技术领域
本发明主要涉及用于分析和测量细胞和生物样品的分析与检测系统。更具体地讲,本发明涉及一种独特的样品室,例如,一种可用于细菌或寄生虫的明视场和荧光成像的薄间隙流体细胞计数室,及其制造和使用方法。
发明背景
通过测试生物样品来检测、识别、量化和表征相关生物分子或者细胞是医学诊断和生物医学研究领域的重要内容。生物溶液(如血液、脊髓液、细胞培养液和尿液)的微观粒子浓度经常需要被检测。
现有细菌细胞计数方法包括廉价的但需要手工操作的细胞计数器和昂贵的大型仪器如流式细胞仪、微孔板读板器以及细菌菌落测量。手工细胞计数装置极其繁琐且不稳定,并受人为误差和清洗程序制约。流式细胞仪需要大量的样品,并且因为利用激光而非常昂贵且需要良好培训的专业人员来操作设备以获得精确结果。通过光密度的细菌菌落测量也被用于细胞计数,但制备过程繁琐且易受污染。除此之外,为了精确测量,需要数天甚至数周的时间用于细菌培养。
最近,Nexcelom Bioscience利用微流体计数器开发了一款可自动计数直径大于2微米的细胞的自动化细胞计数器。
但是,对用于直径小于2微米例如细菌或寄生虫的生物微粒的高效、稳定的成像和计算设备和方法的需求是迫切的。
发明概述
本发明在一定程度上是基于发现了一种新颖的薄间隙流体室设计及其高效、经济的装配和制造方法。为了稳定可靠地成像并计算微粒,需要一种薄间隙流体样品室来最小化多重相面效应。根据微粒大小,该新颖装配方法可以用于产生合适的室间隙高度。本发明的细胞室可用于细菌、寄生虫、蛋白质聚集体,或更大的哺乳动物、昆虫或植物细胞类的明视场和荧光成像。
一方面,本发明一般涉及适于承载用于光学成像的液体样品的样品室。所述样品室包括:顶层;粘结层;底层;位于粘结层和底层之间并具有约0.5微米到约1,000微米之间的固定深度的样品承载腔;用于将样品放入或移出所述室的存取口;其中至少部分粘结层在约250纳米到约10,000纳米的波长范围内光学透明,且其中至少部分顶层和底层在约300纳米到约10,000纳米的波长范围内光学透明。
样品室的底层还包括基膜。
在某些实施方式中,所述样品室的样品承载腔具有约1微米到约100微米之间的固定深度。在某些优选的实施方式中,所述样品承载腔具有约2微米到约50微米之间的固定深度。在某些优选的实施方式中,所述样品承载腔具有约2微米到约20微米之间的固定深度。在另一些优选的实施方式中,所述样品承载腔具有小于约2微米的固定深度。在另一些优选的实施方式中,所述样品承载腔具有小于约1微米的固定深度。
在一些实施方式中,至少部分粘结层在约300纳米到约10,000纳米的波长范围内光学透明,且其中至少部分顶层和底层在约300纳米到约10,000纳米的波长范围内光学透明。
所述样品室的顶层可由选自如下的材料制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯(或PMMA)、聚苯乙烯、环烯烃共聚物,以及其他光学透明的塑料薄膜。
所述样品室的粘结层可为包含选自丙烯酸或丙烯酸酯胶粘剂材料的压敏粘结层。
所述底层可包含辐射固化或热固化材料。例如,底层可为紫外固化的聚合材料。在某些优选的实施方式中,所述紫外固化材料选自紫外固化的丙烯酸酯、聚氨酯和环氧树脂。
在某些优选的实施方式中,样品室还包括粘附于底层的衬层。所述衬层可包括选自如下的材料:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯、环烯烃共聚物,以及其他光学透明的塑料薄膜。
