“改善/增强联想学习”表示在训练环境下学习信号、符号、规则的意义以及如何对刺激作出反应的能力得到改善。
“改善/增强压力下的学习”表示当新任务发生在压力条件(例如作为日常生活/工作压力的一部分所经历的压力条件)下时,受试者学习新本领或新信息的能力得到改善。
“改善/增强压力下的记忆”表示:当经历有压力的环境或日常生活时,受试者回忆信息的能力得到改善。一个实例是在考试情况下回忆答案。
“改善/增强空间学习或记忆”表示:受试者学习目标相对于环境的其他部分在何处的能力得到增强。例如,一个人可能更容易地学会购物中心周围的路。另一个实例是一个人能更好地记住其停车或放钥匙的位置。
“唯一的增强学习的化合物”表示:该组合物中不包含有助于认知的其他成分。具体排除的成分的实例包括:银杏(提取物或单独的活性成分)、乙酰左旋肉碱、维生素C和E、维生素B、胞磷胆碱、CoQ10(辅酶-Q10)、人参、石杉碱甲A、Ω-3脂肪酸(例如DHA/EPA)、磷脂酰丝氨酸、松树树皮提取物(Pycnogenol碧萝芷)或Bacopa monieri植物提取物。
“观察”增强的认知方面能力表示观察者注意到受试者有改善或受试者本身注意到其有改善。这些改善可以通过使用标准化的测量方法来量化,但不需要这样做,可以仅根据主观感受或经验。
“处治”包括共处治和预防。“预防”并不限于未来完全不存在这些症状,而是意指包括:减少个体或群体表现症状的风险,减轻与特定病症有关的症状,减少特定病症的发病时间,减轻病症的严重性,减少无症状的个体在未来显示出病症的可能性。
为了实现这些改善,建议连续几天施用(例如至少6天或10天),通常优选在数周内每天施用。
在优选的实施方式中,叶黄素或叶黄素和玉米黄质被健康的受试者摄取。“健康的”表示受试者没有遭受任何危害其心理健康的病症的影响,即,没有遭受特征为记忆恶化(例如痴呆、阿尔茨海默病等)、抑郁的病症或影响记忆和学习的其他精神病症(例如精神分裂症)的影响。
因而,本发明的另一方面将涉及叶黄素或叶黄素和玉米黄质在改善联想记忆中的用途。
本发明的另一方面将涉及叶黄素或叶黄素和玉米黄质在改善受试者处于压力下时的记忆的用途。具有改善的处于压力下的记忆的受试者将观察到下列中的至少一项:
●在工作中不会忘记执行分配的任务(当几项任务待解决时);
●在演讲中记住事实等;
●在考试条件或需要快速回忆的条件下表现更好。
本发明的另一方面将涉及叶黄素或叶黄素和玉米黄质在改善空间记忆中的用途。具有改善的空间记忆的受试者将观察到下列中的至少一项:
●较少“放错的”项,例如某人的钥匙、眼镜、移动电话或电视遥控器;
●在周围环境(例如在房子里)中和驾驶时容易确定自己的方向的能力,或者能够简单地记住某人将汽车停放在大的停车场的何处。
除了可用于人类之外,本发明的组合物在兽医界具有额外的用途。动物将受益的状况具体为特定目的的训练过程和培养,例如对于猎犬、导盲犬、警犬、或电影业中所用的动物。可以从增强的认知功能中受益的动物包括那些经受压力条件的动物。这些条件出现在例如捕获或运输之后,或者可能是由于居住条件(例如住所或主人的改变),当动物产生类似的失调或变得痛苦或具有攻击性时,或者表现出刻板行为(stereotypicbehaviou)、或焦虑和强迫行为时。经受压力的动物也包括那些赛跑的动物(例如狗、马、骆驼)或各种运动中所用的动物、表演动物(例如马戏团的动物以及舞台、电视或电影中出现的那些动物)和表演盛装舞步和其他高度纪律性成套动作的马。
