶活性被维持在高于起始值的99%,特别地,枯草杆菌蛋白酶活性被维持在高于起始值的99.5%。
多元醇
根据本发明,可以使用任何多元醇。然而,优选的是,选自1,2-丙二醇(单丙二醇或MPG)、1,3-丙二醇、丙三醇、乙二醇、木糖醇、阿拉伯糖醇、卫矛醇、甘露醇、赤藓醇、纤维二糖以及山梨糖醇,特别是MPG的多元醇。
多元醇可以以10%(v/v)至60%(v/v)的浓度添加,优选以20%(v/v)至50%(v/v)的浓度添加,特别地,以30%(v/v)至50%(v/v)的浓度添加。
下列实施例进一步描述了本发明,所述实施例并非意图以任何方式限制本发明所要求保护的范围。
实施例1
灭活获得自发酵液的枯草杆菌蛋白酶溶液中的RNase活性
测定:
使用修改的Ambion测定方法分析枯草杆菌蛋白酶样品的RNase活性,并使用Protazyme AK(交联和染色酪蛋白)测定分析枯草杆菌蛋白酶样品的蛋白酶活性。
RNase活性测定:
该测定与来自Ambion Inc.的RNase检测试剂盒:RNaseAlert QC系统(目录#1966)相比稍有修改。
底物:RNaseAlert底物,与荧光体和淬灭剂偶联的RNA(和试剂盒一起提供)。
温度:室温。
测定缓冲液:10×NucleaseAlert缓冲液和TE缓冲液(均和试剂盒一起提供)。
无RNase的水(和试剂盒一起提供)。
RNase去污染:就在进行RNase活性测定之前,用RNaseZap(和试剂盒一起提供)来清洁吸量管和实验台。
荧光计:PolarStar,激发滤光片=485-P,发射滤光片=520-12,参数:输出值=50,循环数=20,瞬时发光=4,迟延=0.3,循环时间=30秒。
底物(工作溶液):将1管冻干的RNaseAlert底物溶解于1ml TE缓冲液(涡旋)并使用无RNase的水稀释5倍。
将20微升10×NucleaseAlert缓冲液和80微升稀释的枯草杆菌蛋白酶样品(在无RNase的水中稀释)置于黑色微量滴定板的微孔中。通过添加100微升底物(工作溶液)启动测定,通过PolarStar设备监测荧光的增加,作为对RNase活性的检测值。缓冲液盲(无RNase的水)包括在该测定中(代替酶)。
枯草杆菌蛋白酶活性测定:
底物:Protazyme AK片(来自Megazyme)。
温度:37℃。
测定缓冲液:50mM H3BO3/NaOH,0.01%Triton X-100,pH 9.0。
通过轻柔搅拌将Protazyme AK片(来自Megazyme)悬浮于2.0ml 0.01%Triton X-100中。将500微升该悬浮液和500微升测定缓冲液混合于Eppendorf管中,并置于冰上。添加20微升稀释的枯草杆菌蛋白酶样品(在0.01%TritonX-100中稀释)。通过将Eppendorf管转移至Eppendorf热混合器来启动测定,将所述Eppendorf热混合器设置到测定温度。于Eppendorf热混合器上以最高振动速率(1400r.p.m.)温育管15分钟。通过将管转移回冰浴来终止温育。然后,在冰冻离心机中离心该管几分钟,将200微升上清液转移至微量滴定板。读取OD650作为蛋白酶活性的检测值。缓冲液盲包括在该测定中(代替酶)。
试验A:
如本领域已知对枯草杆菌蛋白酶147进行发酵。
如本领域已知对发酵液进行絮凝、离心以及超滤。超滤液的pH为大约6.5,超滤液的温度为20-25℃(室温)。
使用27%(w/w)NaOH将超滤液的pH调节到pH 9.0(RNase活性或枯草杆菌蛋白酶活性未丧失)。
将200微升调节过pH的超滤液转移至Eppendorf管。在时间=0分钟时,将管转移至被预温至70℃和1400r.p.m.的Eppendorf混合器。通过从Eppendorf混合器移除管而终止温育,并使得管达到室温(几分钟)。然后,将管转移至冰浴,并保持于冰上直至检测RNase和枯草杆菌蛋白酶活性。在温育0分钟、1分钟、2分钟、4分钟、6分钟以及8分钟之后检测RNase和枯草杆菌蛋白酶活性。结果示于表1中。