另一方面,本发明一般涉及适于承载用于光学成像的生物样品的样品室。所述样品室包括:光学透明的底层;光学透明的顶层;一面与顶层粘结而在另一面与辐射固化或者热固化聚合物层形成样品承载室的光学透明粘结层,其中所述样品承载室具有从粘结层至辐射固化或者热固化聚合物层之间约0.5微米到约1,000微米固定深度;用于向所述室引入样品的入口;用于将样品取出所述室(样品承载腔)的出口。
在一些实施方式中,所述样品承载腔具有约1微米到约100微米之间的固定深度。在某些优选的实施方式中,所述样品承载腔具有约2微米到约50微米之间的固定深度。在某些优选的实施方式中,所述样品承载腔具有约2微米到约20微米之间的固定深度。在另一些优选的实施方式中,所述样品承载腔具有小于约2微米的固定深度。在另一些优选的实施方式中,所述样品承载腔具有小于约1微米的固定深度。
对于某些应用,至少部分粘结层在约250纳米到约10,000纳米的波长范围内光学透明是优选的。
所述顶层可由选自如下的材料制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。

又一方面,本发明一般涉及装配样品室的方法。所述方法包括:提供光学透明衬层;在衬层上均一放置辐射或热固化层;在辐射固化或热固化层上压印预成型结构模具,其中预成型结构模具伸入辐射固化或者热固化层中;通过辐射或热处理固化辐射或热固化层;移去预成型结构模具,从而在辐射或热固化层中形成反映模具的预成型结构的压印结构;在辐射或者热固化层的压印结构上放置具有光学透明粘结层的光学透明顶膜,从而形成具有压印结构的室。
所述方法还可包括:在所述室上形成一个或多个存取口,以允许引入或移除样品。
所述光学透明衬层可由选自下列材料的制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。
所述辐射层可由选自紫外固化丙烯酸酯、聚氨酯或环氧树脂的紫外固化材料制备。
所述光学透明顶膜可由选自下列材料制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。
在一些优选的实施方式中,所述预成型结构模具由铝模加工而成。
又一方面,所述本发明一般涉及制造适于承载生物样品的样品室的方法。所述方法包括:在辐射固化或热固化层上压印预成型结构模具,所述预成型结构模具伸入辐射固化或者热固化层中;对所述辐射固化或热固化层进行固化以形成光学透明层从而形成具有压印结构的室;移去预成型结构模具;在辐射固化或者热固化层的压印结构上粘附光学透明顶膜,从而形成样品室;在所述室上形成一个或多个存取口以允许引入或移除样品。
因此,在又一方面,本发明一般涉及制造适于承载生物样品的样品室的方法。所述方法包括:在熔融塑料层上压印预成型结构模具;冷却所述熔融塑料层以在其上获得压印结构;移去所述预成型结构模具;在压印结构上粘附光学透明顶膜,从而形成样品室;在所述室上形成一个或多个存取口以允许引入或移除样品。
在又一方面,本发明一般涉及制造适于承载生物样品的样品室的方法。所述方法包括:在塑料层上压印热的预成型结构模具,在其上获得压印结构;移去所述预成型结构模具;冷却所述塑料层;在压印结构上粘附光学透明顶膜,从而形成样品室;在所述室上形成一个或多个存取口以允许引入或移除样品。

发明详述
本发明在一定程度上是基于发现了独特的样品室设计和高效且经济的装配和制造方法,所述方法可以使用明视场和荧光对细菌或寄生虫进行精确、稳定与高效成像。
细菌的形状和大小多种多样。细菌细胞的大小通常是真核生物细胞的十分之一,其长度通常在0.3到5.0微米之间。细菌中最小的是支原体属,只有0.3微米,和最大的病毒一样小。有些细菌甚至更小。虽然存在其他形状,但大部分细菌种类都是球形或棒状的。许多细菌种类是简单的单细胞,其他的连结成对、链、束等特征形状。细菌也可被拉长形成丝状。
