Description
技术领域
本发明涉及与膜联蛋白-A1结合的特异性结合分子和产生这种特异性结合分子的杂交瘤细胞系,特别涉及该分子的抗体和片段。这种特异性结合分子可用于治疗T-细胞介导的疾病。
背景技术
由于糖皮质激素(GCs)能够同时阻止固有免疫应答和适应性免疫应答,因此经常被用于治疗各种慢性自身免疫疾病。通过本发明人和其他研究组过去约10年的研究已经表明GCs对固有免疫应答的某些炎症效应是由被称作膜联蛋白-1(Anx-A1)的蛋白介导的。已经证明该蛋白在许多细胞种类包括中性粒细胞、巨噬细胞和内皮细胞的自我平衡控制中发挥着作用。然而,经常被忽视的方面是Anx-A1在适应性免疫应答中的作用。令人惊讶的是考虑已经将Anx-A1提议作为GCs的药理作用的第二信使之一。
本发明人先前已经证明,Anx-A1通过调节T细胞受体(TCR)信号的强度在T细胞中发挥自我平衡作用(D’Acquistoetal.,Blood109:1095-1102,2007)。
而且,本发明人已经证明,高水平的Anx-A1降低T细胞活化的阈值,并促进其分化成Th1细胞,但Anx-A1基因缺陷型小鼠却表现出T细胞活化受损和分化成Th2细胞的增加(D’Acquistoetal.,Eur.J.Immunol.37:3131-3142,2007)。
WO2005/027965记载了凋亡中性粒细胞将抗炎信号传递到树突细胞的机制的发现过程,并识别了干扰该过程的抗体。
发明内容
本发明人已经识别出一种在T细胞活化的特异性抑制方面性能良好且没有任何不利的细胞毒性效应的单克隆抗体。该抗体可用于治疗T细胞介导的疾病,例如风湿性关节炎或多发性硬化症。
因此,本发明提供一种针对具有图2A所示的氨基酸序列的人类Anx-A1蛋白而产生的特异性结合分子。
定义
本文中使用的“特异性结合分子”是一对彼此具有结合特异性的分子中的成员。特异性结合对中的成员可以是天然来源的或其整个或部分是合成产生的。成对分子中的一个成员的表面具有一个能与成对分子中的另一成员的特定空间和极性结构特异性结合从而互补的区域,该区域可以为突起或凹陷。因此,每对成员具有彼此特异性结合的特性。各种类型的特异性结合对的实例为抗原-抗体、生物素-亲和素、激素-激素受体、受体-配体、酶-基质。本发明主要考虑抗原-抗体类型的反应。本发明的特异性结合分子与Anx-A1的结合亲和力大于与其它分子的结合亲和力,即该特异性结合分子与Anx-A1特异性结合。与Anx-A1结合的特异性结合分子包括抗Anx-A1抗体和适体。本发明的特异性结合分子通常为抗体。本发明的抗Anx-A1抗体通过阻止T细胞活化而发挥功能,因此,当被给药时,该抗体可以用于治疗T细胞介导的疾病,该疾病通常是由异常的T细胞活化引起的。

具体实施方式
膜联蛋白是一类结合钙和磷脂的细胞蛋白,也被称作脂皮质蛋白。膜联蛋白家族有13个成员,包括膜联蛋白A1、膜联蛋白A2和膜联蛋白A5。膜联蛋白A1也被称作膜联蛋白-1,在本文中被称作“Anx-A1”。膜联蛋白-1(Anx-A1)是一种分子量为37kDa的蛋白,并且最初被认为是糖皮质激素活动的介导物。在过去的几年中,已经有证据表明Anx-A1通过调节T细胞受体(TCR)信号的强度在适应性免疫系统,特别是在T细胞中发挥着自我平衡的作用。Anx-A1在体内固有免疫系统的细胞中发挥着炎症内源性下调物的作用。图1A是显示Anx-A1三维结构的带状图。
人类有8个编码Anx-A1的核苷酸序列。其中,仅有四个被翻译,因此有四种Anx-A1的亚型,所述四个亚型被命名为ANXA1-002、ANXA1-003、ANXA1-004和ANXA1-006。这些序列可以从数据库网站(www.ensembl.org)中得到,且被命名为OTTHUMT00000052664(ANXA1-002)、OTTHUMT00000052665(ANXA1-003)、OTTHUMT00000052666(ANXA1-004)和OTTHUMT00000052668(ANXA1-006)。图2A显示了一种人类膜联蛋白-1(Anx-A1)的ANXA1-003亚型的氨基酸和核苷酸序列。图2B、2C和2D分别显示了亚型ANXA1-002、ANXA1-004和ANXA1-006的氨基酸序列。从图2可以看出,亚型ANXA1-002、ANXA1-004和ANXA1-006是ANXA1-003的短剪接变体或是少量氨基酸发生改变的ANXA1-003的变体。

