Abstract
一种吸附剂、固相载体及精制方法。本发明的课题在于提供一种具有高亲和性与难劣化性,同时可在温和条件下利用简便的程序,且廉价、安全地精制血液凝固因子或细胞粘附因子的吸附剂、及血液凝固因子或细胞粘附因子的精制方法。本发明是将具有式(1)所表示的肽片段的多肽作为用于血液凝固因子或细胞粘附因子的吸附剂。另外,使用所述吸附剂来精制血液凝固因子或细胞粘附因子。?-(Pro-Hyp-Gly)n-??(1)?(式(1)中,n为2~9000的整数)。

技术领域
本发明涉及一种用于血液凝固因子(bloodcoagulationfactor)或细胞粘附因子(celladhesionfactor)的吸附剂及精制方法。
背景技术
作为蛋白质(protein)等生物分子的精制方法,一般已知有色谱法(Chromatography),作为其具体例,有阳离子交换色谱法(CationExchangeChromatography)、阴离子交换色谱法(AnionExchangeChromatography)、凝胶过滤色谱法(GelfiltrationChromatography)、疏水性作用色谱法(HydrophobicInteractionChromatography)、亲和色谱法(AffinityChromatography)。由于通过组合这些方法可高度地精制对象蛋白质,所以在对血液、血浆或细胞等的培养液中含有的例如成为生物医药品的红细胞生成素或免疫球蛋白等的精制中,色谱法逐渐成为重要技术。尤其是亲和色谱法重要的是如下方面:与其他方法相比,对于对象物质的选择性非常高,因此可通过1次操作高度地精制,而可凭借短操作时间获得高生产性。
亲和色谱法是利用与对象物质具有相互作用的配位基(ligand)来进行,有时进行使用蛋白质作为该配位基的精制。例如,已知有利用蛋白质A固定化填充剂来精制免疫球蛋白(Immunoglobulin)而制造抗体医药,或者利用明胶(Gelatin)固定化填充剂来精制纤维连接蛋白(Fibronectin)等血液成分等。
虽然以蛋白质作为配位基的亲和色谱法能够实现高选择性的效率良好的精制,但存在由如下原因引起配位基劣化的问题:由碱洗引起的配位基的改性或分解、由蛋白质分解酶引起的配位基的分解、或配位基本身的脆弱性。
另外,以蛋白质作为配位基的亲和色谱法存在其配位基材料中会混入来自动物的材料的情况,也存在安全性问题。即,近年来,基于对疯牛病等人畜共通传染病的担忧,例如需要避免使用来自牛的明胶作为配位基,谋求一种代替来自动物材料的配位基材料。
此外,以蛋白质作为配位基的亲和色谱法存在如下情况:由于配位基与对象物质的高亲和性,所以对象物质的回收条件必须采用温和条件。例如,在自所述的蛋白质A固定化凝胶中洗脱出免疫球蛋白的情况下,需要酸性条件(例如pH值为3),在自明胶固定化凝胶中洗脱出纤维连接蛋白的情况下,有时使用8M的尿素、1M的溴化钠(NaBr)或精氨酸(Arginine)等(非专利文献1)。在此种严格的洗脱条件的基础上,对象物质的蛋白质改性的担忧、或洗脱所使用的尿素或精氨酸等试剂为高浓度而耗费成本成为问题,或者洗脱步骤后需要进行中和等处理而使成本或繁杂度成为问题。
如此,在蛋白质的精制中,业界寻求一种具有与对象物质的高亲和性与难劣化性,同时可在温和条件下利用简便的程序,且廉价、安全地精制蛋白质的亲和配位基。
此处,作为生物分子而熟知的胶原蛋白(Collagen)是具有在相同方向上聚合3条包含Gly-X-Y(X、Y为各种氨基酸,X为Pro、Y为Hyp的情形居多)的重复结构的多肽(Polypeptide)所形成的三重螺旋结构的蛋白质的统称。
已知有天然胶原蛋白会与各种生物分子结合,或具有血小板聚集能力。目前为止,得知对于天然胶原蛋白中的特定氨基酸序列而言会结合特定的蛋白质等分子(非专利文献3)。利用此种知识见解,也对创造出一种与特定的对象分子结合的胶原蛋白样肽进行研究,例如揭示了与血管性血友病因子(vonWillebrandFactor)等胶原蛋白结合分子结合的具有特定氨基酸序列的胶原蛋白样肽(非专利文献2)。