结论:从表1可以看出,如果没有添加多元醇,可以灭活RNase(6分钟之后0%),但是同时枯草杆菌蛋白酶活性被降低至起始值的70%。
试验B:
如本领域已知对枯草杆菌蛋白酶147进行发酵。
如本领域已知对发酵液进行絮凝、离心以及超滤。超滤液的pH为大约6.5,超滤液的温度为20-25℃(室温)。
使用27%(w/w)NaOH将超滤液的pH调节到pH 9.0,并添加MPG至最终50%(v/v)的浓度。
将200微升调节过pH的超滤液转移至Eppendorf管。在时间=0分钟时,将管转移至被预温至70℃和1400r.p.m.的Eppendorf混合器。通过从Eppendorf混合器移除管而终止温育,并使得管到达室温(几分钟)。然后,将管转移至冰浴,并保持于冰上直至检测RNase和枯草杆菌蛋白酶活性。在温育0分钟、1分钟、2分钟、4分钟、6分钟以及8分钟之后检测RNase和枯草杆菌蛋白酶活性。结果示于表2中。

结论:从表2可以看出,通过使用本发明的方法,可以完全灭活RNase(2分钟之后0%),同时维持所有枯草杆菌蛋白酶活性。
试验C:
如本领域已知对枯草杆菌蛋白酶147进行发酵。
如本领域已知对发酵液进行絮凝、离心以及超滤。超滤液的pH为大约6.5,超滤液的温度为20-25℃(室温)。
使用27%(w/w)NaOH将超滤液的pH调节到pH10.0,并添加MPG至最终50%(v/v)的浓度。

结论:从表3可以看出,通过使用本发明的方法,可以完全灭活RNase,同时维持所有枯草杆菌蛋白酶活性(通过使用与试验B相比较高的pH但较低的温度)。
Claims (12)
1.一种用于降低获得自发酵液的枯草杆菌蛋白酶溶液中核糖核酸酶活性的方法,其包括:
a)将所述枯草杆菌蛋白酶溶液的pH调节到pH8.5至pH10.5的范围;
b)添加多元醇;
c)将所述枯草杆菌蛋白酶溶液的温度调节到55℃至75℃的范围;
借此,所述核酸酶活性被降低至低于起始值的5%,且所述枯草杆菌蛋白酶活性维持在高于起始值的80%。
2.根据权利要求1的方法,其中,所述枯草杆菌蛋白酶选自枯草杆菌蛋白酶Carlsberg、枯草杆菌蛋白酶BPN′、枯草杆菌蛋白酶147、枯草杆菌蛋白酶309以及枯草杆菌蛋白酶I168。
3.根据权利要求1的方法,其中,所述枯草杆菌蛋白酶由芽孢杆菌产生。
4.根据权利要求1的方法,其中,所述枯草杆菌蛋白酶溶液是超滤过的发酵液。
5.根据权利要求1的方法,其中,将所述枯草杆菌蛋白酶溶液的pH调节到pH9.0至pH10.0的范围。
6.根据权利要求1的方法,其中,将所述枯草杆菌蛋白酶溶液的温度调节到60℃至70℃的范围。
7.根据权利要求1的方法,其中,所述核酸酶活性被降低至低于起始值的1%。
8.根据权利要求1的方法,其中,所述枯草杆菌蛋白酶活性维持在高于起始值的90%。
9.根据权利要求1的方法,其中,所述多元醇选自1,2-丙二醇、1,3-丙二醇、丙三醇、乙二醇、木糖醇、阿拉伯糖醇、卫矛醇、甘露醇、赤藓醇、纤维二糖以及山梨糖醇。
10.根据权利要求1的方法,其中,温育所述枯草杆菌蛋白酶溶液持续达10分钟的一段时间。
11.根据权利要求1的方法,其中,温育所述枯草杆菌蛋白酶溶液持续达5分钟的一段时间。
12.根据权利要求1的方法,其中,在步骤a)之前进行步骤b)。