本发明的特异性结合分子可与膜联蛋白-1(Anx-A1)结合。特异性结合分子结合的Anx-A1是具有图2A所示的多肽序列的人类Anx-A1。
在第一方面中,本发明提供一种针对具有图2A所示的氨基酸序列的人类Anx-A1蛋白而产生的特异性结合分子。

CDRs是根据保守序列定义(Kabatetalin“SequencesofProteinsofImmunologicalInterest”,Nat’l.Inst.Health,Bethesda,MD(1987))和结构化定义(ChothiaandLeskJ.MolBiol.196:901-17(1987))的结合指定的。后来在Carteretal,ProcNat’lAcadSciUSA.89:4285-9(1992)中也记载了这些定义。

也可以对上述蛋白如融合蛋白的氨基酸序列进行氨基酸的缺失或插入。因此,例如,对多肽活性没有实质作用的氨基酸或至少没有消除该活性的氨基酸是可以缺失的。这种缺失可能是有益的,因为在保持活性的同时,降低了该多肽的整体长度和分子量。这可以减少特定目的所需的多肽量,如可以减少剂量水平。
也可以对上述的融合蛋白进行氨基酸插入。进行该过程可以改变本发明物质的性质(如有助于识别、纯化或表达,如上文与融合蛋白有关的解释)。
可以使用任何合适的技术进行与上述序列有关的氨基酸改变,如通过使用定点诱变或固态合成。
应该理解的是本发明范围内的氨基酸置换或插入可以使用天然存在的或非天然存在的氨基酸进行。无论使用的是天然氨基酸还是合成氨基酸,优选仅存在L-氨基酸。
本领域中已知的“同一性”是指两个或多个多肽序列或两个或多个多核苷酸序列之间的关系,这是通过比较序列确定的。在本领域中,同一性也可以指多肽或多核苷酸序列之间的序列关联度,视具体情况而定,这是通过比对序列的字符串确定的。虽然有多种方法都可以测量两个多肽序列或两个多核苷酸序列之间的同一性,但是用于确定同一性的一般方法是在计算机程序中进行编码。优选的用于确定两个序列之间同一性的计算机程序包括但不局限于GCG程序包(Devereux,etal.,NucleicAcidsResearch,12,387(1984),BLASTP,BLASTN和FASTA(Atschuletal.,J.Molec.Biol.215,403(1990))。
人们可以使用程序如CLUSTAL程序比较氨基酸序列。所述程序比较氨基酸序列,并通过在任一适合的序列中插入空格找到最优比对。用最优比对能够计算出氨基酸的同一性或相似性(同一性加上保守氨基酸的类型)。程序如BLASTx将比对最长的一段相似序列,并赋给一个合适的值。因此,可能获得发现多个相似区域的比较,每个区域都有不同的评分。本发明中考虑了两种类型的同一性分析。
两个氨基酸序列或两个核酸序列的同一性百分比是通过以实现最佳比对为目的的序列比对(例如,为了与序列最佳比对,可以在第一个序列中引入缺口)并比较相应位置的氨基酸残基或核苷酸确定的。“最优比对”是指两个同一性百分比最高的序列的比对。同一性百分比是通过被比较的序列中同源氨基酸残基或核苷酸的数量确定的(即,同一性百分比=同源位置的数量/位置的总数量×100)。
使用本领域技术人员已知的数学算法能够完成两个序列之间同一性百分比的确定。用于比较两个序列的数学算法的一个实例是KarlinandAltschul(1990)Proc.Natl.Acad.Sci.USA87:2264-2268中的算法,后修改为KarlinandAltschul(1993)Proc.Natl.Acad.Sci.USA90:5873-5877。Altschul,etal.(1990)J.Mol.Biol.215:403-410中的NBLAST和XBLAST程序已经纳入了该算法。用分数=100、字长=12的NBLAST程序可以进行BLAST核苷酸检索,以得到与本发明核酸分子同源的核苷酸序列。用分数=50、字长=3的XBLAST程序可以进行BLAST蛋白质检索,以得到与本发明蛋白分子同源的氨基酸序列。为了得到用于比较目的的间隙比对,可以使用Altschuletal.(1997)NucleicAcidsRes.25:3389-3402中记载的间隙BLAST。或者,可以使用PSI-Blast进行重复检索,该重复检索可检测分子之间的远缘关系(Id.)。当使用BLAST、间隙的BLAST和PSI-Blast程序时,可以使用各个程序(例如XBLAST和NBLAST)的默认参数。参见http://www.ncbi.nlm.nih.gov。序列比较使用的数学算法的另一个实例是CABIOS(1989)中的Myers和Miller算法。属于CGC序列比对软件包的ALIGN程序(版本2.0)已经纳入了该算法。本领域中已知的其他序列分析算法包括TorellisandRobotti(1994)Comput.Appl.Biosci.,10:3-5中记载的ADVANCE和ADAM;以及PearsonandLipman(1988)Proc.Natl.Acad.Sci.85:2444-8中记载的FASTA。在FASTA内,ktup是一个设置灵敏度和检索速度的控制选项。