另外,作为以胶原蛋白样肽的形式创造出的肽,存在具有Pro-X-Gly(X为Pro或Hyp)序列的重复结构的多肽。得知该多肽具有血小板聚集能力(非专利文献4),并揭示有一种含有该多肽的血小板聚集引发物质(专利文献1)。然而,尚未知晓具有所述序列的重复结构的多肽片段是以高亲和性与纤维连接蛋白或血管性血友病因子等进行结合。

发明内容
本发明鉴于所述情况,课题在于提供一种具有与对象物质的高亲和性与难劣化性,同时可在温和条件下利用简便的程序,且廉价、安全地精制血液凝固因子或细胞粘附因子的吸附剂、及血液凝固因子或细胞粘附因子的精制方法。
本发明发明人们为了解决所述问题进行努力研究,结果发现血液凝固因子或细胞粘附因子等是以高亲和性吸附在具有包含Pro-Hyp-Gly序列的重复结构的肽片段的多肽上,还发现利用提升盐浓度的温和方法进行脱附。
即,本发明如下所述。
[1]一种用于血液凝固因子或细胞粘附因子的吸附剂(以下,记为本发明的吸附剂),其特征在于:含有具有式(1)所表示的肽片段的多肽,
-(Pro-Hyp-Gly)n-(1)
式(1)中,n为2~9000的整数。
[2]根据[1]所述的吸附剂,其中所述血液凝固因子或细胞粘附因子是选自血管性血友病因子、纤维连接蛋白、糖蛋白VI(GPVI(GlycoproteinVI))、整合素(Integrin)、热休克蛋白(Heatshockprotein)47、及玻璃粘连蛋白(Vitronectin)之中。
[3]根据[1]或[2]所述的吸附剂,其中所述多肽的重均分子量为570~700万。
[4]一种固相载体,其固定化有根据[1]至[3]中任一项所述的吸附剂。
[5]一种精制样品溶液中的血液凝固因子或细胞粘附因子的精制方法(以下,记为本发明的精制方法),其包括:
使根据[1]至[3]中任一项所述的吸附剂或根据[4]所述的固相载体与所述样品溶液接触,而使所述吸附剂吸附所述血液凝固因子或细胞粘附因子的吸附步骤。
[6]根据[5]所述的精制方法,其还包括:使所述吸附步骤后的吸附剂或固相载体与含有洗脱剂的洗脱液接触,而洗脱所述血液凝固因子或细胞粘附因子的洗脱步骤。
[7]根据[5]或[6]所述的精制方法,其中血液凝固因子或细胞粘附因子是选自血管性血友病因子、纤维连接蛋白、糖蛋白VI(GPVI)、整合素、热休克蛋白47、及玻璃粘连蛋白之中。
[8]根据[5]至[7]中任一项所述的精制方法,其中所述样品溶液中的样品为血浆。
[9]根据[6]至[8]中任一项所述的精制方法,其中洗脱剂是选自氯化钠(NaCl)、氯化钾(KCl)、氯化镁(MgCl2)、氯化钙(CaCl2)、硫酸钠(Na2SO4)、溴化钠(NaBr)、溴化钾(KBr)、乙酸钠(sodiumacetate)、三(羟甲基)氨基甲烷盐酸盐(TrisHydrochloride,Tris-HCl)、磷酸盐、胍盐(Guanidinesalt)、精氨酸及其盐、以及尿素之中。
由于在本发明中,使用化学合成的多肽作为吸附剂,所以与天然胶原蛋白等相比,不易因清洗液而发生劣化、不易被蛋白质分解酶分解、另外其为简单的重复序列,所以可容易地制造、获得,此外还能够消除对使用来自动物的材料的担忧。另外,针对本发明的吸附剂,能够仅以盐或尿素的浓度变化而吸附脱附血液凝固因子或细胞粘附因子,因此,可抑制成本,防止血液凝固因子或细胞粘附因子的改性。
因此,根据本发明,可提供一种具有与对象物质的高亲和性与难劣化性,同时可在温和条件下利用简便的程序,且廉价、安全地精制血液凝固因子或细胞粘附因子的方法。

另外,本说明书中的肽链的氨基酸序列是依据惯用方法,将N末端的氨基酸残基置于左侧,且将C末端的氨基酸残基置于右侧来记载。
<1>本发明的吸附剂
本发明的吸附剂中所含有的多肽具有下述式(1)所表示的肽片段(以下,记为聚PHG)。
-(Pro-Hyp-Gly)n-(1)
此处,Hyp例如为4Hyp,优选为反式-4-羟基-左旋-脯氨酸。