当细菌与其他有机体形成寄生关系后,它们就变成了病原体。病原体细菌是引起人类死亡和疾病的主要原因之一,并能导致多种感染如破伤风、伤寒、白喉、梅毒、霍乱、食源性疾病、麻风病、肺结核。
通过使用手工细胞计数器、流式细胞仪、微孔板读卡器以及多种荧光和电子显微术和原子力显微术已经对细菌和寄生虫进行成像。一个实例为Cole Palmer公司的细菌计数器(图1)所示的传统手工细胞计数方法。它利用毛细管作用将细胞分散在H型的槽中以形成两个具有改良Neubauer规则的计数区域。位于负荷端的V形切口提供了流体负荷。当用显微镜观察时,计数器在黑色背景上显示出白色的Neubauer标记。盖玻片在刻画表面0.1毫米上方覆盖槽区。
最近,已经开发出一些可用于成像并计数细胞和生物材料(包括某些细菌和寄生虫)的设备。一个实例为Nexcelom Bioscience开发的一种设备(Cellometer Vision 10X),其是一种利用微流体计数室的自动化细胞计数设备,可以自动计数直径大于2微米的细胞。
已有设备和方法的一个重要的限制在于它们都不能提供高效的自动样品成像和细胞计数并在具有成本效益的同时提供精确、稳定的结果。该限制的一个主要原因在于缺乏有效的样品(或细胞)室设计和相应高效、经济的装配和制造方法。
本发明提供了样品室设计及其装配方法,其允许利用明视场和荧光两者有效地对小型生物粒子(如细菌或寄生虫)进行成像。
参考图2,其描述了本发明的一个实施方式中的细胞计数室的横截面图。所述细胞计数室200含有可为紫外固化材料层的底层210。在所述底层210上,存在预先设计的腔230(展现于底层210的上表面215上)。光学透明的粘结层220位于底层210和顶层240之间,粘附于底层210的上表面215和顶层240的下表面245上(除了预先设计的腔230处)。另外,存在至少一个(且优选两个(图3))样品孔(例如圆形开口)以允许通向所述腔230。
参考图4,其描述了本发明的另一个示例实施方式的横截面图。此处,样品室400含有两个各自具有固定高度h1和h2的样品承载腔,432和438。固定高度h1和h2可以相同也可以不同。相应的样品孔(图5)允许单独地将样品引入两个腔中,因此能够对两份不同的样品同时成像。
为了开发用于成像细胞计量术的经济高效的装配方法,必须解决一个非常重要的问题,即计数室的封闭。所述室的深度可以通过多种方式装配,但此处公开的最有效的方法是利用压印技术。该方法利用模具,当在热固化或光固化溶液中压印时,在塑料薄片上形成预先设计的结构。为了避免昂贵的设备和多步骤,此处公开了一种利用光学透明的粘结剂来覆盖整个上表面以在提供清晰成像能力的同时封闭所述室的独特方法。
图6描述了本发明的一个装配方法的示例实施方式。紫外固化溶液610被涂覆在塑料表面620上,结构模具630被压印在溶液610的表面615上。紫外光用于固化溶液610。模具630被移去后,形成所需的腔结构640。之后,包含粘结层660的顶层650被放置在含有结构640的紫外固化层610之上。
本发明的一个重要方面在于利用光学透明转移胶粘剂作为薄间隙室的封闭层,其从利用昂贵且繁琐的墨水转移系统或胶粘剂涂层简化为简单地加盖顶部薄片以在模具结构上形成封闭的室。
通过使用复制成型技术,可以制造间隙尺寸不同的结构,从而提供特定样品浓度的室。所述独特的装配方法可以引入生产程序中,以制造在顶部和底部有两个保护膜的载片(图7)。此外,可以使用示踪孔来移动片状加工的载片,类似于模拟照像机的行进胶片(图8)。另外,载片可以圈在塑料卷上,从而进一步降低成本和生产时间(图9)。通过使用塑料卷和胶粘剂可以大量生产所述计数室,从而显著降低工艺时间和成本。