优选的“动物”是宠物或伴侣动物(companion animals)以及农场动物。宠物的例子为狗、猫、鸟、观赏鱼、豚鼠(guinea pigs)、(长腿)兔((jack)rabbit)、野兔和雪貂、农场动物的例子为水产养殖的鱼、猪、马、反刍动物(牛、绵羊和山羊)和家禽。
营养药物的用途/制剂/剂量
在本文中使用时,术语“营养药物”表示在营养和医药应用领域均可用。因而,新颖的营养药物组合物可用作食品/饲料和饮料的补充物,和可以作为肠内或肠胃外应用的药物制剂,其可以为固体制剂,例如胶囊或药片,或者为液体制剂,例如溶液或悬浮液。
根据本发明的营养药物组合物可以进一步包含保护性水胶体(例如树胶、蛋白质、改性淀粉)、粘结剂、成膜剂、成胶囊剂/材料、壁/壳材料、基质化合物、包衣、乳化剂、表面活性剂、增溶剂(油、脂肪、蜡、卵磷脂等)、吸附剂、载体、填料、复合物(co-compound)、分散剂、润湿剂、加工助剂(溶剂)、流动改性剂、掩味剂、增重剂、凝结剂(jellyfyingagent)、凝胶剂、抗氧化剂和抗微生物剂。
此外,多维生素和矿物质补充剂可以被添加到本发明的营养药物组合物中以得到足够量的某些膳食中缺少的必需营养。多维生素和矿物质补充剂还可以用于疾病预防和保护免受因生活方式而导致的营养损失和缺乏的影响。
根据本发明的营养药物组合物可以是含有常规载体材料的、适于施用至身体的任何盖仑形式(galenic form),特别是常用于口服施用的任何形式,例如固体形式,诸如食品或饲料(用于食品或饲料的添加剂/或补充剂)、食品或饲料预混料、强化食品或饲料、药片、药丸、颗粒、糖衣丸、胶囊和泡腾片剂型(例如粉末和片剂);或者液体形式,例如溶液、乳液或悬浮液,诸如饮料、糊和油悬浮液。所述糊可以包在硬壳或软壳胶囊中,其中该胶囊具有例如(鱼、猪、家禽、奶牛)明胶、植物蛋白或木素磺酸盐的基质。其他应用形式的例子是用于经皮施用、肠胃外施用或注射施用的那些。膳食和药物组合物可以是受控(延缓)释放制剂的形式。
食品的例子为乳制品,包括例如人造奶油、涂抹酱(spread)、黄油、芝士、酸奶或奶饮料。
强化食品的例子为面包、谷物棒、烘烤食品(例如蛋糕和曲奇饼干)以及薯条或油炸马铃薯片。
宠物食品的例子为湿的罐头食品、零食和压缩饲料(pellets)。
饮料包括非酒精饮料和酒精性饮料以及添加到饮用水和液体食品中的液体制剂。非酒精饮料为例如软饮料、运动饮料、果汁、柠檬饮料、茶和奶饮料。液体食品为例如汤和乳制品。含有叶黄素或叶黄素和玉米黄质的营养药物组合物可以被添加到软饮料、能量棒或糖果中,从而使成年人消耗每天每kg体重至多2mg。
如果营养药物组合物是药物制剂,该组合物进一步包含药物可接受的赋形剂、稀释剂或佐剂。其配制可以使用标准技术,如例如Remington′sPharmaceutical Sciences,20th edition Williams&Wilkins,PA,USA中公开的。对于口服施用来说,优选使用药片和胶囊,其含有适当的粘结剂(例如明胶或聚乙烯基吡咯烷酮)、适当的填料(例如乳糖或淀粉)、适当的润滑剂(例如硬脂酸镁)以及任选的其他添加剂。
为了本发明的目的,对于动物(包括人类)来说,叶黄素或叶黄素和玉米黄质的合适的每日剂量可以在每天每kg体重0.15mg到约每天每kg体重10mg的范围内。
下面所示的非限制性实施例将更好地说明本发明。
实施例1