另外,式(1)中,重复数n为2~9000的整数。通过使n在该范围内,能够以高亲和性与血液凝固因子等或细胞粘附因子等吸附。就形成三重螺旋结构的配位基稳定性的观点而言,n优选5~1000,更优选10~500。
在本发明的吸附剂中所含有的多肽可以重复数n为5以上而具有三重螺旋结构。由于产生与血液凝固因子或细胞粘附因子等的吸附作用,所以多肽并非必须为三重螺旋结构,也可具有无规卷曲结构,但如果多肽具有三重螺旋结构,则如所述那样,作为配位基的稳定性提高。形成三重螺旋结构的多肽链可为直线状或也可具有1个以上的分支。在具有分支的情况下,可在分支点之后形成三重螺旋结构,此外,也可在该三重螺旋结构之后具有分支。
另外,可通过对多肽溶液测定圆二色性光谱来确认多肽是否具有三重螺旋结构。具体而言,认为在波长220~230nm下显示出正考顿效应(PositiveCottonEffect)、及波长195~205nm下显示出负考顿效应(NegativeCottonEffect)的情况下,该多肽具有三重螺旋结构。
另外,在本发明的吸附剂中所含有的多肽链彼此之间也可以相互交联。
在本发明的吸附剂中所含有的多肽的重均分子量并没有特别限定,就与血液凝固因子或细胞粘附因子的高亲和性及三重螺旋结构的稳定性的观点而言,优选570~700万,更优选2850~30万。
此处,多肽的重均分子量例如可通过记载在日本专利特开2003-321500号公报中的如下方法进行测定:使用色谱柱:Superdex200HR10/30(通用电气医疗日本股份有限公司制造)、流速:0.5mL/min、洗脱液:含有150mM氯化钠的10mM磷酸盐缓冲液(pH值7.4)作为分子量标准的凝胶过滤低分子量LMW标准试剂盒(GelFiltrationLMWCalibrationKit)及凝胶过滤高分子量HMW标准试剂盒(GelFiltrationHMWCalibrationKit)(通用电气医疗日本股份有限公司制造)的凝胶渗透色谱法的方法;或者使用色谱柱:SuperdexpeptidePE7.5/300(通用电气医疗日本股份有限公司制造)、流速:0.25mL/min、洗脱液:含有150mM氯化钠的10mM磷酸盐缓冲液(pH值7.4)作为分子量标准的凝胶过滤低分子量LMW标准试剂盒(GelFiltrationLMWCalibrationKit)(通用电气医疗日本股份有限公司制造)、人胰岛素(humaninsulin)及甘氨酸的凝胶渗透色谱法的方法。作为其他方法,可通过HPLC(HighPerformanceLiquidChromatography,高效液相色谱法)凝胶渗透色谱法进行测定,该HPLC凝胶渗透色谱法是使用色谱柱:TSK-GEL6000PWXL-CP8.0×300mm(东曹制造)、流动相20mM的KH2PO4·H3PO4(pH值3.0):MeOH=8:2、色谱柱温度40℃、流速0.5mL/min、检测利用UV(ultraviolet,紫外线)监测仪的215nm及示差折射检测系统、作为分子量标准的分子量50000至1600000的普鲁兰多糖(pullulan)昭和电工制造)及分子量11900000的右旋糖酐(dextran)(德国PSS公司(PolymerStandardsServiceGmbH))的方法。另外,通过在该HPLC凝胶渗透色谱法的检测器中使用怀雅特技术(WyattTechnology)公司的DAWNHELEOS、OptolabrEX,也可利用凝胶浸透色谱仪/多角度激光光散射检测器(GelPermeationChromatography-Multi-angleLightScattering,GPC-MALS)法进行测定。在本说明书中,多肽的重均分子量是通过这些方法测得的值。
在本发明的吸附剂中所含有的多肽可为仅包含聚PHG的多肽,但在无损三重螺旋结构的稳定性、另外无损本发明效果的范围内,除聚PHG以外也可含有氨基酸残基或肽片段、或者亚烷基(alkylene)。