通常,使用HGMP(人类基因组图谱计划)提供的BLAST计算机程序(Atschuletal.,J.Mol.Biol.215,403-410(1990))中的默认参数时,本发明特异性结合分子的CDRs的氨基酸序列在氨基酸水平上与上述CDRs的氨基酸序列具有至少70%的同一性。更典型地,CDR序列在氨基酸水平上与上述序列具有至少75%、80%、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%或至少99%的同一性。通常,本发明中特异性结合分子的每个CDR序列与上面列出的CDRs的氨基酸序列都有该水平的同一性。或者,本发明的特异性结合分子的任何1个,2个,3个,4个或5个CDRs与上面列出的CDRs的氨基酸序列有该水平的同一性。
本发明的特异性结合分子通常是抗体,更典型的是单克隆抗体。在一种实施方式中,本发明的单克隆抗体是人源化的。
通过修饰抗体的氨基酸序列可以将本发明的单克隆抗体人源化。降低本发明特异性结合分子的免疫原性的方法包括例如通过定位诱变或其它常用的分子生物学方法(RoguskaetalProteinEng.9895-904(1996))将CDR移植到适合的抗体主链骨架上或重塑可变表面残基。
其它可用的方法可以包括分子内潜在的T细胞表位的识别和随后如通过定点诱变移除这些表位(去免疫法)。当将特异性结合分子用作药物时可能需要所述特异性结合分子的人源化。也可以按照需要进行CDR区域或骨架序列周围的人源化。
可以使用单克隆抗体和其它抗体并通过重组DNA技术来产生保持原抗体特异性的其它抗体或嵌合分子。这类技术可以包括将编码抗体的免疫球蛋白可变区或互补决定区(CDRs)的DNA引入不同的免疫球蛋白的恒定区或恒定区加上骨架区。参见如EP-A-184187、GB2188638A或EP-A-239400。产生抗体的杂交瘤细胞或其它细胞可进行基因突变或其它改变,这可能改变或不改变所得抗体的结合特异性。
由于可以在多个方面对抗体进行修饰,术语“抗体”应该被解释为涵盖具有所需特异性结合结构域的任何特异性结合成员或物质。因此,该术语涵盖抗体片段、衍生物、功能等效物和抗体类似物,人源化抗体,包括任何包括免疫球蛋白结合结构域的多肽,不论是天然的还是整体或部分合成的。因此,涵盖了与另一种多肽融合的含有免疫球蛋白结合结构域或其等效物的嵌合分子。EP-A-0120694和EP-A-0125023中记载了嵌合抗体的克隆和表达。人源化的抗体可以是被修饰的具有非人如小鼠抗体的可变区和人类抗体的恒定区的抗体。例如,专利号为No.5225539的美国专利中记载了制备人源化抗体的方法。
本发明的特异性结合分子可以是抗体的片段。已经证明一个完整抗体的片段能够发挥结合抗原的作用。结合片段的实例有(i)由VL,VH,CL和CH1结构域组成的Fab片段;(ii)由VH和CH1结构域组成的Fd片段;(iii)由单一抗体的VL和VH结构域组成的Fv片段;(iv)由VH结构域组成的dAb片段(Ward,E.S.etal.,Nature341:544-546(1989));(v)分离的CDR区域;(vi)F(ab’)2片段,包括两个联合的Fab片段的二价片段;(vii)单链Fv分子(scFv),其中VH结构域和VL结构域是通过肽连接物连接的,所述肽连接物使两个结构域相连接以形成抗原结合位点(Birdetal.,Science242:423-426(1988);Hustonetal.,PNASUSA85:5879-5883(1988));(viii)双特异性单链Fv二聚体(PCT/US92/09965)和(ix)“双体抗体”,通过基因融合构建的多价或多特异性片段(WO94/13804;P.Hollingeretal.,Proc.Natl.Acad.Sci.USA90:6444-6448(1993))。通常,所述片段为Fab、F(ab’)2或Fv片段或scFv分子。