在一些实施方式中,所述样品承载腔具有约1微米到约100微米的固定深度。在某些优选的实施方式中,所述样品承载腔具有约2微米到约50微米的固定深度。在某些优选的实施方式中,所述样品承载腔具有约2微米到约20微米的固定深度。在另一些优选的实施方式中,所述样品承载腔具有约2微米的固定深度。在另一些优选的实施方式中,所述样品承载腔具有小于约1微米的固定深度。
对于某些应用,至少部分粘结层在250纳米到10,000纳米的波长范围内光学透明是优选的。
所述顶层可由选自下列材料制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。
所述粘结层可为含有选自丙烯酸或丙烯酸酯胶粘剂的材料的压敏粘结层。
所述底层可包含辐射固化或热固化材料。在某些优选的实施方式中,所述底层包含紫外固化聚合材料,例如选自丙烯酸酯、聚氨酯或环氧树脂的材料。
在某些优选的实施方式中,所述样品室还包括粘附于底层的衬层。所述衬层可由选自下列材料制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。
又一方面,本发明一般涉及装配样品室的方法。所述方法包括:提供光学透明衬层;在衬层上均一放置辐射固化或热固化层;在辐射固化或热固化层上压印预成型结构模具,其中预成型结构模具伸入辐射固化或者热固化层中;通过辐射或热处理固化辐射固化或热固化层;移去预成型结构模具,从而在辐射固化或热固化层中形成反映模具的预成型结构的压印结构;在辐射固化或者热固化层的压印结构上放置包含光学透明粘结层的光学透明顶膜,从而形成具有压印结构的室。
所述方法还可包括:在所述室上形成一个或多个存取口,以允许引入或移除样品。
所述光学透明衬层由选自下列材料的制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。
所述辐射层可由选自紫外固化丙烯酸酯、聚氨酯和环氧树脂的紫外固化材料制备。
所述光学透明顶膜由选自下列材料制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。
在一些优选的实施方式中,所述预成型结构模具由铝模加工而成。
在一些实施方式中,胶粘剂被涂覆在顶层(例如,膜)以提供支持,而所述顶层和所述粘结层一同被直接施用于底层的结构表面上以形成所述室。
又一方面,所述本发明一般涉及制造适于承载生物样品的样品室的方法。所述方法包括:在辐射固化或热固化层上压印预成型结构模具,所述预成型结构模具伸入辐射固化或者热固化层中;对所述辐射固化或热固化层进行固化以形成光学透明层从而形成含有压印结构的室;移去预成型结构模具;将光学透明顶膜粘合在辐射固化或者热固化层的压印结构上,从而形成样品室;在所述室上形成一个或多个存取口以允许引入或移除样品。
可以通过热铸造形成需要的结构(例如,直接在底层上将熔融塑料浇铸在模具表面以形成底部部件)。可以通过热冲压形成所需结构(例如将塑料薄膜进行热冲压以形成所述结构)。这些工艺仅包含利用热使塑料材料“成形”以在塑料材料上形成所需结构,且不需任何化学反应。本发明还涉及底层由热铸造或者热冲压形成的样品室。
因此,在又一方面,本发明一般涉及制造适于承载生物样品的样品室的方法。所述方法包括:在熔融塑料层上压印预成型结构模具;冷却所述熔融塑料层以在其上形成压印结构;移去所述预成型结构模具;在压印结构上粘附光学透明顶膜,从而形成样品室;在所述室上形成一个或多个存取口以允许引入或移除样品。
在又一方面,本发明一般涉及制造适于承载生物样品的样品室的方法。所述方法包括:在塑料层上压印热预成型结构模具,从而在其上形成压印结构;移去所述预成型结构模具;冷却所述塑料层;在压印结构上粘附光学透明顶膜,从而形成样品室;在所述室上形成一个或多个存取口以允许引入或移除样品。