作为氨基酸残基,可列举选自Ala、Arg、Asn、Asp、Cys、Gln、Glu、Gly、His、Hyp、Ile、Leu、Lys、Met、Phe、Pro、Sar、Ser、Thr、Trp、Tyr、Val之中的至少1种。作为肽片段,可列举多个所述氨基酸残基的1种以上结合而成的肽。作为亚烷基,可为直链状、分支状中的任一种,没有特别限定,具体可列举碳数1~18的亚烷基,实用上优选碳数2~12的亚烷基。
本发明的吸附剂中所含有的多肽以重量比计,在聚PHG:其他氨基酸残基或肽片段、或者亚烷基以外的肽片段=1:99~100:0、优选10:90~100:0的范围内具有聚PHG与除聚PHG以外的氨基酸残基或肽片段、或者亚烷基。
本发明的吸附剂中所含有的多肽只要不阻碍血液凝固因子或细胞粘附因子的吸附,则也可为与无机酸(盐酸、硫酸等)、有机酸(乙酸、乳酸、马来酸、草酸、柠檬酸等)、金属(钠、钾等)、有机碱(三甲胺、三乙胺等)的盐。本发明的吸附剂中所含有的多肽的盐化合物可为单独一种或也可为两种以上的组合。
只要在无损本发明效果的范围内,则本发明的吸附剂也可含有除具有聚PHG的多肽以外的物质。其他物质只要考虑到保存性、易操作性、活性稳定性等进行选择确定即可,没有特别限定,例如可列举如下的保存溶剂。
具有聚PHG的多肽可为通过任一方法所获得的多肽。
例如可优选通过如下方法获得:使用通过已知的固相合成法或液相合成法所获得的包含构成聚PHG的氨基酸的肽低聚物,进行缩合反应。
所述肽低聚物的缩合反应通常在溶剂中进行。溶剂只要是能够使成为原料的肽低聚物溶解(溶解一部分或全部)或悬浮的溶剂即可,通常可使用水或有机溶剂。具体而言为水、酰胺类(二甲基甲酰胺、二甲基乙酰胺、六甲基磷酰胺等)、亚砜类(二甲基亚砜等)、含氮环状化合物(N-甲基吡咯烷酮、吡啶等)、腈类(乙腈等)、醚类(二恶烷、四氢呋喃等)、醇类(甲醇、乙醇、丙醇等)、及这些成分的混合溶剂等。在这些溶剂中,优选使用水、二甲基甲酰胺、二甲基亚砜。
另外,所述肽低聚物的缩合反应优选在脱水剂(脱水缩合剂、缩合助剂)的存在下进行。如果在脱水缩合剂与缩合助剂的存在下进行反应,则不用经过反复进行去保护与结合氨基酸的繁杂的处理而抑制二聚化或环化,同时顺利地进行缩合反应。
脱水缩合剂只要可在所述溶剂中效率良好地进行脱水缩合,则没有特别限定,例如可例示:碳二酰亚胺系缩合剂(二异丙基碳二酰亚胺(Diisopropylcarbodiimide,DIPC)、1-乙基-3-(3-二甲基氨基丙基)碳二酰亚胺(1-Ethyl-3-(3-Dimethylaminopropyl)Carbodiimide=WaterSolubleCarbodiimide,EDC=WSCI)、1-乙基-3-(3-二甲基氨基丙基)碳二酰亚胺盐酸盐(WSCI·HCl)、二环己基碳二酰亚胺(DicyclohexylCarbodiimide,DCC)等)、氟磷酸盐系缩合剂(O-(7-叠氮苯并三唑-1-基)-1,1,3,3-四甲基脲六氟磷酸盐、O-苯并三唑-1-基-N,N,N',N'-四甲基脲六氟磷酸盐、苯并三唑-1-基-氧基-三吡咯烷基鏻六氟磷酸盐、苯并三唑-1-基-三(二甲基氨基)鏻六氟磷酸盐(benzotriazol-1-yl-oxy-tris(dimethylamino)phosphoniumhexafluorophosphate,BOP))等)、叠氮磷酸二苯酯(DiphenylPhosphorylazide,DPPA)。
这些脱水缩合剂可单独使用或组合两种以上而以混合物的形式使用。优选的脱水缩合剂是碳二酰亚胺系缩合剂(例如1-乙基-3-(3-二甲基氨基丙基)碳二酰亚胺、1-乙基-3-(3-二甲基氨基丙基)碳二酰亚胺盐酸盐)。
脱水缩合剂的使用量相对于肽片段的总量1摩尔,在使用不含水的非水系溶剂的情况下通常在0.7~5摩尔、优选0.8~2.5摩尔、更优选0.9~2.3摩尔(例如1~2摩尔)的范围内。由于在含有水的溶剂(水系溶剂)中,有时因水会导致脱水缩合剂失去活性,所以脱水缩合剂的使用量相对于肽片段的总量1摩尔,通常在2~500摩尔、优选5~250摩尔、更优选10~125摩尔的范围内。