也可以使用与双特异性整体抗体不同的双特异性双体抗体,因为它们在E.coli中很容易被构建和表达。使用噬菌体展示技术(WO94/13804)能够从文库中很容易的筛选出具有合适的结合特异性的双体抗体(和许多其它的多肽如抗体片段)。例如,如果对抗原X有特异性的双体抗体的一个臂保持恒定,那么在另一个臂变化时就可以制备文库并筛选出具有合适特异性的抗体。

适于吸入给药的药物组合物包括可以由各种类型的压力定量气溶胶、喷雾器或吹药器产生的微粒粉尘剂或雾剂。
适合阴道给药的药物组合物可以以阴道栓、棉球、乳脂、凝胶剂、糊剂、泡沫或喷雾制剂存在。
适合肠胃外给药的药物组合物包括水和非水无菌注射液以及水和非水无菌悬浮液,所述注射液可以含有抗氧化剂、缓冲液、抑菌剂和使该剂型与目标受试者的血液基本上等渗的溶质;所述悬浮液可以含有悬浮剂和增稠剂。用于注射用溶液剂的赋形剂包括例如水、乙醇、多元醇、丙三醇和植物油。所述组合物可以置于单位剂量或多剂量的容器内,例如密封的安瓿和小瓶中,并可以储藏于冷冻干燥(冻干的)的条件下,仅需添加无菌液体载体,例如注射用水,就可以立即使用。可用无菌粉剂、颗粒剂和片剂制备临时的注射溶液剂和悬浮液。

对于哺乳动物特别是人类给药,预期的活性剂的日剂量为1μg/kg体重至10mg/kg体重,通常为10μg/kg体重至1mg/kg体重。在任何情况下,医生将决定最适合个体的实际剂量,该剂量取决于各种因素,包括个体的年龄、体重、性别和应答等。上述剂量是一般情况的示例。当然,也有使用较高剂量或较低剂量的实例,并且这都在本发明的范围内。

因此,在第四方面,本发明提供一种供医药学上使用的本发明的特异性结合分子。
在第五方面,本发明提供一种用于治疗T细胞介导的疾病的本发明的特异性结合分子。因此,本发明的这方面还包括治疗受试者的T细胞介导的疾病的方法,通常是需要被治疗的受试者,所述方法包括向受试者给药本发明的特异性结合分子。因此,本发明还涉及本发明的特异性结合分子在制备用于治疗T细胞介导的疾病的药物中的用途,或还涉及本发明的特异性结合分子在制备治疗T细胞介导的疾病的药物中的用途。所述治疗方法可以用于人类受试者或动物受试者,而且本发明同样涉及在人类药品和/或动物药品中的用途。本发明的特异性结合分子优选以“治疗有效量”向个体给药,该量足以显示出对个体的益处。本文中使用的“治疗”包括能够对人类或非人类动物优选为哺乳动物有益的任何方案。治疗可以是针对现存的病症或可以是预防性的(预防性治疗)。
在本发明的这个方面,本发明的特异性结合分子可以用于治疗由T细胞介导的各种不同疾病。在本文中,“T细胞介导的疾病”是指T细胞在发病或疾病或病症的发展中能够发挥作用的任何疾病或病症。T细胞介导的疾病通常是由异常的T细胞活化引起的。相应地,可以通过阻止Anx-A1的活性而阻止T细胞的活化来治疗这种疾病,本文中已经证明这可以通过单克隆抗体VJ-4B6起作用。通常,本发明中治疗的T细胞介导的疾病是Th1细胞起作用的疾病。
T细胞介导的疾病包括但不局限于移植物抗宿主疾病、移植排斥反应、动脉粥样硬化、HIV和/或AIDS、牛皮癣、流产和一些自身免疫疾病。根据本发明可治疗的自身免疫疾病包括风湿性关节炎(RA)、多发性硬化症(MS)、全身性红斑狼疮(SLE)、艾迪生氏病(Addison'sdisease)、格雷夫斯病(Grave'sdisease)、硬皮症、多肌炎、糖尿病且特别是某些形式的糖尿病(例如,青少年糖尿病)、自身免疫性葡萄膜视网膜炎、溃疡性结肠炎、寻常型天疱疮、炎性肠病、自身免疫性甲状腺炎、葡萄膜炎、贝赛特氏症(disease)和舍格伦式综合症(disease)。典型的T细胞介导的疾病是风湿性关节炎、多发性硬化症、全身性红斑狼疮、动脉硬化、HIV、AIDS或牛皮癣。更典型地,T细胞介导的疾病是风湿性关节炎或多发性硬化症。
本文中限定的T细胞介导的疾病还包括流产。本文中限定的治疗可以是预防性的(预防性治疗)。因此本发明的特异性结合分子可以用来预防流产。已知的是失控的Th1应答与流产有关,并且增加的Th2应答有助于怀孕。
典型的T细胞介导的疾病是风湿性关节炎。在风湿性关节炎(RA)中,认为是滑膜中T细胞识别抗原呈递细胞且与抗原呈递细胞相互作用。这些细胞一旦被活化,就会产生细胞因子和效应物分子;细胞因子的连续放大生产会形成“细胞因子级联反应”,该级联反应会导致巨噬细胞的活化和诱导炎性过程,使软骨和骨头的退化和吸收达到最高。随着时间的推移,会出现骨腐蚀,软骨损坏,以及完全丧失关节的完整性。最终,可能会影响多个器官系统。
在另一种实施方式中,T细胞介导的疾病是多发性硬化症(MS)。