利用本发明的方法分析的样品包括获取于或来源于生物体的生物材料。通常所述样品包括细菌,寄生虫,细胞,组织,或生物分子,例如蛋白质、多核苷酸(如DNA或RNA),有机材料及上述的任意组合。这些样品包括但不限于头发、皮肤、组织、培养的细胞、培养的细胞介质和体液。
组织是大量的连接的细胞和/或细胞外基质物质,例如来源于人类或其他哺乳动物的中枢神经系统(CNS)组织、神经组织、眼组织、肝组织、胎盘组织、乳腺组织、胃肠道组织、肌肉组织、泌尿生殖组织等,并包括与细胞和/或组织相结合的连接的物质和液体物质。体液是来源于人类或其他哺乳动物的液体物质。这些体液包括但不限于血液、血浆、血清、血清衍生物、胆汁、痰、唾液、汗液、羊水、乳腺液和脑脊液(CFS),例如腰部或心室CSF。样品也可以是含有细胞或生物材料的介质。
本发明的系统还可以用于探询细胞系。细胞系是指在给定的适当介质和条件下可以无限生长的特定细胞。本发明的系统可用于探询任何类型的细胞系。细胞系可以是哺乳动物细胞系、昆虫细胞系或植物细胞系。示例细胞系可以包括肿瘤细胞系或干细胞系。
实施例
将铝模具放置在一个30密尔去除绿色衬垫的PC(聚碳酸酯)片上。然后用移液管向铝模具整个表面滴加约5大滴紫外固化聚合物溶液。之后将30密尔(“密尔”为千分之一英寸)的聚碳酸酯片(切成4块)放置在铝制模具的顶部,以使所述紫外固化溶液涂布于整个表面。在紫外线室内将整个装置固化大约12到15秒。将30密尔的顶部聚碳酸酯片移去,所述室的结构被压印在表面上。切割出一个标准的Nexcelom计数载片(Model No.CHT4-PD,CHT4-SD)顶膜并除去底部衬垫。将光学透的转移明胶粘剂置于顶部载片膜的底部。开两个孔以形成流体入口和出口。除去转移胶粘剂的底部衬垫并用金属辊将其卷在固化的压印结构上以防止胶粘剂向下接触所述表面。最后,切除多余的塑料区域。
测试了几个参数。首先,使用测量载片各层反射光的自制激光位置探测器测量室间隙的大小(参见图10)。测得的距离用来计算间隙的大小。第二,测量了所述室内的2.5微米荧光珠粒以测试载片的荧光能力和光学胶粘剂的自身荧光。第三,使用校准好的照明光源以避免出现图像中心亮区。最后,发现薄间隙室中流体流动特性满足要求。
所述薄间隙室载玻片如图11所示。使用激光位置探测器测得的间隙尺寸为大约30微米深。所述激光位置探测器使用集中照射在室上的532纳米绿色激光。不同层的反射光产生了明亮的光,其用来估测室的深度。在绿色和橙色两者荧光发射通道中观察所述薄间隙室内的2.5微米荧光珠粒(线性流动,Invitrogen)。图12显示了明视场(右)和绿光发射时(左)在薄间隙室内的2.5微米珠粒的荧光图像。图13显示了明视场(右)和橙光发射时(左)在薄间隙室内的2.5微米珠粒的荧光图像。图14显示了明视场(右)和蓝光发射时(左)在薄间隙室内的2.5微米珠粒的荧光图像。
另一个实施例使用了2微米珠粒。为了促进溶液流入所述室内,将2微米珠粒与2.5%、5%、10%和20%的IPA(异丙醇)溶液混合,这极大地将流速从多于5分钟提高至少于5秒钟(2.5%、5%、10%和20%的异丙醇各自需约4、3、2和1秒)。图15显示了制造的薄间隙室中2微米珠粒的明视场图像,以显示室内珠粒的分散状况。观察到所述珠粒在沉降下来之前持续流动了一长段时间。用透明胶带把出口和入口封住,沉降时间从约15分钟减为少于约5分钟。光的温度梯度不影响珠粒的流动。