缩合助剂只要促进缩合反应,则没有特别限制,例如可例示:N-羟基多元羧酰亚胺类(例如N-羟基琥珀酰亚胺(N-HydroxySuccinimide,HONSu)、N-羟基-5-降冰片烯-2,3-二甲酰亚胺(N-hydroxy-5-norbornene-2,3-dicarboxylicimide,HONB)等N-羟基二羧酸酰亚胺类)、N-羟基三唑类(例如1-羟基苯并三唑(1-Hydroxybenzotriazole,HOBt)等N-羟基苯并三唑类)、3-羟基-4-氧代-3,4-二氢-1,2,3-苯并三嗪(3-hydroxy-4-oxo-3,4-dihydro-1,2,3-benzotriazine,HOObt)等三嗪类、2-羟基亚胺基-2-氰基乙酸乙酯。
这些脱水缩合助剂也可单独使用或组合两种以上而以混合物的形式使用。优选的缩合助剂是N-羟基二甲酰亚胺类(HONSu等)、N-羟基苯并三唑或N-羟基苯并三嗪类(N-Hydroxybenzotriazine)(HOBt等)。
缩合助剂的使用量与溶剂的种类无关,相对于肽片段的总量1摩尔,通常在0.5~5摩尔、优选0.7~2摩尔、进而优选0.8~1.5摩尔的范围内。
优选适当地组合使用脱水缩合剂与缩合助剂。作为脱水缩合剂与缩合助剂的组合,例如可列举:DCC-HONSu(HOBt或HOOBt)、WSCI-HONSu(HOBt或HOOBt)。
在所述肽低聚物的缩合反应中,也可调节反应溶液的pH值,通常将反应溶液的pH值调整在中性附近(pH值=6~8左右)。pH值的调节通常可使用无机碱(氢氧化钠、氢氧化钾、碳酸钠、碳酸氢钠等)、有机碱、无机酸(盐酸等)或有机酸来进行。
另外,也可在反应溶液中添加不参与缩合反应的碱。作为不参与缩合反应的碱,可例示:叔胺类,例如三甲胺、三乙胺、二异丙基乙基胺等三烷基胺类;N-甲基吗啉、吡啶等杂环式叔胺类等。此种碱的使用量通常可从本肽低聚物的总摩尔数的1~2倍左右的范围内选择。
在以如上方式所获得的多肽中残留有反应所使用的试剂。由于残留试剂会影响本发明的吸附剂的血液凝固因子或细胞粘附因子的吸附能力,所以优选去除该试剂。去除所残留的试剂可使用透析法、色谱柱法、超滤法等已知方法。
另外,如果考虑多肽的稳定性及易操作性,则优选将反应溶剂置换为保存溶剂。自反应溶剂向目标保存溶剂的置换,在透析法中可通过使用透析外液作为目标保存溶剂,在色谱柱法中可通过使用流动相作为目标保存溶剂的方式进行置换。
作为保存溶剂,只要是能够抑制所获得的有效成分多肽的物理性质等发生变化的溶剂,则没有特别限定。例如可列举:水、生理盐水、具有自弱酸向弱碱的缓冲能力的缓冲液。其中,优选不含有对血液凝固因子或细胞粘附因子产生影响的物质。
本发明的吸附剂能够以高亲和性吸附血液凝固因子或细胞粘附因子。尤其对吸附剂的亲和性较高,因此在所述血液凝固因子或细胞粘附因子之中,优选血管性血友病因子(以下,记为vWF)、纤维连接蛋白(以下,记为FN)、糖蛋白VI(以下,记为GPVI)、整合素、热休克蛋白47、玻璃粘连蛋白。
由于本发明的吸附剂可从血液等蛋白质的混合物中精制或去除血液凝固因子或细胞粘附因子,或者精制利用菌体或酵母等所制造的重组蛋白质,所以可适宜地用于目标血液凝固因子或细胞粘附因子的精制或制造中。
可将本发明的吸附剂设为固定化在固相载体上的态样。作为此种固相载体的材料,没有特别限定,可使用纤维素类、聚合物类、二氧化硅类、琼脂糖类等已知的材料。另外,其形态也可为颗粒、平板、纤维等,没有特别限定。
就防止操作中吸附剂的脱附的观点而言,固定化优选以共价键将本发明的吸附剂固定在固相载体上。利用共价键的固定化,例如可通过在所述固相载体的表面导入酯基、醛基、环氧基等反应性基,并在固相载体表面进行与本发明的吸附剂所含有的多肽的胺基或羟基的偶联反应的通常公知的方法进行。另外,由于存在各种市售的表面导入有反应性基的固相载体,所以可适宜地利用这些市售的固相载体。多肽在固相载体上的固定化量只要不阻碍本发明的效果,则可任意地设定。
