Abstract
本申请公开了水溶的亲油性维生素(例如维生素E)的微生物稳定的溶液。所述溶液含有约0.5%至约30%的盐,所述盐选自氯化钾、氯化镁、氯化钙、磷酸钾、磷酸钙、硫酸钾、磷酸钙、磷酸镁、硫酸镁、碘化钾以及这些盐的混合物。本发明还公开了提供所述水溶的亲油性维生素的微生物稳定的溶液的方法,并公开了含有所述微生物稳定的水溶的亲油性维生素的食物和饮料产品。

本申请涉及并要求2009年7月31日提交的序号为61/230,148的美国临时申请的优先权,其通过引用包含于此。
技术领域
本发明涉及微生物稳定的维生素溶液,特别是可以用在食物和饮料产品中的维生素E溶液。
背景技术
当配制食物或饮料组合物时,加入维生素来提高产品的营养价值通常是有用的。维生素是水溶性的(亲水性的)或脂溶性的(亲油性的)。在水基产品(例如保健饮料)中配入脂溶性的维生素(例如维生素A、D、E或K)是特别具有挑战性的。
在水溶液中配入亲油性维生素的一个已知方法是将亲水的侧链加在亲油基分子上。通过该方法,可以将整个分子的亲水亲油平衡(HLB)向着水溶性方向调节,使该分子可溶于水。维生素E(生育酚)的水溶性变体的实例为维生素E聚乙二醇(PEG)1000琥珀酸酯(也被称为维生素E TPGS),其具有下式:
可以通过用其他二羧酸(例如碳酸、草酸、丙二酸、戊二酸、己二酸、苯二酸、乙醇酸、乳酸、苦杏仁酸、柠檬酸及其混合物)取代琥珀酸来制备维生素E的水溶性变体。此外,可以调节聚乙二醇成分以使其含有约200至约2000个(优选约1000个)亚单元。还可以使用除PEG以外的聚醚原料,例如含有约200至约2000个(优选约1000个)亚单元的聚丙二醇。而且,PEG和PPG化合物可以以单月桂酸酯、单肉豆蔻酸酯、单棕榈酸酯或单硬脂酸酯衍生物来提供。具体的可以商业获得的水溶性维生素E原料包括维生素E聚乙二醇(PEG)1000琥珀酸酯。
尽管这些原料可以被置于水溶液中,但是它们有可能引起微生物污染的问题。具体地,在储存时,这些原料可以为酵母、细菌和霉菌的生长提供理想的环境。该污染问题必须在这些原料用于食物和饮料产品之前得到解决。解决该问题的一个方法是将这些原料进行无菌包装。所需的额外工艺使得上述包装方法非常昂贵。解决该问题的另一个方法是在溶液中放置广谱的防腐剂。那样也许是有效的,但是在成品的各个成分中优选不包括防腐剂,因为在成品中使用防腐剂可能并不是最佳的或者人们可能希望不用防腐剂来配制成品。解决该问题的第三种方法是使维生素E溶液的pH值保持极低(即在约2至约4之间)。然而,这样会引起维生素E的酸解,导致维生素损失。
发明内容
发明概述
本发明提供一种水溶液,其包含约0.5%至约30%(重量)的脂溶性维生素的水溶的衍生物和约0.5%至约30%(重量)的盐,所述维生素选自维生素A、D、E和K(特别是维生素E),所述盐选自KCl、MgCl2、CaCl2、K3PO4、Ca3(PO4)2、K2SO4、CaSO4、Mg3(PO4)2、MgSO4、KI及其混合物(特别是KCl或KI)。
发明详述
本发明提供了一种简单、有效且廉价的方法以解决所述微生物稳定性的问题。所述方法的实现是通过将有效量的抗微生物的氯化钾(KCl)置于含有溶解的维生素E原料的水溶液中。可以用其他盐来代替KCl,只要这些盐在溶液中具有相对中性的pH值(pH值为约6至约8,优选约为7),因为如果pH值超过该范围,也许会使亲水侧链中的醚键或酯键水解,因此会使维生素不可溶或无效。所述可以用来代替KCl的盐的实例包括氯化镁(MgCl2)、氯化钙(CaCl2)、磷酸钾(K3PO4)、磷酸钙(Ca3(PO4)2)、硫酸钾(K2SO4)、硫酸钙(CaSO4)、磷酸镁(Mg3(PO4)2)、硫酸镁(MgSO4)、碘化钾(KI)或这些原料的组合。除了使可溶性维生素E成分微生物稳定,如果所述原料用在食物或饮料产品中,所述盐也可以充当矿物质(如钾、镁、钙)的来源。尽管从效果的角度看钠盐也可以用在此处,但是由于一些与饮食中的钠有关的健康问题(例如高血压),因此通常避免使用这种盐。
通常,本发明的水溶液包括浓度为约0.5%至约30%(重量)的溶解的维生素E成分。所述溶液的一个实施方式含有约20%重量的溶解的维生素E成分。尽管溶解的维生素E的包含通常受其临界胶束浓度(CMC)限制,但是即使在CMC以上,所述盐的存在也可以使形成的胶束盐析,使得所述溶液具有更低粘性并且允许在溶液中使用较高维生素E浓度。通常,所述盐用于溶液中的浓度为约0.5%至约30%(重量);在一个实施方式中,所用盐的浓度为约5%至约20%(重量);在另一个实施方式中,所用盐的浓度为约10%至约15%(重量)。
脂溶性维生素的水溶的衍生物在现有技术中是已知的。它们可以通过将亲水的侧链加在亲油基分子上而配制得到。通过该方法,可以将整个分子的亲水亲油平衡(HLB)向着水溶性方向调节,使该分子可溶于水。上述维生素E(生育酚)水溶性变体的实例为维生素E聚乙二醇(PEG)1000琥珀酸酯,其具有下式:

在本发明的溶液中可以包括以本领域公认水平存在的、经常且常规用于维生素和营养补充溶液中的支持微生物的作用的其他原料。
本发明的一个实施方式为包含约20%(重量)维生素E聚乙二醇1000琥珀酸酯和约15%(重量)氯化钾的水溶液。该产品是透明的,具有良好的粘性,并且至少24小时抑制霉菌、酵母和细菌的生长。每克所述溶液向使用者提供2.5-3.0RDI的d-α-生育酚和0.022RDI的钾。
该技术还可以使用除维生素E以外的脂溶性维生素,具体可以使用维生素A、D和K。
从营养效果的角度来看,本文定义的维生素溶液可以加到食物和饮料产品中,以向这些产品提供维生素补充。可以使用传统上已知的食物和饮料产品。
具体实施方式
以下非限制性实施例用于说明本发明。
实施例1(对照)
通过使用加入200克切碎的维生素E TPGS的800克去离子水制备溶液。将该混合物升温至37℃,并搅拌直到所有维生素E溶解,制得均匀的溶液。然后通过利用白色念珠菌(Candida albicans)、黑曲霉菌(Aspergillus brasiliensis)、大肠杆菌(Escherichia coli)和金黄色葡萄球菌(Staphylococcus aureus)对所述溶液进行3周微刺激(microchallenge)来评估该混合物样品的微生物敏感性。结果显示生物体可以在未进行防腐的维生素E溶液中繁殖。
实施例2
在第二测试中,通过使用加入并溶解100克氯化钾的700克去离子水制备溶液。将该混合物升温至37℃。向其中加入200克切碎的维生素E TPGS。搅拌所述混合物,直到所有维生素E溶解,制得均匀的溶液。然后通过利用白色念珠菌、黑曲霉菌、大肠杆菌和金黄色葡萄球菌对所述溶液进行12周微刺激来评估该混合物样品的微生物敏感性。结果显示测试生物体受到控制,因此证明了抗菌性。
实施例3
在第三测试中,通过使用加入并溶解150克氯化钾的650克去离子水制备溶液。将该混合物升温至40℃。向其中加入200克切碎的维生素E TPGS。搅拌所述混合物,直到所有维生素E溶解,制得均匀的溶液。然后通过利用白色念珠菌、黑曲霉菌、大肠杆菌和金黄色葡萄球菌对所述溶液进行12周微刺激来评估该混合物样品的微生物敏感性。结果显示测试生物体受到良好控制,因此证明了抗菌性。
实施例4
通过混合650.0克去离子水、150.0克氯化钾和200.0克维生素ETPGS提前制备出维生素E TPGS的溶液并储存。将该混合物加热至35℃,搅拌至其均匀,冷却至室温并在使用前储存数周。
通过组合并混合以下成分制备饮料产品:

实施例5
通过混合650.0克去离子水、149.5克氯化钾、0.5克碘化钾和200.0克维生素E TPGS制备维生素E TPGS的溶液并储存。将该混合物加热至30℃,搅拌至其均匀,冷却至室温并在使用前储存。

Claims (14)
1.一种水溶液,其包含约0.5%至约30%(重量)的水溶的维生素E原料和约0.5%至约30%(重量)的盐,所述盐选自KCl、MgCl2、CaCl2、K3PO4、CaSO4、Ca3(PO4)2、K2SO4、CaSO4、Mg3(PO4)2、MgSO4、KI及其混合物。
2.根据权利要求1所述的水溶液,其中所述盐选自KCl、KI及其混合物。
3.根据权利要求2所述的水溶液,其中所述盐为KCl。
4.根据权利要求3所述的水溶液,其中所述水溶的维生素E原料为维生素E聚乙二醇1000琥珀酸酯。
5.根据权利要求4所述的水溶液,其包括约20%的维生素E聚乙二醇1000琥珀酸酯和约15%的KCl。
6.一种提供水溶性维生素E原料的微生物稳定的水溶液的方法,包括向所述溶液加入约0.5%至约30%(重量)的盐,所述盐选自KCl、MgCl2、CaCl2、K3PO4、Ca3(PO4)2、K2SO4、CaSO4、Mg3(PO4)2、MgSO4、Kl及其混合物。
7.根据权利要求6所述的方法,其中所述盐选自KCl、KI及其混合物。
8.一种向食物或饮料产品提供维生素E补充的方法,通过向所述食物或饮料产品加入在营养上有效量的权利要求1所述的溶液。
9.一种水溶液,其包含约0.5%至约30%(重量)的脂溶性维生素的水溶的衍生物和约0.5%至约30%(重量)的盐,所述维生素选自维生素A、D、E和K,所述盐选自KCl、MgCl2、CaCl2、K3PO4、Ca3(PO4)2、K2SO4、CaSO4、Mg3(PO4)2、MgSO4、KI及其混合物。
10.根据权利要求9所述的水溶液,其中所述盐选自KCl、KI及其混合物。
11.一种食物产品,其包含在营养上有效水平的权利要求1所述的溶液。
12.一种水基饮料产品,其包含在营养上有效水平的权利要求1所述的溶液。
13.一种食物组合物,其包含在营养上有效水平的权利要求5所述的溶液。
14.一种水基饮料组合物,其包含在营养上有效水平的权利要求5所述的溶液。