对酵母菌和细菌样品进行了成像(图16-18),其结果证明本发明的细胞室和方法的有效性。在图16中,利用20倍物镜和薄间隙室,在明视场下同时观察酵母菌和乳酸菌。用能使所有细胞被染色的细胞核染色剂吖啶橙(AO)将酵母菌和细菌样品染色。酵母菌和细菌的荧光图像如图17所示,信号显示于图像的横截面图中。此外,其他染色剂如SYTO9(Invitrogen)也可用于达到于吖啶橙相同染色质量(图18)。

Claims (48)
1.适于承载用于光学成像的液体样品的样品室,其包括:
顶层;
粘结层;
底层;
位于粘结层和底层之间且具有约0.5微米到约1,000微米的固定深度的样品承载腔;以及
将样品引入或移出所述室的存取口;
其中至少部分粘结层在约250纳米到约10,000纳米的波长范围内光学透明,且其中至少部分所述顶层和所述底层在约300纳米到约10,000纳米的波长范围内光学透明。
2.权利要求1的样品室,其中所述底层包括基膜。
3.权利要求1的样品室,其中所述样品承载腔具有约1微米到约100微米的固定深度。
4.权利要求3的样品室,其中所述样品承载腔具有约2微米到约50微米的固定深度。
5.权利要求4的样品室,其中所述样品承载腔具有约2微米到约20微米的固定深度。
6.权利要求3的样品室,其中所述样品承载腔具有小于约2微米的固定深度。
7.权利要求6的样品室,其中所述样品承载腔具有小于约1微米的固定深度。

17.适于承载用于光学成像的生物样品的样品室,其包括:
光学透明底层;
光学透明顶层;
一面与顶层粘结而在另一面与辐射固化或热固化聚合物层形成样品承载室的光学透明粘结层,其中所述样品承载室具有从粘结层至辐射固化或热固化聚合物层之间约0.5微米到约1,000微米的固定深度;
用于向所述室引入样品的入口;以及
用于将样品取出所述室的出口。
18.权利要求17的样品室,其中所述样品承载腔具有约1微米到约100微米的固定深度。
19.权利要求18的样品室,其中所述样品承载腔具有约2微米到约50微米的固定深度。
20.权利要求19的样品室,其中所述样品承载腔具有约2微米到约20微米的固定深度。

提供光学透明衬层;

通过辐射或热处理固化所述辐射固化或热固化层;

37.权利要求33的方法,其中所述辐射固化或热固化层由选自紫外固化丙烯酸酯、聚氨酯和环氧树脂的紫外固化材料制备。
38.权利要求33的方法,其中所述辐射固化或热固化层由热固化材料制成。
39.权利要求33的方法,其中所述光学透明顶膜由选自下列材料制成:聚酯、聚碳酸酯、聚甲基丙烯酸甲酯、聚苯乙烯和环烯烃共聚物。
40.权利要求33的方法,其中所述预成型结构模具由铝模加工而成。
41.制造适于承载生物样品的样品室的方法,所述方法包括:
在辐射固化或热固化层上压印预成型结构模具,所述预成型结构模具伸入辐射固化或者热固化层中;
固化所述辐射固化或热固化层以形成光学透明层从而形成具有压印结构的室;
移去所述预成型结构模具;
在辐射固化或者热固化层的压印结构上粘附光学透明顶膜,从而形成样品室;以及
在所述室上形成一个或多个存取口以允许引入或移除样品。
42.权利要求41的方法,其中所述辐射固化或热固化层为辐射固化层。
43.权利要求42的方法,其中所述辐射固化层为紫外固化层。

47.制造适于承载生物样品的样品室的方法,所述方法包括:
在熔融塑料层上压印预成型结构模具;
冷却所述熔融塑料层以在其上获得压印结构;
移去所述预成型结构模具;
在压印结构上粘附光学透明顶膜,从而形成样品室;以及
在所述室上形成一个或多个存取口以允许引入或移除样品。
48.制造适于承载生物样品的样品室的方法,所述方法包括:
在塑料层上压印热的预成型结构模具,从而在其上获得压印结构;
移去所述预成型结构模具;
冷却所述塑料层;
在压印结构上粘附光学透明顶膜,从而形成样品室;以及
在所述室上形成一个或多个存取口以允许引入或移除样品。