此种固定化固相载体例如包含在蛋白质精制试剂盒等中,并可适宜地用于下述血液凝固因子或细胞粘附因子的精制方法。
<2>本发明的精制方法
本发明的精制方法包括:使本发明的吸附剂或固定有本发明的吸附剂的固相载体与所述样品溶液接触,而使所述吸附剂吸附所述血液凝固因子或细胞粘附因子的吸附步骤。另外,在进一步的态样中,还包括:使所述吸附步骤后的吸附剂或固相载体接触含有洗脱剂的洗脱液,而洗脱所述血液凝固因子或细胞粘附因子的洗脱步骤。
本发明的精制方法可精制样品溶液中的血液凝固因子或细胞粘附因子。尤其是在所述血液凝固因子或细胞粘附因子之中,可优选地将vWF、FN、GPVI、整合素、热休克蛋白47、玻璃粘连蛋白设为精制对象。
另外,作为所述样品,并无特别限定,可列举:全血、血浆、细胞等的培养液、细胞提取液、血清、体液、重组细胞的产物等,尤其优选血浆。当样品为全血或血浆等来自生物的样品的情况下,可为任何动物的样品,可优选应用人类的样品。
本发明的精制方法可应用于从血液等蛋白质混合物中精制或去除血液凝固因子或细胞粘附因子、或者应用于利用菌体或酵母等所制造的重组蛋白质的精制中,所以可适宜地用于目标血液凝固因子或细胞粘附因子的精制或制造中。
以下,对本发明的精制方法的各步骤进行说明。
在本发明的精制方法的吸附步骤中,样品溶液通常含有0.01~0.5M的盐或尿素。其原因在于:如果是低于0.01M的浓度,则会损害血液凝固因子或细胞粘附因子的稳定性,另外,如果是高于0.5M的浓度,则会阻碍吸附剂吸附血液凝固因子或细胞粘附因子。另外,作为盐的种类,可列举:氯化钠、氯化钾、氯化镁、氯化钙、硫酸钠、溴化钠、溴化钾、乙酸钠、三(羟甲基)氨基甲烷盐酸盐、磷酸盐、胍盐、精氨酸及其盐等,没有特别限定,也可使用2种以上的盐,或组合使用盐与尿素。样品可通过利用Tris-HCl(Tris(hydroxymethyl)aminomethaneHydrochloride(三(羟甲基)氨基甲烷盐酸盐))、磷酸盐、乙酸盐、碳酸盐、柠檬酸、古德(Good's)缓冲盐等公知的缓冲液进行稀释来调整,将其pH值调整在无损血液凝固因子或细胞粘附因子的稳定性的范围内,例如调整至4~9。
在吸附步骤中,通过使样品溶液与本发明的吸附剂接触,而使吸附剂吸附血液凝固因子或细胞粘附因子,该步骤只要使用固定有本发明的吸附剂的固相载体,即可容易地进行操作。例如,也可使样品溶液通过填充有此种固定化载体的色谱柱,在该情况下,可适宜地调整样品溶液的通过速度或通过次数来使吸附充分地进行。另外,也可在装入固定化载体的容器(容器本身也可为固定化载体)中加入样品溶液,并使吸附进行充分的时间后(可进行溶液搅拌也可不进行),通过去除样品溶液等来回收固定化载体。
吸附步骤后,根据需要,也可利用清洗液、例如含有浓度0.01~0.5M的盐或尿素的缓冲液等进行清洗,而去除非特异性吸附至吸附剂上的分子或未吸附的分子。
另外,吸附步骤没有特别限定,优选在2~37℃的温度下进行,就防止吸附剂或蛋白质的改性的观点而言,更优选4~25℃。
在本发明的精制方法的洗脱步骤中,所使用的洗脱液含有洗脱剂,该洗脱剂的浓度通常为0.5~1M。其原因在于:如果是低于0.5M的浓度,则难以使血液凝固因子或细胞粘附因子自吸附剂脱附,另外,如果是高于1M的浓度,则不仅洗脱效率达到极限,而且会损害血液凝固因子或细胞粘附因子的稳定性。另外,作为洗脱剂的种类,可列举:氯化钠、氯化钾、氯化镁、氯化钙、硫酸钠、溴化钠、溴化钾、乙酸钠、三(羟甲基)氨基甲烷盐酸盐、磷酸盐、胍盐、精氨酸及其盐、尿素等,没有特别限定,也可组合使用2种以上的洗脱剂。另外,只要将洗脱液的pH值调整在无损血液凝固因子或细胞粘附因子的稳定性的范围内,例如调整至4~9即可。