叶黄素在体外神经毒性模型中的作用
谷氨酸是中枢神经系统中一种主要的兴奋性氨基酸,并且在正常的生理机能中起到关键作用。谷氨酸功能的正常调节机制中的扰动可能导致大脑中的谷氨酸受体过度的神经毒性活化,这可能不利于正常脑和老化的脑中的学习和记忆表现。使用神经元的原代培养物,可以通过谷氨酸中毒再现这些状态。对培养的神经元的影响通过神经元的神经丝细胞骨架网络的形态变化来示明。
单一化合物或化合物的组合的神经保护作用通过使用特异于成熟神经元神经突的抗神经丝抗体(重链200kD)标记来分析网络密度而评估。
利鲁唑(Riluzole,一种药物神经保护剂)和MK801被用作抵消谷氨酸的神经毒性作用(导致神经突过生长)的抑制化合物。抑制性的MK801(对NMDA型谷氨酸受体完全阻滞)很有效,导致中毒之后的100%恢复。
使用下面所示的时间安排来测试叶黄素的神经保护功能:

在第5天,用对照的培养介质或含叶黄素的介质(在生理范围中的3个浓度,每个浓度下n=6)来替换培养介质。每2-3天更换一半的介质。在第12天,用250μM的谷氨酸使细胞中毒6小时。
对照培养物(谷氨酸中毒且没有任何附加的化合物)的神经突长度被设定为100%。这些实验表明,用叶黄素预处治(所有使用的浓度,p<0.05且<0.01)明显保护了神经元,使其免受兴奋毒性的谷氨酸神经毒性(参见图1)。
实施例2
叶黄素和/玉米黄质在新的、完全自动的、啮齿动物学习和记忆的模型中的
作用
在IntelliCage中,用叶黄素和/或玉米黄质处治的小鼠的认知表现与用赋形剂处治的、年龄相当的对照组相比较,所述IntelliCage是一个能在与笼屋(homecage)类似的环境下自动监测小鼠的自发的学习行为的系统(NewBehavior AG,Zürich,Switzerland,www.newbehavior.com;Galsworthy etal.2005,Behav Brain Res 157:211-217;Onishchenko et al.2007,Toxicol Sci97:428-437;Mechan et al.2009,J.Neurosci.Methods,180:43-51)。各小鼠通过IntelliCage角落内的传感器读取转发器(参考识别标签)来识别,其中,所述转发器植入小鼠的颈部的颈背中。
每个IntelliCage基本上是一个大的笼子(37.5×55×20.5cm),其中放置了包含四个记录(操作)室的金属框。记录室安放在笼子的四个角落中,每一个均占据15x15x21cm的直角三角形的占地面积。笼中天线能自动监测每一只老鼠的角落访问;每一个角落内的照片光束能自动记录每一次探鼻和水瓶喷头舔舐。4个三角形的老鼠躲避处被放置在笼子的中间,其上有食物漏斗,使小鼠能随意获取食物。
每一个记录室均包括:(1)一个塑料环(内径为30mm),其充当进入记录室的入口并容纳登记角落访问的环形天线;(2)格子地板,小鼠一旦进入记录室内便坐在其上;(3)两个圆形开口(直径为13mm),其能接近水瓶喷头;各开口均被登记探鼻的照片光束横穿;(4)两个监测门,其允许(门打开)或禁止(门关闭)接近水瓶喷头;(5)两个水瓶;(6)管子,通过该管子可以输送空气喷气作为厌恶性刺激;(7)不同颜色的发光二极管,其可用于条件反射实验(Mechan等人,2009 J Neurosci Methods 180:43-51)。参见图2。
实验阶段:
本研究包括两个测试组(每一个组n=12-14):赋形剂(对照)、叶黄素(9mg/kg BW)(即FloraGLOLutein,包含约98%的叶黄素(全-E和Z异构体)和约2%的其他类胡萝卜素(其中约0.5-1.5%的玉米黄质)的金盏花提取物)。
叶黄素包含在正常的动物饲料中。在IntelliCage研究之前的12天以及贯穿4周的研究期间将测试物质或空白对照剂以饲料(50mg/kg饲料)施用给所有的小鼠。
在最初的适应阶段(9天),小鼠能自由接近所有的角落、水和饲料,并且能自由地探索笼子。随后,小鼠必须学习使用探鼻(探鼻适应模块,2天);所有的门最初都是关闭的(禁止接近水),小鼠为了打开门并够到水瓶喷头必须执行探鼻。所采集的数据包括几个参数,例如每个个体小鼠最不偏好的角落,其被记下用于编程下一个模块。
在探鼻适应之后,执行下列任务:
-侧面辨别力模式:联想学习和记忆以及记忆和注意力的测试;
-联想记忆模式:利用负强化(压力)的联想学习和记忆的测试;
-方位学习模式:空间学习和记忆以及方向性的测试。
侧面辨别力模式
该模块被设计用于测试注意力和联想记忆。为每只老鼠指定一个正确的角落。在该角落中只有一个侧面(两个侧面中的一个)被指定为正确的,并通过绿色LED向动物指示。动物可以在正确的侧面上探鼻,随后从水瓶中喝水。在该模块中,记录位置错误率(访问错误角落的百分比)和侧面错误率(在正确角落的错误侧面上探鼻的百分比)。
这些数据表明,用叶黄素长期处治之后,注意力得到改善,因为在测试活跃期期间,其位置错误率(图3)和侧面错误率(图4)明显低于赋形剂对照组。
概括地说,自动IntelliCage研究的结果表明,与用赋形剂处治的同窝出生者相比,用叶黄素处治使得小鼠为了获得水而将短时间(2秒)存在的光信号与正确角落和正确的角落侧面联系在一起的学习和记忆得到明显改善。
联想记忆模式
在该模块中,为每只老鼠指定一个正确的角落,并通过3秒的灯光显示来指示。因而,在正确角落的探鼻将打开门,小鼠在该特定的角落中能够获得水,而每一次访问任何错误的角落均将导致空气喷气的惩罚(压力)。结果显示在图5中,表明与用赋形剂处治的同窝出生者相比,用叶黄素处治的小鼠学习和记忆得到水(因而避免惩罚)的位置的表现明显较好。
方位学习模式
在这个学习任务中,小鼠必须进入正确的角落,并且为了得到水必须在角落的任一侧面上进行探鼻。根据之前的探鼻适应模块中动物的访问行为来指定角落,如果合适的话,最不偏好的角落被指定为各老鼠的正确角落。不提供额外的信号(例如灯光),从而指导小鼠基于其环境和/或笼子的空间提示来尝试学习并记住能获取水的角落的位置。图6总结出,该测试的结果表明,与用空白对照剂喂养的同窝出生者相比,用叶黄素处治的小鼠的表现明显较好。
在第二组实验中,在上述学习和记忆模式中测试不同剂量的叶黄素(8mg/kg/d、16mg/kg/d和30mg/kg/d)或叶黄素与玉米黄质的组合(分别为15和16mg/kg/d)。与对照组相比,用中间的叶黄素剂量补充的动物在联想记忆任务中在位置错误率和侧面错误率方面的表现显示出明显的改善。此外,用叶黄素和玉米黄质喂养的动物在侧面错误率方面的表现更优异。
在比第一组实验的动物小2-3个月的动物中进行这些实验,结果表明,根据所使用的年龄和认知模式,叶黄素的有利效果可能不同。
实施例3
软明胶胶囊的制备
可以制备包含下列成分的软明胶胶囊:
成人可以在3个月中每天服用两颗胶囊。
实施例4