在另一实施方式中,T细胞介导的疾病是系统性红斑狼疮(SLE)。
鉴于Anx-A1促进Th1细胞分化的能力,本发明也可以用于例如限制失控的保护性细胞(Th1)对抗细胞内病原体的应答和通过抑制Th1分化和促进Th2分化来治疗细胞外感染(Th2应答)。

单克隆抗体VJ-4B6是由图3A所述的方法产生的。简单地说,将cDNA编码的具有图2A所示序列的全长人类Anx-A1克隆到表达载体中,并确认其细胞表面的表达。将吸附到金微粒的载体DNA通过几次皮内应用后给药小鼠。鼠细胞接受了免疫载体并表达了cDNA编码的蛋白,从而刺激了免疫应答。然后,将鼠淋巴细胞与鼠骨髓瘤细胞融合以产生杂交瘤。然后,进行特异性测试,克隆杂交瘤和并产生单克隆抗体。

本发明的抗体特别可用于治疗T细胞介导的疾病,如风湿性关节炎和多发性硬化症。

实施例1-VJ-4B6特异性抑制含有膜联蛋白-1的T细胞中的T细胞活化
材料和方法
小鼠(图4B、5、6、7、8、9和10中使用的小鼠)
Balb/C小鼠来自B&KUniversal(Grimston,England)。AnxA1-/-小鼠来自发明人的实验室,该小鼠在无菌条件下出生于B&KUniversal。这些研究中使用的所有小鼠的年龄都在6-8周龄。动物实验是根据英国内政部规定(GuidanceontheOperationofAnimals,ScientificProceduresAct1986)和欧洲联盟条款的规定进行的。
小鼠T细胞的提取(图4B、5、6、7、8、9和10中使用的细胞)
脾脏和淋巴结(腋窝淋巴结、腹股沟淋巴结和肠淋巴结)取自6至8周龄的小鼠,并如前人所述地用注射器柱塞和50μm细胞滤网(BD)使组织温和降解而制备。用Ficoll将细胞悬液分层,以获得单核细胞,然后收集细胞并用RPMI培养基(Gibco)洗涤。
T细胞增殖试验(图5和图6中的数据)
将纯化的淋巴结T细胞(105细胞/ml)在96孔板中用包被在培养板上的抗-CD3(克隆145-2C11;eBioscience)或抗-CD3和抗-CD28(克隆37.51;eBioscience)刺激。18-20小时后,将培养物用1μCi的[3H]-胸苷(AmershamPharmaciaBiotech)脉冲12小时,并用自动闪烁计算器测量整合后的放射性。
IL-2的产生(图7和图8中的数据)
将纯化的淋巴结T细胞(105细胞/ml)在96孔板中用包被在培养板上的抗-CD3(克隆145-2C11;eBioscience)或抗-CD3和抗-CD28(克隆37.51;eBioscience)刺激。20-24小时后,收集培养上清液,并按照制造商的说明书使用鼠IL-2ELISA试剂盒(eBioscience)分析上清液,以得到IL-2的含量。
流式细胞术分析(图9和图10)
将细胞在FACS缓冲液(含有1%FCS和0.02%NaN2的PBS)中重悬。将淋巴细胞以1x106/ml的密度在100μl的FACS缓冲液中染色,并用CellQuestTM软件(BectonDickinson)在FACScalibur流式细胞仪中捕获。使用的抗体为抗-CD25FITC(克隆PC61,eBioscience)和PE-缀合的抗-CD69(克隆H1.2F3)。在4℃下将细胞在含有抗-CD16/32的FACS缓冲液中预培养30分钟以排除非特异性结合,然后用合适浓度的缀合抗体在4℃标记30分钟。标记后,洗涤并分析细胞。设定正向散射和侧向散射以排除红细胞和死细胞,每个样品至少分析2×104个淋巴细胞。在所有实验中,被染色的细胞都用FACScalibur流式细胞仪捕获并用FlowJo软件进行分析。