在本发明的精制方法中,也可利用提升洗脱液的盐或尿素的浓度简便的方法,从吸附步骤所使用的溶液中洗脱、回收吸附剂所吸附的血液凝固因子或细胞粘附因子。该方法与现有的亲和色谱法相比,可使各阶段在温和条件下进行精制,且不存在使血液凝固因子或细胞粘附因子或成为配位基的具有聚PHG的多肽劣化的担忧。另外,也不会大量使用昂贵的试剂,因此效率非常好。
洗脱步骤也与所述吸附步骤相同,只要使用经固定化的固相载体,即可容易地进行操作。即,也可使洗脱液通过填充有吸附步骤后的固定化载体的色谱柱,在该情况下,可适宜地调整洗脱液的通过速度或通过次数来使洗脱充分地进行。另外,也可在装入吸附步骤后的固定化载体的容器(容器本身也可以是固定化载体)中加入样品洗脱液,并使洗脱进行充分的时间后(可以进行也可以不进行溶液搅拌),回收洗脱液。
另外,洗脱步骤并没有特别限定,优选在4~25℃的温度下进行。
[实施例]
以下,列举实施例进一步详细地说明本发明,但本发明不限定于这些实施例。
<<配位基的固定化>>
利用公知的方法将作为本发明吸附剂的含有聚PHG的多肽或用于比较的胶原蛋白作为亲和配位基而固定在凝胶载体上。
<实施例1>
利用50mM碳酸钠清洗CellufineFormyl(JNC股份有限公司制造)并进行吸滤,将所获得的清洗凝胶4.3g放入200mL的锥形瓶中,此外添加0.5重量%的含有聚PHG的多肽水溶液16g、50mM碳酸钠6.4mL、水9.6mL并充分地混合后,在40℃下振荡2小时。添加硼氢化钠50mg/mL的水溶液0.7mL,并进一步在40℃下振荡2小时。反应结束后,对反应浆料进行吸滤,回收凝胶。为了去除残留在凝胶中的未反应物等,利用50mM碳酸钠、盐酸水溶液(pH值为3)、纯水加以清洗,并进行吸滤,获得聚PHG固定化凝胶。
对清洗液中残留的含有聚PHG的多肽量进行定量,并根据与用于反应的含有聚PHG的多肽的差而算出经固定化的含有聚PHG的多肽的量。清洗液中的含有聚PHG的多肽量是由210nm的吸光度测定浓度而算出。
此处所使用的含有聚PHG的多肽是仅由式(1)所表示的肽片段构成的重均分子量约为500万的多肽,在以下的实施例、比较例中也相同。
<比较例1>
利用50mM碳酸钠清洗CellufineFormyl(JNC股份有限公司制造)并进行吸滤,将所获得的清洗凝胶4.3g放入200mL的锥形瓶中,此外添加猪皮精制胶原蛋白溶液1%(日本火腿(NipponMeatPackers)股份有限公司制造)8g、50mM碳酸钠24mL并充分地混合后,在40℃下振荡2小时。添加硼氢化钠50mg/mL的水溶液0.7mL,并进一步在40℃下振荡2小时。反应结束后,对反应浆料进行吸滤,回收凝胶。为了去除残留在凝胶中的未反应物等,利用50mM碳酸钠、盐酸水溶液(pH值为3)、纯水加以清洗,并进行吸滤,获得胶原蛋白固定化凝胶。
对清洗液中残留的胶原蛋白量进行定量,并根据与用于反应的胶原蛋白的差而算出经固定化的胶原蛋白的量。清洗液中的胶原蛋白量是由210nm的吸光度测定浓度而算出。
<实施例2>
将ShodexBIOACTgelEPO系列(昭和电工股份有限公司制造)以干燥重量称取2.7g,添加至30mL的0.25重量%的含有聚PHG的多肽、50mM碳酸钠溶液中,在40℃下振荡一晚进行反应。反应结束后,对反应浆料进行吸滤,回收凝胶。为了去除残留在凝胶中的未反应物等,利用50mM碳酸钠、盐酸水溶液(pH值为3)、纯水加以清洗,并进行吸滤,获得聚PHG固定化凝胶。
对清洗液中残留的含有聚PHG的多肽量进行定量,并根据与用于反应的含有聚PHG的多肽的差而算出经固定化的含有聚PHG的多肽的量。清洗液中的含有聚PHG的多肽量是由210nm的吸光度测定浓度而算出。
<实施例3>
将反应温度从40℃变为4℃,除此以外,与实施例2同样地进行。
<比较例2>
将ShodexBIOACTgelEPO系列(昭和电工股份有限公司制造)以干燥重量称取2.7g,添加至30mL的稀释为0.25重量%的猪皮肤精制胶原蛋白溶液(日本火腿股份有限公司制造)、50mM碳酸钠溶液中,在40℃下振荡一晚进行反应。反应结束后,对反应浆料进行吸滤,回收凝胶。为了去除残留在凝胶中的未反应物等,利用50mM碳酸钠、盐酸水溶液(pH值为3)、纯水加以清洗,并进行吸滤,获得胶原蛋白固定化凝胶。