即食调味的软饮料的制备
将所有成分共混并用500μm的筛子过筛。将产生的粉末放入合适的容器中,在管式搅拌机中混合至少20分钟。为了制备饮品,将840g所得到的混合粉末与足量的水混合以产生1升的饮料。
这种即饮饮品的软饮料中每一份(250ml)包含约12mg叶黄素。作为加强剂,正常人每天可以饮用一份(250ml)。发现压力下的记忆得到改善。
实施例5
强化的非烘焙的谷物棒的制备
叶黄素和玉米黄质与脱脂奶粉预混合,放入行星式混合器中。加入玉米片和米脆,并轻轻地混合。然后加入干的苹果碎丁。在一个烹饪锅中,以上面给出的量混合水和盐(溶液1)。在第二烹饪锅中,以上面给出的量混合葡萄糖浆、转化糖浆和山梨醇糖浆(溶液2)。脂肪相由烘焙油脂、棕榈仁油、卵磷脂和乳化剂的混合物组成。溶液1被加热到110℃。溶液2被加热到113℃,然后在冷水浴中冷却。随后,将溶液1和2合并。脂肪相在水浴中在75℃下融化,然后加入溶液1和2的合并混合物中。将苹果香精和柠檬酸加入液态糖/脂肪的混合物中。将该液体物质加入干成分中,并在行星式混合器中混合均匀。将团块放在大理石板上并滚压至期望的厚度。将其切成段。每份(20g)非烘焙的谷物棒包含约10mg叶黄素和2mg玉米黄质。
实施例6
含叶黄素的干的狗粮
用叶黄素(例如FloraGLOLutein 5%Corn Oil)与抗氧化剂的溶液以足以使每kg食物施用约5mg叶黄素的量喷洒市售的狗的基础膳食(例如Mera Dog“Brocken”,MERA-Tiernahrung GmbH,Marienstraβe 80-84,D-47625Kevelaer-Wetten,德国),所述抗氧化物诸如为维生素C(例如来自瑞士Kaiseraugst的DSM Nutritional Products Ltd的ROVIMIXC-EC)或其衍生物,即,抗坏血酸单磷酸酯钠(例如来自瑞士Kaiseraugst的DSMNutritional Products Ltd的STAY-C50)或钠的三-、二-、单磷酸酯/L-抗坏血酸钙的混合物(例如来自瑞士Kaiseraugst的DSM Nutritional ProductsLtd的ROVIMIXSTAY-C35)。对于较重的狗来说,相应地制备饲料混合物。

Claims (5)
1.一种增强健康个体的记忆方面的方法,其中所述记忆方面选自由下列组成的组:联想记忆、空间记忆和压力下的记忆,所述方法包括:
施用由a)有效量的叶黄素或叶黄素与玉米黄质的组合和b)适当的载体组成的组合物;以及
观察所述增强的联想记忆、空间记忆或压力下的记忆。
2.一种增强压力下的学习和记忆的方法,所述方法包括:
施用由a)增强与压力有关的学习和记忆的有效量的叶黄素或叶黄素与玉米黄质的组合和b)适当的载体组成的组合物;以及
观察所述增强的压力下的学习和记忆。
3.一种增强空间学习和记忆的方法,所述方法包括:
施用由a)增强与空间有关的学习和记忆的有效量的叶黄素或叶黄素与玉米黄质的组合和b)适当的载体组成的组合物;以及
观察所述增强的空间学习和记忆。
4.叶黄素或叶黄素与玉米黄质的组合作为唯一的活性化合物用于增强a)联想学习和记忆、b)压力下的学习和记忆、或c)空间学习和记忆中的用途。
5.叶黄素或叶黄素与玉米黄质的组合作为对于增强记忆起作用的唯一的活性化合物在制备用于增强a)联想学习和记忆、b)压力下的学习和记忆和/或c)空间学习和记忆的营养药物或功能性食品中的用途。