用流式细胞术分析确认VJ-4B6(图4)
将人类PBMC和PMN或小鼠的脾细胞和腹膜巨噬细胞先在含有4%多聚甲醛的FACS缓冲液中重悬10分钟,随后在含有4%多聚甲醛和0.02%皂苷的FACS缓冲液中重悬15分钟。将细胞与50ng/ml的生物素化VJ-4B6在4℃培养12小时。然后用链霉亲和素-FITC(稀释度1:200;eBioscience)将细胞在室温下培养1小时。标记后,洗涤并分析细胞。设定正向散射和侧向散射以排除红细胞和死细胞,每个样品至少分析2×104个淋巴细胞。在所有实验中,被染色的细胞都用FACScalibur流式细胞仪捕获并用FlowJo软件进行分析。
人类外周血白细胞(图4A中使用的细胞)
献血者为20-35岁的健康男性和女性,他们的血液测试结果是HIV、乙型肝炎病毒和丙型肝炎病毒均呈阴性。进一步筛选标准是在过去的四周里没有经历明显的感染,发烧、过敏症状、异常的血液细胞计数、肝脏酶的增加或任何药物治疗。从健康的志愿者中收集新鲜的静脉血,并将其立刻转移到含有3.2%柠檬酸钠的试管中(1:10稀释)。然后,使用密度梯度法将细胞分离:将3ml的FicollHistopaque-10771置于3ml的FicollHistopaque-11911上层(都来源于Sigma),以实现分层,并将6ml血液(用RPMI培养基1:1稀释)置于Histopaques上层。在室温下以1500RPM离心30分钟后,使用无菌滴管将PBMCs和PMNs从合适的层中吸出,并用RPMI洗涤三次。
统计分析
所有的统计分析都是用Prism软件进行的(GraphPadsoftware)。所有的数值都用平均值±SE表示。统计分析是通过Student’st检验或适当的单向ANOVA评估的。P<0.05的概率被认为是显著的。
结果
新的抗AnxA1抗体是由图3A中所示的遗传免疫方法产生的(GenovacGmbH,Germany)。测试了来自几只被免疫小鼠的血清,并用AnxA1cDNA对三个IgG识别细胞的结果呈阳性的小鼠进行转染。将这些小鼠的脾细胞与骨髓瘤细胞融合以产生杂交瘤细胞。三个杂交瘤细胞克隆中仅有一个可以成功地被亚克隆和扩增。这些杂交瘤细胞被称作VJ-4B6-E5-B10-D4。从杂交瘤细胞纯化的IgG2b碎片识别转染有AnxA1cDNA的细胞(图3B,绿线;右手峰),但不识别转染有无关cDNA的细胞(图3B,红线;左手峰)。
为了确认VJ-4B6的特异性,通过FACS分析已知表达不同蛋白水平的透化人类和小鼠细胞中的AnxA1表达。如图4A所示,与人类PBMC相比,人类PMN的VJ-4B6染色结果是高度阳性的(图4A,右手峰,分别为右栏和左栏)。这与先前观察到(D’Acquistoetal,结果未公布)的PMN与PBMC相比显著地表达更高的AnxA1水平是一致的。同样地,与脾细胞相比,用VJ-4B6染色的小鼠巨噬细胞表现出更高的AnxA1表达水平(图4B,右手峰,分别为右栏和左栏)。总体来说,这些结果显示了VJ-4B6可识别人类AnxA1和小鼠AnxA1。
然后,测试了VJ-4B6对T细胞活化的作用和特异性。为了这个目的,先用AnxA1+/+或AnxA1-/-小鼠的T细胞测量了VJ-4B6对抗-CD3诱导细胞增殖的作用(以3H-胸苷整合的平均值为指标)。图5显示了AnxA1+/+和AnxA1-/-的T细胞用抗-CD3刺激后,3H-胸苷整合的增加(分别为图5A和图5B)。在刺激后的培养物中加入VJ-4B6引起了AnxA1+/+的T细胞中3H-胸苷整合的浓度依赖性抑制,但对AnxA1-/-的T细胞中3H-胸苷整合没有影响。用抗-CD3和抗-CD28刺激的T细胞也出现类似的结果(分别为图6A和6B)。