对清洗液中残留的胶原蛋白量进行定量,并根据与用于反应的胶原蛋白的差而算出经固定化的胶原蛋白的量。清洗液中的胶原蛋白量是由210nm的吸光度测定浓度而算出。
<比较例3>
将反应温度从40℃变为4℃,除此以外,与比较例2同样地进行。
将所述实施例1~3及比较例1~3的配位基在凝胶载体上的固定化量示于表1。

<<血浆成分的吸附洗脱实验1>>
将实施例1及比较例1的固定化凝胶载体、及作为凝胶载体的原料的6重量%纤维素粒子(JNC股份有限公司制造)各1mL填充至聚丙烯制的色谱柱中。在使5mL以上的100mM的Tris-HCl(pH值7.5)(以下称为TB)液体通过而进行平衡化后,向色谱柱中通入用TB稀释至10倍的人类血浆(西格玛(Sigma)/P9523-5ML)5mL(1次)。其后,通入5mL的TB,冲洗未吸附的血浆成分(1次)。将这些从色谱柱出口流出的液体一并回收作为流通、清洗液。清洗结束后,向色谱柱中通入5mL的含有1M氯化钠的TB(1次),并回收从色谱柱出口流出的液体作为洗脱液。以上的操作均在室温下(25℃)进行。
通过ELISA(Enzyme-LinkedImmunosorbentAssay,酶联免疫吸附分析法),对流通、清洗液及洗脱液中分别含有的FN及vWF的各量进行测定。
表2中表示FN的吸附及洗脱结果,表3中表示vWF的吸附及洗脱结果。
本发明的吸附剂可效率良好地吸附血浆中的FN及vWF,另外,通过含有1M氯化钠的洗脱液,可容易地回收所吸附的FN及vWF。尤其是vWF在吸附剂上的吸附量多于胶原蛋白,而得知本发明的吸附剂对vWF的亲和性较高。另外,尤其得知虽然FN在吸附剂上的吸附量与胶原蛋白的程度相同,但是仅凭使盐浓度提高的简便洗脱操作便可容易地进行回收。

<<血浆成分的吸附洗脱实验2>>
将洗脱液变更为含有0.7M精氨酸及0.3M氯化钠的TB,除此以外,与所述吸附洗脱实验1同样地进行。通过ELISA,对流通、清洗液及洗脱液中分别含有的FN量进行测定。
表4中表示FN的吸附及洗脱结果。
由表2及表4得知,虽然实施例1与比较例1的FN的吸附量的程度相同,但在该洗脱条件下,本发明的吸附剂较为有利。即,比较例1(胶原蛋白)中,仅洗脱出以1M氯化钠的条件所吸附的FN的34.4%,在0.7M精氨酸及0.3M氯化钠的条件下能够洗脱出99.2%。相对于此,实施例1中,即便不使用精氨酸,也可以100%回收在1M氯化钠的温和条件下吸附的FN。

根据本发明,具有与对象物质的高亲和性与难劣化性,同时即便从如血浆那样的复杂蛋白质的混合物中也能够在温和条件下利用简便的程序,且廉价、安全地回收或去除血液凝固因子或细胞粘附因子。由于本发明可应用在血液凝固因子或细胞粘附因子的精制或使用其的生物医药品的制造中,所以在产业上非常有用。
Claims (4)
1.一种精制方法,其精制样品溶液中的血液凝固因子或细胞粘附因子,且包括:
使用于所述血液凝固因子或所述细胞粘附因子的吸附剂或固定化有所述吸附剂的固相载体与所述样品溶液接触,而使所述吸附剂吸附所述血液凝固因子或所述细胞粘附因子的吸附步骤,其中所述吸附剂含有具有式(1)所表示的肽片段的多肽,
-(Pro-Hyp-Gly)n-(1)
式(1)中,n为5~9000的整数;以及
使所述吸附步骤后的所述吸附剂或所述固相载体与含有洗脱剂的洗脱液接触,而洗脱所述血液凝固因子或所述细胞粘附因子的洗脱步骤,其中
所述血液凝固因子或所述细胞粘附因子是选自血管性血友病因子及纤维连接蛋白之中。
2.根据权利要求1所述的精制方法,其中所述多肽的重均分子量为2850~700万。
3.根据权利要求1所述的精制方法,其中所述样品溶液中的样品为血浆。
4.根据权利要求1所述的精制方法,其中所述洗脱剂是选自氯化钠、氯化钾、氯化镁、氯化钙、硫酸钠、溴化钠、溴化钾、乙酸钠、三(羟甲基)氨基甲烷盐酸盐、磷酸盐、胍盐、精氨酸及其盐、以及尿素之中。