为了进一步确认这些结果,测试了VJ-4B6对T细胞活化的其它传统标记即白细胞介素-2(IL-2)的产生和CD25/CD69上调的作用。用抗-CD3单独刺激或用抗-CD3和抗-CD28刺激T细胞可产生大量的IL-2(分别为图7和图8)。VJ-4B6的添加以浓度依赖性方式显著地抑制了AnxA1+/+T细胞中IL-2的产生,但对AnxA1-/-的T细胞中IL-2的产生没有影响。对于CD25/CD69上调也得到了相似的结果。用抗-CD3单独刺激或用抗-CD3和抗-CD28刺激T细胞活化诱导CD25/CD69双阳性T细胞的百分比显著提高(分别为图9和图10中从左起第二栏)。在VJ-4B6存在下的刺激可以浓度依赖性方式显著抑制AnxA1+/+T细胞中CD25/CD69的诱导,而不影响AnxA1-/-的T细胞中CD25/CD69的诱导。
这些结果共同表明VJ-4B6显著抑制T细胞增殖、IL-2的产生和由抗-CD3或由抗-CD3和抗-CD28引起的信号诱导的CD25/CD69的上调。另外,由于在AnxA1-/-T细胞中它的作用消失,因此这种作用应特别地归因于VJ-4B6与AnxA1的中和。这与发明人先前观察到的活化的T细胞释放合适的T细胞活化所需的内源性AnxA1-/-的结果相一致(D’Acquistoetal.,Blood109:1095-1102,2007;D’Acquistoetal.,Eur.J.Immunol.37:3131-3142,2007)。
总之,这些数据表明VJ-4B6以浓度依赖性方式抑制了抗-CD3诱导的CD25和CD69(T细胞活化的标记)的上调。在IL-2的产生和T细胞增殖上也观察到了相似的抑制作用。更重要的是,在膜联蛋白-1-缺乏的T细胞中没有观察到这种作用,这说明这种抑制是特异性的且抗体并没有引起任何不利的毒性作用。
实施例2-VJ-4B6的测序
本实施例的目的是从杂交瘤细胞中克隆抗体重链和轻链可变区域的基因和确定互补决定区(CDRs)的DNA序列和定位以及其他特征。
抗体可变区的克隆和测序
利用QiagenRNeasy迷你试剂盒(CatNo:74104)从1小瓶杂交瘤细胞中制备总RNA。用50μL水洗脱RNA并在1.2%琼脂糖凝胶上检查。
VH和VK(可变的kappa轻链)的cDNAs是用逆转录酶和IgG和kappa恒定区域的引物制备的。使用一大组信号序列引物,可通过PCR扩增出第一条链cDNAs。将扩增的DNAs用硅胶纯化,并克隆到载体TEasy(Promega)中。筛选得到的VH和VK的克隆以确定其中含有合适大小的插入物。通过自动DNA测序在两个方向上测定选定克隆的DNA序列。参照其它抗体序列确定序列中互补决定区(CDRs)的定位(KabatEAetal.,1991)。
结果
VJ-4B6轻链
识别了单一VK序列。图11显示了VJ-4B6VK的DNA序列和推导出的氨基酸序列。图12显示了推导出的蛋白序列,并标注了CDRs。两个单独扩增步骤中的九个克隆(七个是独立的)得到了相同的V区域序列。由杂交瘤融合伴侣产生的非生产性的异常VK序列也在多个克隆中出现,并且还有一个序列中有缺失的克隆。
VJ-4B6重链
识别了单一VH序列。图13显示了VJ-4B6VH的DNA序列和推导出的氨基酸序列。在九个单独的克隆中发现了相同的V区域序列。有两个克隆产生的序列中都有一个单碱基对的改变,有一个克隆产生的序列中有一个单碱基对的缺失和一个单碱基对的改变,还有一个克隆产生的序列中有两个单碱基对的改变。这五个单碱基对的变化中的每一个仅发生在一个克隆中。剩下的五个克隆有相同的序列。图14显示了推导出的蛋白序列,并标注了CDRs。

实施例3-VJ-4B6的体内免疫抑制作用
为了测试VJ-4B6的体内免疫抑制作用,我们选择了两种传统的免疫疾病模型:MOG33-55诱导的实验性自身免疫性脑脊髓炎(EAE;多发性硬化的小鼠模型)和胶原诱导的关节炎(CIA;风湿性关节炎的小鼠模型)。
方法
实验性自身免疫性脑脊髓炎。0天时用溶于PBS的300μg的MOG35-55以及等体积的含有300μg热灭活的结核分枝杆菌(M.tuberculosis)H37Ra的CFA组成的300μl的乳剂对小鼠进行皮下免疫。小鼠身体两侧均注射乳剂,然后在0天和2天时腹腔内注射溶于100μl生理盐水中的百日咳杆菌毒素(B.pertussistoxin)(500ng/100μl)。每天观察小鼠,以得到EAE指征和体重的减少量。将疾病严重性分为6个等级:0=没有疾病;1=尾部部分迟缓;2=尾部全部迟缓;3=正位反射受损;4=局部后肢瘫痪;5=全部后肢瘫痪;6=动物濒临死亡或死亡。
胶原诱导的关节炎。将乳化在弗氏完全佐剂(completeFreund’sadjuvant)(CFA;Hookelabs)中的200μg胶原类型II皮下注射到6-8只雄性DBA/1小鼠(8-12周大)的尾部。在初次免疫后的第21天,给小鼠皮下注射200μg用IFA(Hookelabs)乳化的胶原类型II以增强免疫(s.c.)。使用两种临床参数监测小鼠的关节炎发病的指征,所述参数为爪肿胀和临床评分。爪肿胀是通过用器官血量测量仪(plethysmometer)测量受影响的后爪的厚度来评估的。临床评分是通过制造商推荐的标准(http://hookelabs.com/protocols/ciaInductionDBA1/ciaInduction_DBA1.html)评估的。将四肢进行分级,以得出12只动物中每只动物的最高可能得分。
结果
如前人所述(Paschalidisetal.,JNeuroinflammation.2009;6:33)地用MOG33-55/CFA对雄性C57/BL6小鼠进行免疫。从用MOG33-55/CFA免疫后的第六天开始,每六天给小鼠腹膜内(i.p.)注射VJ-4B6(5、50和100ng/100μl)、IgG对照(100ng/μl)或PBS载体(对照)。如图15所示,与对照小鼠相比,用5、50和100ng的VJ-4B6处理的小鼠显示出统计学上显著的剂量依赖性减少的疾病指征(分别为图15B、C和D),然而,注射对照IgG却没有作用(图15A)。每种治疗的曲线下面积(AUC)和相对于对照(27.29AUC)的抑制百分比如下:IgG100ng,24.17AUC,11.4%;VJ-4B65ng,15.04AUC,44.8%;VJ-4B650ng,5.87AUC,78.5%;VJ-4B6100ng,7.04AUC,74.2%。
对EAE动物模型的研究已经表明疾病的急性期与体重的减轻同步发生,这可能是由于食欲不振和流体摄入不足。处理过的小鼠的重量测量与临床评分的严重度相关,并显示了与对照小鼠相比从第18天开始的VJ-4B6处理过的小鼠中剂量依赖性减少的体重减轻,但是IgG处理过的小鼠则没有这种现象(图16)。
为了确认治疗潜在的VJ-4B6作为体内免疫抑制剂,我们在CIA模型中测试了它的作用。如前人所述地用CFA中的牛科动物类型II胶原对雄性DBA/1小鼠进行免疫(D’Acquistoetal.,Blood.2007;109(3):1095-102)。从注射胶原增强免疫那天开始,每六天给小鼠腹腔内注射VJ-4B6(100ng/100μl)或PBS载体(对照)。与针对EAE所得到的数据一致的是,与对照小鼠(AUC83.50)相比,注射VJ-4B6显著地减少了(AUC26.75;67.9%)疾病指征的发展(图17)。

6.根据权利要求1-3中任意一项所述的抗体或该抗体的片段,所述抗体或该抗体的片段是由2010年6月3日保藏于欧洲细胞培养物保藏中心(ECACC),登记号为No.10060301的杂交瘤细胞系产生的。
7.一种杂交瘤细胞系,所述杂交瘤细胞系产生针对具有图2A所示的氨基酸序列的人类Anx-A1蛋白而产生的抗体或该抗体的片段,所述抗体或该抗体的片段具有互补决定区(CDRs)VLCDR1、VLCDR2、VLCDR3、VHCDR1、VHCDR2和VHCDR3,其中

11.根据权利要求1-3中任意一项所述的抗体或该抗体的片段,所述抗体或该抗体的片段用于治疗T细胞介导的疾病,其中,所述T细胞介导的疾病为风湿性关节炎(RA)或多发性硬化症(MS)。
12.权利要求1-6中任意一项所述的抗体或该抗体的片段在制备用于治疗T细胞介导的疾病的药物中的用途,其中,所述T细胞介导的疾病为风湿性关节炎(RA)或多发性硬化症(MS)。
